Cisplatin delivery by biodegradable polymer implant is superior to systemic delivery by osmotic pump or i.p. injection in tumor-bearing mice.

The use of biodegradable polymer implants to deliver cisplatin was compared with delivery by systemic injection and by osmotic pump. Drug levels in the tumor were found to be higher than those in the blood and kidney when the drug was delivered using the polymer implant. In contrast, for the other two delivery methods blood and kidney cisplatin levels were greater than those in the tumor. It has been previously shown that tumor response, in terms of growth delay, was greatest when drug was delivered by polymer implant and least when treatment was by osmotic pump.

Prolonged therapeutic window for ischemic brain damage caused by delayed caspase activation.

Apoptotic cell death is prominent in neurodegenerative disorders, such as Alzheimer's disease and Huntington's disease, and is found in cerebral ischemia. Using a murine model of delayed cell death, we determined that cleavage of zDEVD-amino-4-trifluoromethyl coumarin (zDEVD-afc) in brain homogenate, a measure of caspase activation, increased initially 9 hours after brief (30 minutes) middle cerebral artery occlusion along with caspase-3p20 immunoreactive cleavage product as determined by immunoblotting. zDEVD-afc cleavage activity was blocked by pretreatment or posttreatment with the caspase-inhibitor N-benzyloxycarbonyl-Asp(OMe)-Glu(OMe)-Val-Asp(OMe)-fluoromethyl-ketone (zDEVD-fmk), and ischemic damage was reduced when the drug was injected up to 9 hours after reperfusion. The protection was long lasting (21 days). Hence, the period before caspase activation defined the therapeutic opportunity for this neuroprotective agent after mild ischemic brain injury. Prolonged protection after caspase inhibition plus the extended treatment window may be especially relevant to the treatment of neurodegenerative disorders.

Prevention of experimental autoimmune neuritis by nasal administration of P2 protein peptide 57-81.

Experimental autoimmune neuritis (EAN) is a CD4+ T cell-mediated inflammatory demyelinating disease of the peripheral nervous system (PNS) that serves as a model for Guillain-Barre syndrome (GBS) in humans. Both EAN and GBS are associated with upregulated T and B cells responses to PNS myelin proteins including P2 protein, and by changes of the Th1/Th2 cell balance in favor of Th1. Here we report that EAN can be prevented by the dominant neuritogenic peptide 57-81 of the PNS P2 protein when given nasally before immunization of Lewis rats with bovine PNS myelin (BPM) + Freund's complete adjuvant (FCA). P2 peptide-tolerized rats were also resistant to EAN relapse after challenge with BPM. Tolerance to EAN in rats receiving high dose (60 microg/day/rat) P2 peptide nasally was associated with specific T and B cell anergy. This was characterized by the failure of T cells to proliferate in response to PNS myelin antigens, while responsiveness to phytohemagglutinin was retained. Numbers of BPM- and P2 peptide-reactive interferon-gamma mRNA expressing lymph node cells were reduced, while levels of P2 peptide-reactive interleukin 4 and transforming growth factor-beta mRNA-expressing cells were markedly upregulated on day 18 post immunization in the rats receiving high dose P2 peptide nasally. Tolerance to EAN was also associated with lower CD4+ cell infiltration, low-grade inflammation, or the absence of histological evidence of EAN, as well as with low IL-2 receptor and MHC class II molecule expression within the PNS. This is the first study showing that mucosal tolerance is applicable to EAN and, as an extension, could be considered in GBS.

Regulation of hepA of Anabaena sp. strain PCC 7120 by elements 5' from the gene and by hepK.

In Anabaena spp., synthesis of the heterocyst envelope polysaccharide, required if the cell is to fix dinitrogen under aerobic conditions, is dependent on the gene hepA. A transcriptional start site of hepA was localized 104 bp 5' from its translational initiation codon. A 765-bp open reading frame, denoted hepC, was found farther upstream. Inactivation of hepC led to constitutive expression of hepA and prevented the synthesis of heterocyst envelope polysaccharide. However, the glycolipid layer of the heterocyst envelope was synthesized. A hepK mutation blocked both the synthesis of the heterocyst envelope polysaccharide and induction of hepA. The predicted product of hepK resembles a sensory protein-histidine kinase of a two-component regulatory system. Analysis of the region between hepC and hepA indicated that DNA sequences required for the induction of hepA upon nitrogen deprivation are present between bp -574 and -440 and between bp -340 and -169 relative to the transcriptional start site of hepA. Gel mobility shift assays provided evidence that one or more proteins bind specifically to the latter sequence. The Fox box sequence downstream from hepA appeared inessential for the induction of hepA.

Analogs of the autoinducer 3-oxooctanoyl-homoserine lactone strongly inhibit activity of the TraR protein of Agrobacterium tumefaciens.

The TraR and TraI proteins of Agrobacterium tumefaciens mediate cell-density-dependent expression of the Ti plasmid tra regulon. TraI synthesizes the autoinducer pheromone N-(3-oxooctanoyl)-L-homoserine lactone (3-oxo-C8-HSL), while TraR is an 3-oxo-C8-HSL-responsive transcriptional activator. We have compared the abilities of 3-oxo-C8-HSL and 32 related compounds to activate expression of a TraR-regulated promoter. In a strain that expresses wild-type levels of TraR, only 3-oxo-C8-HSL was strongly stimulatory, four compounds were detectably active only at high concentrations, and the remaining 28 compounds were inactive. Furthermore, many of these compounds were potent antagonists. In contrast, almost all of these compounds were stimulatory in a congenic strain that overexpresses TraR and no compound was a potent antagonist. We propose a model in which autoinducers enhance the affinity of TraR either for other TraR monomers or for DNA binding sites and that overexpression of TraR potentiates this interaction by mass action. Wild-type A. tumefaciens released a rather broad spectrum of autoinducers, including several that antagonize induction of a wild-type strain. However, under all conditions tested, 3-oxo-C8-HSL was more abundant than any other analog, indicating that other released autoinducers do not interfere with tra gene induction. We conclude that (i) in wild-type strains, only 3-oxo-C8-HSL significantly stimulates tra gene expression, while many autoinducer analogs are potent antagonists; (ii) TraR overexpression increases agonistic activity of autoinducer analogs, allowing sensitive biodetection of many autoinducers; and (iii) autoinducer stimulatory activity is potentiated by TraR overproduction, suggesting that autoinducers may shift an equilibrium between TraR monomers and dimers or oligomers. When autoinducer specificities of other quorum-sensing proteins are tested, care should be taken not to overexpress those proteins.

TNF-alpha modulates expression of the tissue transglutaminase gene in liver cells.

One of several postulated roles for tissue transglutaminase (tTG) is the stabilization and assembly of extracellular matrix via peptide cross-linking. We previously determined that tTG activity increased in an animal model of hepatic fibrogenesis and in human liver disease. To further study the role of tTG in liver disease, we initiated investigations into the effect of a proinflammatory mediator, tumor necrosis factor (TNF)-alpha, on tTG activity in cultured liver cells. Treatment of human Hep G2 cells with 1 ng/ml TNF-alpha increased [14C]putrescine cross-linking to cellular proteins. An increase in tTG mRNA content was observed 1 h after addition of TNF-alpha, and levels of tTG mRNA remained elevated after 24 h. Hep G2 cells, transiently transfected with a luciferase reporter containing 1.67 kb of the human tTG promoter, showed an increase in reporter activity after addition of TNF-alpha. Gel shift experiments using nuclear extracts from TNF-alpha-treated cells and oligonucleotides containing the tTG nuclear factor (NF)-kappa B motif revealed increased binding, concordant with mRNA data. Transient transfections with a truncated reporter construct lacking the tTG NF-kappa B sequence showed an attenuated response to TNF-alpha treatment. Similar responses were seen in stably transfected HeLa cells. Primary hepatocytes isolated from a transgenic mouse line containing the mouse tTG promoter driving the beta-galactosidase reporter, show similar time-dependent increases in promoter activity when treated with TNF-alpha. Furthermore, Hep G2 cells are incapable of upmodulating tTG promoter reporter activity in the presence of TNF-alpha when those cells overexpress a transdominant, negative mutant NF-kappa B subunit. Because TNF-alpha expression is upregulated in hepatic inflammation, the data suggest TNF-alpha-mediated increases in tTG expression may play an important role in the process of hepatic fibrogenesis.

Ischemic preconditioning: antiarrhythmic effects and electrophysiological mechanisms in isolated ventricle.

The objective of this study was to identify cellular electrophysiological mechanisms by which ischemic preconditioning decreases arrhythmias in an isolated ventricular tissue model of ischemia and reperfusion. Electrical activity was recorded with microelectrodes from endocardium and epicardium of paced guinea pig right ventricular free walls. Control preparations were exposed for 15 min to Tyrode solution modified to simulate selected ischemic conditions and then were reperfused for 30 min with normal solution. Preconditioned tissues were exposed to a 2- or 5-min period of simulated ischemia before this same protocol. Neither preconditioning protocol affected incidence of ventricular tachycardia (VT) in ischemia; however, the 5-min protocol significantly decreased premature beats (PVB) and transmural conduction block. Preconditioning for 5 min, but not 2 min, significantly decreased reperfusion-induced VT and PVB. Ischemic preconditioning did not change effects of ischemia or reperfusion on action potential duration, effective refractory period, or endocardial conduction time. However, preconditioning markedly attenuated depression of transmural conduction by ischemia and early reperfusion and thereby prevented conduction delays necessary for transmural reentry.

Epithelial rests colocalize with cementoblasts forming acellular cementum but not with cementoblasts forming cellular cementum.

Epithelial rests of Malassez and cementoblasts were examined in the rat molars during the early stages of root formation using an antilaminin antibody and/or peanut agglutinin (PNA), and an antiosteocalcin (OC) antibody, respectively. The roots of the first molars were used for study. The antilaminin antibody stained the basement membrane surrounding the epithelial root sheath and epithelial rests. The basement membrane of the epithelial root sheath was continuous, but that of the epithelial rests was discontinuous. The cells of epithelial rests and epithelial root sheath were positive for PNA. The structural characteristics of the epithelial rests were seen in the sections stained doubly with PNA and the antilaminin antibody. The cells of epithelial rests were fibroblast-like and formed a fine mesh in 2-week-old rats. In 3-week-old rats, the epithelial rests were also present at the coronal half of root surface, showing typical cell cords, but were not present at the apical part of the root surface where the cellular cementum covered the root dentin. At the root apex of 3-week-old rats, the cells of epithelial rests forming fine meshes were seen near the epithelial root sheath. The anti-OC antibody stained cementoblasts lining acellular and cellular cementum. The sections doubly stained with the anti-OC and the antilaminin antibodies or PNA further revealed the close relation between epithelial rests and cementoblasts. The OC-positive cells lining acellular cementum or dentin were localized very close to the epithelial rests. In contrast, the OC-positive cells lining cellular cementum did not show close association with the epithelial cells, except the cells located most apically where the basement membrane of the epithelial root sheath is disrupted and the initial cellular cementum begins to be formed. The present results suggest that the epithelial rests and/or the discontinuous basement membrane of them may have a role for the acellular cementum formation at least in the early stage of root formation.

Stress and the menstrual cycle: relevance of cycle quality in the short- and long-term response to a 5-day endotoxin challenge during the follicular phase in the rhesus monkey.

The notion that stress activates central and peripheral pathways to inhibit the menstrual cycle is well accepted, but the initial processes through which this occurs have not been investigated. This study uses a relevant nonhuman primate model to document the cyclic endocrine effects imposed by a moderate short-term stress episode in the follicular phase. The stress paradigm is a 5-day inflammatory/immune-like challenge produced by the administration of bacterial endotoxin [lipopolysaccharide (LPS)], which, through the release of endogenous cytokines and other mediators, induces a physiopathological response similar to a bacterial infection. LPS was administered iv twice daily for 5 days starting on days 2-8 of the follicular phase. The stress challenge resulted in a significant lengthening of the follicular phase in all monkeys. Two distinct groups were observed. In group 1 (n = 5), the mean (+/- SE) length of the follicular phase in the LPS-treated cycle was significantly increased, from 10.2 +/- 0.2 in control cycle 2 to 30.8 +/- 4.3 days (except in one monkey that had a 4-month amenorrheic interval). In group 2 (n = 5), the length of the follicular phase significantly increased but not to exceed the duration of the LPS treatment (9.7 +/- 1.1 vs. 13.6 +/- 1.2). Estradiol concentrations decreased significantly after LPS in group 1 (34.8 +/- 5.5 vs. 16.2 +/- 6.5 pg/mL) and remained suppressed after the challenge. In group 2, estradiol levels remained stationary throughout the 5-day LPS treatment (26.0 +/- 6.5 vs. 25.6 +/- 3.9). Compared with control values at a similar stage of the follicular phase, most LH and FSH values during LPS treatment were higher than controls. Estradiol and gonadotropin surges were delayed by LPS treatment for a varying length of time according to each grp. Significant differences in integrated luteal progesterone concentrations characterized control cycles of groups 1 and 2 (group 1: 36.5 +/- 1.5, group 2: 47.5 +/- 2.6). In group 1, there were no further effects of LPS on luteal progesterone during the treatment and two post-LPS cycles. In contrast, in group 2, integrated luteal progesterone concentrations were significantly decreased in post-LPS cycle 1 (to 36.0 +/- 4.4). Cortisol significantly increased at hour 3 after each morning LPS injection but the amplitude of the response decreased over the 5-day period. Progesterone increased significantly by hour 3 after the first LPS injection but remained unchanged after subsequent LPS administration. Our data demonstrate that a 5-day inflammatory-like episode during the follicular phase can delay folliculogenesis and that damage to this process is intensified in individuals who already demonstrate a subtle cyclic degradation, in the form of decreased progesterone secretion in the luteal phases preceding the stress episode. Long-term endocrine effects, in the form of decreased luteal secretory activity in the first poststress cycle, are observed in normally cycling individuals, suggesting that inadequacy of the luteal phase may represent the first stage in the damage that a stress episode can inflict upon the normal menstrual cycle.

In vivo color measurement of 410 maxillary anterior teeth.

OBJECTIVE: To make in vivo color measurements of maxillary anterior teeth. METHODS: A fiber-optic spectrophotometer (FMC-9204, Kunming, China) was used for in vivo color measurements of 410 healthy maxillary anterior teeth of 70 Kunming residents aged 18 to 70. The results were expressed by the CIE (Commission Internationale de L'Eclairage) L*A*B* system. RESULTS: The value of L* was positively correlated with L* = 42.74 + 0.14 x age; the value of A* was negatively correlated with age (A* = 1.73 - 0.03 x age). The color of the maxillary anterior teeth was related to tooth position: the central incisor had the highest value of L*, the canine had the lowest. The canine was also redder, yellower, and more saturated.

[Oxidative DNA damage of cartilage in osteoarthrosis].

In order to study molecular mechanism of osteoarthrosis, we used molecular techniques to detect DNA oxidative damage of cartilage. There were 10 samples from clinical operation including 5 cases of replacement of hip joint, 4 degenerative cartilage of knee osteoarthrosis, and 1 free body of knee joint. DNA from all samples was run out on an agarose gel and appeared classic nucleosomal ladders. (8-hydroxyl-2'-deoxyguanosine)/ml (8-OHdG) in DNA samples was measured by high-performance liquid chromatography with electrochemical detection (HPLC-EC). The results showed that the levels of 8-OHdG were increased to 0.11-0.26 ng.ml-1, average 0.177 ng.ml-1. Normal controls were lower than 0.05 ng.ml-1. The high 8-OHdG in DNA led to a misreading of affected templates and base-pair exchange, which could be a sensitive indicator about oxidative DNA damage, which was produced by free radicals or other DNA damage agents. The result suggests that oxidative DNA damage may be the molecular mechanism of osteoarthrosis.

[Determination of serum copper by atomic absorption spectrometry sensitized with mixed surfactants].

The effects of six kinds of surfactants were studied in the detection of copper in serum by atomic absorption spectrometry. The results demonstrated that the absorption decreased when polyvinyl alcohol was used alone or together with the other one, and the absorption increased when one or two of Tween-20, emulgent-OP, palmityl trimethyl ammonium bromide, arabicgum and sodium laurylsulfate were used. Tween-20 and sodium laurylsulfate used together are most effective. A method of determination of serum copper by atomic absorption spectrometry sensitized with mixture of Tween-20 and sodium laurylsulfate was established. The absorption increased by 50%. The relative standard deviation was 1.35%-2.11%. The recovery was 98.00%-98.13%. The detection limit was 0.0524 mumol/L.

[The effect of tamoxifen on experimental nasal hypersensitivity].

The effect of estrogen on nasal hypersensitivity was observed in female adult guinea pigs with high level of estrogen. The animal were divided into three groups: group A as normal control; in group B, tamoxifen was given before inducing high level estrogen and in group C, estrogen was given but without tamoxifen. The groups B and C were sensitized and then challenge with TDI as described previously. The result showed that tamoxifen may inhibit significantly the development of nasal hypersensitivity and reduce the pathologic changes of nasal mucosa. It is suggested that the mast cells may be the target of estrogen and tamoxifen might be useful in treatment of nasal hyperreaction concerning with the change of estrogen level.

Methylation of p16 gene in hematological malignancies.

OBJECTIVE: To investigate the incidence of p16 gene methylation in hematological malignancies, and the relations between p16 gene methylation and the prognosis of hematological malignancies. METHODS: Fifty-five patients were studied by restriction enzyme polymerase chain reaction (PCR) to detect p16 gene methylation. RESULTS: p16 gene methylation was detected in 6 of 55 patients (10.9%), who were 1 patient with M2a, 1 patient with M5a, 2 patients with chronic myelogenous leukemia (CML) in blast crisis, 1 patient with progressing multiple myeloma (MM), 1 with non-Hodgkin's lymphoma (NHL) accompanied by B-ALL, respectively. p16 gene methylation correlates with adverse prognostic features. The patients with p16 gene methylation had poor response to therapy, and died shortly after p16 gene methylation was detected. CONCLUSION: The methylation of p16 gene plays an important role in the pathogenesis and the development of some hematological malignancies. The patients with p16 gene methylation have poor prognosis. The detection of p16 gene is a useful tool for evaluating prognosis of hematological malignancies.

[Study on androgen receptor mRNA expression on laryngocarcinoma tissue].

Exploring the role of androgen receptor (AR) in development of laryngocarcinoma, expression of AR in larynx squamous cell carcinoma and normal larynx tissue were observed and analysed by Northern blot hybridization. The results showed: AR mRNA was founded in both 27 larynx squamous carcinoma and 6 normal larynx tissue beside carcinoma samples. The intensity of AR mRNA expression in carcinoma was significantly stronger than that in normal larynx tissue, and the expression of AR mRNA was in low degree in normal larynx tissue. The existence and different expression of AR mRNA in both laryngocarcinoma and normal larynx tissue suggest that AR gene can control development of laryngocarcinoma tissue.

[A preliminary study on the in vitro eukaryotic expression of human factor VIII cDNA].

OBJECTIVE: To evaluate the expression efficiency of a cDNA sequence of human clotting factor VIII (4.7 kb, B domain-deleted) in in vitro systems. METHODS: After insertion of the cDNA into several mammalian expression vectors, such as retroviral vector pMSCV, EB virus-based vector pGRE5.2/EBV and eukaryotic expression plasmid pCI, the expression of these constructs were tested in a variety of cells. RESULTS: All the three kinds of constructs-pCI-VIII, pGRE5.2/EBV-VIII and pMSCV-VIII were able to direct FVIII synthesis in NIH3T3, Hela and Bosc23 cells, respectively, while the pMSCV-VIII and pGRE5.2/EBVVIII produced relatively high levels of FVIII activity (up to 0.7 units/ml and 2.0 units/ml from 24 h to 48 h, respectively, after transfection with lipofectamine). The three forms of pMSCV-VIII vector worked in a similar efficacy in Bosc 23 cells, but this function was not detected in NIH3T3, psi-Crip and GP + E86 as well as 32DC13 cells in a transient transfection assay. Moreover, the NIH3T3 and 32DC13 cells infected with culture supernatant from pMSCV-VIII transfected-Bosc23 cells (as packaging cells) unexpectedly did not produce detectable FVIII activity. CONCLUSION: Apart from the design and construction of vectors, target cell selection may play a crucial role in the efficient expression of the FVIII cDNA.
