[Retrospective analysis of misdiagnosed amoebic keratitis].

OBJECTIVE: To analyze the clinical characteristic of amoebic keratitis and the cause of misdiagnosis retrospectively. METHODS: Corneal pathological sections of 269 cases having been diagnosed as herpes simplex keratitis (HSK), bacterial keratitis and other microbial keratitis and then undergone penetrating keratoplasty (PKP) were reexamined to find the misdiagnosed cases of amoebic keratitis. The clinical characteristic efficacy of treatment and cause of misdiagnosis were retrospectively analyzed according to case history and photographs. RESULTS: Based on the pathological examination, 5 cases of amoebic keratitis were recognized which had been misdiagnosed, including HSK in 3 cases, fungal keratitis in 1 case and suppurative keratitis in 1 case previously. The main clinical characteristics were severe radiating neuralgia and ring-shaped stromal infiltrates. After PKP, infection of 3 cases were controlled, 2 cases were recurrent and underwent eyeball enucleation eventually. CONCLUSION: The incidence of amoebic keratitis is not very low in China. It is necessary to pay high attention to this kind of disease, and its early and definite diagnosis is very important for the prevention of serious results.

Pattern of polycyclic aromatic hydrocarbons (PAHs) pollution in communication air of Hangzhou, China.

Nine PAHs were surveyed in communication air of Hangzhou. The results indicated that the PAHs pollution is very serious and the total sum of 9 PAHs, on the average are 3.39-13.82 micrograms/m3. The PAHs signatures for all streets are similar to each other. Multivariate statistical techniques were used to investigate source apportionment for PAHs. A factor analysis/multiple regression model was successfully applied to the study. The most important three PAHs sources in communication air are diesel emission, gasoline engine emission, coal-burning accounting for 61.1 +/- 6.4%, 19.9 +/- 8.3%, 10.8 +/- 10.8% of total PAHs, respectively. The relationship for three source tracers and total PAHs is: [PAHs] = 1.471 (+/- 0.155) [Phen] + 2.538 (+/- 2.522) [1 - Mepy] + 2.254 (+/- 0.943) [Chry] + 1.022 (+/- 1.767).

Safety and bioactivity of intracoronary delivery of naked plasmid DNA encoding human atrial natriuretic factor.

AIM: To evaluate the safety and bioactivity of catheter-mediated intracoronary gene delivery of naked plasmid DNA encoding human atrial natriuretic factor (hANF). METHODS: hANF gene delivery was performed in 12 canines. For each canine, 4 mg of reconstructed naked plasmid DNA encoding hANF (pCR3*hANF, n=6) or pCR3 (n=6, control) in 2 mL normal saline was injected into left coronary artery via a coronary angiographic catheter. The expression of hANF mRNA in left ventricular myocardium, liver, lung, and kidney was identified by reverse transcription polymerase chain reaction and Southern blot analysis 40 d after gene delivery. Plasma ANF levels were determined by radioimmunoassay. RESULTS: The naked pCR3*hANF caused significant expression of hANF mRNA in ventricular myocardium (P <0.01). No hANF mRNA was detected in distal tissues, including liver, lung, and kidney (P >0.05). ANF levels were significantly higher in pCR3*hANF group than those in control group. CONCLUSION: These facts firstly demonstrate that intracoronary delivery of naked pCR3*hANF is probably a safe and feasible method for gene delivery by which hANF gene can be expressed in ventricular myocardium effectively.

Chemotherapy in epithelial ovarian cancer.

The management of epithelial ovarian carcinoma can be the most challenging of all the gynecologic malignancies partly because of the paucity of effective screening tools for early-stage disease. Numerous randomized clinical trials identified several cytotoxic agents that are active against epithelial ovarian cancer, but the current standard of care remains optimal surgical cytoreduction followed by platinum and taxane-based chemotherapy. When disease recurs or progresses, alternative chemotherapy regimens often have efficacy, either as a single agent or in combination. While treatment recommendations should be individualized, the disease-free interval after induction chemotherapy is often the most clinically useful parameter to determine selection of second-line therapy. Investigations into novel chemotherapeutic agents and other biologic molecules have led to advances in the therapeutic armamentarium and have improved survival for women with ovarian cancer.

[Toxicity assessment of soil contaminated by heavy metals using algae growth inhibition test].

Scenedesmus obliquus growth inhibition test was carried out to assess the toxicity of soil contaminated by heavy metals. The results showed that the growth rate of Scenedesmus obliquus was positively related to the concentration of heavy metals added to soil, and it was decreased with the increasing concentration of heavy metals. Two parameters were chosen and the sensitivities were compared. It was found that the cell growth rate was more sensitive than OD growth rate. The orders of general detection limit by EC50 of different metals were Pb > Cu > Zn > Cd by using cell growth rate as the test index and Pb > Cu > Cd > Zn by using OD as the test index. This study also indicated that under the complex pollution condition, the toxicity of heavy metals in soil was stronger than that in the case of soil contaminated by single metal.

Comparative study of catheter-mediated gene transfer into heart.

OBJECTIVE: To investigate the feasibility and features of 3 catheter-mediated approaches of gene transfer into heart, including direct myocardial injection (DMI), coronary artery perfusion (CAP), and intrapericardial cavity injection (ICI). METHODS: Fifteen dogs were used, and 0.3 ml (1 x 10(9) pfu) of an adenovirus (Adex1SR LacZ) was injected into the heart by 3 methods. The dogs were killed 5 days following injection, and gene expressions in heart and liver were evaluated by histochemical analysis. RESULTS: The results showed that (1) the CAP method was relatively less damaging and induced sparse LacZ expression in the myocardium, and the gene expression was also found in both vessels within the myocardium and liver; (2) gene transfer by DMI resulted in intense LacZ expression around the injection accompanied by a local inflammatory response; (3) LacZ expression elicited by ICI was detected in either the inner surface of the parietal pericardium or epicardial surface of the heart, and also in the myocardium underlying the visceral pericardium. CONCLUSION: Three catheter-mediated methods of gene transfer into the heart may be used and a reasonable approach should be chosen according to purpose.

[Effect of clipping at seedling stage on growth and yield of spring wheat].

The compensation for spring wheat (Triticum aestivum) under simulated herbivory stress by clipping was examined in a semi-arid region of Gansu province in 1996. The results showed that clipping at seedling stage reduced spring wheat growth under two irrigation conditions. The yield of spring wheat generally under-compensated the clipping effect. Heavy clipping (cutting all leaves above the ground, T0, T1) resulted in a more serious reduction in yield, compared with light clipping cutting half of the leaves, H0, H1). The yields of all treatments were lower than controls(CK0, CK1), i.e., CK0 > H0 > T0 > CK1 > H1 > T1. Irrigation for once right after clipping increased wheat growth and its compensatory ability, although they were still under-compensation.

[Expression and purification of human papillomavirus type16 L1 protein in a prokaryotic expression system].

This study was intended to establish a method of purification of HPV16 L1 protein expressed in a prokaryotic system and to obtain the purified protein. The prokaryotic expression vector pGEX-4T-1-HPV16 L1 was constructed and transformed into E. coli BL21 cell, and induced by 1 mM IPTG to express HPV16L1 protein. The inclusion bodies were isolated and solubilized with 8 M urea. After the urea was removed by gradual dialysis, the denatured L1 protein were renatured and then were purified by affinity chromatography. The results showed that HPV16L1 protein formed inclusion bodies in bacterial expression system, suggesting that this assay can be used to purify HPV16L1 protein and hence provide a basis for studying the applications of HPV16 L1 protein.

The role of B7 molecules in T cell anergy.

OBJECTIVE: To explore the role and mechanism of B7 molecules in T cell anergy. METHODS: Anti-B7-1 (CD(80)) and anti-B7-2 (CD(86)) monoclonal antibodies were used to induce T cell anergy. T cell proliferation were assayed by mixed lymphocyte reaction (MLR) with (3)H-TdR incorporation, and cytokine mRNA transcripts were analyzed with reverse transcriptase-polymerase chain reaction (RT-PCR). B7-transfected-CHO cells were used as artificial antigen presentation cells (APCs) in MLR to exclude the effects of other costimulatory molecules. RESULTS: MLR results showed that the proliferation of T cells was inhibited to various extents by anti-CD(80) or anti-CD(86) monoclonal antibody, the effect of anti-CD(86) antibody was greater than that of anti-CD(80) antibody, and the proliferation was totally blocked when the two were used together. The results of RT-PCR demonstrated that IL-2 and IFN-gamma mRNA transcripts decreased whereas IL-4 mRNA transcripts increased in T cell after treatment with anti-B7 antibo-dies for 24 hours. In MLR with artificial APC, signal one (DR7) alone could stimulate T cell proliferation at a certain threshold intensity. Costimulator B7-1 molecule could help signal one in T cell proliferation. This effect was blocked by anti-CD(80). CONCLUSION: B7 molecules play an important role in T cell immune response. Blockade of B7 family resulted in T cell anergy. The role of CD(86) may be more important than that of CD(80). The conversion of cytokine profile from Th1's to Th2's reflected that anergetic T cells were differentiated into Th2 cells by anti-B7 suggesting that anergetic blockade of costimulator molecules may be one of the mechanisms of T cell.

[Effects of hexamethylene bisacetamide on related genes expression of apoptosis and cell proliferating in human mucoepidermoid carcinoma cell line MEC-1].

OBJECTIVE: To investigate the effects of hexamethylene bisacetamide (HMBA) on related genes expression of apoptosis and cell proliferating in human mucoepidermoid carcinoma cell line MEC-1. METHODS: MEC-1 was induced 72 hours by 0.002 mol/L HMBA in vitro. The morphologic observation and the expression of PCNA, bcl-2, bax protein and bcl-2 mRNA were determined by microscope, immunohistochemistry, in situ hybridization and image analysis. RESULTS: The expression of bax, bcl-2 protein and bcl-2 mRNA were higher than control group (P < 0.01). The related genes of cell proliferating PCNA were decreased in control group. CONCLUSIONS: HMBA could inhibit the MEC-1 cell proliferation by down-regulation of PCNA expression, it could also induce apoptosis effectively that might be through the way of up-regulation of bax and bcl-2 gene expression.

[The identical effects of B7-1 and B7-2 on regulation of human IL-2 gene transcription factors NF-kappa B and AP-1].

To detect effects of B7 co-stimulation on cytokines, especially on IL-2 mRNA and transcription factors NF-kappa B and AP-1, antiB7-1 McAb, antiB7-2 McAb and C TLA-4 Ig were added into mixture lymphocyte reaction (MLR) system to block B7/C D28 signal transduction, IL-2 mRNA and IL-4 mRNA were determined by using competitive PCR and IFN-gamma mRNA by using semi-quantitative PCR. MHC class II molecules and B7 transfectants were used to stimulate CD28(+) T cell, effects of B7 on NF-kappa B and AP-1 were detected by DNA-protein binding assay. The results showed that IL-2, IL-4 and IFN-gamma mRNA were significantly lower when blockade of B7-2 in MLR than blockade of B7-1. Synergistic effects could be seen with combination of two or three antibodies. One to six hours after MLR, tDR7 alone induced NF-kappa B binding activity; cotransfecting B7 no significantly difference at early time point. After 6 hours, induction of tDR7 was decreased whereas B7 prolonged the induction of NF-kappa B till 72 hours. tDR7 alone also upregulated AP-1 binding activity, on the contrary to NF-kappa B, co-transfecting B7-1 and B7-2 decreased AP-1 binding activity within 24 hours. But during 48 - 72 hours, B7 also prolonged the AP-1 binding activity. It is concluded that after MLR, B 7 increased IL-2 secretion by decreasing the degradation of IL-2 mRNA and upregulating IL-2 transcription factors. B7 also induced several kinds of cytokines secretion. Effects of B7-1 and B7-2 had no significant difference on transcription factors. It is suggested that the different functions between B7-1 and B7-2 maybe related to the difference of cell expression and kinetics. To study the molecular mechanism of B7 mediated T cell immune tolerance can help us to design a better clinic schema to prevent transplantation rejection and GVHD.

[Mesenchymal stem cells and related factors].

Mesenchymal stem cells possess the ability to differentiate into osteoblasts, chondroblasts, lipoblasts, myoblasts and so on, which can be used in the formation of hematopoietic microenvironment, tissue repairing and gene therapy. Growth factors such as TGF-beta, IGF-I, BMP and FGF can influence on the differentiation of MSC and they cooperate with each other. MSCs support hematopoiesis by secreting cytokines including G-CSF, SCF, LIF, M-CSF, IL-6, IL-11 and are related to some diseases. MSC would demonstrate important effect on gene engineering.

[Effect of triadimefon on activity of peroxidase and soluble protein content in detached cucumber cotyledons].

Contents of chlorophyll a, chlorophyll b, soluble protein and proline, as well as the ratio chlorophyll a/b in detached cotyledons of cucumber were distinctly increased by treating with 20 mg/L triadimefon, which suggested that triadimefon can effectively retard the senescence of detached cotyledons in cucumber. Triadimefon could inhibit the increase of peroxide (POD) activity and H2O2 content in different degree. It was found that contents of chlorophyll and protein were markedly negative correlated with the activity of POD and with the content of H2O2 respectively during the senescence of cotyledons. These results indicated triadimefon inhibit the degradation of chlorophyll and protein in cucumber cotyledons through POD- H2O2 enzyme system.
