Sequence Analysis of SOD Gene of Bombyx mori Nuclear Polyhedrosis Virus and Its Expression in E.coli.

Superoxide dismutases scavenge superoxide radicals and protect cells from oxidative stress. SOD gene of Bombyx mori nuclear polyhedrosis virus has been cloned by PCR, and is expressed in E.coli with the activity of SOD being 576.13 u/mL. DNA sequence analysis shows that SOD gene of BmNPV encodes 151 amino acids. The homology between BmNPV and AcNPV of the nucleotide sequences of SOD gene is 97.2%, and 56% between BmNPV and human SOD1.

CYP6B2 Expression in Insecticide-Resistant and -Suspcitible Strains of Helicoverpa armigera.

A fragment of CYP6B2 gene was obtained from the midgut cDNA library of the fifth instar larvae of the ballworm (Helicoverpa armigera) with RT-PCR. The product of 627 bp was cloned and sequenced. The results of dot blotting analysis of mRNA proved that CYP6B2 is expressed at higher level (mRNA) in an insecticide-resistant strain than in a susceptible strain, showing that the regulation was at transcription level. The mRNA is induced by pyrethroid in susceptible, but not in the insecticide-resistant strains. We guess that mutation in the 5' upstream regulator of the insecticide-resistant strain may lead to the difference in the expression level between resistant and susceptible strains.

Two new bioactive triterpene glycosides from the sea cucumber Pseudocolochirus violaceus.

By activity-guided fractionation, two new triterpene glycosides, violaceusides A (1) and B (2), were isolated from the sea cucumber Pseudocolochirus violaceus as active compounds causing morphological abnormality of Pyricularia oryzae mycelia. By extensive 2D NMR techniques and chemical evidence, the structures of the two new glycosides were established as 16beta-acetoxy-3-O-[3-O-methyl-beta-D-glucopyranosyl-(1 --> 3)-beta-D-xylopyranosyl-(1 --> 4)-beta-D-quinovopyranosyl-(1 --> 2)-4-O-sodiumsulphate-beta-D-xylopyranosyl]-holosta-7,24-diene-3beta-ol (1) and 16beta-acetoxy-3-O-[3-O-methyl-beta-D-glucopyranosyl-(1 --> 3)-beta-D-xylopyranosyl-(1 --> 4)-beta-D-glucopyranosyl-(1 --> 2)-4-O-sodiumsulphate-beta-D-xylopyranosyl]-holosta-7,24-diene-3beta-ol (2), respectively. The two glycosides also exhibited significant cytotoxicity against HL-60 and BEL-7402 cancer cell lines.

A new acylated flavonol glycoside from Derris triofoliata.

A new acylated flavonol glycoside, kaempferol 3-O-[(6''''-feruloyl)-beta-D-glucopyranosyl-(1 --> 3)]-[alpha-L-rhamnopyranosyl-(1 --> 6)]-beta-D-glucopyranoside and two known cyclolignan glycosides, (+)-lyoniresinol-3alpha-O-beta-D-glucopyranoside and ( - )-lyoniresinol-3alpha-O-beta-D-glucopyranoside were isolated from n-BuOH extracts of the aerial parts of Derris triofoliata, their structures were determined from spectroscopic and chemical evidences.

Development and validation of a multiwavelength spatial domain near-infrared oximeter to detect cerebral hypoxia-ischemia.

Detection of cerebral hypoxia-ischemia in infants remains problematic, as current monitors in clinical practice are impractical, insensitive, or nonspecific. Our study develops a multiwavelength spatial domain construct for near-infrared spectroscopy (NIRS) to detect cerebral hypoxia-ischemia and evaluates the construct in several models. The NIRS probe contains photodiode detectors 2, 3, and 4 cm from a three-wavelength, light-emitting diode. A construct determines cerebral O(2) saturation based on spatial domain principles. Device performance and construct validity are examined in in-vitro models simulating the brain, and in piglets subjected to hypoxia, hypoxia-ischemia, and hyperoxic conditions using a weighted average of arterial and cerebral venous O(2) saturation measured by CO-oximetry. The results in the brain models verify key equations in the construct and demonstrate reliable performance of the device. In piglets, the device measures cerebral O(2) saturation with bias +/-4% and precision +/-8%. In conclusion, this NIRS device accurately detects cerebral hypoxia-ischemia and is of a design that is practical for clinical application.

Optimization of block-floating-point realizations for digital controllers with finite-word-length considerations.

The closed-loop stability issue of finite-precision realizations was investigated for digital controllers implemented in block-floating-point format. The controller coefficient perturbation was analyzed resulting from using finite word length (FWL) block-floating-point representation scheme. A block-floating-point FWL closed-loop stability measure was derived which considers both the dynamic range and precision. To facilitate the design of optimal finite-precision controller realizations, a computationally tractable block-floating-point FWL closed-loop stability measure was then introduced and the method of computing the value of this measure for a given controller realization was developed. The optimal controller realization is defined as the solution that maximizes the corresponding measure, and a numerical optimization approach was adopted to solve the resulting optimal realization problem. A numerical example was used to illustrate the design procedure and to compare the optimal controller realization with the initial realization.

Treatment of advanced gastric cancer by chemotherapy combined with autologous cytokine-induced killer cells.

The effects of autologous cytokine-induced killer (CIK) (CD3+CD56+) cells together with chemotherapy were investigated in patients who suffered from advanced gastric cancers (stage IV). Fifty-seven patients were divided into two groups: chemotherapy plus CIK biotherapy and chemotherapy alone. CIK cells were induced from autologous peripheral blood mononuclear cells in vitro and separated by flow cytometry and then transfused into the patients. The T-lymphocyte subgroups (CD3+, CD4+ or CD8+), CIK cells and NK cells (CD3-CD56+) were separated and determined by flow cytometry and the serum levels of MG7-Ag, CA72-4, CA19-9 and CEA were determined by ELISA or ECLIA. It was demonstrated that the cytotoxic activity of CIK cells reached a maximum between days 14 to 21 (68.7+/-10.9% and 65.3+/-10.4%, respectively). The amounts of CIK cells were gradually increased from day 0 to day 21 and slightly decreased in the further incubations. Thereafter, the CIK cells on days 14 to 21 (with the highest population of CIK cells) transfused back to the patients. The serum levels of the tumor markers were significantly decreased, the host immune function was increased and the short-term curative effect as well as the quality of life (QOL) were improved in the patients treated by chemotherapy plus CIK cells compared to the patients treated by chemotherapy alone. Moreover, the 2-year life-span was prolonged in the group treated by chemotherapy plus CIK cells compared to the group treated with chemotherapy alone. It is concluded that chemotherapy plus CIK cells has obvious benefits for patients who suffer from advanced gastric cancers.
