Hepatoprotective compounds from Canarium album and Euphorbia nematocypha.

Successive purification of the extract from Canarium album and Euphorbia nematocypha, guided by antihepatotoxic activity in primary cultured rat hepatocytes, led to the isolation of brevifolin (1), hyperin (2), ellagic acid (3) and 3,3'-di-O-methylellagic acid (4) as hepatoprotective compounds. Compounds 1,3 and 4 also reduced carbon tetrachloride (CCl4)-induced liver damage in mice. The hepatoprotective activities of 1, 2, 3 and 4 in vitro and in vivo are apparently due to their antioxidative effects, which were exhibited by further studies using 1,1-diphenyl-2-picrylhydrazyl (DPPH) radicals and CCl4-induced lipid peroxidation systems.

[Magnesium deficiency dermatitis--changes in leukotriene levels and effects of azelastine hydrochloride on the dermatitis and leukotriene B4].

To estimate the pathogenesis of magnesium deficiency dermatitis (MDD) in the hairless rats fed with a hypomagnesic diet, plasma concentrations of leukotrienes (LTB4, LTC4, LTD4) were measured. Inhibitory effect of azelastine hydrochloride (AZ), which is known as an inhibitor of 5-lipoxygenase, on MDD was also evaluated. In the hairless rats with magnesium deficiency condition, plasma level of leukotriene B4 was increased, whereas LTC4 and LTD4 showed no changes. In AZ-administered rats, MDD was depressed with the dose-dependent tendency. However, the decrease of LTB4 level did not follow therapeutic effect of AZ for gross appearance of dermatitis. These results indicated that development of MDD associates with increase of LTB4, but its pathogenetic role still remains unknown, and inhibitory effect of AZ against MDD not based on reduction of LTB4 production.

[An immunohistochemical study on the response of nerve fibers in the periodontium of rat molars during experimental tooth movement].

It is well known that orthodontic forces induce pain during tooth movement. However there have been few neurohistological studies on sensational periodontal tissue response to orthodontic tooth movement. So we have many unknown problems about reactions of periodontal nerve fibers during tooth movement. The present study deals with the response of nerve fibers in periodontal ligament of rat molar during experimental tooth movement by means of immunohistochemical method for calcitonin gene-related peptide (CGRP). The results obtained in this study were as follows: 1. In control sections, CGRP-immunopositive nerve fibers were observed both around the root apex and in the intermediate region of the periodontal ligament. Although most of those fibers was distributed around the blood vessels, a few nerves were recognized to terminate in the periodontal fibers. 2. At three days after tooth movement, the CGRP-positive nerves gradually increased in number around the blood vessels. After that, the number of CGRP-positive nerves decreased and no difference of distribution of CGRP-positive nerves was observed between the control sections and the experimental sections at seven days. 3. Especially, at three days when active remodeling of alveolar bone was taking place, a few nerve fibers showing CGRP-immunoreactivity were observed to concentrate around osteoclasts or to migrate Howship's lacunae. These findings showed the dynamic changes of CGRP-immunoreactive nerves in the periodontal ligament during tooth movement, and suggested that this reaction of the periodontal nerves might induce pain in patients. In addition to neurotransmitter function, it was supposed that CGRP directly or indirectly effected the cellular elements and surrounding tissues on peripheral organs such as vascular system and osteoclasts.

[A long-term study on dentofacial changes after sagittal splitting osteotomies in skeletal Class III malocclusions].

The purpose of this study is to clarify the long-term dentofacial changes after sagittal splitting ramus osteotomy in skeletal Class III patients, and to study the factors involved to find its countermeasures. For this purpose we used the radiographic cephalograms in 16 patients (13 females and 3 males) observed for more than five years after surgery. We measured the postoperative changes in the position of Pogonion and the inclination of maxillary and mandibular incisors. The results were as follows: 1. Pogonion was retruded 9.3 mm on the average by surgery. In the period from the removal of intermaxillary fixation to one year postoperatively Pogonion slipped forwardly and upwardly (average; anteriorly 1.0 mm, superiorly 0.8 mm). In the period from one year postoperatively to the time of the final observation Pogonion slipped to a downward direction. 2. No correlation was found between the amounts of vertical displacement by surgery and postoperative vertical changes of Pogonion. On the other hand, a high correlation was found between the amounts of mandibular setback and the forward displacement of Pogonion. 3. Mandibular incisors were tipped labially by preoperative orthodontic treatments, then maxillary and mandibular incisors moved lingually during intermaxillary fixation. After release of intermaxillary fixation, maxillary incisors moved labially in a small distance. After one year maxillary incisors were inclined slightly in the lingual direction. It is obvious that the position of the mandible and the incisor inclination tend to change even after one year. Because the postoperative changes were small in quantity, we supposed they were mostly caused by occlusal changes. Clinically, occlusal relationship in the incisor area, i.e., overjet and overbite, was maintained to be normal. It was considered that the changes of incisal inclination after release of intermaxillary fixation were of a compensatory or adaptational response to a positional change of the mandible.

[The present states and considerations of orthognathic surgery in Japan. Questionnaire in department of orthodontics of university dental hospital, and department of oral surgery of university medical hospital].

To investigate the present state of orthognathic surgery, questionnaires were sent to 30 departments of orthodontics of university dental hospital and 60 departments of oral surgery of university medical hospital. 1. Orthognathic surgery was carried out in 100% of the departments of orthodontics and 91.8% of the departments of oral surgery examined. 2. There was a difference in the number of patients between individual hospitals. 3. Analyses of cephalogram were used in 96% of the departments of orthodontics and 93% of the departments of oral surgery. 4. Pre- and post-surgical orthodontic treatments were carried out in all of the departments of orthodontics. 5. Orthodontic appliances (83.9%) were used in the departments of orthodontics for intermaxillary fixation, while orthodontic appliances (47.8%) and wire splints (49.2%) were used in the departments of oral surgery. 6. It is pointed out that the criterion of orthognathic surgery, treatment objectives on soft and hard tissue, selection of surgical method, adaptability of soft tissue after surgery, relapse, treatment planning of two jaw surgery, and postoperative stability remain to be solved in future.

[Study on the simulation in planning for orthognathic surgery by using a personal computer].

This study was made in order to develop and evaluate three-dimensional simulation programs in planning for orthognathic surgery by using microcomputer system. Microcomputer system is composed of CPU (NEC, PC-9801), high resolutional color display, color printer, image scanner, and MOUSE. This simulation is composed of four procedures. 1. Data input Three-dimensional coordinate data on tracing obtained from CT films is inputted. One data file is made from one outline of structures. 2. Three dimensional reconstruction A control file is made to arrange data files constructing structures. 3. Simulation This procedure is composed of cutting and movement. The horizontal projection of mandible is displayed. After cutting simulated orthognathic surgery is designed, new data files and new control file are made. A movement consists of parallel translation and rotation. A movement data is combined with movements designed on each of three projections. 4. Three-dimensional graphic display Three-dimensional graphics of the mandible before and after a simulation for orthognathic surgery are displayed by perspective views with treated hidden lines and accurate pictures (COSMOZONE 2SA, NIKON). These simulation programs were carried out with the asymmetrical patient. The simulation was supposed that the operation was combined with sagittal splitting osteotomy of the right mandibular ramus and body ostectomy of the left lower first premolar part. Three-dimensional graphics of the mandible before and after the simulation were presented. Its three-dimensional mandibular position and form after orthognathic surgery were predicted. These three-dimensional simulation programs may be important procedure, when a treatment planning in asymmetrical prognathic case is discussed.

An index for objective evaluation of the soft tissue profile.

Statistical analysis of the relationship between human perceptions and the physical characteristics of a sample of Japanese profiles led to the development of the Profile Aesthetic Index. The Profile Aesthetic Index is a composite of seven physical variables (five angles and two distances) measured on soft tissue profiles. The correlation between the index score and the mean rating score given by orthodontists and dental students was r = .80. Clinical validity of the Profile Aesthetic Index was examined by comparing the index scores before and after the surgical-orthodontic treatment of 50 patients. Potential uses and limitations of the index are discussed.

Determination of the loci of action of phencyclidine on the CNS-pituitary-adrenal axis.

Phencyclidine (PCP) markedly stimulates the pituitary-adrenal axis in the rat, inducing the release of adrenocorticotropin (ACTH) and corticosterone. However, the site or sites where PCP produces these effects is not known. This study sequentially examined the effects of PCP on the different components of the central nervous system-pituitary-adrenal axis. PCP did not produce corticosterone release in dispersed adrenal cells in vitro, nor did it stimulate the release of corticosterone in hypophysectomized rats, showing that PCP-induced corticosterone release in intact animals is secondary to the release of ACTH from the pituitary. PCP failed to alter either the basal or the corticotropin releasing factor-induced release of ACTH from superfused pituitaries in vitro, indicating that PCP does not act directly at the level of the pituitary. PCP increased plasma levels of ACTH in adrenalectomized rats, demonstrating that PCP does not stimulate the release of ACTH only by blocking glucocorticoid negative feedback mechanisms. PCP stimulated the release of both ACTH and corticosterone when given by injection directly into brain via the lateral cerebral ventricles. These results indicate that PCP activates the pituitary-adrenal axis by acting at a site or sites within the central nervous system, leading to the subsequent release of ACTH from the pituitary.

[Hemolytic activity of Mycobacterium kansasii].

Mycobacterium kansasii has a strong virulence as compared with those of "mycobacteria other than tubercle bacilli". In this study, hemolytic activities of several strains of M. kansasii were investigated. Secretion of hemolytic factor from these strains was not observed, since no hemolytic zone was found around the colonies grown on the 7H11 agar plate when 5% rabbit blood agar was overlayed. On the other hand, weak hemolytic activities were detected after treatment of sonication and trypsin-digestion of M. kansasii cells. They were also heat-resistant. These results indicate that endogenous nonproteinaceous factor was involved in the hemolytic activity of M. kansasii. Moreover, these activities were not species-specific in respect to the kind of target erythrocytes used for the assay. However, hemolytic activity of M. kansasii was not increased in spite of the change of culture conditions or supplement of nutrients into the medium. It is unlikely that this hemolytic activity is directly associated with a pathogenesis of M. kansasii, but detailed studies were needed to clarify on this point.

[Effect of biliary obstruction and cholangitis on serum SPan-1 level].

We investigated the effect of biliary obstruction and cholangitis on serum SPan-1 level in 14 patients (5 patients with pancreatic cancer and 9 patients with biliary tract cancer) who were performed PTCD for biliary obstruction. 1 out of 5 cases with pancreatic cancer and 8 out of 9 cases with biliary tract cancer showed a decrease of SPan-1 level with improvement of jaundice. It suggested that the effect of biliary obstruction and cholangitis on serum SPan-1 level was much more in biliary tract cancer than in pancreatic cancer. There was no significant difference in changes of between serum SPan-1 level and serum CA 19-9 level after PTCD. We conclude that the effect of biliary obstruction and cholangitis on serum SPan-1 level is almost same on serum CA 19-9 level.

Orientation of the carboxyl terminus of the Na+/proline symport carrier in Escherichia coli.

The orientation of the carboxyl terminal region of the Escherichia coli proline carrier in the cytoplasmic membrane was studied. The beta-galactosidase moiety of the PutP-LacZ fusion protein [(1987) J. Biol. Chem. 262, 14100-14104] was exposed outside the inside-out vesicles and inside the right-side-out vesicles. A site-directed antibody raised against a synthetic peptide corresponding to the putative carboxyl terminal region of the carrier reacted preferentially with the inside-out vesicles prepared from a wild-type proline carrier overproducing strain and less with the right-side-out vesicles. These results indicate that the carboxyl terminus of the proline carrier is exposed to the cytoplasmic side of the membrane.

Kinetics of velvet tobacco mottle virus satellite RNA synthesis and encapsidation.

Synthesis of circular (RNA 2) and linear (RNA 3) molecules of velvet tobacco mottle virus (VTMoV) satellite RNA (sat RNA) has been studied by incubating strips of tissues excised from systemically infected Nicotiana clevelandii in solutions of [14C]uridine. After a short lag, RNA and virus synthesis proceeded at a constant rate for at least 24 hr, during which time most of the synthesis was directed to the production of RNAs 2 and 3. The kinetics of [14C]uridine incorporation into the sat RNA molecules after increasing times of incubation and during pulses of [14C]uridine followed by chase incubation with excess [12C]uridine suggest that RNA 3 is a percursor of RNA 2. However, not all the RNA 3 synthesized was shown to end up as RNA 2, even after 72 hr of incubation. Several lines of evidence are presented supporting the conclusion that VTMoV-infected cells contain large pools of unencapsidated sat RNA. It is suggested that the sat RNA may have a greater affinity for the VTMoV replicase than the helper viral RNA which results in copious production of the sat RNA.

Modification of strain-specific femoral bone density by bone marrow chimerism in mice: a study on the spontaneously osteoporotic mouse (SAM-P/6).

The role of marrow-derived cells in determining strain differences in femoral bone density in mice was investigated by inducing marrow chimerism in a newly developed, osteoporotic strain of mouse: SAM-P/6. As a normative study prior to the chimeric experiment, bone density of F1 hybrids between SAM-P/6 and SAM-P/2 (strain with a high bone density) was assessed. Microdensitometrically, the F1(P2P6F1) exhibited a significantly higher bone density than did the SAM-P/6. P2P6F1 marrow cells, injected into SAM-P/6 neonates, increased the bone density at age 60 days while the syngeneic cells were without effect. The successful induction and maintenance of chimerism was confirmed by flow cytometry in which a donor-specific H-2K haplotype was used as the marker. This study shows that, at least in certain combinations of strains of mice, strain-specific bone density can be modified by bone marrow chimerism. Also, it suggests that the low bone density observed in the SAM-P/6 might reflect functional characteristics of the marrow-derived cells.

Impairment of the inferior alveolar nerve after sagittal split osteotomy.

The incidence and degree of neurosensory disturbance of the inferior alveolar nerve, as well as its recovery course, were studied on 46 sides in 23 patients who had undergone bilateral sagittal split osteotomies, by means of subjective symptoms, light touch, anaesthesiometer and two-point discrimination. The degree of disturbance was classified into mild, moderate and severe grades by the threshold pressure shown in tests with the anaesthesiometer. The disturbance, which was almost exclusively limited to mild (37%) and severe (28%) grades, was observed in 67% of the sample at one week. The disturbance disappeared completely within one to three months postoperatively in most sites with mild disturbance, and within three months to one year in half of the severely affected sites. Although the recovery was delayed in the other half of the severely disturbed sites, the disturbance was of mild grade at one and a half years. The overall incidence of disturbance at one year was 15%. Computed tomographic examination of the ascending ramus showed that the narrowest width between the mandibular canal and the buccal cortical plate ranged from 0 mm to 3.2 mm with a mean of 1.6 +/- 0.9 mm (SD) and it was less than 1.2 mm in 91% of sites with a severe grade disturbance, whereas it was distributed in a range of 0.9 mm to 3.2 mm in sites with no disturbance or with mild or moderate disturbance. The importance of preoperative computed tomography to indicate the location of the mandibular canal and the use of a thin cement spatula for the osteotomy was stressed, to avoid or reduce postoperative development of neurosensory impairment.

Superoxide anion in polymorphonuclear leukocytes of hairless rats suffering from magnesium-deficiency dermatitis.

Clinical and histological studies were made on magnesium-deficiency dermatitis produced on a new strain of hairless rats. Superoxide anion generation in polymorphonuclear leukocytes (PMNs) and the inhibitory effect of some antioxidants and anti-inflammatory agents against the elicited dermatitis were also examined. From the detection of increased superoxide production and effective actions of inhibitors including superoxide dismutase, cepharanthine, 4-4'-diamono-diphenylsulphone and 5,4,11,14-eicosatetraynoic acid (ETYA), it is indicated that superoxide anion or related activated oxygen may be responsible for magnesium-deficiency dermatitis as an inflammatory mediator and that the dermatitis may be related to lipoxygenase metabolites derived from arachidonic acid.

Morning and evening adrenocortical responses to ACTH stimulation in endogenously depressed patients: a preliminary report.

Adrenocortical stimulation with ACTH both in the morning (M-test) and in the evening (E-test) and the dexamethasone suppression test were carried out in patients suffering from endogenous depression (DEP) and normal controls (NOR). A greater cortisol release in DEP was recognized than in NOR in the M-test, an earlier peak response of DEP was shown in the M-test than in the E-test, and a lack of association between hypersecretion of cortisol during depression and cortisol output after ACTH administration was noted. These findings, together with the results of DST, suggest that excessive activity of the hypothalamic-pituitary-adrenal (HPA) axis in depression may result, partly, from adrenocortical hyperresponsiveness.
