Augmented recession of the medial recti.

Bimedial rectus recession with measurement from the limbus was combined with conjuctival recession 85 children undergoing surgery for esotropia. Using this augmented recession technique, the prism diopters of correction per millimeter of surgery done were 4.8 compared with 3.2 found in 56 control patients. Postoperatively, the conjunctiva had a satisfactory cosmetic appearance and ductions were normal.

In vitro RNA-RNA splicing in adenovirus 2 mRNA formation.

"Splicing" of the precursor to an adenovirus mRNA was accomplished in isolated cell-free extracts. Nuclei were prepared from hypotonically swollen cells that had been labeled with [3H]uridine for 10 min prior to nuclear isolation. Addition of a "cytoplasmic" fraction was required for the splicing to occur. The nuclear precursor, a poly(A)-terminated RNA molecule approximately 5 kilobases long, contained sequences complementary to the 58.5--75.9 region of the adenovirus 2 genome, including those sequences spliced out of the mature mRNA molecule. The in vitro spliced product was a poly(A)-terminated RNA molecule identical in size to the cytoplasmic 72,000 Mr protein mRNA (2 kilobases long) in which the sequences encoded in the 70.7--75.9 region of the viral genome were spliced to those encoded at 58.7--65.6, with the sequences encoded at 66.1--70.7 deleted.

The definition of transcription units for mRNA.

The detailed analysis of TUs with purified Ad2 DNA and the analysis of TU size for the bulk HeLa cell hnRNA compared to the size of mRNA of infected and uninfected cells supports the conclusion that mRNA in mammalian cells is generally derived by the processing of primary transcripts. In the context of this volume, these results indicate that the RNA transcription products of chromatin which are related to mRNA are longer than the mRNA itself. Proper functioning of chromatin in vitro must eventually take these results into account.

Genetic analysis of adenovirus type 2. VIII. Physical locations of temperature-sensitive mutations.

Temperature-sensitive mutations of human adenoviruses can be physically located on the viral genome by determining the DNA structures of recombinants formed in genetic crosses between different members of the same subgroup. We have analyzed the DNA structures of many interserotypic recombinants formed in crosses between temperature-sensitive (ts) mutants of adenovirus type 2 and adenovirus type 5 with the restriction endonucleases BamHI, EcoRI, HindIII, and and Sma I. In this way, we have mapped the physical coordinates of adenovirus type 2 (Ad2) ts1, Ad2 ts3, Ad2 ts4, and Ad2 ts48, and refined the mapping of Ad5 ts1.

Physical mapping of the genes controlling adenovirus paracrystal formation.

The genetic properties and physical mapping of the genes controlling adenovirus-induced paracrystals were examined by means of a new, simplified technique. The induction of paracrystals was virus-strain dependent and blocked by several temperature-sensitive mutations. Using interserotypic recombinants between a paracrystal-producing and a non-producing strain of adenovirus, the principal paracrystal determining factor was mapped between coordinates 30 and 44 on the 100-unit adenovirus genome.

Transformation of human cells by temperature-sensitive mutants of simian virus-40.

Conditions necessary for the establishment and maintenance of transformation of human cells by wild type and temperature-sensitive mutants of SV40 were examined. For both early and late mutants, the frequency of transformation was found to be up to 5-fold higher, and virus yield 100-fold lower, at 39 degrees than at 33 degrees. No such effect was observed with the wild type virus under the same conditions. This observation is apparently at variance with previously published work, but may be explained by the semipermissive nature of the cells that we used. Increasing the temperature to 40.5 degrees caused cells transformed by the early mutant, tsA30, to lose T-antigen as detectable by staining, and also to lose the ability to grow to high density, while it produced no effect on cells transformed by wild type virus.

[Clinical demonstrations: 1. Renal tubular acidosis; 2. Breast neoplasm with bone metastases in a man; 3. Thrombotic thrombocytopenic purpura; 4. Precocious eclampsia in a 22 years old primipara].

Four patients with metabolic and/or circulatory failure are described. The clinical, biological and radiologic features of reversible renal failure with renal tubular acidosis are presented. A case of male breast carcinoma is discussed with reference to the effect of steroids on the hormonal receptors at the malignant cell. The pathophysiology of thrombotic thrombocytopenic purpura and its treatment (splenectomy and platelet aggregation inhibitors) are discussed. An overview of eclampsia and its relationship to underlying kidney disease is presented. It may be concluded that a metabolic and/or ischemic imbalance interferes with the normal activity of the reticular substance and the brainstem, which normally maintain the state of consciousness.

The definition of a large viral transcription unit late in Ad2 infection of HeLa cells: mapping of nascent RNA molecules labeled in isolated nuclei.

Nuclei isolated from HeLa cells 15 hr after infection with Ad2 synthesize an RNA transcript approximately 25 KB long, beginning between 0.2 and 0.3 on the physical map and extending to (or close to) the right end of the genome. The majority (60-92%) of the RNA transcribed from fragments between 0.2 and 1.0 is synthesized as part of this large molecule which represents the major product of late Ad2 RNA synthesis. Smaller transcripts are also formed mainly from the left end of the Ad2 genome.

The definition of a large viral transcription unit late in Ad2 infection of HeLa cells: mapping by effects of ultraviolet irradiation.

The synthesis of Ad2-specific nuclear RNA in HeLa cells 15 hr post-infection has been examined after ultraviolet irradiation. The synthesis of RNA complementary to various segments of Ad2 DNA is decreased differentially, with the most sensitive region at the right end of the physical map and the most resistant in the region from 0 up to 0.2-0.3 (20-30% from the left end of the genome). For a given ultraviolet dose, the amount of labeled RNA homologous to various DNA fragments decreased exponentially from map position 0.2-0.3 to 1.0. These results suggest that a major transcriptional unit late in Ad2 infection is a rightward-transcribing unit between 0.0-0.3 and 1.0.

The initiation sites for RNA transcription in Ad2 DNA.

Six restriction fragments of Ad2 DNA which contain sites for RNA initiation have been identified by their ability to hybridize nascent labeled RNA less than 1 kb in length. Four RNA initiation sites early in infection are identified in regions where previous work (Flint, 1977) had mapped mRNAs. The major late RNA initiation site is the origin of a giant nuclear transcript extending from approximately 16.3 map units to (or close to) 100 units at the end of the genome. This transcription unit encompasses at least four or five mRNA sites; processing of this long transcript appears necessary to generate the mRNA.

[Occlusion phlebography. Technics and possibilities of use].

The technique and use of occlusion phlebography is described. The method is based on temporary occlusion of blood flow by means of a balloon catheter. Various balloon catheter systems were used and their technical properties were evaluated. The distal veins can be demonstrated by using double lumen catheters, distal and proximal veins by using a catheter with three lumina. Occlusion phlebography is a suitable method for demonstrating hepatic and renal veins; it improves the demonstration of the azygos vein, the suprarenal vein and of visceral pelvic veins, including the iliolumbar veins. It also permits demonstration of the inferior vena cava and parietal veins in the pelvis in separate stages. Occlusion phlebography is the method of choice for the direct demonstration of splenorenal shunts.
