OBJECTIVE: To observe the effect of recombinant human erythropoietin (rHuEPO) on immune function of premature rats. METHODS: RHuEPO of 250 IU/(kg.t) or 500 IU/(kg.t) was administered to premature rats every other day for nineteen days. The control premature rats were received normal saline. The changes of hemoglobin (Hb), serum erythropoietin (EPO), red blood cell (RBC) immune function, T lymphocyte proliferative responsiveness, and production of tumor necrosis factor alpha (TNF-alpha) were observed. RESULTS: Premature rats showed lower levels on Hb, RBC immune function, T cell responsiveness and production of TNF-alpha compared with mature rats at birth. The postnatal declines of Hb and RBC immune function were lessened in the treated groups of premature rats, the higher dosage group of 500 IU/(kg.t) was more significant than the lower dosage group of 250 IU/(kg.t). When experiments were over, Hb of control premature rats was (7.72 +/- 0.89) g/dl, Hb of premature rats received 500 IU/(kg.t) was (10.08 +/- 0.90) g/dl (P < 0.01). C3b-R% of control premature rats was (11.00 +/- 0.95)%, C3b-R% of premature rats received 500 IU/(kg.t) was (17.75 +/- 1.04)% (P < 0.01). IC-R% in control premature rats was (12.83 +/- 1.33)%, IC-R% of premature rats received 500 IU/(kg.t) was (10.50 +/- 1.67)% (P < 0.01). The postnatal rise of T cell responsiveness and the production of TNF-alpha in premature rats increased in the treated groups, which was more significant in the higher dosage group of 500 IU/(kg.t) than in the lower dosage group of 250 IU/(kg.t). The OD index of control premature rats was 0.159 +/- 0.014, the OD index of premature rats received 500 IU/(kg.t) was 0.354 +/- 0.050 (P < 0.01). TNF-alpha in control premature rats was (0.270 +/- 0.014) ng/ml, TNF-alpha of premature rats received 500 IU/(kg.t) was (0.415 +/- 0.010) ng/ml (P < 0.01). CONCLUSIONS: (1) Premature rats had lower RBC immune function and T cell responsiveness and underproduction of TNF-alpha at birth. (2) Premature rats had an improvement with the RBC immune function after rHuEPO administration. (3) Premature rats had improvements with T cell responsiveness and TNF-alpha after rHuEPO administration, and there was a positive correction between the RBC immune function and T cell responsiveness with the production of TNF-alpha.

Fungi, which can grow on and degrade pendimethalin, were isolated from activated sludge and soil. The biodegradation characterization in vitro was studied. The results showed that pendimethalin could be degraded by fungi, 16 fungi were isolated by enrichment culture, among which, 10 fungi could degrade more than 60% pendimethalin in 5 days in culture fluid. According to the physiological resistance and the capacity to degrade pendimethalin, three fungi were considered as efficiently degrading strains identified as Aspergillus terreus, Monilochaetes and Aspergillus furnigatus. The efficiency of degradation was affected by culture conditions such as pH, temperature, and concentration of cane sugar and pendimethanlin. The results showed that when the additional carbon source was 0.5%-1.0%, pH value was 6-8, and cultivated temperature was 20-30 degrees C, the growth amount of fungi and the degradation rate were enhanced. The fungi grew quickly, and the biodegradation rate was high under pendimethalin concentration being 100 mg.L-1. The removed amount of pendimethalin increased with increasing pendimethalin concentration. The optimal conditions were proposed, which could provide theoretic basis for prevention and control of pesticides pollution.
OBJECTIVE: To investigate the possibility and efficacy of allograft transplantation in treating patient with huge tissue defect after radical giant malignant melanoma resection. METHODS: A male person received blood type matching was chosen as donor. Immediately after the donor's brain death, allograft was excised with the depth to the layer intervenient between periosteum and epicranial fascia in calvaria, the superficial layer of deep temporal fascia in both sides of temporal regions, close to zygomatic bones and mandibles including masseter and auricles upon in face, and cervical soft tissues including sternocleidomastoid muscles, cervical and external jugular vessels of both sides were excised simultaneously. After being perfused with 4 degrees C UW solution through both common carotid arteries, the homograft was sheared and radiated with X-ray before being preserved in UW solution for further use. During the operation, both sides of external auditory meatus were anastomosed with ears firstly, and vessels were anastomosed end-to-end sequently, at last, the border of skin flap was sutured intermittently. Combined use of MMF, FK506, Prednisone and Zenopax was performed as post-operation immunosuppressive treatment. Clinical observations were made on the signs and symptoms of graft survival or rejection as well as blood FK506 concentrations and immunological indexes were tested in laboratory. Biopsies of graft were also made at 1 h, 4 h, 8 h, 7 d, 14 d and 30 d after operation. RESULTS: The circulation of the graft was satisfactory, and the temperature and color of skin were normal. Primary healing of suture and hair growth about 0.8 cm in a month were observed. Skin Biopsies of every time had no found of hyperacute or acute rejection. The concentration of FK506 was maintained 20 mg/ml 1 month after the operation. CONCLUSION: Allograft transplantation with compound tissue of head skin flap and ears is a kind of effective and safe treatment in repairing huge tissue defect. Good tissue matching and combined use of currently available immunosuppressants can prevent hyperacute and acute rejection efficiently.
Four soils with different fertility were treated with different concentrations of atrazine. The results showed that the urease activity was stimulated in the soils treated with low concentration of atrazine at the first day, while inhibited after treated with high concentration of atrazine during the whole period of the experiment. There was a significant difference in urease activity in soils with different fertility after treated with high concentration atrazine. The urease activity in CK and in NPK was significantly inhibited, the inhibition rate reached 30.35% and 28.89%, respectively, but in NPK + S and NPK + M, the inhibition rate was only 21.35% and 16.86%, respectively. The maximum inhibition rate appeared later in soils with high than with low fertility, showing that soils with high fertility could endure the effect of atrazine.

OBJECTIVE: To study the production technology of extraction in combination hydrolysis in situ for isolating diosgenin from Dioscorea nipponica by orthogonal design. METHOD: The optimum production conditions were investigated with the recovery of diosgenin as an index by extraction in combination hydrolysis in situ, and were compared with the traditional method. RESULT: Extraction in combination hydrolysis in situ conducted in 1.5 mol x L(-1) sulfuric acid of water containing 75% isopropanol at 100 degrees C for 4.5 h could get higher recovery of diosgenin than traditional methods. CONCLUSION: This production technology can get higher recovery of diosgenin, and it is simple, time and money saving.
OBJECTIVE: To observe the clinical characteristics of different patterns of the left ventricule in hypertensive patients. METHODS: Echocardiography was used in 228 essential hyventricule of heart in hypertensive patients and 60 healthy individuals. RESULTS: In the hypertensive group, the rate of normal left ventricle, concentric remodeling, concentric hypertrophy and eccentric hypertrophy were 21.9%, 36.4%, 32.9% and 8.8%, respectively. The aortic root dimension and left atrial dimension were higher in the hypertensive group than those in the normal group (P < 0.05), especially for concentric hypertrophy and eccentric hypertrophy. Cardiac index and stroke volume index in eccentric hypertrophy were higher than those in the normal group and concentric remodeling (P < 0.05). Ejection fraction and fractional shortening in concentric remodeling were the highest and ejection fraction in eccentric hypertrophy was the lowest among all patterns (P < 0.05). E/A rate was lower in the hypertensive group than in the normal group (P < 0.05), especially for concentric hypertrophy and eccentric hypertrophy. The total peripheral resistance was the highest in concentric remodeling and the lowest in eccentric hypertrophy (P < 0.05). CONCLUSION: Different patterns in the left ventricle have their own characteristics and patients should be treated differently according to the patterns.
A 209-base pair (bp) crocodilian-specific sequence characterized amplified region (SCAR) was identified from a 425-bp randomly amplified polymorphic DNA (RAPD) fragment. The 209-bp SCAR was produced from amplifications of DNA extracted from fresh and/or dry meat samples from at least three species of Crocodylus, Caiman crocodylus, and Alligator mississippiensis. No amplification was observed from DNA of other common animal species. The use of SCAR opens the way for quick authentication of crocodilian samples for conservation biology and trade regulation.
The National Institutes of Health Mammalian Gene Collection (MGC) Program is a multiinstitutional effort to identify and sequence a cDNA clone containing a complete ORF for each human and mouse gene. ESTs were generated from libraries enriched for full-length cDNAs and analyzed to identify candidate full-ORF clones, which then were sequenced to high accuracy. The MGC has currently sequenced and verified the full ORF for a nonredundant set of >9,000 human and >6,000 mouse genes. Candidate full-ORF clones for an additional 7,800 human and 3,500 mouse genes also have been identified. All MGC sequences and clones are available without restriction through public databases and clone distribution networks (see http:mgc.nci.nih.gov).
Epidemiological reports suggest that Asians consuming a diet high in soy have a low incidence of clinically manifested prostate cancer. We have tested the hypothesis that life-time exposure to genistein, the primary isoflavone component of soy, is responsible for this protective effect. Lobund-Wistar rats were exposed to 0, 25 and 250 mg genistein/kg AIN-76A diet, starting at conception and continued until necropsy at 11 months. Male offspring were injected s.c. with Flutamide on days 50-66 and with testosterone on days 67-69, injected with N-methylnitrosourea (NMU) into the dorsal prostate on day 70, and given testosterone implants, starting at day 77. Genistein in the diet inhibited the development of NMU-induced prostate invasive adenocarcinomas, in a dose-dependent manner. Genistein did not alter body, prostate and testes weights. Male rats fed 0, 25 and 250 mg genistein/kg diet had serum genistein concentrations of 9, 60 and 861 pmol/ml, and prostate genistein concentrations of 85, 230 and 775 pmol/g tissue. We conclude that lifetime dietary genistein protected against chemically induced prostate cancer development in rats.
Due to a diffusive nearest-neighbor coupling, phase-synchronized states can emerge in two-dimensional chaotic coupled map lattices. By defining a direction phase (like a spin with up or down direction) as the direction of two sequential iterations of the logistic map, we find several novel kinds of phase synchronization which correspond to four different regions in a phase diagram. For the phase with partial phase synchronization, as the coupling strength epsilon increases to a critical threshold epsilon(c), a percolationlike transition is found in the cluster feature of the direction phases relating to the pattern formation. In addition, a scaling of the percolation probability rho approximately (epsilon-epsilon(c))(beta) with beta=2.1 is obtained. The spatial and time correlation functions of the phase clusters are also discussed.
A new cationic polymer (PPA-SP), polyphosphoramidate bearing spermidine side chain, was prepared as a non-viral vector for gene delivery. PPA-SP was synthesized from poly(1,2-propylene H-phosphonate) by the Atherton-Todd reaction. The weight average molecular weight of PPA-SP was 3.44x10(4) with a number average degree of polymerization of 90, as determined by GPC/LS/RI method. The average net positive charge per polymer chain was 102. PPA-SP was able to condense plasmid DNA efficiently and formed complexes at an N/P ratio (free amino groups in polymer to phosphate groups in DNA) of 2 and above, as determined by agarose gel electrophoresis. This new gene carrier offered significant protection to DNA against nuclease degradation at N/P ratios above 2, and showed lower cytotoxicity than PLL and PEI in cell culture. The LD(50) of PPA-SP was 85 microg/ml in COS-7 cells, in contrast to 20 and 42 microg/ml for PLL and PEI, respectively. The complexes prepared in saline at N/P ratios of 5 approximately 10 had an average size of 250 nm and zeta-potential of 26 mV. PPA-SP mediated efficient gene transfection in a number of cell lines, and the transfection protocol was optimized in HEK293 cells using a luciferase plasmid as a marker gene. Gene expression mediated by PPA-SP was greatly enhanced when chloroquine was used in conjunction at a concentration of 100 microM. Under the optimized condition, PPA-SP/DNA complexes yield a luciferase expression level closed to PEI/DNA complexes or Transfast mediated transfection. In a non-invasive CNS gene delivery model, PPA-SP/DNA complexes yielded comparable bcl-2 expression as PEI/DNA complexes in mouse brain stem following injection of the complexes in the tongue.
Previous studies indicated that calcitonin (CT), a peptide hormone involved in calcium homeostasis, is transiently expressed in the receptive rat and human endometrial epithelia within the window of implantation. Attenuation of uterine CT expression using antisense methods severely impaired implantation in the rat. The molecular pathway of CT in the pregnant uterus, however, remains unknown. In the present study, we investigated the cellular events following the binding of CT to its membrane receptors in human endometrial epithelial cell line Ishikawa. We observed that CT treatment triggers a transient rise in intracellular calcium in these cells. Most interestingly, CT treatment also led to the disappearance of E-cadherin, a critical cell adhesion molecule, from cell-cell contact sites. Blockade of intracellular calcium release by BAPTA-AM (1,2-bis(2-aminophenoxy)ethane-N,N,N',N'-tetraacetic acid-acetoxymethyl) prevented the CT-induced disappearance of E-cadherin. Our studies further revealed that CT treatment markedly down-regulates the level of E-cadherin mRNA in Ishikawa cells. We also examined whether CT influences the expression of E-cadherin mRNA in intact rat uterine tissue during implantation. In pregnant rats, high levels of E-cadherin mRNA were expressed during the first 3 days of gestation when the CT mRNA in uterine epithelial cells is undetectable. Concomitant with a transient burst of CT expression during days 4-5 of pregnancy, the level of E-cadherin mRNA declined sharply. Furthermore, administration of exogenous CT to animals on day 2 of pregnancy led to a premature suppression of E-cadherin mRNA level on day 3, indicating a direct link between elevated levels of uterine CT and the down-regulation of E-cadherin expression in the surface epithelium. Collectively, our results are consistent with the hypothesis that CT-induced reduction in E-cadherin expression may remodel the adherens junctions between epithelial cells, and this change in epithelial cell phenotype might be a critical event during the implantation of the blastocyst.
Parallel activation of Ca(2+)-dependent K(+) channels and volume-sensitive Cl(-) channels is known to be responsible for KCl efflux during regulatory volume decrease (RVD) in human epithelial Intestine 407 cells. The present study was performed to identify the K(+) channel type. RT-PCR demonstrated mRNA expression of Ca(2+)-activated, intermediate conductance K(+) (IK), but not small conductance K(+) (SK1) or large conductance K(+) (BK) channels in this cell line. Whole cell recordings showed that ionomycin or hypotonic stress activated inwardly rectifying K(+) currents that were reversibly blocked by IK channel blockers [clotrimazole (CLT) and charybdotoxin] but not by SK and BK channel blockers (apamin and iberiotoxin). Inside-out recordings revealed the existence of CLT-sensitive single K(+)-channel activity, which exhibited an intermediate unitary conductance (30 pS at -100 mV). The channel was activated by cytosolic Ca(2+) in inside-out patches and by a hypotonic challenge in cell-attached patches. The RVD was suppressed by CLT, but not by apamin or iberiotoxin. Thus we conclude that the IK channel is involved in the RVD process in these human epithelial cells.
Determination of individual genotypes in DNA mixture remains a challenge in forensic science. Using an approach of mixture of distributions, this article provides formula for calculation of paternity index (PI) in cases where only tissue mixture of the mother and alleged father, the genotypes of the mother and child, but not that of the alleged father are available. The formula has been used to solve a real case using mother's vaginal tissue contaminated with semen from alleged father.
