Neuroma-in-continuity model in rabbits.

The present study presents a model for creation of neuroma-in-continuity in the rabbit, confirmed by histologic study, electrophysiology, and examination of molecular markers. Twelve New Zealand rabbits were used. The lateral fascicule of the peroneal nerve with 15-mm in length was resected. In the intervals from 4 to 6 weeks postoperatively, the histology showed the typical pathologic changes of neuroma by hematoxylin-eosin (HE), Luxol fast blue, and Van-Gieson staining. As compared with the healthy nerve, the motor nerve conduction velocity (MCV) and compound motor action potential (CMAP) were found significantly slowed and reduced. The expression of ciliary neurotrophic factor (CNTF) in nerve and calcitonin gene-related peptide (CGRP) mRNA in L7, S1 dorsal root ganglia were downgraded and upgraded, respectively, with formation of neuroma-in-continuity. These electrophysiologic and molecular markers' expression data can be used as reliable parameters for evaluation of management of the neuroma-in-continuity.

Transcytosis of Human immunodeficiency virus 1 across the placenta is enhanced by treatment with tumour necrosis factor alpha.

The human placenta is relatively resistant to Human immunodeficiency virus 1 (HIV-1), but obstetric complications associated with inflammatory processes, including chorioamnionitis and spontaneous preterm delivery, are associated with increased rates of vertical transmission. It was hypothesized that the pro-inflammatory mediator tumour necrosis factor alpha (TNF-alpha), which promotes HIV-1 transmission across endothelial membranes, increases HIV-1 transmission across the placenta. Flow cytometry and immunostaining studies were performed, which demonstrated that the HIV-1 receptors CD4, CCR5 and CXCR4 were not expressed by villous trophoblast cells. Consequently, primary villous trophoblast cells were not infected with cell-free HIV-1 isolates, as measured by in situ PCR and quantitative PCR, but villous trophoblast cells were infected by HIV-1-infected peripheral blood mononuclear cells (PBMC). HIV-1 from infected PBMC was rapidly transported across confluent transformed trophoblast cell monolayers by transcytosis, and TNF-alpha significantly upregulated transcytosis of HIV-1 across the trophoblast layer without disrupting cell viability or confluency. Inhibitors of TNF-alpha (antibodies against TNF-alpha and TNF-alpha receptors) and an anti-inflammatory drug (tenidap) significantly reduced transcytosis rates. It was concluded that the villous trophoblast is resistant to infection by cell-free HIV-1 but susceptible to transcytosis of HIV-1 from infected PBMC, and inflammatory mediators such as TNF-alpha may play a critical role in promoting maternal-fetal transmission of HIV-1.

A discriminative learning framework with pairwise constraints for video object classification.

To deal with the problem of insufficient labeled data in video object classification, one solution is to utilize additional pairwise constraints that indicate the relationship between two examples, i.e., whether these examples belong to the same class or not. In this paper, we propose a discriminative learning approach which can incorporate pairwise constraints into a conventional margin-based learning framework. Different from previous work that usually attempts to learn better distance metrics or estimate the underlying data distribution, the proposed approach can directly model the decision boundary and, thus, require fewer model assumptions. Moreover, the proposed approach can handle both labeled data and pairwise constraints in a unified framework. In this work, we investigate two families of pairwise loss functions, namely, convex and nonconvex pairwise loss functions, and then derive three pairwise learning algorithms by plugging in the hinge loss and the logistic loss functions. The proposed learning algorithms were evaluated using a people identification task on two surveillance video data sets. The experiments demonstrated that the proposed pairwise learning algorithms considerably outperform the baseline classifiers using only labeled data and two other pairwise learning algorithms with the same amount of pairwise constraints.

Determination of structure and transcriptional regulation of CYSLTR1 and an association study with asthma and rhinitis.

Pharmacologic studies have revealed that cysteinyl leukotrienes (CYSLTs) act through two receptors, cysteinyl leukotriene receptor 1 (CYSLTR1) and CYSLTR2. CYSLTR1 antagonists are widely used to treat asthma and rhinitis. In this study, we characterized the genomic structure and transcriptional regulation of CYSLTR1 and examined associations between CYSLTR1 polymorphisms and asthma/rhinitis. The experiment of rapid amplification of cDNA end revealed that CYSLTR1 contains three exons and that the entire open reading frame is located in exon 3. Reverse transcriptase-polymerase chain reaction showed that there were multiple splice variants of CYSLTR1 and that the transcript expression patterns differed from tissues and cell types. The promoter region of CYSLTR1 is from -665 to -30 bp relative to the transcription start site. We identified four polymorphisms (c.-618-434T/C, c.-618-275C/A, c.-618-136G/A, and 927C/T), and transmission disequilibrium tests revealed that none of these polymorphisms was associated with the development of asthma/rhinitis. However, the TCG and CAA haplotypes in the promoter region caused different transcriptional activity. Our findings indicate that CYSLTR1 polymorphisms are not likely to be involved in the development of asthma/rhinitis, but it is possible that these polymorphisms could influence drug responses in individuals with atopic diseases.

Design, synthesis and antitumor evaluation of a new series of N-substituted-thiourea derivatives.

AIM: To design and synthesize a novel class of protein tyrosine kinase inhibitors, featuring the N-(2-oxo-1,2-dihydroquinolin-3-yl-methyl)-thiourea framework. METHODS: First, compounds 1 and 2 were identified using the virtual screening approach in conjunction with binding assay based on surface plasmon resonance. Subsequently, 3 regions of compounds 1 and 2 were selected for chemical modification. All compounds were characterized potent inhibitory activities toward the human lung adenocarcinoma cell line SPAC1. RESULTS: Forty new compounds (1-2, 3a-g, 4a-w, and 5a-l) were designed, synthesized and bioassayed. Six compounds (1, 3e, 4l, 4w, 5a, and 5b) were found to show promising inhibitory activity against the SPAC1 tumor cell line. The inhibitory activity of compound 5a increases approximately 10 times more than that of the original compound 1. CONCLUSION: This study provides a promising new template with potential antitumor activity.

Construction and activity assay of the activating transcription factor 3 reporter vector pATF/CRE-luc.

Activating transcription factor 3 (ATF3), a member of the activating transcription factor/cAMP responsive element binding protein (ATF/CREB) family of transcription factors, is induced by many physiological stresses. To investigate the activity of ATF/CREB in cells with physiological stresses, we developed a practical reporter vector, the plasmid pATF/CRE-luc, bearing activating transcription factor/cAMP responsive element (ATF/CRE) binding sites. This plasmid was constructed by inserting three repeats of the ATF/CRE binding element into the plasmid pG5luc, replacing the GAL-4 binding sites. The plasmids pACT/ATF3 and pATF/CRE-luc were transfected into HeLa and NIH3T3 cells, respectively, and the results showed that the expression of luciferase was increased in a dose-dependent manner on plasmid pACT/ATF3. The data suggested that the plasmid pATF/CRE-luc could be used as a sensitive and convenient reporter system of ATF3 activity.

Variation in chemical composition and antibacterial activities of essential oils from two species of Houttuynia THUNB.

Houttuynia THUNB. (Saururaceae) has been used for dozens of years in China for the treatment of cough, leucorrhea and ureteritis. The essential oils from the two species: Houttuynia emeiensis and Houttuynia cordata sold in China under one trade name 'Yuxingcao', obtained by hydrodistillation, were analyzed by GC-MS. The results show that fifty-five components were identified and methyl nonyl ketone (2.10-40.36%), bornyl acetate (0.4-8.61%) and beta-myrcene (2.58-18.47%) were the most abundant components in oil, but the percentage of most of compounds in different species and parts varied greatly. The two fold broth dilution and agar dilution method were used to study essential oil of two Houttuynia THUNB. species for their antibacterial properties against microorganisms, Staphylococcus aureus and Sarcina ureae. The two fold dilution method was allowed to determine the minimum inhibitory concentration (MIC) of essential oil from different parts and species. Results showed that all essential oils possessed antibacterial effect, with MIC values in the range of 0.0625 x 10(-3) to 4.0 x 10(-3) ml/ml. However, essential oil from different parts and species differed clearly in their antibacterial activities. The essential oil from the aboveground part of the cultivated Houttuynia emeiensis exhibited higher activity than both parts of the wild and cultivated Houttuynia cordata when used on Staphylococcus aureus (MIC = 0.25 x 10(-3) ml/ml) and Sarcina ureae (MIC = 0.0625 x 10(-3) ml/ml), and had the same activity as the positive control ampicillin sodium.

Apolipoprotein E polymorphism influences lipid phenotypes in Chinese families with familial combined hyperlipidemia.

BACKGROUND: Apolipoprotein E (apoE) polymorphism is associated with changes in the lipoprotein profile of individuals with familial combined hyperlipidemia (FCHL), but its effects on the lipoprotein profiles of members of Chinese families with FCHL remain uncertain. METHODS AND RESULTS: 43 FCHL families (n=449) and 9 normolipidemic families (n=73) were recruited to assess the influence of apoE polymorphism on plasma lipids. The relative frequency of the epsilon4 allele in affected and unaffected FCHL relatives, spouses and normolipidemic members was 13.8%, 5.3%, 9.1% and 6.8%, respectively, with a significantly higher frequency in affected FCHL relatives, compared with unaffected FCHL relatives or normolipidemic members (p=0.0002 or p=0.029). In FCHL relatives, the apoE4 subset (E4/4 and E4/3) exhibited significantly higher levels of apoB, total cholesterol and low-density lipoprotein-cholesterol (LDL-C) than did the apoE3 (E3/3) subset, especially in women (all p<0.05), and there was significant elevation of LDL-C concentrations in men only (p<0.05). In men, the apoE2 (E3/2) subset indicated a decreased level of apoB and increased apoA1 compared with those in the apoE3 subset (p<0.05). CONCLUSIONS: ApoE polymorphism appears to be associated with variance of the lipoprotein phenotype in Chinese families with FCHL.

Positive pressure ventilation treatment reduces plasma levels of amino terminal-pro brain natriuretic peptide in congestive heart failure patients with sleep apnea.

BACKGROUND: The purpose of the present study was to assess the short-term effects of positive pressure ventilation (PPV) on plasma amino terminal-pro brain natriuretic peptide (NT-proBNP) levels in patients with congestive heart failure (CHF) and sleep apnea (SA). METHODS AND RESULTS: Polysomnography was performed in 105 CHF patients. Twenty-six patients with CHF and SA were randomly assigned to a control group or a PPV group. Patients in the higher New York Heart Association classes had higher plasma levels of NT-proBNP. The plasma NT-proBNP concentration in the PPV group decreased. CONCLUSION: Plasma NT-proBNP levels reflect the severity of CHF, and PPV can decrease plasma NT-proBNP levels in patients with CHF and SA.

Mice lacking cathepsin K maintain bone remodeling but develop bone fragility despite high bone mass.

UNLABELLED: Bone microstructural and biomechanical properties were analyzed in mice genetically lacking cathepsin K (CatK). CatK deficiency (CatK(-/-)) produced mild osteopetrosis, elevated numbers of osteoclasts, regions of disorganized bone microstructure, and increased bone fragility, showing how chronic alteration of enzyme activity during skeletal development dramatically affects bone organization and function. INTRODUCTION: Mouse models of CatK deficiency recapitulate the osteopetrosis of human pyknodysostosis and allow study of clinically relevant issues: how inhibition of this enzyme activity affects bone integrity structurally and biomechanically. To address these questions, we generated CatK-deficient mice by targeted disruption of the Ctsk gene and compared their bone structural and mechanical properties with wildtype (WT) controls. MATERIALS AND METHODS: Standard histomorphometric and biomechanical analyses were performed on femora from C57BL/6J male and female CatK(-/-), CatK(+/-), and WT mice. RESULTS: CatK(-/-) femora exhibited the mild metaphyseal osteopetrosis, a greater cortical bone area and thickness, normal bone strength, but a high degree of brittleness (nearly 50-70% decrease in postyield displacement versus WT) and a 30-40% reduction in the work-to-failure. In cancellous bone, osteoclast numbers and resorption surface were increased markedly (approximately 150% and 50%, respectively), despite the overall decrease in net bone resorption for CatK-deficient mice. Bone formation indices were altered in CatK(-/-) mice as well, with significant increases in mineral appositional rate, but not in bone formation surface; these data suggest difference in osteoblast work but not in their recruitment in CatK deficiency. CatK-deficient cortical bones had large areas of woven bone and intracortical resorption spaces within the disorganized tissue. Bone phenotype in CatK(-/-) was similar in males and females. CONCLUSIONS: Genetic CatK deficiency in mice results not only in the impairment of osteoclast function and osteopetrosis, but also altered osteoblast function, defective tissue organization, and very brittle bones. Whether this bone fragility in CatK deficiency results entirely from indirect effects of suppressed bone turnover because of impaired osteoclast function or perhaps represents a previously unappreciated more direct role for CatK in bone formation remains to be established.

Widespread and stable pancreatic gene transfer by adeno-associated virus vectors via different routes.

Diabetes is a disease of epidemic proportions and is on the rise worldwide. Gene therapy has been actively pursued but limited by technical hurdles and profound inefficiency of direct gene transfer to the pancreas in vivo. Here, we show that, for the first time, appropriate serotypes of adeno-associated virus (AAV), coupled with a double-stranded vector DNA cassette, enable extensive and long-term in vivo gene transfer in the adult mouse pancreas by three different delivery methods. Intraperitoneal and intravenous delivery of AAV8 effectively transduced exocrine acinar cells as well as endocrine beta-cells, while local pancreatic intraductal delivery of AAV6 showed the best efficiency in the beta-cells among all AAV serotypes tested in this study. Nearly the entire islet population showed gene transfer but with distinct gene transfer efficiency and patterns when different delivery methods and vectors were used. Importantly, localized gene delivery coupled with an insulin promoter allowed extensive yet specific gene expression in the beta-cells. These effective new methods should provide useful tools to study diabetes pathogenesis and gene therapy.

Adeno-associated virus-2 (AAV-2) causes trophoblast dysfunction, and placental AAV-2 infection is associated with preeclampsia.

Shallow invasion by extravillous trophoblast cells into the uterine wall reduces placental perfusion and causes placental dysfunction, but the one or more causes of shallow placental invasion are unknown. We hypothesized that infection with adeno-associated virus-2 (AAV-2) inhibits trophoblast invasion and is associated with preeclampsia, which is a common obstetric complication resulting from placental dysfunction. We determined that transformed extravillous trophoblast (HTR-8/SVneo) cells were susceptible to AAV-2 infection in the presence or absence of adenovirus, which provides helper function for AAV-2 replication, and that AAV-2 infection reduced invasion of HTR-8/SVneo cells through an extracellular matrix before cytopathic effects were detected. In a case-control study, AAV-2 DNA was found more frequently in trophoblast cells from cases of severe preeclampsia (22/40) than from normal term deliveries (5/27, P = 0.002). These results indicate that AAV-2 infection is a previously unidentified cause of placental dysfunction. Additional studies to determine the susceptibility of extravillous trophoblast to other viruses, and the mechanisms by which viral infection impairs placental function, are warranted.

[Quantitation of antigen specific T lymphocytes in peripheral blood of patients with primary biliary cirrhosis using tetramer technology].

AIM: To quantitate antigen specific T lymphocytes in peripheral blood from patients with primary biliary cirrhosis(PBC) and study the role of antigen specific T lymphocytes in the development of PBC. METHODS: Using tetramers and CD8 monoclonal antibody staining, PDC-E2 159-167aa and PDC-E2 165-174aa specific CD8(+) T lymphocytes were determined respectively in the peptide-induced cytotoxic T cell lines prepared from peripheral blood mononuclear cells(PBMC) of 15 PBC patients. The frequencies of these two kinds of antigen specific T lymphocytes in HLA-A*0201 positive (A2(+)) PBC were compared with those in A2(-) PBC patients, patients with other A2(+) chronic liver diseases and healthy controls. RESULTS: PDC-E2 159-167aa/HLA-A*0201 and PDC-E2 165-174aa/HLA-A*0201 tetramer positive CD8(+) T lymphocytes were detected in all of A2(+) PBC patients with average percentages of 0.42%+/-0.24% (0.17%-1.08%) and 0.27%+/-0.17% (0.05%-0.56%), respectively. The frequencies of the two kinds of antigen specific CD8(+) T lymphocytes from peripheral blood were significantly higher in earlier stages I and II of PBC as compared with stage III (P<0.001), while no difference was found between PDC-E2 159-167aa and PDC-E2 165-174aa specific CD8(+) T lymphocytes at the same stages. In addition, there existed no statistical difference between frequencies of antigen specific T lymphocytes in AMA or anti-PDC positive and negative PBC patients (P>0.05). CONCLUSION: This study suggests that HLA-A*0201 restricted PDC-E2 165-174aa and PDC-E2 159-167aa specific CTL play important roles in the development of PBC, and there might be a similar mechanism of T cell-mediated damage between AMA or anti-PDC positive and negative PBC patients.

[Fusion expression, purification and bioactivity detection of human defensinalpha (HDalpha)].

AIM: To obtain recombinant human defensin alpha(HDalpha) and detect its biological activity, so as to facilitate further research. METHODS: The HDalpha gene fragment with hydroxylamine cleavage site was synthesized, and then cloned into pBV220-IL-4 vector to construct pBV220-IL-4-HDalpha. The constructed vector which was confirmed to be correct by sequencing was transformed into E.coli DH5alpha and the IL-4-HDalpha fusion protein was expressed under temperature induction. After fusion protein was cleaved to remove IL-4 by hydroxylamine, purification and renaturation was performed. HDalpha's characteristics were identified by SDS-PAGE and bioactivity detection. RESULTS: After temperature induction, the expressed fusion protein which accounted for about 20% of total bacterial protein existed mainly in the form of inclusion body. After cleaving by hydroxylamine, the purity of obtained HDalpha was about 99.8%. Bacteriostatic test and clone forming test showed that recombinant HDalpha could obviously inhibit the growth of bacteria. CONCLUSION: The recombinant expression plasmid for HDalpha gene has been constructed successfully and obtained engineering bacteria can stably express target protein. Furthermore, techniques of purification and renaturation was set out, which lays the foundation for further functional study and application of HDalpha.

[Relationship between the expression of fragile histidine (FHIT) and the development of vulvar carcinoma].

OBJECTIVE: To explore the relationship between the reduction of FHIT expression of fragile histidine (FHIT) and the development of vulvar carcinoma. METHODS: The expression of the FHIT product was detected by immunochemistry in the tissue samples of 20 normal vulvas, 22 vulvar intraepithelial neoplasias (VINs), and 60 primary vulvar carcinomas. RESULTS: The expressive rates of FHIT protein in the squamous epithelium of normal vulvas, VIN I - II, VIN II, and noninvasive and invasive vulvar carcinoma were 100% (20/20), 72.7% (8/11), 45.5% (5/11), and 21.7% (13/60) respectively (P<0.05). The expressive rates of FHIT protein in the well differentiated, intermediately differentiated and poorly differentiated invasive vulav carcinoma were 60.0% (9/15), 20.0% (3/15), and 3.3% (1/30) respectively (P<0.05). The expressive rate of the impaired FHIT protein in the invasive vulva carcinoma with lymphnode metastasis (10%) was lower than that without lymphnode metastasis (27.5%). CONCLUSION: Abnormal FHIT expression may play an important role in the progression of vulvar carcinoma. The expression of FHIT may provide important information for the prognosis of vulva carcinoma.

[Mitotic arrest of gastric cancer cells induced by silencing of STK15 gene].

OBJECTIVE: To investigate the role of STK15 in regulating mitosis of gastric cancer cells (MKN45) by gene silencing through RNA interference mechanism. METHODS: RNA interference technique was used to inhibit STK15 expression in MKN45 cells. The expression levels of STK15 mRNA and protein were measured by real-time quantitative RT-PCR and Western blot respectively and cell morphological changes were investigated by reverse microscopy. In addition, cell cycle distribution and cellular proliferation were determined by flow-cytometry and MTT assay respectively. Finally, the mitotic phenotype of MKN45 cells was studied by immunofluorescence staining and confocal microscopy. RESULTS: Silencing of STK15 gene by RNA interference was confirmed by marked decrease of STK15 mRNA and protein levels in the treated MKN45 cells. This silencing correlated with rounding of the cells, decreasing of DNA content in G(2) phase (P < 0.05) and a lowered proliferation index (P < 0.05), along with alterations of mitotic phenotype of MKN45 (P < 0.05). CONCLUSION: STK15 gene may play a key role in regulating cellular mitosis and its inhibition by RNA interference leading to mitosis arrest in MKN45 cells.

[Construction of Tet-on inducible CXCR1 eukaryotic expression plasmid and identification of the expression character in NIH3T3 cells].

AIM: To construct tetracycline (Tet)-controlled inducible vector CXCR1-pTREhyg, and then detect the expression character of CXCR1 under the regulation of Dox in NIH3T3 cells. METHODS: A full length cDNA of human CXCR1 was cloned from sample of fibroblast like synovium (FLS) in rheumatoid arthritis (RA) patient by RT-PCR and then sub-cloned into the pTREhyg plasmid after sequence analysis. CXCR1-pTREhyg and pTet-on was co-transfected with Lipofect2000 to NIH3T3 cells, and the expression of IL-8RA was detected by Western blot after given different concentration of Dox. RESULTS: Western blot showed that CXCR1 could be induced by Dox in NIH3T3 cells, and the phosphorylated Erk-1/2 level was significantly increased after IL-8 stimulation. CONCLUSION: Tet inducible recombinant vector of CXCR1-pTREhyg was successfully constructed, and it could be expressed in NIH3T3 cells. The stimulation of IL-8 obviously changed the activity of Erk-1/2 in the transfected NIH3T3 cells. This work has laid foundations for further study on the relationship between CXCR1 and RA disease.
