A survey of rotavirus infection in the topics.

The age distribution of rotavirus antibody in the serum of inhabitants of Laos and Indonesia was studied by the neutralization test using antigenically related calf rotavirus (NCDV) as a substitute for non-cultivable human rotavirus. The results revealed that both the rate of antibody-positives and the modal titers of antibody distribution by each age group in these countries were higher than those in respective are groups of Japanese, which suggest higher endemicity of rotavirus infection in these countries. A survey of rotavirus in diarrheic patients' stools by enzyme-linked immunosorbent assay indicated that the virus infection occurred most frequently among infants between 4 and 12 months of age.

A new sesquiterpene antibiotic, heptelidic acid producing organisms, fermentation, isolation and characterization.

A new sesquiterpene antibiotic, heptelidic acid, was found in the culture filtrate of three different strains of fungi isolated from soil samples. These strains were identified as Gliocladium virens, Chaetomium globosum and Trichoderma viride. Heptelidic acid was produced by conventional submerged culture and purified by successive column chromatography on silica gel and Sephadex LH-20 and finally by preparative TLC on silica gel. The molecular formula of heptelidic acid was determined as C15H20O5 on the basis of elementary analysis and high resolution mass spectrometry of its monomethyl ester. The antimicrobial spectrum of the antibiotic revealed its specific activity against anaerobic bacteria, especially against Bacteroides fragilis.

[Primary endodermal sinus tumor of the fourth ventricle (author's transl)].

A case, a 18-year-old male, of an endodermal sinus tumor (yolk sac tumor) in the fourth ventricle, was reported. The patient had a month history of headache, vomiting and gait disturbance prior to the hospitalization, when he admitted to our service he was in lethargic condition with left cerebellar ataxia and horizontal nystagmus. Lumbar tap revealed clear CSF under normal pressure of 110 mm H2O with the CSF protein of 432.5 mg/dl and cell count of 147/3. The vertebral angiography demonstrated space occupying lesion in the posterior fossa. Plain CT demonstrated only disappearance of the fourth ventricle and slightly dilated bilateral ventricles and third ventricle. However diffuse high density area around the fourth ventricle was demonstrated and the wall of bilateral anterior horn was slightly enhanced, after injection of contrast media. There was no other abnormal findings around the pineal region. Suboccipital craniectomy was performed and the tumor was totally removed macroscopically. The tumor was situated in th floor of the fourth ventricle and infiltrated into the fourth ventricular wall and th adjacent cerebellar tissue. The tumor was with soft, greyish color and extremely vascular. Histologically the tumor was diagnosed as endodermal sinus tumor according to Teilum's classification. There were stellate cells arranged in a loose with vacuolated network which formed cystic cavities and a complicated network of honeycomb appearance with a system of communicating cavities and channels. Various size of intra- and extracellular PAS-positive hyaline globules were also seen. Glomerular-like structure (Schiller-Duval body) was not observed. Immunoperoxidase study clearly demonstrated the presence of intra- and extracytoplasmic alpha-fetoprotein granules in the tumor tissue. The amount of the serum alpha-fetoprotein, measured by radioimmunoassay, showed 400 ng/ml. After irradiation in the posterior fossa (5000 rad) the patient was discharged. Three months later, follow up CT demonstrated small high density area in the anterior horn of the left lateral ventricle, so he was rehospitalised. Irradiation in the whole brain was again administered. The tumor was very radiosensitive. CT, after 800 rad, demonstrated complete disappearance of the tumor. After irradiation totally (3000 rad), he discharged with left cerebellar ataxia.

Clinico-hematologic and serological comparison of Japanese and Russian strains of Theileria sergenti.

The Japanese and Russian strains of Theileria sergenti were compared clinico-hematologically and serologically. They were subjected to passage in splenectomized calves 4 approximately 8 months old by the infectation of infected ticks or the inoculation of infected blood. As a result, the average of incubation of the Japanese strain was proved to be 23 days by the tick infection and 21 days by the inoculation of infected blood stored in a frozen state. The rate of detection and the average size of various forms of Theileria in the proliferative stage of the Japanese strain were as follows: Comma-shaped and bacilli forms, 86.8%; bacilli from 2.15 x 0.73 micron; oval and spherical forms, 11.3% and 1.97 x 1.22 and 1.46 micron, respectively; tetracoccal form and anaplasma-like form, 0.9 and 1.0%, respectively, and 0.54 micron in diameter. The highest parasitic rate of the Japanese strain was 11.9% on the average, and the rate of decrease in erythrocyte count 70.6% on the average. Rods were seen in protozoa particularly rich in cytoplasm. Such symptoms as pyrexia, depression, and anorexia appeared as Theileria propagated. No hemoglobinuria occurred or no schizonts were detected. Essentially the same results were obtained from the Russian strain as from the Japanese. Besides, Haemaphysalis longicornis produced in Japan acted also as a vector for the Russian strain. There was a good serological agreement between the Japanese and the russian strain in the results of the cross complement fixation test and the cross indirect fluorescent antibody test. There was no difference in the results of the complement fixation with the eight strains of the Japanese strain.

Carcinogenicity of 1,2-dimethylhydrazine dihydrochloride in BALB/c mice. Influence of the route of administration and dosage.

The carcinogenicity of 1,2-dimethylhydrazine dihydrochloride (DMH) by oral, intragastric and subcutaneous administration was examined in 339 BALB/c mice. Subcutaneous injection of DMH induced intestinal tumors in the lower colon of all mice. After oral administration it induced a high incidence of vascular tumors in the liver and soft tissues, but colon tumors were found in only 2 mice when given at a high dosage. On intragastric administration, it induced a fairly high incidence both of colon and vascular tumors. The sites and incidences of vascular tumors and squamous cell carcinomas of the perianal glands were also described.

Immune responses in rabbits treated with tetracycline.

Cellular and humoral immunological phenomena in rabbits treated with tetracycline were investigated by lymphocyte transformation, leucocyte migration inhibition, and indirect hemagglutination tests. Cellular and/or humoral immunity against tetracycline and/or homologous or autologous rabbit gamma globulin was acquired by about two-thirds of the rabbits, with some also acquiring immunity against homologous tissue antigen after prolonged tetracycline administration. Based on these results, the significance of immune responses to autoantigens by drug administration was discussed in relation to potential agents for the induction of autoimmune diseases.

Demonstration of antibodies to chlorophenothiazine derivatives.

Antibody to 2-chloro-10-(2-carboxyethyl)-phenothiazine (CECP) was produced in rabbits by immunization with the hapten-BSA conjugate. The titer of antibody was determined by passive hemagglutination (PHA) using erythrocytes coupled with the CECP-protein carrier. Results from inhibition tests using structurally related (CECP, CPZ, PCPZ, CMP) and almost unrelated haptens (DDA, RF) strongly suggested that both the side chain and the nucleus of the haptens are important in determining the specificicity of immunological reactions. 18 sera from psychiatric patients administered phenothiazine derivatives for a long time as well as 20 control sera from healthy individuals were then tested for their reactivity to CECP. Of patients' sera, 12 had detecable hemagglutination titers (1:8 or greater), while all of the normal human sera were negative. The PHA activity was contained in euglobulin fraction and was specific in the inhibition tests. From these results, the activity to CECP in patients' sera was supposed to be specific antibody.

Demonstration of antibody activity to clioquinol in sera of experimental rabbits and patients with subacute myelo-optico-neuropathy.

Antibody to clioquinol(5-chloro-7-iodo-8-hydroxyquinoline:CIHQ) was detected by passive hemagglutinating reaction in rabbits receiving a prolonged administration of Emaform which was once a commercial preparation of CIHQ. The antibody was shown to be in the immunoglobulin fractions by separation with a specific immunoadsorbent, and it had relatively broad antigenic specificity. The antibody was also demonstrated in sera from patients who suffered from Subacute Myelo-Optico-Neuropathy (SMON) and from normal healthy individuals. However, its titer and frequency of positive reactors in the former were higher than those in the latter, and the two groups could be differentiated each other in frequency distribution patterns of the antibody proprietors. From these results, we discussed on desirable application of sero-epidemiological study to assessment of effects of chemical pollutants on living systems.

A radioimmunoassay of plasma unconjugated and conjugated estetrol.

A radioimmunoassay for the measurement of both unconjugated and conjugaged estetrol in plasma has been developed. The antiserum obtained after 6 months of immunization with 6-oxoestetrol-6-(O-carboxy-methyl)oxim-BSA was used at a final dilution of 1:90,000 and showed almost no cross reaction with other steroids except for estriol at 1.24%. Esterol-glucosiduronate was synthesized by incubating with adrenalectomized rat liver homogenate and uridine diphosphoglucuronic acid. Then, plasma estetrol-glucosiduronate was measured in the same manner for unconjugated estetrol after hydrolysis with beta-glucuronidase. Sephadex LH-20 column chromatography (7X110 mm, benzene:methanol, 85:15) was employed for accurate assessment. The sensitivity was 10 pg and the smallest amount measurable was 40 pg/sample. The method bland was consistently negligible. The intra and inter assay precision was 11.8% and 14.2% for unconjugated estetrol and that for estetrol-glucosiduronate was 13.5% and 17.1%.

Radioimmunoassay of 16alpha-hydroxy-dehydroepiandrosterone and its sulfate.

A simple and reliable radioimmunoassay for plasma 3beta, 16alpha-dihydroxy-5-androsten-17-one(16alpha-OH-DHEA) and its sulfate has been developed. The antiserum against 16alpha-OH-DHEA and its sulfate (16alpha-OH-DHEA-3-sulfate) was produced in rabbits immunized with 16alpha-OH-DHEA-3-succinate-bovine serum albumin. This antiserum reacted well with both 16alpha-OH-DHEA and its sulfate and only slightly cross reacted with DHEA and its sulfate. The coefficient of variation (C.V.) of the intra assay was 10.26% for 16alpha-OH-DHEA and 12.32% for 16alpha-OH-DHEA-S. The C.V. of the interassay were 14.34% for 16alpha-OH-DHEA and 15.64% for 16alpha-OH-DHEA-S. The umbilical artery concentrations for 16alpha-OH-DHEA and 16alpha-OH-DHEA-S were 7.20 +/- 6.71 ng/ml and 4490 +/- 2140 ng/ml, and the umbilical vein concentrations were 14.20 +/- 11.27 ng/ml and 2970 +/- 1450 ng/ml respectively.
