Uptake of amino acids and glucose by microfilariae of Dirofilaria immitis in vitro.

The uptake of amino acids and glucose by microfilariae of Dirofilaria immitis maintained for 8 days in vitro was studied. The quantity of glutamic acid, histidine, lysine, alanine, tryptophan, and cystine in the medium did not change. Isoleucine, serine, valine, phenylalanine, tyrosine, methionine, and cystathionine decreased slightly; both asparatic acid and leucine decreased considerably. Glutamine, threonine, and proline decreased markedly, while glycine increased to some extent. Ornithine increased and arginine decreased; however, their changes were smaller than the changes of respective amino acids in the control medium free of microfilariae. Glucose decreased markedly.

Density gradient centrifugation of cells separated from multicellular tumor spheroids.

Cells from a murine fibrosarcome (FSa) have been grown in vitro as multicell tumor spheroids (MTS). The growth rate of these MTS was determined. Following selected periods of growth, MTS were made into a single cell suspension and separated on linear density gradients of Renografin. While only 1 population of cells were separated from small spheriods (400 mum diameter), at least 3 subpopulations of tumor cells were separated and isolated from large spheroids (800 mum in diameter).

A histochemical study of the innervation of the cerebral blood vessels in the domestic fowl.

Adrenergic and cholinergic nerves innervating the cerebral arteries of the domestic fowl were examined by specific histochemical techniques. The adrenergic nerve plexuses of the cerebral carotid system are markedly denser than those of other vertebrates observed by similar techniques. They form longitudinally elongated meshworks of fine fibres in the vascular wall of the arterial branches. Those innervating the vertebro-basilar system are less dense and more elongated, and, as the size of the artery diminishes, the fibres of the plexus become coarser. In the small pial and parenchymal arteries they are reduced to a few fibres running parallel to, or spiralling around the vascular axis. The cholinergic nerve plexuses are not as dense as the adrenergic system. The acetylcholinesterase activity is very weak, except in the plexuses innervating the cerebral carotid artery and proximal portion of the anterior and posterior rami. In the vertebro-basilar system, a few thick nerve bundles run alongside the blood vessels of the vertebral and basilar arteries. Cholinergic nerves enter the cranial cavity along the internal carotid, the vertebral and possibly the cerebroethmoidal arteries. Intracerebral capillaries and some arterioles are not innervated with cholinergic and adrenergic fibres of peripheral origin, but with ones arising from parenchymal nerve cells.

Trialwise tracking method for measuring drug-affected sensory threshold changes in animals.

Rats and rhesus monkeys were trained under a multiple schedule, the components of which were random ratio schedules for food presentation and for shock presentation. The discriminative stimulus for the shock presentation component was a pure tone for the rats and a light for the rhesus monkeys. In the test session under the extinction condition for the shock presentation component, the intensity of the discriminative stimulus was successively either decreased by fixed units when the conditioned suppression was observed or increased when the conditioned suppression was not observed. The levels finally oscillated within a narrow range around the threshold. The auditory thresholds of rats were increased by intramuscular administration of quinidine at 20 mg/kg and also by repeated intramuscular administration of kanamycin at 250 and 500 mg/kg/day. In rhesus monkeys, visual thresholds were raised by application of pilocarpine at 0.02-0.16 mg/kg to the eyes and also by subcutaneous administration of LSD-25 at 4-8 micrograms/kg in one monkey and at 20-30 micrograms/kg in another. The method used for tracking the animals' sensory thresholds was sensitive enough to test the selective effect of the drugs and was also a relatively easy way to obtain a stable behavioral baseline for experimental purposes.

Juvenile type of distal and segmental muscular atrophy of upper extremities.

Seventy-one cases of distal and segmental muscular atrophy of the upper extremities with juvenile onset were studied. The clinical features consisted of: juvenile onset, male preponderance, unique distribution of the muscular atrophy in the hand and forearm, tendon reflexes hypoactive in most cases but hyperactive in some, no definite sensory disturbances, no involvement of the cranial nerves, and autonomic nerve disorders in the affected region. There was rapid progression during the 2 to 3 years after onset with a slowly progressive course thereafter. There were no abnormal laboratory findings except for electrophysiological and morphological findings of the affected muscles. The site of lesion was surmised to be from the C5 to T1 spinal segments with intramedullary involvement. The cause is unknown.

Funiculosin, a new antibiotic. II. Structure elucidation and antifungal activity.

Structure elucidation and some biological properties of an antiviral and antifungal antibiotic, funiculosin, are summarized. Funiculosin is a derivative of N-methyl-4-hydroxy-3, 5-disubstituted-2-pyridone containing a novel substituent, cyclopentanetetrol moiety, that is reported here for the first time in a natural product. The antibiotic protects guinea pigs efficiently against experimental trichophytosis. Toxicity of funiculosin is unique and highly selective for animal species.

[A case of cerebral vascular disease associated with prosopagnosia and minor hemisphere syndrome (author's transl)].

A case of "apractognosia", a syndrome of minor hemisphere, and prosopagnosia was described. A 68 years old, right handed, hypertensive man who had experienced an episode of left hemiparesis of several weeks' duration developed a syndrome of minor hemisphere, metamorphopsia, visuoconstructive disability, spatial dysgraphia, topographical and geographical disorientation and dressing apraxia, with associated left homonymous hemianopsia. A right occipital lesion was disclosed by an isotopic brain scan and a CT scan. The CT scan also disclosed a generalized cerebral atrophy. Memory disturbance, both verbal and visual, was observed. Verbal memory was examined by coupled words test, digit span and 10-words learning test, and visual memory by spatial pointing span and memorizing unfamiliar faces. The patient was able to differentiate unfamiliar faces, but was not able to memorize them correctly. Although the patient has complained of a disturbance of recognizing familiar faces (prosopagnosia), but actually, sometimes he was able to recognize the members of his family by the faces and sometimes he was not. He has stated that he could not recall familiar faces. The authors suggested prosopagnosia is, as Benton (1975) has supposed, considered to be the limited disorders of memory and association.

Cytocidal and cytostatic ability of Corynebacterium liquefaciens in mouse squamous cell carcinoma in vivo.

The cytostatic and cytocidal action of Corynebacterium liquefaciens was studied in a mouse squamous cell carcinoma in vivo. Kinetic analysis of tumor cells 4 and 8 days after a single i.p. dose of 2.0 mg C. liquefaciens showed a marked prolongation of the mean cell generation time (TG). This prolongation affected most the G1 and, to a lesser extent, the S phases of the cell cycle. The tumor growth factor was decreased, and the mean value of the cell loss factor was increased. Assays to determine the number of tumor cells needed to produce the tumor in one-half of the transplant recipients showed that peritoneal exudate cells collected from mice given injections of C. liquefaciens exerted tumor cell killing that depended on the peritoneal exudate cell tumor:cell ratio. This cell killing was not found with peritoneal exudate cells from normal or proteose peptone-treated mice.
