Analysis of cocaine-positive fatalities.

A review of all autopsy and toxicology reports for persons dying in New York City in an 11-month period found 935 persons dying with cocaine in their bodies. Cocaine-positive fatalities were more likely in the young black and Hispanic and male population. In addition to cocaine and its metabolites, heroin and other opiates were found in 39% of persons and ethanol in 33% and barbiturates and minor tranquilizers in only 2% of the deceased. Cocaine overdose was responsible for 4% of the deaths and overdose with heroin and cocaine for 12% of the deaths. Violence was often the cause of death. Thirty-eight percent died of homicide, seven percent of suicide, and eight percent from accidents. Of particular interest were 6 persons who died of acute cardiac events directly related to cocaine as well as 4 cases of ruptured dissections of the ascending aorta, and 9 cases of cerebral hemorrhage. Autopsy findings for these individuals are described, and possible mechanisms of death are discussed.

The determination of selenomethionine in selenium yeast by cyanogen bromide gas chromatography.

A new method for the determination of bonded selenomethionine in selenium yeast by gas chromatography is established. Bonded selenomethionine is rapidly and precisely determined by measuring CH3SeCN released during the reaction of selenoprotein with CNBr (CNBr-GC method). Conditions for the reaction and chromatography are described. The results of CNBr-GC are compared with those obtained by acid hydrolysis/ion exchange chromatography. This new method has the advantage of being accurate, sensitive and selective.

D10S20, a previously unmapped RFLP (OS-3), is located on 10q near D10S4.

The locus recognized by the probe OS-3 is assigned to chromosome 10 both by Southern blot analysis of a panel of somatic cell hybrid DNAs and by genetic linkage to markers already assigned to chromosome 10. In Caucasians this probe recognizes a three-allele TaqI RFLP as well as two-allele BanII and RsaI RFLPs which are both in strong linkage disequilibrium with each other and with the TaqI RFLP. The D10S20 locus defined by this probe maps 5.5 cM distal to D10S4 on the long arm of chromosome 10. Because this human clone hybridizes with mouse genomic DNA, it will be useful in comparative mapping studies.

Structure of the medium-chain acyl-CoA dehydrogenase from pig liver mitochondria at 3-A resolution.

The three-dimensional structure of the medium-chain acyl-CoA dehydrogenase (EC 1.3.99.3) from pig liver mitochondria has been determined to 3.0-A resolution by the x-ray diffraction method. The enzyme is a tetramer of four identical 43-kDa subunits and contains one equivalent of flavin adenine dinucleotide (FAD) per subunit. The polypeptide is folded into three domains. The N-terminal and the C-terminal domains are composed mainly of alpha-helices, and the middle domain is packed with orthogonal beta-sheets. The FAD has an extended conformation: the flavin ring lies between the N-terminal and the beta-sheet domains, and the adenine moiety is found at the junction between the C-terminal and the beta-sheet domains of one subunit and the C-terminal domain of a neighboring subunit. The polypeptide chain folding near the FAD binding site is different from those observed in other flavoproteins, such as glutathione reductase and glycolate oxidase.

Biological vulnerability to depression: replication of MAO and evoked potentials as risk factors.

In previous work, we have reported that specific combinations of platelet (MAO) activity and evoked potential augmenting/reducing (AR) are associated with risk for affective disorders. This new study screened 271 college freshmen solely on MAO and AR and selected a sample with extreme values on both measures. These students were interviewed with the Diagnostic Interview Schedule and they completed a family history questionnaire and psychosocial scales. Following the previous work, the same MAO and AR combinations were related to the frequency of major depression and a family history of psychiatric disorder.

[Alpha-hANP-like immunoreactivity in lung cancer].

Specific radioimmunoassay and immunohistochemical method of alpha-human atrial natriuretic polypeptide (alpha-hANP) was established to diagnose one case of large cell lung carcinoma (clear cell carcinoma) proved by pathology. It was found that there was alpha-hANP-like immunoreactivity in the cancer cells at a concentration of 147.5 pg/mg protein. Chromatography analysis showed that alpha-hANP-like immunoreactive materials had multiple molecular forms. The significance of these phenomena remains to be further investigated.

Articulation proficiency and error pattern of cleft palate children with delayed hard palate closure.

Prior to 1982, children with cleft palate received delayed hard palate closure at Chang Gung Memorial Hospital. In this treatment, soft palate is usually repaired with Widmaier palatoplasty at about 18 months with surgery; hard palate is not closed until the age of 6 or 7. Our previous study indicated that the articulation skill of the cleft palate children before delayed hard palate closure is generally far inferior to the non-cleft children of the same age. Further investigation was carried out to look into the articulation proficiency and error patterns of the cleft palate children undergoing delayed hard palate closure. Data of 47 children (32 boys, 15 girls) were analyzed. Articulation proficiency was compared in four phoneme categories: nasals, plosives, fricatives and affricates. The results indicated that affricates are most difficult for Chinese cleft palate children to pronounce. At age 5, just before the hard palate closure, none of the phonemes is produced correctly except nasals.
