Reciprocal action between BMP-2 and BMP-3 in cultured fibroblast in vitro.

OBJECTIVE: To explore reciprocal action between BMP-2 (bone morphogenetic protein-2) and BMP-3 for better understanding of the mechanism of BMP during bone fracture union. METHODS: rhBMP-2 was added into the cultured fibroblasts with the concentration of 1,200 ng/ml. The expression of BMP-3 in fibroblasts was detected by immunohistochemistry. Eukaryotic expression vector pcDNA3-BMP-3 was transfected into the fibroblasts. After the effective expression of BMP-3 was identified, BMP-2 was also detected by immunohistochemistry in BMP-3 expression cells. The fibroblasts transfected with empty vector pcDNA3 were used as the control. RESULTS: Exogenous rhBMP-2 could promote the expression of BMP-3 in fibroblasts. BMP-3 also could be detected in these cells. CONCLUSIONS: BMP-2 and BMP-3 could reciprocally adjust the expression in fibroblasts.

[A pilot study on changes of posterior available spaces in mandible dentitions for adolescents between the ages of 11 and 20 years in Chengdu area].

OBJECTIVE: The purpose of this study was to study the changes of available spaces of posterior arch segments in mandible dentitions (ASPAS) from ages of 11 to 20 years, which would provide information on normal growth and development of the dental arches in teenagers in Chengdu area. METHODS: The data were derived from systematically collected preorthodontic lateral cephalograms of 526 patients, including 232 males and 294 females from the Orthodontic Department of West China College of Stomatology, Sichuan University during the period of 1999-2000 (aged 11-20 years). The values of ANB angle, FH-MP, ASPAS were obtained using the cephalometric analysis. Afterwards, the growth changes of ASPAS from the subjects and their correlations with age, gender, angle malocclusion types and vertical skeletal patterns were analyzed. RESULTS: ASPAS was related to age and class II angle malocclusion. Though gender was not associated with ASPAS, significant differences in curves of ASPAS were found between males and females. The growth changes of ASPAS in the patients with skeletal Class III malocclusion were more obvious than in patients with Class I and II malocclusion. CONCLUSION: Our results reveal that significant differences were demonstrated in growth change values in posterior available spaces of patients with different races and angle malocclusion types.

[Expression of apoptosis and proliferation related genes in carcinogenesis of lung squamous cell carcinoma in rats].

BACKGROUND & OBJECTIVE: The activation of caspase-3 protein, located the downstream of apoptosis, is the key of cell apoptosis signal conduction. Caspase-3 is the most important performer in accelerating apoptosis in the caspase family. Much progress has been achieved in the study of caspase-3 and human non-small cell lung cancer. This study was designed to investigate the effect of cellular proliferation and apoptosis related genes caspase-3 and proliferating cell nuclear antigen (PCNA), and to seek whether they could be chosen as molecular biology markers for lung cancer. METHODS: 3-Methylcholanthrene (MCA) and diethyinitrosamine (DEN) were used to induce lung squamous cell carcinoma by intra-left lobar-bronchial instillation in 50 Wistar rats. The other 10 rats instilled with iodized oil were regarded as control group. The expression of caspase-3 and PCNA were evaluated by SP immunohistochemistry during carcinogenesis. TUNEL(TDT-mediated dUTP nick end labeling) method was used to examine apoptotic cells. RESULTS: For the rat bronchial epithelium cells in control group, precancerous lesions, and lung squamous cell carcinoma, positive coefficient values of caspase-3 protein were 3.10+/-0.99, 2.25+/-1.13, and 1.38+/-0.95 on average, respectively; the means of PCNA-labeling index (PCNA-LI)were 14.10+/-5.02, 28.13+/-8.72, and 41.88+/-14.24, respectively; the means of apoptotic index(AI) were 0.60+/-0.52, 2.06+/-0.85, and 2.26+/-1.14, respectively.Significant differences in caspase-3 protein expression were observed between bronchial epithelium in control group and lung squamous cell carcinoma (P< 0.01). Caspase-3 expression was showed stronger in precancerous lesions than that in lung cancer (P< 0.05). Low proliferation index and low AI were detected in rat bronchial epithelium region in control group, which were significant differences in precancerous lesions and lung cancer, respectively (P< 0.01). In 34 rats with lung squamous cell carcinoma, there was negative relationship between caspase-3 and PCNA-LI (r=-0.7306, P< 0.01), so did it in AI and PCNA-LI(r=-0.8127,P< 0.01), but there was no relationship between caspase-3 expression and AI(P >0.05). CONCLUSION: Loss of caspase-3 expression may be associated with the development of lung squamous cell carcinoma in Wistar rats, but it is not associated with AI. PCNA-LI is an important marker for malignant progression of lung cancer.

The effects of land use and its patterns on soil properties in a small catchment of the Loess Plateau.

Due to relatively strong human activities in the hilly area of Loess Plateau, the natural vegetation has been destroyed, and landscape pattern based on agricultural land matrix was land use mosaic composing of shrub land, grassland, woodland and orchard. This pattern has an important effect on soil moisture and soil nutrients. The Danangou catchment, a typical small catchment, was selected to study the effects of land use and its patterns on soil moisture and nutrients in this paper. The results are as follows: The comparisons of soil moisture among seven land uses for wet year and dry year were performed; (1) the average of soil moisture content for whole catchment was 12.11% in wet year, while it was 9.37% in dry year; (2) soil moisture among seven land uses significantly different in dry year, but not in wet year; (3) from wet year to dry year, the profile type of soil moisture changed from decreasing type to fluctuation-type and from fluctuant type to increasing type; (4) the increasing trend in soil moisture from the top to foot of hillslope occurred in simple land use along slope, while complicated distribution of soil moisture was observed in multiple land uses along slope. The relationship between soil nutrients and land uses and landscape positions were analysed: (1) five nutrient contents of soil organic matter (SOM), total N (TN), available N (AN), total P (TP) and available P (AP) in hilly area were lower than that in other area. SOM content was less than 1%, TN content less than 0.07%, and TP content between 0.05% and 0.06%; (2) SOM and TN contents in woodland, shrub land and grassland were significantly higher than that in fallow land and cropland, and higher level in soil fertility was found in crop-fruit intercropping land among croplands; (3) soil nutrient distribution and responses to landscape positions were variable depending on slope and the location of land use types.

[Effects of plastic film mulching on soil temperature and moisture and on yield formation of spring wheat].

Field experiments were carried out to study the effects of plastic film mulching on soil temperature and moisture, and on yield formation of spring wheat. The results showed that plastic film mulching could increase soil temperature with "U" variances in the whole growth period of spring wheat. Plastic film mulching could also increase soil available water by restraining evaporation and elevating deep water to the layer useable for roots. The promotion of soil temperature and soil water content under plastic film mulching was favorable to the development and water use of spring wheat in its earlier stage. Root developments were restrained when mulching in later stage of spring wheat, with decreases in crop water consumption and water use efficiency, and there was no significant effects on yield formation. The spring wheat yields under control (CK), pre-sowing irrigation (W), mulching for whole stage (M), pre-sowing irrigation adding mulching for 30 days (WM30), pre-sowing irrigation adding mulching for 60 days (WM60), and pre-sowing irrigation adding mulching for whole stage (WMw) were 2 554, 2,424, 2,750, 3,138, 3,305, and 3,123 kg.hm-2, respectively, and the optimum mulching time was at 40-60 days.

[Lung squamous cell carcinoma-related genes cloned by using suppression subtractive hybridization].

OBJECTIVE: To identify genes differentially expressed in human lung squamous cell carcinoma (LSCC). METHODS: A subtracted cDNA library of human LSCC constructed by using suppression subtracted hybridization (SSH) method was screened. Clones representing mRNAs that were truly differentially expressed in LSCC but not in its adjacent non-cancerous tissues were identified by semi-quantitative RT-PCR in 12 patients with LSCC. Partial novel genes were detected by Northern blot. RESULTS: Ten differentially expressed gene cDNA fragments of LSSC were obtained by SSH. Among them six were known genes; two sequences were identified but their functions were unknown (hypothetical protein); two were novel (GenBank accession numbers AF363068 and AY032661, respectively). The results from semiquantitative RT-PCR showed that the transcription expression level of PPP1CB, calumenin, S100A2, HSNOV1, OCIA and AY032661 was down-regulated in some LSCC cases, while the transcription of HSP90, ferritin, gp96 and AF363068 was up-regulated in others. CONCLUSIONS: Six known genes identified by SSH technique have been implicated in the pathogenesis of lung carcinogenesis, or they are involved in immunological defense mechanism in human body. Two hypothetical proteins probably also play an important role in the pathogenesis of lung cancer. The function of the two novel genes in lung carcinogenesis are under investigation.

[Expression of nm23 genes in acute myeloid leukemia and its clinical significance].

OBJECTIVE: To explore nm23 gene mRNA expression and its clinical significance in acute leukemias (AML). METHODS: The levels of nm23-H1 and nm23-H2 transcripts in 22 patients with acute myeloid leukemia (AML), 9 AML in complete remission (AML-CR), 12 acute lymphoblastic leukemia (ALL) and 4 chronic myeloid leukemia in chronic phase (CML-CP) were assayed by reverse transcriptase-polymerase chain reaction (RT-PCR). RESULTS: The expression of nm23-H1 in AL especially in AML-M4 and AML-M5 was significantly higher than that in normal blood cells. An analysis of correlation between nm23 expression and clinicopathological parameters showed that increased nm23-H1 mRNA levels were associated with some poor-prognostic factors such as extramedullary infiltration, high white blood cell count (WBC), high lactate dehydrogenase (LDH) activity and high CD(7) expression, while inversely correlated with t(8; 21) and t(15; 17) which had a good-prognostic effect. The expression of nm23-H1 in AML patients in CR was significantly decreased compared with those untreated. CONCLUSION: nm23-H1 was overexpressed in AL, especially in AML-M4 and AML-M5. High expression of nm23-H1 may be a poor prognostic factor.

[Perioperative continuous positive airway pressure therapy for severe obstructive sleep apnea hypopnea syndrome patients].

OBJECTIVE: (1) To investigate the severe obstructive sleep apnea hypopnea syndrome (OSAHS) patients' perioperative variations of their polysomnographic indices, to discuss the necessity for their perioperative treatment; (2) To investigate the effects of continuous positive airway pressure (CPAP) on severe OSAHS patients during their perioperative period. METHODS: (1) 21 cases severe OSAHS patients were selected for at least 7 days preoperative CPAP therapy, and this group of patients were also given 3 continuous nights autotitrated CPAP (AutoSet) therapy postoperatively, i.e., the first 3 nights after operation. The 21 cases were remonitored with PSG during the second night after operation with their AutoSet ON. And all the 21 cases had revised uvulopalatopharygoplasty, in which the uvula is reserved completely. All patients apnea hypopnea index(AHI), lowest SaO2 (LSaO2), and sleep structure indices were calculated. (2) Another 24 cases of severe OSAHS patients without preoperative CPAP therapy or tracheotomy were selected as the control group, all patients received polysomnography (PSG) on the second night postoperatively. RESULTS: (1) 6 cases' condition of the control group got worse during the second night after operation, their LSaO2 are lower and their AHI got higher than pre-operation. For the another 18 cases, their condition got better than before operation. (2) 21 cases' AHI and LSaO2 are 61.1 +/- 9.9, 65.0% +/- 9.6% respectively before CPAP treatment, and the AHI and LSaO2 are 2.2 +/- 1.4, 94.5% +/- 2.9% during CPAP therapy. P < 0.001 (TTEST). All the 21 cases main symptoms disappeared after 1 week CPAP therapy. All 21 cases could tolerate AutoSet treatment well for the first 3 nights after operation. During the 2nd night with AutoSet therapy, the AHI and LSaO2 are 3.6 +/- 1.8 and 93.7% +/- 3.4% respectively. (3) For the 2nd night after operation, the CPAP and AutoSet treating group's AHI is lower than that of the control group, also the LSaO2 is higher, the deep and REM sleep stages are longer, and the S1, S2 sleep stages are shorter than that of the latter. CONCLUSION: For severe OSAHS patients, the postoperative condition has the possibility of getting worse. Perioperative CPAP therapy can have OSAHS patients severe condition alleviated, so the perioperative risks could be lowered; During the first 3 postoperative nights, AutoSet can be well tolerated by severe OSAHS patients.

[Study on the effect of NaFeEDTA on preventing lead poisoning in rats and its impact on zinc, copper].

In order to explore the effect of sodium iron ethylenediaminetetraacetate (NaFeEDTA) on preventing rats from lead poisoning and reducing the lead toxicity, and at the same time to explore its impact on zinc and copper, sixty Wistar rats were randomly divided into six groups including blank control group, model control group, positive control group and three different level NaFeEDTA (high, middle and low) groups. With the exception of the blank control group, which only received double distilled water treatment throughout the experiment, all the other rats were continuously treated with lead solution. At the same time, with the exception of the blank and model control groups, the other four groups were treated with acidi dimercaptosuccinici (DMSA), high, middle and low level NaFeEDTA solutions respectively. At the end of the study, several indicators were determined. The results showed that the hemoglobin levels in the three different level NaFeEDTA groups were significantly higher than those of the other four groups and at the same time the lead levels in blood and organs were lower than those of model group significantly. There were significant reverse correlations between iron supplement and the lead levels in liver, kidney and tibia among the three different level NaFeEDTA groups, and the correlation coefficients were -0.4432, -0.6134 and -0.3878 respectively. There were no significant differences in zinc levels and copper levels between the three level NaFeEDTA groups and the negative control group except the brain zinc levels in the middle and high level NaFeEDTA groups were lower and the kidney copper level in the low level NaFeEDTA group was higher than those of the model control group. It is concluded that the NaFeEDTA can effectively improve hemoglobin level, prevent rats from lead poisoning and reduce the lead toxicity. At the same time, NaFeEDTA does not have significant influence on zinc and copper levels in rats.

[Studies on molecular epidemiology of Y chromosome azoospermia factor microdeletions in Chinese patients with idiopathic azoospermia or severe oligozoospermia].

OBJECTIVE: To determine microdeletion loci and the characteristic of Y chromosome azoospermia factor(AZF) associated with Chinese idiopathic azoospermia or severe oligozoospermia and hence to provide a theoretic basis for gene diagnosis of AZF microdeletion in Chinese infertility men. METHODS: The subjects of this study included 134 cases of azoospermia and 118 severe oligozoospermia, and 210 healthy male controls. With multiplex PCR technique, an analysis of 15 sequence tag sites(STS) in AZFa, AZFb and AZFc microdeletion was performed. RESULTS: No microdeletion was detected in controls whereas 13 STS loci microdeletions existed in cases, including sY84 and sY86 in AZFa; sY121, sY123, sY124, sY127, sY134 and sy133 in AZFb; sY152, sY242, sY254, sY255 and sY157 in AZFc. Five azoospermia patients were involved in AZFa microdeletions, 7 azoospermia and 3 severe oligozoospermia patients in AZFb, and 14 azoospermia and 18 severe oligozoospermia patients in AZFc. The prevalence rates of microdeletion in AZFa, AZFb and AZFc were 2.0%,4.0% and 12.7% respectively. The microdeletion rate showed statistically significant difference between the cases and controls. CONCLUSION: This study revealed an association between the microdeletion of 13 STS loci of AZF region and the idiopathic azoospermia or severe oligozoospermia in Chinese. There is no evidence to identify the genetic polymorphism of the above STS loci. The frequency and distribution characteristic of AZF microdeletion are similar to those of Caucasians. As candidate loci the 13 STS are useful in clinical gene diagnosis for the detection of AZF microdeletion in Chinese idiopathic azoospermia and severe oligozoospermia.

[Effects of exercise training during 21 d -6 degrees head down bed rest on dynamic posture equilibrium and motor coordination].

OBJECTIVE: To study the effects of 21 d -6 degrees head down bed rest (HDBR) with exercise training on dynamic posture equilibrium and motor coordination. METHOD: Ten healthy young men were randomly divided into HDBR control groups and HDBR exercise groups, with 5 in each group. The subjects in the exercise group performed exercise with a bicycle ergometer in the supine position 1 h/d during bed rest. Dynamic posture equilibrium as well as isokinetic concentric muscular strength were determined before and after 21 d HD-BR. RESULT: After HDBR, average dynamic proprioception score and dynamic motor coordination decreased significantly, relative peak torque (peak torque /body weights PT/BW) also decreased significantly and accompanied with increase of the peak torque ratio between hamstrings and quadriceps (H/Q) in HDBR control groups. Average dynamic proprioception score and dynamic motor coordination in the HDBR exercise groups remained above the HDBR control groups, but the H/Q ratio showed no significant change. CONCLUSION: The results showed that exercise training during 21 d -6 degrees head-down tilt bed-rest do have beneficial effects on retaining the dynamic posture equilibrium and dynamic motor coordination of the subjects after bed rest.

[Effect of absorbable poly-DL-lactide rods on experimental fracture healing].

This study was designed to assess the effect of a home-bred poly-DL-lactide (PDLLA) rods on experimental fracture healing. An operation was performed to create bilateral ulnar and radial fractures of forearm in 15 rabbits. The right sides were fixed with 2 PDLLA (Mv = 34 x 10(4), 1.6 mm x 1.6 mm x 30 mm) rods, and the contralateral sides were fixed with 2 Kirschner wires to be used as controls. After operation, casts were used for external fixation, and the joint above and the joint below the fracture were immobilized. The casts were removed at the 6th week after operation. The specimens were removed at specific intervals between 2 and 12 weeks. Histologically, the light microscopy and scanning electron microscopy evaluation of the rabbit ulnar and radial fractures fixation revealed that, the fractured bone gradually healed and the PDLLA rods properly fixed on the fracture area during the implanted period. However, results suggest that internal fixation by absorbable material made the course of fracture healing slower than that in the control group, but the osteogenesis appeared to be normal. In the absorbable group, the collagenous fiber was characterized by order and orientation, and calcified normally. In conclusion, this kind of PDLLA rods have an effect on delaying the process of the normal union of fracture and should be applicable to fractures where the tissues are rich in blood supply.

[Effect of plastic film mulching on soil microbial biomass in spring wheat field in semi-arid loess area].

This paper studied the effect of different periods of plastic film mulching (M0-no mulching, M30-mulching for 30 days, M60-mulching for 60 days, and M-mulching for whole growth period) on soil microbial biomass carbon (SMBC) of spring wheat field in semi-arid loess plateau. The mean SMBC in 1999 and 2000 was 335.3 and 259.3 mg.kg-1 dry soil, respectively. It was 29.3% higher in 1999 than in 2000. The highest SMBC was recorded at the harvest stage in M treatment for the two years. In 1999, a wet year with more rainfall, the SMBC of M60 and M treatments was significantly higher than those of M0 and M30 in the mid-period of growth, reached its top at the end of the growth period. The highest grain yield was also achieved in M60. It was a dry year in 2000, but rainfall was rich in the latter growth period of spring wheat. SMBC increased at the beginning period of growth, and did not increase during the mid-growth period. It increased again during the latter period of growth, and showed a significant difference among the treatments. At the harvest stage of 2000, SMBC in M0 was the highest among all the treatments. It was similar between M and M60, and lower than that of M30. In the two years, the ratio of C/N ranged between 7.732-9.042, being lower than the threshold of 11.3, and the ratio of C/P was 300.8-719.6, being higher than the threshold of 300. The two parameters showed that the increase of SMBC was inhibited because of the competition of soil available nutrients between soil microbes and crops. These indicated that soil organic matter content was so shortage that it became the key factor to restrict crop productivity. Under this condition, increasing crop productivity through the input of chemical fertilizers would conceal the problem of soil degradation, and result in a further decrease of soil quality. A long term plastic film mulching would make the problem more serious.
