Identification of a diuretic hormone of Locusta migratoria.

We have isolated a peptide from brains and corpora cardiaca of Locusta migratoria which is immunologically related to the diuretic hormone of Manduca sexta. We determined its structure as a 46 amino acid linear peptide with 43-50% identity to the M. sexta hormone. Moreover, we showed that the new peptide functions as a diuretic hormone in L. migratoria, stimulating urine production by Malpighian tubules and elevating levels of cAMP in tubules.

Oscillating activity of a calcium-activated K+ channel in normal and cancerous mammary cells in culture.

Calcium-activated potassium channels were the channels most frequently observed in primary cultured normal mammary cell and in the established mammary tumor cell, MMT060562. In both cells, single-channel and whole-cell clamp recordings sometimes showed slow oscillations of the Ca2(+)-gated K+ current. The characteristics of the Ca2(+)-activated K+ channels in normal and cancerous mammary cells were quite similar. The slope conductances changed from 8 to 70 pS depending on the mode of recording and the ionic composition in the patch electrode. The open probability of this channel increased between 0.1 to 1 microM of the intracellular Ca2+, but it was independent of the membrane potential. Charybdotoxin reduced the activity of the Ca2(+)-activated K+ channel and the oscillation of the membrane current, but apamin had no apparent effect. The application of tetraethylammonium (TEA) from outside and BaCl2 from inside of the cell diminished the activity of the channel. The properties of this channel were different from those of both the large conductance (BK or MAXI K) and small conductance (SK) type Ca2(+)-activated K+ channels.

Mixed tumor of salivary gland type arising in the bronchus.

During bronchoscopy for atelectasis of the middle lobe, a 61-year-old man was found to have a polypoid mass obstructing the bronchus. Right middle and lower lobectomy disclosed a tumor growing into the lumen of the middle lobe bronchus. The histological features of the tumor were indistinguishable from those of the mixed tumor of the salivary gland. A review of the literature reveals only 7 cases of this extremely rare tumor.

An established cell line of larval Echinococcus multilocularis.

A cell line of larval Echinococcus multilocularis has been established from an echinococcal cyst excised surgically from a patient with alveolar hydatid disease. A standardized procedure established for the preparation and continuation of primary cultures was applied to isolate the E. multilocularis cells from the tissue fragments. Trypsin was used for the enzymatic release of the monodispersed cells from the tissue fragments and for dispersing monolayers. The culture medium was RPMI 1640 with 10% fetal calf serum. Cell supports were collagen-coated plastic dishes and flasks. The morphological features of the cultured cells showed spindle-like cells during the first few subcultures, and then polygonal or star-like cells. Population doubling time at passage 34 was approximately 23 h and plating efficiency at the same passage was 15%. Chromosome numbers obtained from 70 metaphase plates at passage 40 ranged between 14 and 104 and cells with 91-100 chromosomes were clearly predominant. The chromosomes could be morphologically classified into telocentric, subtelocentric, and metacentric types. Over 90% of the chromosomes were of the telocentric type. Cells collected at passage 57 were intraperitoneally inoculated into two cotton rats (Sigmodon hispidus) at a cell concentration of 10(7) and the rats were sacrificed 100 days later. It was found that the two rats had echinococcal cyst masses in the peritoneal cavity. This result indicates that our isolated cells are germinal cells with ability to differentiate into cystic structures in vivo.

Positional dislocation of the pisiform.

Positional dislocation of the pisiform repeatedly occurred in the right wrist of a 19-year-old man following a motorcycle accident. The pisiform repeatedly dislocated distal to the triquetrum when the wrist was extended to 45 degrees or more, and the pisiform was reduced to proper joint congruity after wrist flexion of 55 degrees or more. The patient had a background of generalized joint laxity. Resection of the pisiform resulted in a pain-free, fully functional wrist.

Clinical application of sapphire pins as an internal fixation device for the upper extremity.

Since June 1980, sapphire pins (monocrystalline alumina ceramic pins) have been used in 22 patients as an internal fixation device for hand and elbow problems. In our follow-up studies, good bone healing was observed in all cases except for one delayed union in a fracture of the diaphysis of the proximal phalanx. Radiographs showed no pin migration or osteolytic reaction around the pins. We therefore conclude that our device and its insertion technique can be successfully applied to intraarticular fractures and fractures in the vicinity of the joint.

Desmoplastic fibroma in the thoracic spine.

Desmoplastic fibroma of the spine is an extremely rare, benign tumor. So far, only 7 cases have been reported in the world. We have treated 1 case of desmoplastic fibroma in the thoracic spine. Due to insufficient removal of the tumor, local recurrence was observed, and a second surgery posteriorly and anteriorly was performed for complete removal 8 years following the first operation. After resection of the tumor, Luque instrumentation was used to reconstruct the destroyed spine. At present, 7 years after the second operation, there are no signs or symptoms. Therefore, complete resection of the tumor is most important in the treatment, and rigid spinal instrumentation and bone grafts must be used after removing the tumor in spine cases.

Localization of [125I]endothelin-1 in rat tissues observed by electron-microscopic radioautography.

[125I]Endothelin-1 (ET-1) was administered intravenously into rats, and binding sites in various tissues were examined by electron-microscopic radioautography. Labeling was observed predominantly on the glomerular fenestrated endothelial cells of kidney, alveolar capillary endothelial cells of lung, fat-storing cells in the sinusoids of liver, and subepithelial myofibroblasts of the small intestine. At first, silver grains were observed mostly on the plasmalemma of these cells and with time translocated into the cytoplasm of these cells. Myofibroblasts of the small intestine were cultured to study the cellular effects of ET. By fura-2 fluorescence method, transient increase and oscillation of intracellular Ca2+ level were observed in the cultured myofibroblasts following the application of ET-1 or endothelin-3 (ET-3). These results suggest that ET induces diverse physiological reactions in specific cells of various tissues.

Postoperative magnetic resonance imaging in patients with cervical myelopathy.

Magnetic resonance imaging using T1-weighted sagittal images was performed on 81 postoperative patients with cervical myelopathy. Cord compression was observed after surgery in 32 (40%) of the patients. Compression was demonstrated in the decompressed area in 9 patients, at the upper or lower edge of decompression in 11 patients, and at adjacent levels of fusion in 11 patients. The remaining patient had cord compression in both the decompressed area and at the adjacent level. From their operative results, cord compression was proved to be functionally innocuous in 13 (41%) patients and the cause of neurologic deterioration in 3 (9%); it was not clear whether or not the compression was innocuous in the remaining 16 (50%) patients.

Intraoperative monitoring for tethered spinal cord syndrome.

Functions of the lower extremities and bowel and bladder must be monitored during releasing surgery for tethered spinal cord syndrome, because neural elements are embedded in lipoma or anomalous tissues. Evoked muscle action potentials of the external anal sphincter muscle, external urethral sphincter muscle, and lower extremity muscles can indicate promptly whether or not neural elements are involved before the surgeon releases or cuts the tissue for cord release. It is also important to monitor vesical pressure because of different types of innervation from the external urethral and anal sphincters, in spite of slow reaction times of vesical pressure elevation. Evoked muscle action potentials indicate only motor function; therefore, it is more appropriate to record evoked spinal cord potentials from the spinal cord above a lumbosacral operative field, because sensory function can also be monitored. Evoked muscle action potentials are an easy and extremely sensitive monitoring system; nevertheless, monitoring of vesical pressure and evoked spinal cord potentials should also be done to achieve optimum monitoring. For the past 5 years, 10 patients have undergone cord-release surgery with such a monitoring system, and the results indicate no exacerbating case.

Enhanced expression of CD2 antigen on lung T cells.

CD2 molecules not only function in the adhesion of T cells to other cell types but also in the activation of T cells via an alternative pathway. To investigate the possible roles of CD2 molecules in the activation and accumulation of lung T cells, using monoclonal antibodies and a flow cytometer we evaluated CD2 antigen expression on lung and blood lymphocytes in 10 normal subjects, 30 patients with pulmonary sarcoidosis, 7 patients with interstitial pneumonia associated with collagen vascular disease, 5 patients with farmer's lung disease, and 8 patients with Crohn's disease. The mean fluorescence intensity (MFI) of CD2 on lung T cells obtained by bronchoalveolar lavage (BAL) was significantly higher than that on blood T cells in all study groups except the control group. In patients with pulmonary sarcoidosis this enhancement of the expression of CD2 antigen was observed only on CD4+ T cells. Since ligands for CD2 are present on immunocompetent cells of other types in the local pulmonary milieu, these results suggest that increased expression of CD2 molecules could facilitate the communication and interaction of lung T cells with their environment in the lung and thereby possibly contribute to the local accumulation of T cells.

Cornexistin: a new fungal metabolite with herbicidal activity.

Cornexistin, a new compound demonstrating promising herbicidal activity, was purified from the culture filtrate of a newly-isolated fungus identified as Paecilomyces variotii SANK 21086. The compound was extracted with organic solvents from the culture filtrate, purified using column chromatography on Sephadex LH-20 and finally crystallized from methylene chloride. Following analysis of its physico-chemical properties it was identified to be a new compound belonging to the nonadride group. Chemical structure elucidation was conducted by analyses of various spectral data and the structure was finally confirmed by means of X-ray crystallographic analysis. Based on its herbicidal characteristics cornexistin may be classified as a postemergence herbicide active against certain young annual and perennial monocotyledonous and dicotyledonous plants with selective protection for corn.

Synerazol, a new antifungal antibiotic.

Synerazol, a new antifungal antibiotic, was isolated from cultured broth of Aspergillus fumigatus SANK 10588. The structure was determined based on NMR and mass spectral evidences. Synerazol was found to be a related substance to pseurotin A. Synerazol was active against Candida albicans and other fungi, and showed marked synergistic activity with azole-type antifungal agents.

Phosphoinositides and synaptic function in NG108-15 neuroblastoma x glioma hybrid cells.

1. The second-messengers system of bradykinin (BK) receptors was examined in NG108-15 neuroblastoma x glioma hybrid cells. 2. An application of BK induced an immediate outward (K+) current and acetylcholine (ACh) release, which are generated through inositol 1,4,5-trisphosphate (InsP3)-dependent Ca2+ ions. 3. Application of phorbol dibutyrate (a protein kinase C activator) produced a voltage-dependent inward current and inhibited another K+ (M)-current. 4. A similar current response has been produced by ACh in NG108-15 cells transfected with rodent muscarinic ACh receptor I and III subtype genes. 5. These results suggest a dual and time-dependent role for these two intracellular messengers in the control of neuronal signalling by BK and ACh.

[Genetic factors and smoking habits].

To investigate whether genetic factors are involved in the maintenance of smoking habits, we examined restriction fragment length polymorphisms of cytochrome P-450 gene (P450 IIB1), one of the metabolizing enzymes of nicotine. The presence or the absence of polymorphic bands detected after digestion with Bam HI or Msp I was not related to smoking habits, the subjective degree of fondness for smoking and the subjective strength of addiction to smoking. Thus, we found no evidence for the involvement of this gene or adjacent gene(s) in the determination of smoking habits.
