[Willingness to pay for ecosystem services of urban river in Shanghai].

The contingent valuation method (CVM) is one of the most dominating and standard techniques for eliciting the willingness to pay (WTP) for the ecosystem services provided by environment and resources. Taking Zhangjiabang Creek, a model project in the comprehensive management of urban river in China, as a case study, 800 payment card CVM questionnaires were surveyed and 646 as useful questionnaires were feed back in August 2003, then, the mean WTP, 195.07 - 253.04 Yen per person every year in the future 3 years, to the ecosystem services of the urban river was got in the payment card CVM study, and the figure was compared to some other river CVM studies. Distribution shape and range of WTP were probed, and the regression relationship between respondents WTP and their socioeconomic information such as income, education level, donation experience, and environmental attitude was discussed, what's important, an environmental economics explanation was given to the regression results. Some biases which influence the mean WTP of payment card CVM are inspected and corresponding countermeasures were seriously noticed and implemented in the survey. Finally the critiques and controversies that CVM is faced with were advanced, but the reliability of CVM is also given based upon environmental decision-making theory.

Expression of inducible nitric oxide synthase in transplanted rat liver and its inhibitory effect.

BACKGROUND: It is reported that inducible nitric oxide synthase (iNOS) plays an important role in rejection. This study was designed to observe the expression of iNOS in the transplanted liver tissues and the effect of aminoguanidine(an inhibitor of iNOS) and FK506(immunosuppression) on acute rejection after liver transplantation. METHODS: All rats were divided into four groups: homogeneous (LEW-LEW), acute rejection (BN-LEW), aminoguanidine (BN-LEW), and FK506 (BN-LEW). In the last two groups the drugs were used in heterogeneous rats after liver transplantation. The expression of iNOS in liver tissues of all groups was investigated immunohistochemically. RESULTS: In the acute rejection group, the expression of iNOS was extremely positive compared to the other three groups. The difference was statistically significant. CONCLUSIONS: During the process of acute rejection in rat orthotopic liver transplantation, the enhanced expression of iNOS correlates to the severity of rejection. Aminoguanidine and FK506 can inhibit the expression of iNOS and palliate acute rejection.

[Simultaneous measurement of muscle energy metabolism by MRS and frequency-domain NIR spectroscopy].

The frequency-domain near-infrared spectrometry (NIRS) is capable of measuring the absolute absorption and reduced scattering coefficient of tissue noninvasively. This allows the quantitation of tissue hemoglobin concentration which reflects the balance between oxygen delivery and oxygen utilization of the skeletal muscle. Phosphorus magnetic resonance spectroscopy (31P-MRS) has become a gold standard of noninvasive measurement of human skeletal muscle metabolism. The rate of phosphocreatine (PCr) resynthesis during recovery is an indicator of the rate of oxidative metabolism. The purpose of the present study was the effect of lower intracellular pH (pHi) on PCr and oxygenation recovery. The preliminary results of plantar flexion experiment on a healthy male subject show that both PCr resynthesis and reoxygenation were very much prolonged by severe acidosis (pHi = 6.42).

[3-D FEA of the stress state of abutment and its periodontal tissue with three different retention styles in the absence of mandibular anterior teeth].

PURPOSE: To study the stress of the abutment and its periodontal tissue with different designs of retainer (type of retainer, place)in the same situation of dentural defect. METHODS: Based on the 3-D FEA database of RPD, the stress state of the abutment and its periodontal tissue was analyzed to choose the optimal retainer with the same situation of dentural defect. RESULTS: With three different retainer designs, the retainer on the second premolar and the first molar exerted the smallest stress on the abutment and its periodontal tissue, while the retainer on the first premolar and the first molar had the largest stress. CONCLUSION: Analysis for the health condition of abutment and its periodontal tissue is imperative for the selection of different designs of retainers.

[Comparison of clinical and surgico-pathological TNM stage of 2007 lung cancer patients].

OBJECTIVE: An accurate clinical TNM staging of lung cancer is essential for the precise determination of the extent of the disease in order that an optimal therapeutic strategy can be planned. This is especially true in patients with marginally resectable tumors. Clinical over-staging of the disease may deny a patient the benefit of surgery, whereas under-staging may oblige a patient to accept a fruitless or even harmful surgery. We aimed to analyze preoperative clinical (c-TNM) and postoperative surgico-pathologic staging (p-TNM) of lung cancer patients in order to evaluate the accuracy of our clinical staging and its implications on the surgical strategy for lung cancer. METHODS: We did a retrospective comparison of c-TNM and p-TNM staging of 2007 patients with lung cancer surgically treated from January 1999 to May 2003. Preoperative evaluation and c-TNM staging of all patients were based on physical examination, laboratory studies, routine chest X-ray and CT scan of the chest and upper abdomen. Other examinations included sputum cytology, bronchoscopy, abdominal ultrasonography, bone scintiscan, brain CT/MRI, and mediastinoscopy whenever indicated. RESULTS: In the present study the comparison of c-TNM and p-TNM staging of 2007 patients with lung cancer revealed an overall concurrence rate of only 39.0%. In the entire series the extent of disease was clinically underestimated in 45.2% and overestimated in 15.8% of the patients. Among all c-TNM stages the c-IA/B stage of 1105 patients gave the highest rate (55.2%) of underestimating the extent of disease. Clinical staging of T subsets was relatively easy with an overall accuracy rate of 72.9%, while that of N subsets was relatively more difficult with an overall accuracy rate of 53.5%. Analysis also showed that c-IV stage may not be an absolute contraindication to surgery, because in half of the patients, c-M1 turned out to be p-M0, providing the possibility of resectional surgery depending on the status of T and N. CONCLUSION: For reasons to be further determined, the present preoperative clinical TNM staging of lung cancer remains a crude evaluation. Further efforts to improve its accuracy are needed.

[Plasma level and prognostic significance of VEGF, bFGF and MMP-9 in patients with advanced non-small-cell lung cancer].

OBJECTIVE: To measure plasma levels of VEGF, bFGF and MMP-9 in advanced non-small-cell lung cancer (NSCLC) patients and evaluate their prognostic value. METHODS: The plasma levels of VEGF, bFGF and MMP-9 in 46 cases with advanced NSCLC were measured by ELISA before chemotherapy and in 30 cases after chemotherapy. RESULTS: The plasma levels of VEGF, bFGF and MMP-9 in patients before chemotherapy were significantly higher than those in healthy control persons (P < 0.05). The correlation between plasma levels of VEGF and bFGF was significant, Spearman's r = 0.329 (P = 0.027). No correlation was found among the levels of angiogenic factors studied above and the following clinical parameters such as age, sex, histological subtype, differentiation of tumor cells, TNM-stage and also blood leukocyte, hemoglobin and platelet counts. The plasma level of MMP-9 in patients with extensive metastasis (including bone metastasis) was significantly higher than that in patients with bone metastasis only (P = 0.013). Reduction of plasma bFGF level after chemotherapy was a unique independent good prognostic factor (RR = 11.737, P = 0.02). CONCLUSION: The measurement of plasma levels of such angiogenic factors as VEGF, bFGF and MMP-9 in advanced NSCLC is helpful for prediction of metastasis tendency and evaluation of prognosis.

Development of ion drift-chemical ionization mass spectrometry.

An ion drift-chemical ionization mass spectrometry (ID-CIMS) technique has been developed to detect and quantify trace gases, including volatile organic compounds and inorganic species. The trace species are chemically ionized into positive or negative product ions with a well-controlled ion-molecule reaction time. The ID-CIMS method allows for quantification of the trace gases without the necessity of performing calibrations with authentic standards for the trace gases. Demonstrations of the ability of ID-CIMS to accurately quantify isoprene and HNO3 in a laboratory setting are presented. The results illustrate that the ID-CIMS technique facilitates detection and quantification of organic and inorganic species in laboratory kinetic investigations and field measurements.

Atmospheric new particle formation enhanced by organic acids.

Atmospheric aerosols often contain a substantial fraction of organic matter, but the role of organic compounds in new nanometer-sized particle formation is highly uncertain. Laboratory experiments show that nucleation of sulfuric acid is considerably enhanced in the presence of aromatic acids. Theoretical calculations identify the formation of an unusually stable aromatic acid-sulfuric acid complex, which likely leads to a reduced nucleation barrier. The results imply that the interaction between organic and sulfuric acids promotes efficient formation of organic and sulfate aerosols in the polluted atmosphere because of emissions from burning of fossil fuels, which strongly affect human health and global climate.

Rap1 regulates E-cadherin-mediated cell-cell adhesion.

The small GTPase Rap is best characterized as a critical regulator of integrin-mediated cell adhesion, although its mechanism of action is not understood. Rap also influences the properties of other cell-surface receptors and biological processes, although whether these are a consequence of effects on integrins is not clear. We show here that Rap also plays an important role in the regulation of cadherin-mediated cell-cell adhesion in epithelial cells. Expression of constitutively active Rap1A restored cadherin-mediated cell-cell contacts in mesenchymal Ras-transformed Madin-Darby canine kidney cells, resulting in reversion to an epithelial phenotype. Activation of endogenous Rap via the Rap exchange factor Epac1 also antagonized hepatocyte growth factor-induced disruption of adherens junctions. Inhibition of Rap signaling resulted in disruption of epithelial cell-cell contacts. Rap activity was required for adhesion of cells to recombinant E-cadherin extracellular domains, i.e. in the absence of integrin-mediated adhesion. These findings suggest that Rap signaling positively contributes to cadherin-mediated adhesion and that this occurs independently of effects on integrin-mediated adhesion. Our results imply that Rap may function in a broader manner to regulate the function of cell-surface adhesion receptors.

Quantification of hydroxycarbonyls from OH-isoprene reactions.

Hydroxycarbonyls arising from OH-initiated reactions of isoprene have been quantified by the technique of a flow reactor coupled to proton-transfer reaction mass spectrometry (PTR-MS) detection. The yields of C5- and C4-hydroxycarbonyls are (19.3 +/- 6.1)% and (3.3 +/- 1.6)%, respectively, measured at a flow tube pressure of about 100 Torr and at a temperature of 298 +/- 2 K. A yield of (8.4 +/- 2.4)% is obtained for the unsaturated carbonyl C5H8O, confirming that internal OH addition represents the minor channel in the initial OH-isoprene reaction. The results show that those carbonyl compounds account for the most previously unquantified carbon, enabling the isoprene carbon closure. The study also reveals novel aspects of the delta-hydroxyalkoxy radical degradation mechanism, which is essential for modeling tropospheric O3 formation. In addition, this work demonstrates the application of PTR-MS for quantification of products of hydrocarbon reactions, which should have profound impacts on elucidation of the chemistry of atmospheric anthropogenic and biogenic hydrocarbons.

Cyclopiazonic acid decreases spontaneous transient depolarizations in guinea pig mesenteric lymphatic vessels in endothelium-dependent and -independent manners.

Guinea pig mesenteric lymphatic vessels exhibit vasomotion through a pacemaker mechanism that involves intracellular Ca(2+) release and resultant spontaneous transient depolarizations (STDs) of the smooth muscle membrane potential. This study presents a detailed characterization of the effects of cyclopiazonic acid (CPA) on this pacemaker activity. Microelectrode recordings from smooth muscle in vessel segments revealed that application of CPA (1-10 microM) caused a hyperpolarization accompanied by a decrease in the frequency and amplitude of STDs. The CPA-induced hyperpolarization was abolished after destruction of the endothelium and in the presence of N(G)-nitro-L-arginine (100 microM) or 1H-[1,2,4]oxadiazolol-[4,3-a]quinoxaline-1-one (10 microM), which suggests a contribution of endothelium-derived nitric oxide (EDNO) in this response. In the absence of EDNO-induced effects, CPA decreased the frequency and amplitude of STDs recorded before and in the presence of the thromboxane A(2) mimetic U-46619, norepinephrine, or thimerosal. CPA abolished U-46619-induced vasomotion as determined by measurement of constriction-associated intracellular Ca2+ concentration using the ratiometric Ca2+ indicator fura-2. The endothelial actions of CPA were compared with those of ACh, which is known to cause EDNO release in this preparation. Although CPA and ACh both increased endothelial intracellular Ca2+ concentration and depolarized the membrane potential, the kinetics of action for both parameters were markedly slower for CPA than ACh. These results suggest that CPA first hyperpolarizes the lymphatic smooth muscle and decreases STD frequency and amplitude through endothelial release of EDNO, and second, consistent with the action of CPA to inhibit sarcoplasmic reticulum Ca2+-ATPase and deplete Ca2+ stores, it further reduces STD activity. Inhibition of the lymphatic smooth muscle pacemaker mechanism is thought to abolish agonist-induced vasomotion.

In vivo evaluation of [11C]N-(2-chloro-5-thiomethylphenyl)-N'-(3-methoxy-phenyl)-N'-methylguanidine ([11C]GMOM) as a potential PET radiotracer for the PCP/NMDA receptor.

The development of imaging methods to measure changes in NMDA ion channel activation would provide a powerful means to probe the mechanisms of drugs and device based treatments (e.g., ECT) thought to alter glutamate neurotransmission. To provide a potential NMDA/PCP receptor PET tracer, we synthesized the radioligand [11C]GMOM (ki = 5.2 +/-0.3 nM; log P = 2.34) and evaluated this ligand in vivo in awake male rats and isoflurane anesthetized baboons. In rats, the regional brain uptake of [11C]GMOM ranged from 0.75+/-0.13% ID/g in the medulla and pons to 1.15+/-0.17% ID/g in the occipital cortex. MK801 (1 mg/kg i.v.) significantly reduced (24-28%) [11C]GMOM uptake in all regions. D-serine (10 mg/kg i.v.) increased [11C]GMOM %ID/g values in all regions (10-24%) reaching significance in the frontal cortex and cerebellum only. The NR2B ligand RO 25-6981 (10 mg/kg i.v.) reduced [11C]GMOM uptake significantly (24-38%) in all regions except for the cerebellum and striatum. Blood activity was 0.11+/-0.03 %ID/g in the controls group and did not vary significantly across groups. PET imaging in isoflurane-anesthetized baboons with high specific activity [11C]GMOM provided fairly uniform regional brain distribution volume (VT) values (12.8-17.1 ml g(-1)). MK801 (0.5 mg/kg, i.v., n = 1, and 1.0 mg/kg, i.v., n = 1) did not significantly alter regional VT values, indicating a lack of saturable binding. However, the potential confounding effects associated with ketamine induction of anesthesia along with isoflurane maintenance must be considered because both agents are known to reduce NMDA ion channel activation. Future and carefully designed studies, presumably utilizing an optimized NMDA/PCP site tracer, will be carried out to further explore these hypotheses. We conclude that, even though [11C]GMOM is not an optimized PCP site radiotracer, its binding is altered in vivo in awake rats as expected by modulation of NMDA ion channel activity by MK801, D-serine or RO 25-6981. The development of higher affinity NMDA/PCP site radioligands is in progress.

Expression of MRP14 gene is frequently down-regulated in Chinese human esophageal cancer.

Migration inhibitory factor-related protein 14 (MRP14) is one of calcium-binding proteins, referred as S100A9. The heterodimeric molecule formed by MRP14 with its partner MRP8 (S100A8) is the major fatty acid carrier in neutrophils. The MRP8/14 complex has been also implicated in the intracellular transport of arachidonic acid and its precursors in keratinocytes. We show here the involvement of MRP14 in human esophageal cancer. In an initial study, mRNA differential display-reverse transcription polymerase chain reaction (DD-PCR) was performed with two esophageal carcinomas, one esophageal adenocarcinoma and matched normal adjacent mucosa. DD-PCR with the arbitrary primer OPA3 showed that one cDNA band was highly expressed in normal tissues, but disappeared or substantially decreased in tumor counterparts. It was later identified to be the 3'-end of migration inhibitory factor-related protein 14 (MRP14). Northern blotting, RT-PCR and Western blotting corroborated the down-regulation of MRP14 in 58/64 squamous cell carcinomas and 2/2 adenocarcinomas as compared with adjacent normal epithelia of the esophagus. MRP14 was undetectable in 3/3 esophageal-carcinoma cell lines. Immunochemistry demonstrated that expression of MRP14 was restricted to normal esophageal epithelia. No mutation was found in the genomic DNA of the MRP14 gene by PCR and directed DNA sequencing. Our finding suggested that the reduction of MRP14 expression is a frequent event in Chinese human esophageal cancer.

Detecting cAMP-induced Epac activation by fluorescence resonance energy transfer: Epac as a novel cAMP indicator.

Epac1 is a guanine nucleotide exchange factor for Rap1 that is activated by direct binding of cAMP. In vitro studies suggest that cAMP relieves the interaction between the regulatory and catalytic domains of Epac. Here, we monitor Epac1 activation in vivo by using a CFP-Epac-YFP fusion construct. When expressed in mammalian cells, CFP-Epac-YFP shows significant fluorescence resonance energy transfer (FRET). FRET rapidly decreases in response to the cAMP-raising agents, whereas it fully recovers after addition of cAMP-lowering agonists. Thus, by undergoing a cAMP-induced conformational change, CFP-Epac-YFP serves as a highly sensitive cAMP indicator in vivo. When compared with a protein kinase A (PKA)-based sensor, Epac-based cAMP probes show an extended dynamic range and a better signal-to-noise ratio; furthermore, as a single polypeptide, CFP-Epac-YFP does not suffer from the technical problems encountered with multisubunit PKA-based sensors. These properties make Epac-based FRET probes the preferred indicators for monitoring cAMP levels in vivo.

[Study of the causes of fetal growth restriction with unclear etiologies].

OBJECTIVE: To investigate different factors related to fetal growth restriction (FGR) and to find out the possible causes of FGR with unclear etiologies. METHODS: Sixty-three women who were suspected of FGR during pregnancy between March 2002 and March 2003 were included in this study. Their age, body mass index (BMI) before pregnancy, and gestational weeks were recorded at the time when they were first diagnosed. Haemoglobin levels, haematocrit (HCT), TORCH, anticardiolipin antibody (ACA), 50 gram glucose challenge test (50g GCT), 75 gram oral glucose tolerance test (75g OGTT), leptin levels, systolic/diastolic (S/D) ratio by color doppler monitor and chlamydia trachomatis (CT) were detected and urine culture was done in these groups during the same period. The gestational week, birth weight, body length and the gender were recorded at the delivery period. The FGR group was then divided into two subgroups according to the birth weights: study A group whose birth weights were lower than 10th% of the birth weights at the given gestational weeks (29 cases) and study B group whose birth weights were beyond 10th% (34 cases). The chromosome, leptin, C-peptide, insulin and TORCH of umbilical blood were measured at delivery. The other 25 normal pregnant women were included as control and the same tests were performed accordingly. RESULTS: The fasting glucose and the third hour's glucose of 75 gram oral glucose tolerance test of study A were (3.8 +/- 0.6) mmol/L and (4.5 +/- 1.1) mmol/L. The fetal leptin, C-peptide, and insulin were (7.3 +/- 5.2) ng/ml, (0.5 +/- 0.3) nmol/L and (2.3 +/- 1.3) mU/L. The S/D ratio of umbilical artery, maternal and fetal infection rate of CMV, positive rate of ACA-IgM and the rate of asymptomatic bacteriuria were 3.06, 20.7%, 24.1%, 44.8% and 62.1% respectively. The fasting glucose and the third hour's glucose of 75 gram oral glucose tolerance test of study B were (4.4 +/- 0.7) mmol/L and (4.6 +/- 1.1) mmol/L. The fetal leptin, C-peptide, and insulin were (13.2 +/- 11.3) ng/ml, (0.7 +/- 0.4) nmol/L and (4.3 +/- 3.3) mU/L. The S/D ratio of umbilical artery, maternal and fetal infection rate of CMV, positive rate of ACA-IgM and the rate of asymptomatic bacteriuria were 2.63, 2.9%, 0%, 5.9% and 44.1% respectively. The fasting glucose and the third hour's glucose of 75 gram oral glucose tolerance test in control were (4.3 +/- 0.7) mmol/L and (5.3 +/- 1.2) mmol/L. The fetal leptin, C-peptide, and insulin were (20.5 +/- 12.0) ng/ml, (1.0 +/- 0.4) nmol/L and (6.3 +/- 4.0) mU/L. The S/D ratio of umbilical artery, maternal and fetal infection rate of CMV, positive rate of ACA-IgM and the rate of the asymptomatic bacteriuria were 2.80, 0, 0, 0 and 24.0% respectively. All these items were significantly higher in study A than those in the control (P < 0.05). There was no significant difference between the study B and the control in all the items. CONCLUSIONS: Many factors may play a role in the pathogenesis of FGR, including the maternal blood glucose level, the ability for fetus to use the glucose, the infection of some microorganisms, insufficiency of the blood supply and the autoimmunity of the mother. Finding out the possible causes of FGR and managing them accordingly may improve the outcomes of the fetus.

[Expression of hepatocyte growth factor and its receptor c-met gene in the endometrium of women with endometriosis].

OBJECTIVE: To study the role of hepatocyte growth factor (HGF) and its receptor (c-Met) in the pathogenesis of endometriosis. METHODS: In situ hybridization and immunohistochemistry method was used to examine HGF mRNA and protein expression in eutopic and ectopic endometrium obtained from 54 women in reproductive age with endometriosis confirmed by laparoscopy and histological examination (including 28 cases of stages I/II and 26 of stages III/IV) and from the endometrium of 24 healthy fertile women. RESULTS: In patients with endometriosis, the positivity rate of HGF/c-Met expression was similar in the eutopic and ectopic endometria, but the number of positive cells and expression intensity were much higher in ectopic endometrium. The positive expression rate varied little between the proliferative and secretory stages of the endometrium. Compare with the healthy controls, patients with endometriosis of stages I/II and III/IV had significantly elevated positivity rate of HGF and c-Met expressions in either eutopic and ectopic endometrium, but without significant difference between the two patient groups (P<0.01). The rate of strong c-Met protein expression was significantly higher in III/IV endometriosis than in the controls, without statistical difference between other groups. CONCLUSION: The expressions of HGF and c-met gene are correlated with the pathogenesis of endometriosis, which is preceded by eventful changes in the biological behavior of the eutopic endometrium.

Modified Sugiura procedure for the management of 160 cirrhotic patients with portal hypertension.

BACKGROUND: Portal hypertension is a common disease with a high mortality and serious effect on the life quality of patients. Presently, shunt and disconnection are commonly used for surgical treatment of portal hypertension. The aim of this study was conducted to analyze the results of a modified Sugiura procedure for the management of 160 cirrhotic patients with portal hypertension. METHODS: The results of a modified Sugiura procedure for the treatment of 160 cirrhotic patients with portal hypertension from January 1991 to July 2002 were retrospectively analyzed. RESULTS: The operative mortality for the procedure was zero. Postoperative intra-abdominal bleeding was noted in 2 patients, drowned lung in 1, pneumonia in 1, and splenic venous thrombosis in 4. Of the 160 patients, 157 (98%) were followed up from 6 months to 11.5 years. Of the 157 patients, only one died of hepatic coma 6 years after operation, and 3 of rebleeding. The absolute and relative survival rates were 97.5%(156/160) and 99%(159/160), respectively. The absolute and relative occurrence rates of hepatic coma were 2.5%(4/160) and 0.6%(1/157), respectively. The absolute and relative occurrence rates of rebleeding were 3.8%(6/160) and 1.9%(3/157), respectively. In 96 of 116 Child B patients (82.8%), liver function improved from preoperative class B to A 3 months after operation. Sixty-five patients were subjected to gastroscopy and 22 patients, esophageal barium photography 6 months after operation. Gastro-esophageal varices disappeared in 56 patients (64.4%, 56/87), obviously improved in 30 (34.5%, 30/87), and unchanged in 1 (1.2%, 1/87). The occurrence rate of portal hypertensive gastropathy (PHG) was 13.9%(9/65). CONCLUSION: Our results showed that the modified Sugiura procedure is effective in the treatment of portal hypertension, with a low rate of operative complication, bleeding recurrence, and hepatic coma.
