Localization of dehydropeptidase-I, an enzyme processing glutathione, in the rat kidney.

An antibody prepared against dehydropeptidase-I, one of the glutathione processing enzymes, from the renal membrane fraction of rats was employed to localize at the light microscopic level the enzyme in the kidney using the avidin/biotin-peroxidase complex method. Dehydropeptidase-I was found to be present on both the brush border and the basolateral membranes of proximal tubular cells. Furthermore, the enzyme activity in the isolated brush border and basolateral membrane fractions was also determined. Distribution profiles of the enzyme activity showed good agreement with the results of the immunohistochemical observations.

High prevalence of isolated sleep paralysis: kanashibari phenomenon in Japan.

In Japan, a set of experiences called kanashibari is considered identical with isolated sleep paralysis. We investigated this phenomenon by means of a questionnaire administered to 635 college students (390 men and 245 women). Of all subjects, about 40% had experienced at least one episode of kanashibari [subjects of K(+)]. Therefore, isolated sleep paralysis is apparently a more common phenomenon than is usually appreciated. About half of the subjects of K(+) reported that they had been under "physical or psychological stress" or in a "disturbed sleep and wakefulness cycle" immediately before the episode. Many subjects of K(+) experienced the first episode in adolescence. In the distribution of age of first attack, the peak occurred at an earlier age in women subjects than in men subjects. These findings suggest that two factors influence the occurrence of the phenomenon. One is exogenous physical or psychological load and the other is endogenous biological development.

Differences in sleep-wake habits and EEG sleep variables between active morning and evening subjects.

This article is a survey study, followed by an experimental study, examining the differences of sleep-wake habits and sleep electroencephalographic (EEG) variables between morning and evening type subjects (Ss). In the survey study, the Japanese version of the Horne and Ostberg Morningness-Eveningness Questionnaire and Life Habits Inventory (LHI) were administered to approximately 1,500 university students. The survey results showed that the two types were significantly different from each other in terms of retiring and arising time, sleep latency, mood on arising, nap, adequate amount of sleep, number of times of staying awake all night, and variability in bedtime, arising time, and sleep length. These results suggested that evening type Ss had more irregular and/or flexible sleep-wake habits than morning type Ss. In the experimental study, 10 morning and 11 evening type Ss were selected from the population included in the survey study, and polysomnograms were obtained. The results showed that only in rapid eye movement (REM) latency did morning type Ss significantly differ from evening type Ss. REM latency might be related to personality factors, particularly to neuroticism and anxiety.

Fetal ascites. A report of 3 autopsy cases.

Three rare autopsy cases of fetal ascites were presented and the etiology of each case was described. Case 1 was a male neonate, delivered by cesarean section at 32 weeks' gestation, and died of respiratory failure. The abdomen was remarkably distended with 1020 ml of ascites. The etiology of Case 1 remained unknown even after macroscopic and microscopic examinations. We considered this as "idiopathic" fetal ascites. Case 2 was a female neonate, delivered at 31 weeks' gestation, with marked abdominal distension and cyanosis. Autopsy revealed 435 ml of ascites, and she was considered to have had "polysplenia syndrome" with cardiovascular malformations. Intrauterine heart failure due to cardiac anomalies was thought to be the cause of this ascites. In case 3 embryotomy was carried out under the diagnosis of fetal ascites by ultrasound examination at 22 weeks' gestation. An urachal cyst connected to the dilated urinary bladder and deficiency of musculature of the abdominal wall composed of loose connective tissue with calcification were observed. The abdominal wall was ruptured and 1,960 ml of ascites was measured. Polycystic kidney with renal dysplasia was also found. Case 3 showed "Prune-Berry syndrome" and fetal ascites may have arisen from these anomalies.

Squamous cell cancer of the maxillary sinus in Hokkaido, Japan: a case-control study.

A case-control study of squamous cell cancer of the maxillary sinus was performed in Hokkaido with 106 cases and 212 controls matched for sex, age (within five years), and residence (same health centre region). Univariate analyses showed that a history of chronic sinusitis (relative risk, RR = 3.2), nasal polyps (RR = 5.0), an occupational history of being a carpenter, joiner, furniture worker, or other woodworker (RR = 2.9), and current or past smoking habits (RR = 3.0) were statistically significant risk factors for men. No single item was a significant risk factor for women.

Accumulation of immunoreactive opsin on plasma membranes in degenerating rod cells of rd/rd mutant mice.

Immunoreactive opsin was detectable in the apical portion of normally developing photoreceptor cells on postnatal day 3 by the indirect enzyme-labeled antibody method. Immunoreactivity increased and had extended from the central retina to the periphery by the advanced stages of development. In the rd mutant retinas, accumulated opsin was present in the apical portion and in the outer nuclear layer on postnatal day 8. Immunoreactive opsin mainly was present in the outer nuclear layer by day 14, even being detectable on day 28. No immunoreactivity was present in the remaining cones. Electron microscopic immunocytochemistry confirmed the association of immunoreactive opsin with the persistent rod cell plasma membrane. Molecular weight of immunoreactive opsin in 14-day-old rd mutant mouse retina, as estimated by gel filtration chromatography, was large and did not seem to be degraded. These findings indicate that accumulated rhodopsin continues to function in the plasma membrane because an electroretinogram could be made after day 14 for the rd mutant mouse retina.

Intracellular free calcium concentration, Ca++ channel and calmodulin level in experimental hypertension in rats.

Using fluorescent calcium indicator quin2, we studied intracellular free calcium concentration in platelets that have a number of features similar to vascular smooth muscle cells. Intracellular free calcium concentration in platelets of male SHR was significantly higher at 4, 11 and 28 weeks old compared with age-matched male WKY. However, no significant difference was observed in platelets cytosolic free calcium level of DOCA-salt hypertensive and two-kidney, one clip hypertensive rats in the chronic stage. Cardiac Ca++ channels were estimated by means of radioligand binding method with [3H]-nimodipine. No significant changes were observed in the concentration and affinity of cardiac Ca++ channel in SHR, DOCA-salt hypertensive and two-kidney, one clip hypertensive rats. Calmodulin levels in mesenteric arteries of SHR were significantly decreased in comparison with those of WKY. However no significant differences were observed in DOCA-salt hypertensive rats in the chronic stage. These results indicate that the increase in intracellular free calcium concentration of SHR is not the secondary change caused by high blood pressure. It is impossible to detect the ratio of the three states (open, resting and inactivated) of Ca++ channel. Therefore, there remains a possibility of the changes in the ratio of the states of Ca++ channel. The observed abnormalities of Ca++ regulation may contribute to the pathogenesis of hypertension.

Pathological substratum for clinical features of fatal myocarditis.

Heart autopsies in six cases of fatal myocarditis were examined clinicopathologically. Included were 2 cases in the acute stage, 1 in the subacute stage and 3 in the chronic stage. As to microscopical dating changes, at the acute stage, numerous inflammatory cells were found in the edematous interstitium after 2 days from onset and degeneration, i.e., destruction of myocardial cells without fibrous replacement, was also noted after 11 days. At the subacute stage, irregular patchy and slightly loose fibrosis with inflammatory cells and vascularization was found after 50 days. At the chronic stage, loose fibrosis with inflammatory cells and tight fibrosis without the cells, that is, active and healed myocarditis co-existed. All cases showed heart failure of varying degrees, and also showed, respectively, complete AV block and ventricular fibrillation at the acute stage, and complete AV block and sustained ventricular tachycardia at the chronic stage. In myocarditis, which often was difficult to diagnose clinically, the pathological findings corresponded well to the clinical features retrospectively.

Tumor necrosis factor inhibiting angiogenesis in vitro.

The effect of recombinant human tumor necrosis factor (TNF) on the capillary growth was evaluated using an in vitro angiogenesis model recently developed. Sprague-Dawley rat microvessel fragments, from epididymal fat pads, seeded onto the confluent culture of myofibroblastic cells from the same tissue origin, gave rise to microvascular networks on and in the multilayered myofibroblastic cells. In contrast, the capillary growth from the vessel fragments was markedly inhibited in the presence of 10-1,000 U TNF/ml. Monoclonal antibody against TNF completely neutralized the capillary growth inhibitory activity of TNF. The mode of angiogenesis inhibitory action of TNF was also examined by use of bovine capillary endothelial (BCE) cells and rat myofibroblastic cells. TNF exerted growth inhibitory and cytotoxic actions against BCE cells cultivated on the various basement membrane components, such as extracellular matrix secreted by BCE cells, fibronectin, laminin, and type IV collagen. An irreversible damage to most of the BCE cells was observed ultrastructurally after 60 hours' exposure to TNF. TNF, however, did not injure the myofibroblastic cells but rather stimulated their growth. These findings indicate that TNF inhibits in vitro capillary growth by its direct cytostatic and cytotoxic actions to microvascular endothelial cells.

Effects of nicardipine on blood pressure and platelet cytosolic free calcium concentration in SHR.

Intracellular free calcium concentration was measured by the fluorescent calcium indicator quin2 method in platelets of spontaneously hypertensive rats (SHR) and Wistar-Kyoto (WKY) rats, and the effects of nicardipine on platelet cytosolic free calcium concentration were investigated. Cytosolic free calcium concentration was significantly increased in platelets of 4-, 11-, and 14-week-old SHR compared with age-matched WKY rats. Platelet cytosolic free calcium concentration in 14-week-old male SHR was significantly decreased 1 h after the administration of nicardipine (5 mg/kg p.o.), while that in WKY rats remained unchanged. These results suggest that transmembrane Ca2+ influx is genetically increased in SHR.

[Immunohistochemical study of new monoclonal antibodies--E29, 115D8 --to an epithelial membrane antigen (EMA) in ovarian tumors].

Mouse monoclonal antibodies E29 and 115D8 have been raised in the human milk fat globule membrane. The distribution of the antigen detected by these two monoclonal antibodies has been examined in formalin-fixed paraffin-embeded human ovarian tumors by an indirect immunoperoxidase technique. The highest positive rate of E29 and 115D8 was obtained in the malignant group suggesting that there is relationship between this positive rate and the histological malignancy. As for serous cystadenocarcinomas, a strong staining area of E29 was observed in the diffuse cytoplasm in poorly differentiated cases. These results suggest that the presence of E29 is a useful tumor marker for ovarian cancer.

[Metastatic brain tumor after chemotherapy of ovarian cancer].

A case of brain metastasis in ovarian cancer is reported. Recently cis platin-containing combination chemotherapeutic regimens have improved the prognosis of patients with advanced ovarian carcinoma. Metastasis to the brain in ovarian cancer is very rare. However, the incidence of cerebral metastasis after chemotherapy for ovarian carcinoma seems to be increasing, and this may be related to prolonged survival due to chemotherapy. Thus, neurological examination after chemotherapy with cis platin is necessary to detect metastases to the brain.

Low-dose irradiation to head, neck, or chest during infancy as a possible cause of thyroid carcinoma in teen-agers: a matched case-control study.

A matched case-control study was performed to identify the etiologic factors for thyroid carcinoma in teen-agers. Twenty-seven cases and 69 controls were investigated to assess the significance of various maternal and subject factors. Irradiation during infancy was the only factor which showed a statistically significant association with the incidence of thyroid cancer in teen-agers (summary X2 = 8.040; d.f. = 1; P less than 0.005). The estimated dose ranged from 0.2 to 40 rads on the head, neck, or chest during infancy.
