Adenoid cystic carcinoma cells cultivated in monolayer and sponge matrix culture, or implanted on the chorioallantoic membrane (CAM) of embryonated eggs, were observed morphologically, and the glycosaminoglycan components in the tumor tissue were analyzed. This tumor tissue contained a large amount of glycosaminoglycans, composed of chondroitin 4- and 6-sulfate, heparan sulfate, hyaluronic acid, and a small amount of dermatan sulfate. In monolayer culture spindle cells proliferated vigorously as multilayer, and secreated mucinous material. In sponge matrix culture, the proliferating cells became embedded in the material produced by the cells themselves. A trace of fine fibers stained with orceine was observed in the intercellular material in culture. Histologic sections of the implants grown on CAM showed that the tumor cells arranged in various structures produced a large amount of mucinous material that spread into the stromal area without any contribution from the mesenchymal element. The morphologic and biologic characteristics of these tumor cells are quite similar to those of pleomorphic adenoma.
The effect of acute denervation of the kidney on renal sodium and water excretion, and hemodynamics including intrarenal blood flow, was studied in anesthetized dogs. The intrarenal blood flow was measured by the radioactive microsphere method. In all experiments denervation natriuresis and diuresis was observed without significant change in glomerular filtration rate, renal blood flow and distribution of intrarenal blood flow. There was, however, an associated increase in potassium excretion. We suggest that denervation natriuresis and diuresis may be caused by the elimination of a direct nervous control of sodium and water reabsorption.
A case of radionecrosis in the brain simulating a tumor was presented. Two courses of radiotherapy were given to a right intraorbital tumor over an interval of ten years. Four years after the second course of therapy, symptoms simulating a right frontal lobe tumor developed. A frontal lobectomy including the necrotic area improved the patient's condition.
The correlation between the content of individual glycosaminoglycans and the histological patterns are studied on breast tumor tissues. The myxomatous stroma of intracanalicular fibroadenoma contained a large amount of glycosaminoglycans, which were mainly hyaluronic acid. The chondroitin 4- and 6-sulfate level was also high. As the supporting stroma of this tumor became denser and more fibrous, the level of hyaluronic acid content was reduced. In the case of pericanalicular fibroadenoma, glycosaminoglycans were small in amount and the levels of hyaluronic acid and chondroitin sulfate were low, but the ratio of dermatan sulfate content was higher. In the case of gynecomastia, the conent was almost the same as that of pericanalicular fibroadenoma. Scirrhous carcinoma tissues contained a relatively large amount of hyaluronic acid and chondroitin sulfate. No remarkable differences in heparan sulfate content were observed in any one of the breast tumors tested. Dermatan sulfate-chondroitin sulfate copolymers were detected in all the tumors. The presence of dermatan sulfate seemed to have an intimate relation with the fibrogenesis in the interstitial stromal element of the tumor tissues.
A series of cases of astrocytoma of differing cell composition and degree of malignancy was examined for the presence of reactive perivascular cuffing. Only surgical biopsy material was used. A correlation was made between perivascular lymphocyte cuffing and the presence of neoplastic astrocytes of the gemistocytic type.
Pleomorphic adenoma of the salivary gland is classified into two types based on the morphology of the cells and biochemical analysis of glycosaminoglycans. Type 1: the tumors consisted mainly of spindle cells, producing a large amount of mucinous material, and contained not only chondroitin 4- and 6-sulfate and hyaluronic acid but also heparan sulfate. When the tumors of this type were dissociated and cultivated, spindle cells proliferated vigorously as mulitlayer and secreted mucinous substance into the intercellular space. The cells looked like non-epithelial cells in a conventional culture bottle, but showed an epithelial-like arrangement when they were cultivated in sponge matrix or inoculated subcutaneously into mice. Fine fibres stained with orceine or resorcin-fuchsin were observed in the intercellular material both in resected tissue and in culture. Type 2: the tumors consisted mainly of ductal cells, having no mucinous area, and contained little heparan sulfate. When the tumors were cultivated, the growth of epithelial-like polygonal cells was observed in monotonous pattern, producing scanty intercellular substance, and proliferating, as monolayer, much less rapidly than the cells of Type 1.

Carcinogens injected into the excretory canal of submandibular gland of Donryu rats revealed the following histologic changes in salivary glands. 20-Methylcholanthrene induced squamous cell metaplasia, fibrosis in the early stages, and "benign lymphoepithelial lesion"-like pattern after 3 months. Dense hyalinization occurred after 4-5 months with so-called "mixed tumor"-like pattern. In the later stages epidermoid carcinoma and fibrosarcoma were observed. 9, 10-Dimethylbenzanthracene caused degenerative change, metaplasia, fibrosis and cell infiltration, and later carcinoma and sarcoma appeared at a high rate. 4-Nitroquinoline-N-oxide led to dense hyalinization and so-called "mixed tumor"-like pattern was observed in many specimens. N-nitroso-N-methyl urethane and N-methyl-N-nitroso-N'-nitroguanidine revealed metaplastic changes, fibrosis and lymphoid infiltration. Scarlet red induced remarkable infiltration and aggregation of lymphoid cells, showing benign "lymphoepithelial lesion"-like pattern.
Deviation of systolic time intervals (STI) from the regression lines obtained from 122 normal subjects was studied in 22 healthy adults (Group 1), 18 N.Y. functional class I cardiac patients (Group II) with ischemic (IHD) or primary myocardial disease (PMD), and 15 similar patients (pts) but N.Y. functional class II with prior heart failure (Group III). STIc (corrected for heart rate) were normal in Groups I and II. Supine exercise caused shortening of pre-ejection period PEPc and prolongation of left ventricular ejection time ETc in both groups. Group III pts had a significantly longer PEPc and shorter ETc at rest. Supine exercise caused further prolongation of PEPc and a slight prolongation of ETc in this group. In 8 mongrel dogs, the effect of controlled changes of hemodynamic variables on STI was studied with intact hearts and repeated after myocardial impairment has been induced by pentobarbital. Doubling of venous return while the heart was intact produced changes in STI similar to the effects of supine exercise in Groups I and II human subjects. With myocardial impairment, comparable increase in venous return had an effect on STI similar to the effect of supine exercise Group III patients. Controlled increase in each of heart rate or blood pressure, with other hemodynamic variables kept constant, produced changes in STI different from the effect of supine exercise on human subjects. The study suggests that the value of supine exercise induced changes in STI in reflecting left ventricular performance is attributable primarily to increased volume load. In this resepct supine exercise is probably superior to other forms of exercise in disclosing impaired left ventricular performance.
The present investigation was designed to study the relationship between mechanical and electrophysiological actions on the heart of digitalis and the effects on it of K administration. Nineteen anesthetized dogs were given acetylstrophanthidin (AS) (30 gamma/Kg, followed by 1 gamma/Kg/min. When ectopic tachycardia occurred, sinus rhythm was restored with an infusion of K (5 mEq/Kg/hour, for 5 min). As the dose of AS was increased beyond 30 approximately 40 gamma/Kg, ventricular rate during electrically induced atrial fibrillation was reduced and PQ interval prolonged, while PP interval, QT interval and QRS duration showed little changes. Ectopic tachycardia occurred at an average dose of 56.9 gamma/Kg of AS. The indices of mechanical properties, dp/dt/IIT, max dp/dt and dp/dt (50), began to increase as soon as AS infusion was started and continued to increase during the administration of AS. The dp/dt (50) and PP interval or ventricular rate during atrial fibrillation did not change in parallel. Inotropic activity was not depressed even during ectopic tachycardia. The administration of K eliminated ectopic tachycardia, but caused no changes in the other electrophysiological and mechanical properties of the digitalized heart. The following conclusions were derived from the experiment. 1) The 2 actions of digitalis, mechanical and electrophysiological ones, are not necessarily related to each other. 2) Digitalis intoxication, represented by ectopic tachycardia, does not mean the depression of the inotropic activity. 3) The administration of K does not affect the inotropic activity of digitalis.
Optic glioma associated with bilateral subdural effusion in a four-month-old male infant is reported. Review of the cases with subdural effusion or hematoma and brain tumor in the world literature was made. When subdural effusion is found in a newborn or an older infant, the possibility of an associated brain tumor must be considered. Care also should be taken to search for an underlying brain tumor and to exclude dural metastasis when we deal with chronic subdural hematoma in an adult.
The hemodynamic effects of aortic regurgitation are characterized by an increase of left ventricular volume load due to the diastolic regurgitation and a decrease of aortic diastolic pressure or coronary perfusion pressure. It is presumed that an increase of heart rate in such a situation diminishes the regurgitant flow and elevates the coronary perfusion pressure so far as it is not accompanied by an improper increase of myocardial oxygen consumption. The present study, although it was the one performed on dog heart with an aortoventricular shunt rather than aortic valvular destruction, demonstrated that the increase of heart rate, if not beyond 160 beats per min, minimizes regurgitant flow as well as regurgitant ratio and maintains effective cardiac output without impairing coronary circulation. Thus it is suggested that it would be of beneficial therapeutic means in aortic regurgitation to keep the heart rate high or, at least, to avoid its excessive deceleration.
