[The serum levels of soluble interleukin-2 receptor in patients with malignant lymphocytic proliferative disorders].

OBJECTIVE: To explore the changes of serum levels of soluble interleukin-2 receptor (sIL-2R) in lymphocytic malignancies and its clinical implication. METHODS: The serum levels of sIL-2R were measured in 60 cases of patients with various malignant lymphocytic proliferative disorders by using ELISA method. At the same time, the ratio of CD4+/CD8+ of peripheral blood mononuclear cells was also determined in 43 patients by using fluorescence activated cell sorter(FACS). RESULTS: The serum levels of sIL-2R in patients with multiple myeloma(MM), non-Hodgkin's lymphoma (NHL) or acute lymphoblastic leukemia(ALL) were significantly higher than that of normal value. The ratio of CD4+/CD8+ measured in 43 patients were markedly lower than that of normal value. In MM, NHL and ALL patients, the higher values of sIL-2R were revealed in severe cases or late stage of these diseases. The levels of sIL-2R were independant of the infection in these cases. CONCLUSION: sIL-2R levels significantly elevated as the disease progressing, and the immune function decreasing, and the sIL-2R level may be used as an effective clinical marker.

Glycophorin variants and contents of sialic acid and total sulfhydryl groups on erythrocyte membranes of residents in a malaria hyperendemic area.

OBJECTIVE: To conduct a screening survey of glycophorin (GP) variants and observe the content changes of sialic acid (SA) and total sulfhydryl (SH) groups on the erythrocyte membranes among residents in a tertian malaria hyperendemic area of Guizhou Province. METHODS: GP variants were detected in the erythrocyte hemolysates of 173 local residents at two villages of Libo County by SDS-PAGE on 10% to 15% gradients gel and Western immunoblotting. Their SA and total SH group contents were estimated in erythrocyte membranes by spectrophotometric methods. 114 healthy subjects in Changsha and 49 individuals at a neighbouring village of the above area showing low morbidity of malaria served as normal and endemic controls respectively. RESULTS: Three distinct types of GP variants were found among 19 propositi in this hyperendemic area. The incidence of GP variants was 7.9% (8/101) at Yaolu Village whose population was mainly composed of Yao ethnic group; while that of Buyi ethnic group at Maolan Village was higher (15.3%; 11/72). The erythrocyte membrane contents of SA in residents at both villages exhibited a very significant tendency of decline (P < 0.01), whereas those of total SH groups increased prominently in residents of Yaolu Village only (P < 0.05). CONCLUSIONS: The frequency of GP variants in this hyperendemic area does not depend upon the severity of malarial prevalence. The evident reduction of SA contents in the residents may be related to the breaking down of the SA residues on membrane GPs by the invasion of Plasmodium vivax.

[Effect of inhaled-corticosteroids on the mRNA expressions of endothelin-1 and endothelin converting enzyme in bronchial asthma].

OBJECTIVE: To investigate the expressions of endothelin-1(ET-1) and endothelin converting enzyme (ECE) mRNA in bronchial mucosal biopsis from asthmatics, and the effect of inhaled-corticosteroids on the expressions of ET-1 and ECE genes. METHOD: The expressions of ET-1 and ECEmRNA in bronchial mucosal tissue were evaluated by reverse transcription DNA polymerase chain reaction (RT-PCR); With fibro bronchoscopy, the naked-eye inflammatory severity and inflammatory score of bronchial mucoses were recorded. RESULT: (1) The expression level of ET-1 mRNA was 0.86 +/- 0.06 in non-steroids-treated group and 0.14 +/- 0.06 in control non-asthmatic group. The expression of ECE mRNA in non-steroid group was 0.31 +/- 0.04, and 0.30 +/- 0.05 in control group, P = 0.238. (2) The ET-1 and ECE mRNA expressions were 0.22 +/- 0.01 and 0.16 +/- 0.01 respectively in steroid treated group. (3) The naked-eye bronchial mucosal inflammatory score was 6.0 +/- 1.9 in non-steroids treated group, moreover, the expression of ET-1 mRNA was positively correlated with the score (r = 0.78, P < 0.05). CONCLUSION: There was markedly higher ET-1 mRNA expression in the bronchial mucosa in asthmatics, but the ECE mRNA expression was unchanged significantly; Inhaled corticosteroids obviously inhibited both ET-1 mRNA and ECE mRNA expressions in bronchial mucosa, which may be one of the anti-inflammatory mechanisms of inhaled-corticosteroid in management of bronchial asthma.

[Detecting coronary artery disease with dobutamine 99mTc-MIBI myocardial perfusion tomography].

OBJECTIVE: To assess the safety and accuracy of dobutamine stress 99mTc-MIBI perfusion tomography (DST) for detecting coronary artery disease(CAD). METHODS: Seventy nine patients with suspected CAD without previous myocardial infarction underwent DST. Forty eight of the 79 patients had coronary angiography and CAD was defined as greater than 50% reduction in diameter. RESULTS: Seventeen of 20 patients with CAD demonstrated perfusion abnormalities by DST (sensitivity 85%), 20 of 24 with normal or non-significant stenosed coronary arteries had normal scan (specificity 83.3%). All 4 patients with coronary ectasia showed abnormalities by DST. Fifty seven percent of the patients experienced some discomfort, but no serious side effects occurred. CONCLUSIONS: It shows that DST is a safe and accurate alternative to exercise stress perfusion study for detecting CAD in patients unable to perform exercise testing.

[Studies on the morphology of fruit-body developing of Polyporus umbellatus].

OBJECTIVE: Studying the fruit-body development of polyporus umbellatus to reveal the life-cycle of this medicinal fungus. METHODS: Parts of materials treated by paraffin section, and examined under a light microscope; parts of materials by ultrathin section and observation under transmission electron microscope (TEM). RESULTS: The fruit-body consists of trimitic hyphae, including generative hyphae, skeletal hyphae and binding hyphae. Generative hyphae are characterized by thin walls, dense cytoplasmic contents. Skeletal hyphae are unbranched, thick-walled, and of a narrow lumen. Binding hyphae are much-branched, tending to weave themselves among the other hyphase of flesh. The morphology of basidiospore development was also discussed in this paper. CONCLUSIONS: The trimitic hyphae are the main component parts in the different developing stages of fruit-body of polyporus umbellatus.

[The changes of EG1 and EG2 positive eosinophils and their clinical significance in asthmatics].

OBJECTIVE: To evaluate the significance of EG1 or EG2 positive eosinophils in relating to the severity of asthma. METHOD: EDTA-anticoagulated venous blood were treated by FOG method, and FTTC-conjugated monoclonal antibodies were added. Flow cytometry technique was used. RESULT: EG1 and EG2 positive eosinophils increased significantly in patients with acute asthma (n = 29) as compared with control (n = 13) and COPD (n = 12), P < 0.05. There were significant correlation of the mean fluorescence intensity(MFI) between EG1, EG2 and the degree of the symptom. CONCLUSION: There is a significant increase of activated eosinophils in peripheral blood in asthmatics, the expression of EG1 and EG2 positive eosinophils correlated well with the severity of symptom in asthmatics. EG1 and EG2, as the marker for eosinophils activity, may be sensitive and specific in evaluating the severity of asthmatics symptom.

[Study on relativity between oxygen free radical and thromboxane B2, 6-keto-PGF1 alpha during ligustrazine treatment of intrauterine growth retardation].

OBJECTIVE: To study the pathophysiological changes and effective treatment of intrauterine growth retardation (IUGR). METHODS: Eighty-five cases with normal pregnancy and 58 IUGR women maternal red blood cell superoxide dismutase (SOD), glutathione peroxidase (GSH-Px), serum lipid peroxide (LPO) and thromboxane B2 (TXB2), 6-keto-PGF1 alpha(6KP) were examined. IUGR group was divided into Ligustrazine group (LIG, 47 cases) and amino acid group (AAG, 11 cases) and treated accordingly. RESULTS: In IUGR group, LPO increased and SOD, GSH-Px activity, 6KP value decreased abnormally, TXB2/6KP ratio significantly increased, which was negative correlated with SOD, GSH-Px activity and positive correlated with LPO level markedly. After treatment, all the changes were improved and almost reached normal range. The fetal outcome showed that the total effective rate was 95.7% in LIG, markedly higher than that in AAG (81.8%, P < 0.05), there was significant difference between LIG and AAG. CONCLUSIONS: The balance of body oxidate/antioxidate system was disturbed and the uteroplacental-fetal circulation was obstracted in IUGR. Ligustrazine could inhibit oxygen free radical, rise SOD, GSH-Px activity, regulate TXA2/PGI2 balance, promote fetal growth.

[Analysis on quality control of drug susceptibility testing in the surveillance of drug resistance in tuberculosis in Henan].

OBJECTIVE: To evaluate the working quality of Henan province reference laboratory(PRL) in the surveillance of drug resistance (DRS) in Henan. METHOD: West Pacific Regional (WPR) Supranation Reference Laboratory(SRL) in South Korea conducted three times of quality control of drug susceptibility testing (DST) from SRL to PRL and to national reference laboratories(NRLs) in the region at the same time, and once of which from PRL to SRL. The exchanging strains of M. tuberculosis, which were clinical isolated ones containing both drug susceptible and drug resistant cultures, were tested for resistance to SM, INH, RFP and EMB. The DST procedure was proportion method, and L-J medium was used. RESULT: It was shown that the sensitivities for most of the four drugs in the three times of DST quality control from SRL to PRL were higher than their specificities, and that the sensitivities for both INH and RFP were 100%. And the accuracy, or agreement, for both INH and RFP was more than 90%, except for that of RFP in the first time. Compared with the results from SRL to NRLs (including Henan PRL), except for the specificity for SM in WPR being higher than that in Henan (84% versus 70%, P < 0.05), there was no significant difference between Henan and WPR on the specificity and sensitivity of the other drugs. Similar characteristics can be found in the results from PRL to SRL. And the efficiency for SM, INH and RFP was all higher than 90%. CONCLUSION: The quality control on Henan PRL meet the requirement of the WHO/IUATLD guidelines for surveillance of drug resistance in tuberculosis(1997).

[Effect of Acorus tatarinowii Schott. on gastrointestinal myoelectric activity in rats].

The effect of Acorus tatarinowii on the gastrointestinal myoelectric activity and mechanism in rats was studied electrio-physiologically. The result indicates that Acorus tatarinowii helps inhibit the gastrointestinal myoelectric activity. This inhibitory effect is believed to take place through the blocking of cholinergic M and non-cholinergic M receptors, and has nothing to do with adrenergic alpha and beta receptors.
