[Genistein exerts multiple suppressive effects on human breast carcinoma cells].

OBJECTIVE: To study the suppressive effects of genistein on progression of breast carcinoma. METHODS: In vitro cell culture, animal studies, zymography, Northern and Western blot were used in this study. RESULTS: Genistein inhibited in vitro invasion of MCF-7 and MDA-MB-231 cells. This inhibition was associated with down-regulation of MMP-9 and up-regulation of TIMP-1. These effects were also demonstrated in vivo. Genistein inhibited growth of the breast cancer cells transplanted in nude mice. The growth inhibitory effect was associated with increased tumor cell apoptosis, and up-regulated p21WAF1/CIP1 expression. Furthermore, genistein inhibited angiogenesis as shown by decreased vessel density and decreased production and release of VEGF and TGF-beta 1. CONCLUSION: Genistein exerts multiple suppressive effects on breast carcinoma. It may help halt its progression.

[Clinical implication of apoptosis of breast cancer induced by preoperative chemotherapy].

OBJECTIVE: To investigate whether apoptosis could be induced by preoperative chemotherapy in breast cancer patients, and determine if it is correlated with clinical response and patients' survival. METHODS: Apoptosis was assayed by in situ DNA end labeling and receptors for estrogen and progesterone were examined immunohistochemically. Comparison of the differences in apoptotic index (AI) and clinical response was made between patients received preoperative chemotherapy and those did not, using the one tailed Student's t-test. The Kaplan-Meier method was utilized to calculate relapse-free and overall survival time. Cox proportional hazards model was used to conduct multivariate analysis of prognostic factors and differences in overall survival and relapse-free survival after adjustment for other variables. RESULTS: Apoptosis was induced by preoperative chemotherapy, especially in lymph node positive patients. Post-chemotherapy AI correlated with better clinical response and increased patient survival, including relapse (disease) free survival and overall survival. CONCLUSION: The extent of apoptosis induced by preoperative chemotherapy of primary breast cancer may possibly predict an individual patient's overall response to treatment.

[The experimental research of the rhBMP-2-induced apoptosis in human glioma cells].

OBJECTIVE: To investigate the effects of recombinant human bone morphogenetic protein (rhBMP-2) on the biological characteristics of human glioma cells (SHG44) in vivo and in vitro. METHODS: The growth curve of SHG44 cells was ploted with or without rhBMP-2. The proliferation quantity of SHG44 cells with rhBMP-2 was determined by MTT method. The cell cycle, ultrastructure and DNA fragments of SHG44 cells were detected respectively by flow cytometry (FCM), electron microscope and agarose gel electrophoresis. Tumor growth state of SHG44 gliomas, which grew under the skin of nude mice, was observed with local injection of rhBMP-2. RESULTS: FCM showed that rhBMP-2 inhibited the growth of SHG44 cell in vitro and its effect depended on dosage. DNA content of SHG44 cells in G1 phase was obviously increased (5 mg/L group, 77.4%; control group, 55.2%, P < 0.05) and the content of its S phase was reduced (5 mg/L group, 15.2%; control group, 33.3%, P < 0.01). The apoptosis spike (12.3%) occurred in the 5 mg/L group. The nucleolus chromatin condensation, bound of aggregation and caryorrhexis were observed under the electron microscope and the apoptotic cells appeared. DNA electrophoresis showed a ladder strap (DNA ladder). SHG44 glioma growth was relatively slower and its volume was smaller with the biquadratic local injection of rhBMP-2 (tumor end volume in average: experimental group 894 mm(3) +/- 145 mm(3), control group 5 163 mm(3) +/- 1 028 mm(3), P < 0.01 CONCLUSION: rhBMP-2 can suppress the proliferation of SHG44 gliomas and induce its apoptosisN:

[Apoptosis and related gene expression in lacrimal gland of cases with Sjögren's syndrome].

OBJECTIVE: To assess the roles of apoptosis and expression of the related genes in lacrimal glandular destruction of patients with Sjögren's syndrome (SS). METHODS: Small pieces of palpebral lobes of lacrimal glands were obtained from 12 patients (10 with primary Sjögren's syndrome, 2 with secondary Sjögren's syndrome) and 7 normal controls. They had been investigated by the in situ end labeling and immunohistochemical staining to detect the apoptotic cells and the expression of Fas, FasL, bcl-2 and Bax. RESULTS: The number of epithelial apoptotic cells in lacrimal glands of patients with SS was significantly higher than that in normal glands, and SS epithelial cells showed increased expression of Fas, FasL and Bax. There was significant positive correlation between the number of the apoptotic cells and that of the cells expressing Fas, FasL and Bax, respectively, and there was significant negative correlation between the number of the apoptotic cells and that of the cells expressing bcl-2. CONCLUSIONS: The apoptosis of the epithelial cells in the lacrimal gland may be one of the mechanisms leading to the glandular destruction found in SS. In SS lacrimal glands, the expression of Fas, FasL and Bax may promote apoptosis, and the expression of bcl-2 may inhibit apoptosis.

[Analysis on genetic pattern of familial type II diabetes mellitus].

OBJECTIVE: To analyse hereditary familial type II diabetes mellitus (type II DM), including its pattern and the transmitted expression on different sex of type II DM. METHODS: Familial analysis, segregation analysis, tests were used for multifactorial inheritance and threshold analysis. One hundred seventy-seven pedigrees of familial type II DM were studied. RESULTS: The heritability of familial type II DM was 143.06% +/- 3.3%. One dominant major gene might influence the genesis of type II DM. There were 214 nuclear families in 3 different mating types: U x U, U x A and A x A. The segregation ratios of U x U, U x A and A x A were 0.4703, 0.4838 and 0.6712 respectively. The results of tests for multifactorial inheritance and segregation analysis indicated that the genetic pattern of U x A and A x A nuclear families was not multifactorial inheritant and autosomal recessive (AR) but autosomal dominant (AD) inheritance. The genetic pattern of U x U nuclear families was neither AD nor AR inheritance. CONCLUSION: There was genetic heterogeneity in type II DM discovered with subgroupes of autosomal dominant inheritance in type II DM.

[A study on relationship between plasma heat stress protein 70 and cytokine in patients with heat apoplexy].

OBJECTIVE: To study the change of levels of plasma heat stress protein 70 (HSP70) and cytokine and their interrelationship in pathogenicity of heat apoplexy. METHODS: Plasma HSP70 was determined with Western blot, and plasma interleukin-2 (IL-2), soluble interleukin-2 receptor (sIL-2R) and interleukin-6 (IL-6) were determined with enzyme-linked immunosorbent assay (ELISA). RESULTS: Plasma level of HSP70 was 4 211.2 +/- 1,286.2 (integral optical density) in average in the patients of heat apoplexy group, 4,137.8 +/- 1, 207.5 in those of severe heat apoplexy group, and 6,043.5 +/- 1,354.8 in control group, with very significant difference (P < 0.01). Plasma IL-2 level was (64.3 +/- 32.3) pg/ml and (200.0 +/- 50.0) pg/ml and sIL-2R (54.9 +/- 33.3) U/ml and (167.5 +/- 89.0) U/ml in the patients with heat apoplexy and in controls, respectively, with very significant difference. But, there was no significant difference in IL-6 between varied groups. CONCLUSIONS: The above-mentioned results suggest that establishment of heat acclimatization correlates to the interaction of network of neuroendocrine-cytokine-HSP, in which decrease in plasma IL-2 was one of the important factors causing reduction of HSP70 expression. Balance of network regulation was broken by pathological factors, finally resulting in heat apoplexy.

[Preliminary study on relationship between antibody against heat stress protein 70 and hypertension].

OBJECTIVE: To preliminarily explore the relationship between main antibody against heat stress protein 70 (anti-HSP70) and hypertension. METHODS: Serum anti-HSP70 was determined with Western blotting technique in hypertensive and normotensive groups. Logistic regression model was used to analyze the relationship between anti-HSP70 and hypertension. RESULTS: It was found that level of antibody against the main heat shock protein 70 was significantly higher in patients with hypertension (based on criteria of 160/95 mm Hg, 1 mm Hg = 0.133 kPa) than in controls, after adjustment for the generally recognized risk factors for hypertension (such as age, sex, length of employment, body mass index, job, et al.). There still was stronger association between anti-HSP70 and hypertension (based on criteria of 140/90 mm Hg), but its strength of association was weaker than the former. CONCLUSIONS: Anti-HSP70 may potentially serve as a serum biomarker to assess hypertension, or anti-HSP70 itself may be involved in the development of hypertension.
