Effects of macromolecular compounds on human leukocyte beta-galactosidase activity.

The enzyme properties of leukocyte beta-galactosidase and the effects of macromolecular compounds on its activity were studied in detail. It was demonstrated that various macromolecular compounds markedly influence the activity of beta-galactosidase. These phenomena should be taken into account when considering the biochemical abnormalities of tissues of patients with mucopolysaccharidoses and mucolipidoses.

Infantile neuroaxonal dystrophy. Schwann cell inclusion in the peripheral nerve.

Abnormal Schwann cell inclusion is reported in biopsied peripheral nerve in a case of infantile neuroaxonal dystrophy. In addition to non-specific dystrophic changes of the axons, the Schwann cells contained several distinct bodies which were composed of stacks of irregularly disposed membranes; in some instances, transformed Schwann cell cytoplasm was distined with similar bodies. This change has not been reported on this condition and its significance is briefly discussed.

Relationship between epidemiologic and endocrinologic aspects of cervical cancer.

In search of etiologic relevancy of the steroid deviations in urine, we compared the reproductive activities of patients having cervical cancer with those of normal controls. A premenopausal patient experienced the birth of her first child significantly earlier than did the urban control of corresponding age, despite their similarity of age at menarche. However, when the same patient was compared with the premenopausal rural control, this differences was not found. The same parameter also failed to differentiate a postmenopausal patient from the corresponding control of urban origin. The premenopausal patient who was indistinguishable from the rural control by age at first delivery was distinguished from the rural control by reduced excretions of adrenal steroids including 11-deoxy-17 ketosteroids and some corticosteroid metabolites. The spectrum and the degree of deviation of urinary steroids for a patient coincided with the spectrum and the degree of age dependency of the urinary steroids for a normal woman during and after adolescence. These findings indicated that the inability of the reproductive parameter to discriminate a patient was associated with growth retardation in a population, and a maturation deficiency of the adrenal gland (arrest of adrenarche) was implicated in the genesis of cervical cancer.

Crystallization of arginine-, formylmethionine-tyrosine-, and glycine-transfer RNAs from Escherichia coli.

Crystals as large as 0.5 X 0.3 X 0.2 mm of purified arginine-transfer RNA from Escherichia coli have been prepared by a vapour diffusion method. X-ray diffraction photographs showed that the crystals gave reflections up to 3.7 A spacing. They have a trigonal space group P31 2 1 (OR P32 2 1) and cell-dimensions a=97.2, b=97.2, c=94.8A. Crystals of a mercury derivative of this transfer RNA have also been obtained, and an X-ray diffusion photography of one of them is presented. Formylmethionine-transfer RNA from E. coli was crystallized in various forms, and the appearance of the polymorphs was found to depend upon the amount of spermine in the solution from which the crystallization took place. Crystals of tyrosine-transfer RNA and glycine-transfer RNA have also been obtained.

A study of endotoxemia in ulcerative colitis and Crohn's disease. I. Clinical study.

Endotoxin (lipopolysaccharide, LPS) and LPS antibody in the blood were studied in 61 cases of ulcerative colitis (U.C.) by radioimmunoassay. Lysozyme (LZM) concentration was also studied by the turbidimetric method. As a result, it was found that the blood LPS value as well as serum LZM concentration reflects the clinical observations. The case of endotoxemia in the active phase group showed a positive correlation between the LPS value and LZM concentration. LPS antibody which could not be detected in many cases of the active phase, had a high titer in cases of remission with a long history of the disease. These results would suggest that in U.C. with damaged intestinal mucosal barrier, LPS originating from intestinal flora enters into the blood and aggravates the disease and further that this invading LPS releases LZM into the blood. The same studies were performed on 7 cases of Crohn's disease and the same result was obtained.

A study of endotoxemia in ulcerative colitis and Crohn's disease. II. Experimental study.

The intestinal mucosal barrier of rabbits was damaged by carrageenan-induced ulceration of the colon, superior mesenteric artery occlusion (SMAO) and hemorrhagic shock and the values of Endotoxin (lipopolysaccharide, LPS) were determined by radioimmunoassay and the concentration of lysozyme (LZM) by the turbidimetric method. As a result, endotoxemia was observed in 13 out of 15 carrageenan rabbits, and in all of the SMAO and hemorrhagic shocked rabbits. Serum LZM concentration rose with time in all cases. As to the correlation of LPS and LZM, they changed almost in parallel in carrageenan rabbits, SMAO and hemorrhagic shock. LPS value and LZM concentration in blood were also determined in LPS injected rabbits. It was confirmed that injected LPS increased the LZM concentration of blood. On the basis of these results, it can be concluded that destruction of intestinal mucosal barrier permits an invasion of LPS into blood and then releases LZM into the blood stream.
