The interactions of bromcresol green with various components isolated from heat treated bovine serum albumin at 65 degrees C and pH 9.0 were studied spectrophotometrically. The difference spectra of the dye induced by native albumin and component 1, which has the same electrophoretic mobility as native albumin, had a trough at 616 nm and peak at about 668 nm, whereas those induced by the denatured monomer (component 1'), denatured dimer (component 2) and denatured trimer (component 3) had a trough at 610 nm and peak at 655 nm. From these difference spectra, it was concluded that component 1 is a mixture of at least two components (native and modified native forms) and that components 1', 2 and 3, formed during the process of denaturation, have essentially the same conformation in the environment of the binding sites for bromcresol green. A possible mechanism for the heat denaturation of serum albumin is proposed.
Crystal structure analysis of the zinc(II) complex with cytosine 5'-phosphate, (C9H12N3O8P)Zn(H2O), shows that the complex is two-dimensionally polymeric, and the zinc atom is tetrahedrally coordinated to N(3) of the pyrimidine, to two phosphate oxygen atoms and to one water molecule. The zinc atom also forms weak intramolecular interaction with O(2) of the pyrimidine.

1. Serum dopamine beta-hydroxylase activity was determined in normotensive control subjects and patients with labile or established essential hypertension. The enzyme activity was 25-9 +/- 1-9 (SEM) 29-6 +/- 2-5 and 25-1 +/- 1-9 micronmol min-1 1-1, for control, labile and established hypertensive subjects respectively. 2. Neither blood pressure nor serum dopamine beta-hydroxylase activity was changed in normotensive control subjects by administration of phentolamine; however, in patients with essential hypertension blood pressure was significantly decreased (P is less than 0-01) and serum dopamine beta-hydroxylase activity was slightly increased. With propranolol administration, blood pressure and the serum enzyme activity were not significantly changed in normotensive or hypertensive subjects. 3. Our results suggest that there is no correlation between serum dopamine beta-hydroxylase activity and blood pressure.
The abdominal skin of guinea pigs was irradiated by a Xenon short arc lamp with glass filters. Acid phosphatase and beta-glucuronidase activity increased in the irradiated skin. The change of acid phosphatase is slow and mild in intensity, while that of beta-glucuronidase is quick and high in degree. The activity of acid phosphatase is influenced equally by light with filters of UV-25, UV-27, UV-29, and UV-31, while that of beta-glucuronidase is affected more mildly by light with a UV-31 filter than with any of the other three filters. These findings suggest that there is a close relationship between cutaneous changes induced by ultraviolet light and changes in lysosome activity, and that energy required to induce changes in enzyme activity is much less than energy required to produce erythema.

Sarcoplasmic reticulum (SR) isolated from rat aorta exhibited ATP dependent Ca2+ uptake in the presence of Mg2+. The maximum capacity and apparent binding constant were 23n moles/mg and 1.8 X 10(4) M (-1), respectively. The energy source of this active Ca2+ accumulation would be Mg-ATPase associated with the membrane. Ca2+ which was taken up inside of sarcoplasmic reticulum was released rapidly by washout with the medium containing no Ca2+. These facts suggest that sarcoplasmic reticulum in vascular smooth muscle cell may play and important role in the regulation of intracellular Ca2+ concentration.

The Ca2+ antagonistic coronary vasodilator, Nifedipine, was sublingually administered by a dose of 30 mg to 19 patients with hypertension. Blood pressure of patients with with essential hypertension (n=14) decreased from 177 +/- 24 to 123 +/- 13 mmHg systolic and from 108 +/- 12 to 80 +/- 11 mmHg diastolic (mean +/- SD) (p less than 0.01). Plasma renin activity (PRA) increased significantly from 0.73 +/- 0.62 to 1.50 +/- 1.02 ng/ml/h (p less than 0.05). The same tendency was observed in malignant and renovascular hypertension. In primary aldosteronism (n = 2), blood pressure decreased but PRA did not increase. Hypotensive action and increased plasma renin activity by Ca2+ antagonist, Nifedipine, were clearly demonstrated in patients with hypertension.

Thirty patients with Behcet's disease were treated with immunosuppressive drugs, 25 with Imuran, 5 with Leukerin and 2 with both these drugs. Daily administration of 50-75 mg of Imuran over a period of 1 year alleviated the ocular symptoms in 16% of patients, was slightly effective in 20%, and ineffective in 64%. Imuran therapy was also seen to alleviate the extraocular pathology, such as stomatitis, erythema, and arthralgia. Leukerin proved difficult to prescribe over a long period because of severe side effects.
A striking chemotherapeutically curative effect on tumor was obtained by means of temporary interruption of regional blood flow combined with local hyperthermia. By analyzing various basic conditions required for this system using Ehrlich tumor implanted in the hind limbs of mice, the following were found to be essentially indispensable to obtain satisfactory chemotherapeutic effects: (a) a time interval of 1 to 3 min after systemic i.v. administration of drug to the mice, (b) use of a tourniquet on the tumor-bearing mouse limb to stop blood flow, and (c) warming at 37-41 degrees (d) for a period of at least 30 to 60 min. Among the chemotherapeutic drugs tested in the present study, Carbazilquinone (NSC 134679) was the most effective because it revealed the strongest antitumor effect despite its relative innocuousness to nontumorous adjacent normal tissues. Applying the present method, a large syngeneic mouse sarcoma transplanted to the limb 7 days before the experiment also completely regressed in 6 of 9 mice.
