Analysis of tRNA gene sequences by neural network.

The quantitative similarity among tRNA gene sequences was acquired by analysis with an artificial neural network. The evolutionary relationship derived from our results was consistent with those from other methods. A new sequence was recognized to be a tRNA-like gene by a neural network on the analysis of similarity. All of our results showed the efficiency of the artificial neural network method in the sequence analysis for biological molecules.

Effects of sodium metabisulphite on guinea pig contractile airway smooth muscle responses in vitro.

BACKGROUND: Sodium metabisulphite (MBS) is known to induce bronchoconstriction in asthmatic patients. The effects of MBS on guinea pig airway smooth muscle and on neurally mediated contraction in vitro have been examined. METHODS: Tracheal and bronchial airway segments were placed in oxygenated buffer solution and electrical field stimulation was performed in the presence of indomethacin (10(-5) M) and propranolol (10(-6) M) for the measurement of isometric tension. Atropine (10(-6) M) was added to bronchial tissues. RESULTS: Concentrations of MBS up to 10(-3) M had no direct effect on airway smooth muscle contraction and did not alter either tracheal smooth muscle contraction induced by electrical field stimulation at all frequencies or acetylcholine-induced tracheal smooth muscle contraction. There was a similar response in the absence of epithelium, except for potentiation of the response induced by electrical field stimulation at 0.5 Hz (24 (10)% increase). However, MBS (10(-5), 10(-6) and 10(-7) M) augmented neurally-mediated non-adrenergic non-cholinergic contractile responses in the bronchi (13.3 (3.2)%, 23.8 (9.6)%, and 6.4 (1.6)%, respectively). MBS had no effect on the contractile response induced by substance P, but at higher concentrations (10(-3) M and 10(-4) M) it caused a time-dependent attenuation of responses induced by either electrical field stimulation or exogenously applied acetylcholine or substance P. CONCLUSIONS: MBS had no direct contractile responses but enhanced bronchoconstriction induced by activation of non-cholinergic neural pathways in the bronchus, probably through increased release of neuropeptides. At high concentrations MBS inhibited contractile responses initiated by receptor or neural stimulation.

Cardiovascular effects of N-methyl leukotriene C4, a nonmetabolizable leukotriene C4 analogue, and the antagonism of leukotriene-induced hypotension by Ro 23-3544, in the American bullfrog, Rana catesbeiana.

Although some leukotriene antagonists have been reported to block leukotriene (LT) C4 responses in vivo, it is difficult to determine whether those antagonists block the effect of LTC4 directly or act via blocking the action of LTD4, as LTC4 is metabolized to LTD4 rapidly in vivo. In this study, the dose-response curves of N-methyl LTC4 (NMLTC4), the nonmetabolizable LTC4 analogue, and the peptidoleukotrienes (LTC4, LTD4, and LTE4) were obtained in the absence and presence of the leukotriene antagonist Ro 23-3544 in cannulated frogs. The more potent effect of NMLTC4 suggests that receptors that preferentially bind LTC4 exist in frog vascular smooth muscle and the previously reported LTC4 effect is a combination of LTC4 and its less potent metabolite LTD4. The NMLTC4- and LTC4-induced hypotensive effects were antagonized by Ro 23-3544. Ro 23-3544 also antagonized the effects induced by high doses of LTD4 and LTE4. Ro 23-3544 had no effect on duration of response and did not affect heart rate responses to LTC4 at low dose of the antagonist. The data suggest that receptors that preferentially bind LTC4 in bullfrog vascular smooth muscle regulate the hypotensive effect and that they can be antagonized by Ro 23-3544.

Ozone-induced airway hyperresponsiveness: role of superoxide anions, NEP, and BK receptors.

We investigated the role of reactive oxygen species in ozone-induced airway hyperresponsiveness (AHR) in Brown Norway rats. Airway responsiveness to inhaled acetylcholine (ACh) and bradykinin (BK) and inflammatory cell recruitment in bronchoalveolar lavage fluid (BALF) were measured in vivo. Neutral endopeptidase (NEP) activity assay and measurement of BK-receptor binding sites in Brown Norway rat lungs were carried out in vitro. Apocynin (5 mg/kg), an inhibitor of superoxide anion-generating NADPH oxidase, was administered perorally 30 min before a 3- or 6-h exposure to 3 ppm of ozone, and the animals were studied 18-24 h postexposure. Ozone induced increases in airway responsiveness to ACh and BK and in neutrophil counts in BALF. Apocynin inhibited the increase in airway responsiveness to BK but not to ACh without affecting the neutrophil counts in BALF. The antioxidants allopurinol and deferoxamine prevented ozone-induced AHR to both ACh and BK but did not reduce neutrophil counts. To further examine the mechanisms of ozone-induced AHR to BK, we measured NEP activity and the density of BK receptors in vitro after ozone exposure. Ozone exposure had no significant effect either on NEP activity or on the affinity and the number of BK receptors in lungs from rats treated with or without apocynin. We conclude that superoxide anions released from inflammatory cells in the airway may be involved in ozone-induced AHR. Inactivation of NEP or upregulation of BK receptors do not appear to be involved, but the possibility of localized changes cannot be excluded.

Role of neutral endopeptidase in bronchial hyperresponsiveness to bradykinin induced by IL-1 beta.

Interleukin-1 beta (IL-1 beta) induces bronchial hyperresponsiveness (BHR) to bradykinin but not to acetylcholine. We examined whether this was mediated through the inhibition of neutral endopeptidase (NEP) activity and/or through the enhancement of airway microvascular leakage (AML) by IL-1 beta. We administered human recombinant IL-1 beta (500 U) or saline intratracheally and 24 h later measured the airway responses to bradykinin (1 mM; 45 breaths). IL-1 beta-treated rats showed a decrease of 18.5 and 21.1% of NEP activity in the lungs and tracheobronchial tree, respectively (P < 0.05), associated with an augmented response in total lung resistance to bradykinin but with no increase in Evans blue dye extravasation used as a marker of AML. Phosphoramidon (0.1 and 1 mM; 90 breaths), an NEP inhibitor, induced a dose-dependent increase in lung resistance to bradykinin without further enhancing BHR induced by IL-1 beta. Bradykinin-induced AML was not enhanced by phosphoramidon in either saline- or IL-1 beta-treated rats. Similarly, after captopril (1 mM; 90 breaths), an inhibitor of angiotensin-converting enzyme, there was no further enhancement of BHR to bradykinin induced by IL-1 beta. BHR to bradykinin induced by IL-1 beta may result from an inhibition of peptidase activity, such as NEP and angiotensin-converting enzyme, and is not associated with an enhancement of AML.

Empirical estimation of a distribution function with truncated and doubly interval-censored data and its application to AIDS studies.

In this paper we discuss the non-parametric estimation of a distribution function based on incomplete data for which the measurement origin of a survival time or the date of enrollment in a study is known only to belong to an interval. Also the survival time of interest itself is observed from a truncated distribution and is known only to lie in an interval. To estimate the distribution function, a simple self-consistency algorithm, a generalization of Turnbull's (1976, Journal of the Royal Statistical Association, Series B 38, 290-295) self-consistency algorithm, is proposed. This method is then used to analyze two AIDS cohort studies, for which direct use of the EM algorithm (Dempster, Laird and Rubin, 1976, Journal of the Royal Statistical Association, Series B 39, 1-38), which is computationally complicated, has previously been the usual method of the analysis.

[Observation on the development of Coelomomyces indica in Culex tritaeniorhynchus larvae].

The present paper reports the development of Coelomomyces indica in Culex tritaeniorhynchus larvae. Experimental observations indicated that the first instar of the mosquito larvae can be infected with fungus. The hyphal bodies developed and produced sporangia in the haemocoele of the mosquito larvae. The sporangia were released in water from the dead mosquito larvae. The development of mature sporangia with zoospores was in water at 34 degrees C within 8-12 hours. The zoospores released from the mature sporangia within 1/2-1 hour under optimal conditions were very active in water and their activities decreased after 1 hour. The zoospores act as the infective stage for the Cyclops (host).

[Operative treatment for cervical vertigo caused by foramen arcuale].

The foramen arcuale of vertebral artery of altas is one of the causes of vertigo. Leven cases of the cervical vertigo caused by foramen arcuale were operated on. The patients were followed up for 3-6 years. The results were satisfactory. The mechanism of inducing vertigo by foramen arcuale was discussed. We demonstrated that excision of the foramen arcuale is available for cervical vertigo.

Airway hyperresponsiveness, elevation of serum-specific IgE and activation of T cells following allergen exposure in sensitized Brown-Norway rats.

T lymphocytes may play a regulatory role in the development of allergic airway hyperresponsiveness (AHR). We have studied the relationship between airway responsiveness and a number of immunological changes in Brown-Norway rats sensitized intraperitoneally and repeatedly exposed to ovalbumin (OVA) aerosol. Acetylcholine provocation concentration (PC)150 (the concentration of acetylcholine causing a 150% increase of base-line lung resistance) was measured and peripheral blood and bronchoalveolar lavage (BAL) cells were collected 18-24hr after the final exposure. Total and OVA-specific IgE in serum was measured by enzyme-linked immunosorbent assay (ELISA). Mononuclear cells were analysed by flow cytometry after labelling with monoclonal antibodies against CD2 (pan T-cell marker), CD4, CD8 (T-cell subsets) or CD25 (interleukin-2 receptor). There were significant differences in PC150 (P < 0.05) and in OVA-specific IgE levels in serum (P < 0.002); CD4+ T cells expressed a significantly increased level of CD25 immunoreactivity in BAL, but not in peripheral blood, of rats sensitized and exposed to OVA, compared with saline-exposed controls (P < 0.02). There was a significant correlation between CD25 expression and BAL eosinophil numbers (r = 0.74, P < 0.001), PC150 (r = 0.63, P < 0.003) and OVA-specific IgE (r = 0.77, P < 0.001). These data suggest that activated T cells may be involved in the regulation of allergen-induced AHR in a relevant animal model of allergic asthma.

Calcium channel antagonists: cardiovascular selectivity of action.

The activities of a series of calcium antagonists including nifedipine, verapamil, diltiazem and second-generation 1,4-dihydropyridines were determined in isolated vascular and cardiac preparations by tissue pharmacology and radioligand binding methods. The inhibitory effects against responses induced by 80 mM K+ depolarization in rat tail artery and against contractions evoked in electrically paced rat papillary muscle were determined. Comparison of these IC50 values defines a vascular/cardiac ratio as an index of relative activities in these tissues. Competition studies of the 1,4-dihydropyridines with [3H]PN 200,110 binding in neonatal rat myocytes under polarized (5.8 mM K+) and depolarized (50 mM K+) states yields an index of voltage-dependent binding. The 1,4-dihydropyridine calcium antagonists exhibit significantly higher vascular/cardiac ratios (31-877) than do the nondihydropyridines (1.9-2.1); with the exception of amlodipine, the second-generation 1,4-dihydropyridines have a higher vascular selectivity than the first-generation nifedipine. The ratios of voltage-dependent binding and vascular selectivity correlate well, consistent with a relationship between the two processes. A similar relationship is also observed in a small series of amlodipine derivatives bearing heterocyclic substituents at the 2-position of the 1,4-dihydropyridine ring. These results suggest that calcium antagonists, especially the 1,4-dihydropyridines do show different degrees of vascular selectivity and that voltage-dependent binding, influenced by chemical structure, is a major, but probably not exclusive, determinant of this selectivity.

[Confocal images of cochlear outer hair cells in the guinea pigs].

The effects of ACh, ATP and high K+ on intracellular calcium concentration Ca2+ in outer hair cells (OHC) of the guinea pig cochleae were observed with the use of Ca2+ sensitive dye Fluo-3 and laser scanning confocal microscope. Ca2+ increase induced by these chemicals had individual temporal and spatial features. Moreover, high K+ solutions induced contraction and bending of OHC too. Taking advantage of the "optical sectioning" facility of confocal microscopy, morphography of Fluorescein-labelled living OHC and PI-dyed nuclei was reconstructed three-dimensionally. The application of confocal microscopy in OHC physiological and morphological research was discussed.

[Hepatic artery chemotherapy-embolization and biological response modifiers for late hepatic carcinoma].

Since 1989, 63 cases of far-advanced cases of primary hepatic carcinoma were treated by alternating courses of hepatic artery infusion chemotherapy-embolization (HAIE) and biotherapy. For chemotherapy, mitomycin C, epiadriamycin (with lipiodol) and 5-Fu were used. Biotherapy consisted of systemic administration of gamma-interferon, interleukin 2 and transfer factor. Forty-four patients with late hepatic carcinoma were treated with HAIE alone, HAIE plus biotherapy resulted in more marked reduction in tumor size and serum AFP level as compared to that in the HAIE-treated patients. The 1- and 2-year survival rates of the two groups were 50.8% and 22% versus 40.9% and 6.8%, respectively.

[Adenocarcinoma of the cervix--analysis of 363 cases].

Form 1959 through to May, 1993, 363 patients with adenocarcinoma of uterine cervix were initially treated which accounted for 2.5% of all initially treated cases of cervical carcinoma. Patients' mean age was 50.5 years. Forty-six percent of the patients were post-menopausal. The main symptom was irregular vaginal and/or discharge. Over 88% of the cases were in stage II and III of the disease. They were treated with radiotherapy alone. The overall 5-year survival rate was 51.8%. The recurrence rate was 17.4% with a mean time to recurrence of 33.3 months. Thirteen patients recurred beyond 5 years. Recurrence was accompanied with lung metastasis in 17 patients and supraclavicular eymph node metastases in 11. Among patients whose tumor recurred the 1- and 2-year survival rate were 35.2% and 19.2% respectively. The most significant prognostic factors were the stage of disease, the size of the tumor and the histologic subtypes
