Solubilization and characterization of opioid binding sites from frog (Rana esculenta) brain.

Active opioid receptors were solubilized from frog (Rana esculenta) brain membrane fractions by the use of 1% digitonin. It was found by kinetic as well as by equilibrium measurements that both the membrane and the solubilized fractions contain two binding sites. For the membrane preparations, KD values were 0.9 and 3.6 nM, and Bmax values were 293 and 734 fmol/mg protein. For the solubilized preparations, KD values were 0.4 and 2.6 nM, an Bmax values were 35 and 266 fmol/mg protein. The stereospecificity of the binding did not change during solubilization. Both the membrane-bound and the solubilized receptors showed weak binding of enkephalin and mu-specific drugs, suggesting that they are predominantly of the kappa-type. The membrane-bound and the soluble receptors showed the same distribution of subtypes, i.e., 70% kappa, 13% mu, and 17% delta for the membrane-bound and 71% kappa, 17% mu, and 12% delta for the soluble receptors.

Shunt nephritis: a report on five children.

Of 5 children with glomerulonephritis with infected ventriculoatrial shunt, 3 had improved renal function after antibiotic therapy and removal of the infected shunt. 1 patient with endoextracapillary proliferative glomerulonephritis with 70% glomerular crescents developed a rapidly progressive renal insufficiency. Renal failure was successfully managed by hemodialysis and kidney transplantation. 1 patient died from extrarenal causes in the course of a septic episode.

Effects of daily corticosterone injections upon plasma glucose, insulin, uric acid and electrolytes and food intake pattern in the chicken.

The effect of daily injections of corticosterone (about 0.85 or 4.3 mg/day) on corticosterone-insulin interactions were investigated in ad libitum-fed chickens. The short term (0-5 hr after the injection) hyperglycaemic effect of corticosterone was accompanied by an increase in plasma uric acid levels but was not associated with consistent changes of alpha-amino non protein nitrogen, potassium, sodium or insulin. On the 8th and 15th days of experiment and after an overnight fast (i.e. about 20 hrs after an injection of corticosterone), the responses to a test-meal were measured and glucose tolerance was estimated. Birds receiving 4.3 mg corticosterone showed increased plasma glucose levels when fasting and just after (15 min) the test-meal and the glucose load. Glucose tolerance was slightly improved in birds injected with 0.85 mg corticosterone. In both tests, plasma levels of alpha-amino non protein nitrogen and possibly, uric acid (initial post-stimulation stages in birds treated with 4.3 mg corticosterone) were increased by corticosterone treatment, suggesting catabolism of amino acids. Additionally, in both tests, plasma insulin levels were increased in corticosterone-treated animals. Fasting plasma potassium levels were only decreased by the 4.3 mg dose of corticosterone. Plasma sodium levels were decreased 45-90 min following the test-meal, in all animals including controls. This effect remains unexplained. Following both the test-meal and the glucose tolerance test, plasma potassium levels decreased transiently. This effect may be compared to the hypokalaemic effect of exogenous insulin in mammals.(ABSTRACT TRUNCATED AT 250 WORDS)

[A new ophthalmologic refractometer assisted by a microprocessor].

A new, automated, objective refractometer has been developed in which the examiner looks at the ophthalmoscopic image, while a microprocessor analyzes the data automatically. The results can be displayed as well as printed. The reliability and accuracy of this instrument were evaluated. For spherical or cylindrical powers, 80 to 85% of the refractometer measurements differed from those of subjective refraction by only 0.50 diopter or less. For cylinder axis of eyes with 1.5 diopters or more of astigmatism, the automated and subjective refractions differed by no more than 5 degrees.

[Endoscopic teflon injection in the treatment of female stress urinary incontinence].

Fifty six women with urinary stress incontinence were treated with transurethral teflon injections. Forty eight (86 per cent) were completely cured, or showed considerable improvement. No abscesses or other complications were noted, and hospitalization was reduced to two or three days. The procedure is easy to perform and does not preclude subsequent surgery, if required. It is particularly suitable for obese or debilitated patients, or patients who have previously undergone surgery without success.

Povidone-iodine: its efficacy as a preoperative conjunctival and periocular preparation.

Even though advances in aseptic care have reduced the incidence of endophthalmitis, the conjunctiva still is not sterilized for surgery. This article presents a simple preoperative preparative procedure which, in a series of 225 surgical cases, reduced the incidence of positive postoperative cultures to 9.6%, a much lower percentage than has been reported previously.

[Efficacy of a new antisecretory agent (40 749 RP) on human gastric secretion induced by a meal (intragastric titration)].

An antisecretory drug of a new series, 40 749 RP, without anticholinergic or H2 receptor antagonist activities, was tested on meal-induced gastric acid secretion in 6 healthy volunteers. Gastric acid secretion and emptying of liquid were measured using intragastric titration. Oral dosages tested were 1, 2 and 4 mg/kg versus placebo. Inhibitions obtained were dose-related and expressed in percentage of the placebo values: 36 +/- 10 p. 100 for 1 mg/kg; 51 +/- 13 p. 100 for 2 mg/kg and 83 +/- 5 p. 100 for 4 mg/kg. Statistically significant correlation (p less than 0.001) was observed between maximal blood concentration of 40 749 RP and the percentage of secretory inhibition during the 90 min of the test. No change in gastrin response or in gastric emptying was observed whatever the dose.

A temperature-dependent block in spore differentiation is transduced intercellularly by Dictyostelium discoideum.

Strain HH31 of Dictyostelium discoideum is abnormal in the sense that at 27 degrees C it fails to produce spores and all or nearly all of the cells become stalk cells. In interstrain chimeras produced at the restrictive temperature, this strain shares with strains A3 or M28 the potential to control spore differentiation in neighboring cells. If HH31 is in numerical excess in slugs, then spore formation by A3 or M28 is suppressed by a factor of 20 or more. In contrast, an excess of A3 cells stimulates the formation of spores by HH31; the rescue affects fewer than one in 1000 HH31 cells though this represents a rescue coefficient of up to 2 X 10(3). The transcellular influence mediated by HH31 is rendered locally since it cannot traverse a thin membrane. It is not an artifact of unfavorable positioning of the test cells because in the slug they accumulate in the prespore region. The temperature-dependence of the self-differentiation of HH31 and its effect on A3 or M28 are altered coordinately in a spontaneous variant of HH31. We suggest that HH31 exerts, and is receptive to, a local effect on and by neighboring cells. This may be mediated by plasma membrane wheat germ agglutinin receptors, since their expression is also found to be under coordinate temperature control in HH31 and its variant.

Solubilization and separation of divalent cation dependent adenosinetriphosphatase in native gradient polyacrylamide slab gel.

A new method for the simultaneous solubilization and separation of different cation dependent adenosinetriphosphatases after non-denaturing polyacrylamide gel electrophoresis is described. Using a gradient system, 3 distinct divalent cation dependent adenosinetriphosphatase bands (Mg2+-, Ca2+-, and Mg2+ + Ca2+-dependent) could be separated on the same gel from NP-40 solubilized brain microsomal preparations. This indicates that these 3 adenosinetriphosphatases represent distinct molecular species.

Dynamics of propranolol dosing schedules.

Kinetic and dynamic data from 27 healthy male subjects were evaluated in a double-blind, randomized, double-crossover study to test the hypothesis that 180 mg/day propranolol twice and three times a day would provide much the same plasma levels and beta 1-blockade. The data indicate that propranolol twice rather than three times a day should be favored when beta 1-blockade is needed in therapy. The dynamic efficacy of the two schedules was the same and maximum concentration, AUC0-24, and 0-hr plasma propranolol values were higher after twice-than after three-times-daily dosing. The degree of beta 1-blockade (reduction in exercise tachycardia) was much the same on both dosing regimens at trough concentrations. These data indicate that both twice- and three-times-a-day dosing schedules provide well-sustained 24-hr beta-blockade and are probably interchangeable for therapeutic purposes.

Effects of chronic alcohol consumption and iron deficiency on maternal folate status and reproductive outcome in mice.

The influence of chronic alcohol consumption with or without iron deficiency on reproductive performance and folate status was studied. Female CBA/J mice were fed isocaloric liquid diets prior to and during pregnancy. A 2 X 2 factorial design was employed with ethanol-derived calories (EDC) and iron (Fe) as dietary variables. Groups received 0% EDC and 30 ppm Fe (CA); 0% EDC and 2 ppm Fe (CD); 20% EDC and 30 ppm Fe (EA); and 20% EDC and 2 ppm Fe (ED). Animals were killed on day 18 of gestation. Mean body weights were reduced in CD, EA and ED groups, while daily caloric intakes reduced only in CD and ED groups. Maternal values for hemoglobin, transferrin saturation and red cell folates decreased with iron deficiency and/or 20% EDC; hematocrit, serum iron and serum folates decreased only with iron deficiency. Blood ethanol levels were similar in EA and ED groups. Maternal liver total lipids and alcohol dehydrogenase activity increased only with 20% EDC, while dihydrofolate reductase activity decreased with iron deficiency and/or 20% EDC. Iron deficiency and/or 20% EDC adversely affected gestational performances in mice as indicated by increased resorptions and decreased percentages of live fetuses/litters and live fetal weights. Data indicate that iron deficiency and/or chronic ethanol consumption induce adverse effects on maternal reproductive performance of CBA/J mice possibly via alteration of folate metabolism.

The extent of volunteer faculty integration in teaching hospital settings.

In a survey conducted by the authors, volunteer faculty members were found to be generally satisfied with their role in medical education, despite their acknowledgement of problems involving respect and responsibility when working with academic colleagues. These problems and the indifference of the volunteers to them are delineated. The structural reasons for simultaneous recognition of an indifference to problems are discussed in relation to the erosion of the utilization and authority of volunteer faculty members.
