Expression of glutathione S-transferase-pi in malignant skin tumors.

GST-pi has been known to be markedly increased in human (pre) neoplasms of several organs. In this paper, the significance of immunohistochemical detection of GST-pi in human malignant tumors of the skin was studied. In specimens from 40 patients with various skin cancers, malignant melanoma, Paget's disease and undifferentiated squamous cell carcinoma showed strong reactivity in GST-pi staining. The reactions were negative or weak in Bowen's disease, basal cell epithelioma and solar keratosis. In normal melanocytes, eccrine, apocrine, and breast gland cells stained positively but not in keratinocytes, sebaceus gland and fibroblasts. While immunohistochemical detection of GST-pi in the skin was not specific for malignancies, it contributed to aid the distinction of squamous cell carcinoma from other keratinocytic tumors. GST-pi might provide potentially useful information on chemosensitivity of skin cancer, and might serve as a biomarker of disease activity.

Three-dimensional analysis of condylar hyperplasia with computed tomography.

Three-dimensional surface reconstruction imaging from CT scans was used to study the deformity of the mandible in six patients with mandibular asymmetry. High-resolution axial CT scans of the mandible were obtained using Somatom-DR3 (Siemens). COSMOZONE-2SA (Nikon) with PC-9801VX21 (NEC) was used to reconstruct the three-dimensional images. The six patients were divided into two groups. One group was classified as unilateral hybrid forms and the other group was classified hemimandibular elongation on the diagnostic criteria of Obwegeser and Makek (1986). In the three-dimensional surface reconstruction, exact location and the degree of the deformity in the region from the ascending ramus to the condylar head and the lingual aspect from the ascending ramus to the mandibular body were accurately represented. In addition, the three-dimensional images could be easily rotated arbitrarily, precise evaluation could be done at every part of the mandible. On diagnosis, the mandibular morphology classified into the unilateral hybrid forms was presumed to vary from case to case even in the same classification.

Effect of glutathione depletion on sunburn cell formation in the hairless mouse.

Cutaneous protection against ultraviolet B (UVB) radiation damage by endogenous glutathione (GSH) was evaluated in the epidermis of the hairless mouse by measuring the influence of GSH depletion on sunburn cell (SBC) formation. Cellular GSH exerts antioxidant effects and recent studies have suggested a role for oxygen radicals in the production of SBC. Hairless mice (Skh/h 1) received oral treatment with buthionine S,R-sulfoximine (BSO), an irreversible inhibitor of gamma-glutamylcysteine synthetase, to deplete cutaneous GSH; 4 d later their ears were exposed to UVB radiation. BSO treatment significantly reduced GSH levels in the epidermis to 10-15% of control levels. Twenty-four hours after UVB exposure, SBC counts in the ears of animals with and without BSO treatment were measured, and those exposed to UVB were found to have increased. Greater numbers of SBC were found in the ears of BSO-treated mice exposed to 15 or 20 mJ/cm2 UVB, than in non-BSO-treated mice exposed to the same UVB doses. At higher UVB doses, there were no statistically significant differences between the groups. The results show that endogenous GSH provides the epidermis with measurable protection against injury by low or moderate UVB doses.

[Dental anthropological study of Mongoloid in China].

The purpose of this study was to evaluate the craniofacial and occlusal characteristics of the inhabitants in China. The material consisted of cephalograms, pantomograms, dental casts and anthropological measurements taken from 517 inhabitants: 416 Han Chinese in Beijing, Shanghai, Guangzhou and Changchun and 101 Mongolians in Hohhot. The data were compared among the above five regions and also with the Japanese data. The findings were summarized as follows: I. Craniofacial characteristics; a) The Han Chinese and the Mongolians showed brachycephaly. b) The inhabitants in Guangzhou showed a tendency of bimaxillary protrusion with a smaller facial height, and the inhabitants in Changchun showed a larger gonial angle with a larger facial height than those of the other regions. c) The Mongolians had a larger value of the saddle angle and of the bi-gonial breadth. d) Compared with the Japanese, their craniofacial complex was characterized by a larger facial angle, smaller mandibular plane angle and a smaller gonial angle. II. Occlusal characteristics; a) The inhabitants in China had a similar tendency in the dental arch form and in the dental trait frequency. b) The inhabitants in Beijing had a larger mesiodistal crown-diameter of the anterior teeth than those in the other regions. c) Compared with the Japanese, their teeth were characterized by a smaller mesiodistal crown-diameter (especially on the upper first molar), higher frequency of shovel-shape and lower frequency of Carabelli's tuberculum.

Protective effects of cadmium chloride against UVB injury in mouse skin and in cultured human cells: a possible role of cadmium-induced metallothionein.

Metallothionein (MT) is a cysteine-rich protein with antioxidant and metal-chelating activities that is readily inducible by exposure to a variety of stimuli including heavy metals such as cadmium (Cd2+). We have investigated the protective effects of Cd2+ treatment on sunburn cell (SBC) induction in mouse skin in vivo and human cell survival in vitro after UVB exposure. The number of SBC in mouse ear skin was significantly reduced in Cd(2+)-treated mice (10 mumol CdCl2/kg) with each UVB dose (25, 50 and 100 mJ/cm2) compared with controls. Delay of UVB exposure after Cd2+ administration (24 h) and higher doses of CdCl2 (10-30 mumol/kg) were more efficient in reducing SBC formation. Human bladder cancer cells (MGH-U1) made tolerant to Cd2+ by repeated low dose exposure, or cells acutely exposed to high Cd2+ concentration showed increased tolerance (cell survival) to UVB injury. Electrophoretic-autoradiographic analysis of [35S]-cysteine-labeled protein synthesized by cultured cells after Cd2+ treatment revealed increased MT-like protein content. These results suggest that MT is inducible by exposure to Cd2+ in our system and may be a photoprotective agent against UVB-induced oxidative damage in mammalian skin.

Immune responses in newly developed short-lived SAM mice. IV. Chromosomal location of a gene controlling defective helper T-cell activity.

Short-lived SAMP-P/1 mice are low responders in in vitro antibody responses because of a selectively impaired helper T(Th)-cell activity. After crossing with high responders (B10.BR mice), about 12% of (B10.BR x SAM-P/1) (BRP)F2 mice showed low responsiveness, as did SAM-P/1 mice, against two T-dependent antigens, sheep and horse red blood cells (RBC), both of which were not cross-reactive to each other at helper T- and B-cell levels. The immune activities against the two antigens in individual BRPF2 mice showed a good correlation (r = 0.81), thereby suggesting that SAM-P/1 mice have an antigen non-specific Th cell dysfunction. Based on the incidence of the low responders in F2 generation and statistical analyses, the hypo-responsiveness was postulated to be controlled by two genes. To survey the location of these genes, linkage analyses were performed in the F2 mice using a large set of genetic markers. Low responders in the F2 generation showed a significantly higher incidence of SAM-P/1 genotype at the Gpi-1 as well as c locus on chromosome 7 (Chr.7). However, no linkage of low responsiveness to the Hbb locus was evident, an area present at a more distal site to the centromere on the same chromosome. These results suggest that one of the genes controlling the hypo-responsiveness of SAM-P/1 mice is linked to both Gpi-1 and c loci and that it locates at a more proximal site on Chr.7.

A temperature-sensitive mammalian cell mutant with thermolabile serine palmitoyltransferase for the sphingolipid biosynthesis.

We devised an in situ assay method for the activity of serine palmitoyltransferase (SPT) that catalyzes the first step in sphingolipid biosynthesis and isolated a temperature-sensitive mutant of Chinese hamster ovary cells with thermolabile SPT. This mutant stopped growing at 40 degrees C after several generations, although the cells grew at 33 and 37 degrees C at rates similar to those of the parent. The SPT activity in cell homogenates of the mutant grown at low temperatures was 4-8% of that in the parent homogenates. When the cells were cultured for several generations at 40 degrees C, the activity in the mutant homogenate became negligible. When cell homogenates were incubated at 45 degrees C before enzyme assay, mutant SPT was more markedly inactivated than parental SPT, indicating that mutant SPT had become thermolabile. The rates of de novo synthesis of sphingolipids in the mutant were much slower at 40 degrees C than at lower temperatures, in contrast to those in the parent. The sphingomyelin content in the mutant cultivated at 40 degrees C for several generations was also less than that at low temperatures. These results indicate that SPT functions in the main pathway for sphingolipid biosynthesis. The temperature-sensitive growth of the mutant defective in sphingolipid synthesis suggests that sphingolipid(s) plays an essential role in cell growth.

The importance of Val-157 hydrophobic interaction for papain inhibitory activity of an epoxysuccinyl amino acid derivative. A structure-activity relationship based on the crystal structure of the papain-E-64-c complex.

Based on the crystal structure of the papain-E-64-c complex, 3-dimensional binding modes of a series of epoxysuccinyl amino acid derivatives to the papain active site have been constructed and the structure-inhibitory activity relationship has been analyzed using the accessible surface area and nonbonded energy parameters. The result indicates the importance of the hydrophobic interaction between the amino acid side chain of the inhibitor and the papain Val-157 residue for revealing the potent inhibitory activity.

Circadian rhythms in depression. Part I: Monitoring of the circadian body temperature rhythm.

In order to develop a sampling strategy and a method for analyzing the circadian body temperature pattern, we monitored estimates of the temperature in four ways using rectal, oral, axillary and deep body temperature from the skin surface every hour for 72 consecutive hours in 10 normal control subjects. Data obtained were analyzed with the least-squares method to estimate the circadian rhythm. Deep body temperature from the skin surface correlated better with rectal temperature than with either oral or axillary temperature in all subjects. The correlation coefficient between rectal and deep body temperature from the skin surface ranged between 0.66 and 0.89. The acrophases calculated from the deep body temperature from the skin surface every 2 h for 48 h were consistent with those based on the rectal temperature every 10 min for 72 h. We believe that monitoring of the deep body temperature from the skin surface, in this manner, at intervals of 2 h for a period of 2 days, is a promising method for investigating circadian body temperature rhythm, because it causes less discomfort than other measures and has greater statistical validity for data computation.

Circadian rhythms in depression. Part II: Circadian rhythms in inpatients with various mental disorders.

We investigated longitudinally the circadian body temperature rhythms in 65 inpatients with various mental disorders, 38 of whom had major depressive episodes, by estimating the deep body temperature from the skin surface every 2 h for a consecutive 48-h period. To estimate the circadian rhythm, the data obtained were analyzed by the least-squares method and maximum entropy spectral analysis (MEM). Circadian rhythm disturbances in patients with depression were likely to be manifested in phase variability rather than in phase advance. The amplitude of body temperature of the depressed group was significantly less than that of the control group. A positive correlation between the mesor and the severity of the depressive symptoms was found. Moreover, the body temperature of the patients with affective disorders, both in the depressive and in the manic state, tended to fluctuate so much as not only to fit poorly to sinusoid curves, but also to reduce the periodicity of the circadian rhythm. On the basis of these findings, we conclude that the essential feature of the rhythm disturbances in affective disorders is not the phase shift but the instability of the circadian temperature rhythm.

Three-dimensional analysis of facial morphology before and after orthognathic surgery.

A method for three-dimensional analysis of the facial hard- and soft-tissue morphologies is described. The soft-tissue analysis consisted of calculating three-dimensional values of reference points on the face by perspective transformation of their values in two pairs of photographs, taken simultaneously, from the right and left sides of the face. The shape of the mandible was analyzed three-dimensionally by the simultaneously taken frontal and lateral cephalograms. The hard- and soft-tissue changes were analyzed with the method in 28 patients in whom mandibular prognathism had been corrected by orthognathic surgery. The magnitude of the surgically-produced soft tissue volumetric changes in the anterior mandibular region was proportional to the posterior movement of the mandible. Asymmetry of the face also improved in response to correction of lateral deviation of the mandible and a close correlation between the directional indices of asymmetry of the hard and soft tissues was observed. Thus, the method was found to be quite useful for the analysis of facial morphology in jaw deformity.

Effects of dietary restriction on age-related immune dysfunction in the senescence accelerated mouse (SAM).

The effects of age and dietary restriction on immune response were investigated using an animal model of accelerated senescence (senescence accelerated mouse, SAM). The experimental groups consisted of control (ad libitum fed) and restricted groups (fed 60% of energy intake of the controls). Spleen weight and total number of splenic cells were significantly lower in the food-restricted group at 8 mo of age. Percentages of T (Thy-1.1+) and B (surface Ig+) cells in the splenic cells were not significantly different between the two groups. The number of direct hemolytic plaque-forming cells per 10(6) spleen cells 4 d following immunization with sheep red blood cells and dinitrophenyl-Ficoll was significantly greater in the 8-mo-old mice in the food-restricted group than in the control group. In the latter group, antibody responses Progressively decreased with age. Mitogen responses to concanavalin A and lipopolysaccharide were maintained in the food-restricted group but were depressed in the control group at 8 mo. In addition, though autoantibody to single-stranded DNA increased in the control group with advancing age, there was a steady decrease in the food-restricted group until 8 mo. Serum immunoglobulin (IgA and IgM) concentrations were significantly lower in the food-restricted group than in controls at 8 mo of age. Therefore, our results suggest that when senescence accelerated mice are subjected to food restriction, there may be a modulatory effect on the immune dysfunction associated with advancing age.

The susceptibility of Campylobacter pylori to antiulcer agents and antibiotics.

The antibacterial activities of antiulcer agents and antibiotics against Campylobacter pylori were studied. The MIC90 values of three kinds of antibiotics--macrolides, beta-lactams, and metronidazole--were 0.05-0.78, 0.39-1.56, and 12.5 micrograms/ml, respectively. They were more active than antiulcer agents such as H2-blockers and cetraxate with MIC90 values of greater than or equal to 1,600 and greater than 1,600 micrograms/ml, respectively. Especially, clarithromycin, a new derivative of erythromycin, showed an MIC90 of 0.05 microgram/ml. However, the other antiulcer agents such as sofalcone and tripotassium dicitrate bismuthate (TDB) also had MIC90 values of 50 and 6.25 micrograms/ml, respectively. Clarithromycin, sofalcone, and TDB showed bactericidal activity against C. pylori CLO2. The bactericidal actions of these drugs could be observed under electron microscopy.

In vitro messenger properties of a satellite RNA of cucumber mosaic virus.

The nucleotide sequence of a naturally occurring satellite RNA (satRNA) of cucumber mosaic virus strain F (CMV-F) isolated from Petasites japonicum Miq. has been determined. F-satRNA is 338 nucleotides long and possesses a single open reading frame (ORF; residues 11 to 91) in its 5-terminal region. Analysis of in vitro translation products of the RNA revealed that the major peptide synthesized had an Mr of 2800, corresponding to the value (Mr 2874) calculated from the amino acid sequence predicted from the ORF. Furthermore, a ribosome-binding fragment of the RNA in the initiation step of an in vitro translation system was found to contain the first AUG codon (residues 11 to 13). These results suggest that of the F-satRNA sequence, the coding region for the peptide can be assigned to the region from AUG to GGU (residues 11 to 91), accompanied by a termination codon UGA.

Differential diagnosis and pathophysiology of Cushing's syndrome and primary affective disorder.

Most patients with major depression have increased 24-hour urinary free cortisol and cortisol nonsuppression after dexamethasone administration, which are cornerstones of a diagnosis of Cushing's syndrome. Similarly, Cushing's syndrome patients often suffer from major psychiatric syndromes, most often depression. These similarities between the two conditions sometimes make it difficult to differentiate them and have led some investigators to suggest they are two points on a spectrum of endocrinologic dysfunction. This article reviews the literature comparing Cushing's syndrome and primary affective disorder and presents two cases that illustrate just how closely these diseases may resemble one another.
