Mesenchymal determination of mechanical strain-induced fetal lung cell proliferation.

Fetal breathing movements play an important role in normal fetal lung growth. We have previously shown that an intermittent mechanical strain regimen (60 cycles/min, 15 min/h), simulating normal fetal breathing movements, stimulated growth of mixed fetal rat lung cells in organotypic culture. In the present study, we examined the individual responses of the two major fetal lung cell types, fibroblasts and epithelial cells, to mechanical strain. Also, we investigated the effect of mesenchymal-epithelial interactions on strain-induced cell proliferation during fetal lung development. Fibroblasts and epithelial cells from day 18 to day 21 fetal rat lung (term = 22 days), cultured alone or as various recombinants, were subjected to either a 48-h static culture or to strain, and DNA synthesis was measured. Both cell types responded individually to strain with enhanced DNA synthesis throughout late fetal lung development. Independent of the recombination ratio, there was no additive response to strain when fibroblasts and epithelial cells from the same gestation were recombined. In contrast, strain-induced DNA synthesis was suppressed when cells from different gestations were recombined. The ontogenic response pattern of recombinants to mechanical strain was similar to that of fibroblasts but not of epithelial cells. Strain-induced proliferation increased and peaked at the early canalicular stage of lung development at 19 days of gestation and declined thereafter. We conclude that strain-enhanced growth of the fetal lung is gestation dependent and that the gestational response to mechanical force is regulated by the mesenchyme.

Ontogeny of extracellular matrix gene expression by rat lung cells at late fetal gestation.

Lung epithelial and mesenchymal cells are separated by a basement membrane. At late fetal gestation, this basement membrane in fenestrated, allowing epithelial cytoplasmic extensions to reach in close proximity of the interstitial fibroblast. The enzymes responsible for this focal basement membrane remodelling, and their cellular origin, remains to be defined. Basement membrane remodelling generally involves a special class of matrix-degrading enzymes, called metalloproteinases. Herein, we report that fetal lung cells originating from both tissue layers, mesoderm and endoderm, express the metalloproteinase genes, MMP-1 or interstitial collagenase, and MMP-3 or stromelysin. The inhibitor of metalloproteinases, TIMP-1, is mainly expressed in fetal lung fibroblasts. During late fetal development, MMP-1 mRNA expression in both cell types increases close to term (day 21, term = 22 days), while that of stromelysin and TIMP-1 remain constant. Both fibroblasts and epithelial cells express fibronectin (FN) mRNA. The expression of the FN gene in epithelial cells decreases slightly at the canalicular stage of lung development (days 19-20), whereas FN expression in fibroblasts is not changed with advancing gestation. Procollagen alpha 1 (I) mRNA is predominantly detected in fibroblasts whereas message for laminin B1 chain is primarily found in epithelial cells. Expression of procollagen alpha 1 (I) mRNA decreases in fibroblasts during the canalicular stage of fetal lung development compared to the pseudoglandular stage (day 18) but increases thereafter at the saccular stage (day 21) of development. Laminin B1 expression in epithelial cells declines with advancing gestation. These data are consistent with a process of basement membrane thinning during the canalicular stage, followed by metalloproteinase-mediated penetration. Further, a progressive reduction in laminin expression is consistent with progressive epithelial differentiation.

Dopamine D1 receptor and protein kinase C isoforms in spontaneously hypertensive rats.

-Dopamine, via D1-like receptors, stimulates the activity of both protein kinase A (PKA) and protein kinase C (PKC), which results in inhibition of renal sodium transport. Since D1-like receptors differentially regulate sodium transport in normotensive and hypertensive rats, they may also differentially regulate PKC expression in these rat strains. Thus, 2 different D1-like agonists (fenoldopam or SKF 38393) were infused into the renal artery of anesthetized normotensive Wistar-Kyoto rats (WKY) and spontaneously hypertensive rats (SHR) (n=5 to 6/drug/strain). Ten or 60 minutes after starting the D1-like agonist infusion, both the infused kidney and the noninfused kidney that served as control were prepared for analysis. The D1-like agonists produced a greater diuresis and natriuresis and inhibited Na+,K+-ATPase activity in proximal tubule (PT) and medullary thick ascending limb (mTAL) to a greater extent in WKY (Delta20+/-1%) than in SHR (Delta7+/-1%, P<0.001). D1-like agonists had no effect on PKC-alpha or PKC-lambda expression in either membrane or cytosol but increased PKC-theta expression in PT in both WKY and SHR at 10 minutes but not at 60 minutes. However, membranous PKC-delta expression in PT and mTAL decreased in WKY but increased in SHR with either 10 or 60 minutes of D1-like agonist infusion. D1-like agonists also decreased membranous PKC-zeta expression in PT and mTAL in WKY but increased it in PT but not in mTAL in SHR. We conclude that there is differential regulation of PKC isoform expression by D1-like agonists that inhibits membranous PKC-delta and PKC-zeta in WKY but stimulates them in SHR; this effect in SHR is similar to the stimulatory effect of norepinephrine and angiotensin II and may be a mechanism for their differential effects on sodium transport.

Elastic protein-based polymers in soft tissue augmentation and generation.

Five elastic protein-based polymers, designed as variations of polymer I, (GVGVP)251, elicited different responses when injected as subcutaneous implants in the guinea pig, a preclinical test used to evaluate materials for soft tissue augmentation and specifically for correction of urinary incontinence. All six polymers, prepared using recombinant DNA technology, expressed at good levels using transformed E. coli fermentation. These E. coli-produced polymers were purified for the first time to the exacting levels required for use as biomaterials where a large quantity could disperse into the tissues in a few days. Time periods of 2 and 4 weeks were used. Polymer I functioned as a bulking agent around which a fine fibrous capsule formed. Inclusion of (GVGVAP)8, a chemoattractant toward monocytes and elastin-synthesizing fibroblasts in the sequence of polymer I, resulted in an appropriate tissue response of invasion of macrophages. Inclusion of lysine residues, for lysyl oxidase cross-linking, suggested a possible remodeling of the implant toward fibers. Most promising however, when the cell attachment sequence, GRGDSP, was added to polymer I, the implant elicited tissue generation with a normal complement of collagen and elastic fibers, spindle-shaped histiocytes and angiogenesis. If this response is retained over time, the desired soft tissue augmentation and generation will have been achieved. Our working hypothesis is that on formation of elastin, with a half-life of the order of 70 years, a long lasting soft tissue augmentation would result rather than scar tissue as occurs with Contigen, the currently approved injectable implant for soft tissue augmentation.

Bipolar disorder and panic disorder in families: an analysis of chromosome 18 data.

OBJECTIVE: The authors performed an analysis of their published chromosome 18 linkage data on 28 families in which there was bipolar disorder to test the potential of comorbid panic disorder to define a genetic subtype of bipolar disorder. METHOD: Families ascertained through probands with bipolar I disorder were stratified into three groups based on a history of panic disorder, panic attacks, or no panic attacks in the probands. Multipoint nonparametric linkage analysis was performed on data from bipolar I and II family members in each group. RESULTS: Linkage scores for five consecutive 18q marker loci were highest in the families of the probands with panic disorder and lowest for the families of the probands without panic attacks. CONCLUSIONS: This study supports the authors' previously reported clinical hypothesis of a genetic subtype of bipolar disorder identified by comorbid panic disorder. The hypothesis merits prospective testing.

Transformation of human osteoblast cells to the tumorigenic phenotype by depleted uranium-uranyl chloride.

Depleted uranium (DU) is a dense heavy metal used primarily in military applications. Although the health effects of occupational uranium exposure are well known, limited data exist regarding the long-term health effects of internalized DU in humans. We established an in vitro cellular model to study DU exposure. Microdosimetric assessment, determined using a Monte Carlo computer simulation based on measured intracellular and extracellular uranium levels, showed that few (0.0014%) cell nuclei were hit by alpha particles. We report the ability of DU-uranyl chloride to transform immortalized human osteoblastic cells (HOS) to the tumorigenic phenotype. DU-uranyl chloride-transformants are characterized by anchorage-independent growth, tumor formation in nude mice, expression of high levels of the k-ras oncogene, reduced production of the Rb tumor-suppressor protein, and elevated levels of sister chromatid exchanges per cell. DU-uranyl chloride treatment resulted in a 9.6 (+/- 2.8)-fold increase in transformation frequency compared to untreated cells. In comparison, nickel sulfate resulted in a 7.1 (+/- 2.1)-fold increase in transformation frequency. This is the first report showing that a DU compound caused human cell transformation to the neoplastic phenotype. Although additional studies are needed to determine if protracted DU exposure produces tumors in vivo, the implication from these in vitro results is that the risk of cancer induction from internalized DU exposure may be comparable to other biologically reactive and carcinogenic heavy-metal compounds (e.g., nickel).

Mismatch cleavage detects base deletion in cystic fibrosis gene.

The delta F508 is the most common defect in the cystic fibrosis (CF) gene; it involves in a 3-base deletion in codon 508 and results in the loss of a phenylalanine residue at amino acid position 508. Our previous results have shown the mismatch enzyme cleavage at the mismatch of a DNA duplex in identifying a specific DNA sequence or a point mutation. The assay is simple and reliable. By manipulating the melting temperature (Tm) for the hybrids of the DNA targets and the deoxynucleotide probes, the mismatch cleavage assays are able to detect the most common defective CF gene, delta F508. The assays with a delta F508 and a normal wild-type probe can differentiate the three genotypes, i.e., delta F508/delta F508, delta F508/normal and normal/normal. Furthermore, the addition of ammonium acetate amplifier to the assay for recycling the target DNA can increase the sensitivity to a level that is sufficient to detect the mutated target in a few micrograms of genomic DNA without the aid of PCR amplification. The detection of the base deletion, the amplification of sensitivity and the differentiation among the genotypes of normal, carrier delta F508 and mutant delta F508 suggest the useful application of mismatch cleavage in genetic diagnosis at the DNA level.

A diagnostic polymerase chain reaction assay for species A and D of the Anopheles Dirus (Diptera: Culicidae) species complex based on ribosomal DNA second internal transcribed spacer sequence.

A polymerase chain reaction assay based on differences in the internal transcribed spacer regions of ribosomal DNA was developed for distinguishing 2 members of the Anopheles dirus sibling species complex. This assay distinguished An. dirus species A from species D by producing diagnostic bands, 374 base pairs (bp) in length for species A and 663 bp in length for species D. Both laboratory colonies and field collections from Hainan and Yunnan provinces of China were identified with 100% accuracy.

Oxygen transfer characteristics in the compact callus aggregates of Rhodiola sachalinensis.

The effective diffusivity of oxygen in compact callus aggregates (CCA) of Rhodiola sachalinensis was extremely small, varying from 0.34 x 10(-11) to 5.4 x 10(-9) m2/s and increasing with particle diameter. The calculated results indicated that severe oxygen limitations occurred in the CCA aggregates. However, the direct determination of the viability of the cells demonstrated that the CCA aggregates contained a high fraction of viable cells. It was suggested that the tissue differentiation or plasmodesmata within the CCA aggregates may provide capillaries for improving transport of oxygen and other nutrients.

[A molecular epidemiologic study on the mechanism of intrauterine transmission of hepatitis B virus].

A case-control study with examination of placentas using immunohistochemistry stain was reported in this paper. In the Maternal and Children Health Hospital of Shanxi Province, 242 consecutive HBsAg positive mothers and their babies were selected as subjects and 110 placentas of HBsAg positive mothers and 25 placentas of HBsAg negative mothers during the different period of pregnancy were collected for laboratory test. The results showed that maternal HBeAg positivity (OR = 32.63) and history of threatened premature labor (OR = 22.80) were important risk factors. Among full-term placentas with HBsAg positivity, HBsAg (biomarker of HBV infection) positive rates were 100% in decidual cell, 59.38% in trophoblastic cell, 65.50% in villous mesenchyme cell, and 39.38% in villous capillary endothelial cell (VCEC) with a decreasing trend (trend test, chi 2 = 30.5, P < 0.01) from mothers to fetus whereas HBsAg positive in VCEC was significantly related to intrauterine infection (OR = 20.86, P < 0.01). Results suggested that there might be two transmission routes on the mechanism of HBV intrauterine transmission, hemogenous by damage of placental vessels and cellular through placental cellular transfer of HBV.

[Application of the free parascapular flap in children].

OBJECTIVE: To improve surgical results we have applied the free parascapular flap in children for facial or cervical burn scar contracture. METHOD: Eight children aged from 6 to 9 years received free flap transfer. Based on the defect and the distribution of the cutaneous branches of the circumflex scapular vessels, the flap was designed with its size ranging from 15 cm x 8 cm to 22 cm x 6.5 cm. RESULT: Free flap transferring with microvascular anastomosis was successful in all cases. The average operation time was 5 hours with average blood loss being about 100 ml. CONCLUSION: Free flap transplantation is safe and beneficial in children when the technique and skill of microsurgery and plastic and reconstructive surgery are mastered and intraoperative blood loss is controlled under 1% of the total blood volume. The vertically oriented parascapular flap can not only provide proper tissue for reconstraction but also minimise the donor site morbidity.

[Color Doppler monitoring the utero-placental-fetal circulation variety of normal pregnancy and intrauterine growth retardation].

OBJECTIVE: To study the utero-placental-fetal circulation (UPFC) in normal pregnancy and intrauterine growth retardation (IUGR) cases. METHODS: Color doppler ultrasound was used to detect UPFC in 150 second and third trimester pregnant women, of which 89 cases were normal pregnancy and 58 cases were IUGR. 3 cases were IUGR with chronic renal failure. Hemodynamical value of the umbilical artery (UmA), umbilical vein (UmV) and uterine artery (UtA) were examined directly. The indices included time average velocity (TAV), pulsatility index (PI), resistance index (RI), systolic/diastolic (S/D) ratio, blood flow volume (Q). The maternal serum estriol (E3), human placental lactogen (HPL) and plasma thromboxane B2 (TXB2)/6-keto-PGF1 alpha (6-KP) were measured simultaneously. RESULTS: The result shows that in normal pregnancy group UPFC is abundant gradually with increasing gestational age. In IUGR group 92.53% of cases showed that TAV and Q of UmA, UmV markedly decreased and PI, RI and S/D ratio of UmA elevated at 20 weeks of gestation. There were significant difference between the two groups, maternal serum E3, HPL level in IUGR group were significantly lower than that of the normal pregnancy group, 6-KP level reduced, and TXB2/6-KP ratio significantly increased. CONCLUSION: Using color doppler ultrasound examining hemodynamical changes of UmA, UmV and UtA could observe UPFC function directly. It is one of the best method to early diagnose and predict the prognosis of IUGR.

[Influence of isoflurane and sevoflurane on metabolism of oxygen free radicals in cardiac valve replacement].

To investigate effects of isoflurane and sevoflurane on the levels of lipid peroxides (LPO) in plasma and superoxide dismutase (SOD) in red blood cells during myocardial ischemia/reperfusion, and to study myocardial protection of these two volatile anesthetcs, 30 patients undergoing cardiac valve replacement were randomly divided into isoflurane (Group I), sevoflurane (Group S), and controlled group (Group C). Patients of Group I were anesthetized with isoflurane mainly, Group S with sevoflurane, and Group C with fentanyl. LPO and SOD levels were measured at five points before and during the operation. The results were as follows: SOD levels in all groups did not change significantly. LPO levels in Group C increased during the operation, while in other two groups didn't. Rates of automatical rebeating of hearts in Group C were lower than that of hearts in other two groups. It is suggested that isoflurane and sevoflurane can inhibit increasing of LPO levels to attenuate myocardial ischemia/reperfusion injuries mediated by oxygen free radicals during open-heart surgery.

[Protein status and antioxidant capability of residents in endemic and nonendemic areas of Keshan disease(KD)].

From a survey of residents in endemic and nonendemic areas of Keshan disease(KD), it was found that the plasma cystine, tryptophan and lysine as well as blood selenium content were lower, while the plasma lipid peroxide was higher in endemic areas than those in non-endemic areas. These results suggests that the selenium deficiency coexisting with cystine and tryptophan insufficiency decreases the antioxidant defense of the organism and may be one of the important factors for the development of KD. The role of sulfur-containing amino acid in antioxidant defense is discussed emphatically.

[The combined effects of MCYST and water organic pollutants on the co-induction of SHE cell transformation and expression of Ras P21 protein].

The authors studied the combined carcinogenic activities of MCYST and organic pollutants in water of Dianshan Lake, using in vitro Syrian hamster embryo (SHE) cell transformation and immunohistochemical assays. The results showed that MCYST alone could not induce SHE cell transformation, but it increased the effects of low dose organic pollutants in cell transformation in a dose-dependent manner. In the cells isolated from type II transformed foci, ras P21 protein showed a positive immunohistochemical staining. It suggests MCYST may be a tumor promoter, which has a synergistic carcinogenic activity organic pollutants in water and the activation of ras oncogene may be one of the factors inducing malignant transformation.

[Study on the transformation of mammal cells induced by glycidyl methacrylate in vitro].

Glycidyl methacrylate(GMA) can induce the phenotype transformation of human embryonic lung fibroblasts. The transformed cells prolifeated rapidly with a higher in the ratio of nucleus to cytoplasm, and their life span extended notably. The transformed colonies exhibited in extensive random orientation and the cells crossing-over. The transformed cells could be agglutinated by lower concentration of ConA and could grow in semisolid agar. The chromosome abberations could occur in transformed fibroblasts. These results suggest that GMA is able to induce malignant transformation of mammal cells, and it may be carcinogenic to human.

[Proximal bronchial invasion of lung cancer: a clinicopathological study].

OBJECTIVE: To investigate the characteristics of proximal bronchial invasion of lung cancer of different types. METHODS: Proximal bronchus of 151 operatively resected specimens of hilar type lung cancer was selected for cross-sectional pathological study. Forty one specimens were obtained from total pulmonectomy, and 110 from pulmonary lobectomy. RESULTS: Evidence showed that the direct invasion of tumour cells through the submucosal layer or multiple layers was the major form of lung cancer spread; 96.6% of the cancerous invasion occurred at the proximal bronchial wall less than 1.5 cm apart from the margin of the cancer. The extension of invasion was correlated with the histopathologic type of cancer, mode of invasion and TNM classification(pT, pN). Besides, the invasion in the bronchial wall by metastatic lymph nodes was also an important way for the cancer spread. CONCLUSION: In order to achieve radical removal of tumor, it is imperative to keep a distance of 1.5 cm or more between the excision margin of the bronchus and the tumor, and to completely resect the hilar and mediastinal lymph nodes.
