Purification and characterization of adult oligodendrocyte precursor cells from the rat optic nerve.

Oligodendrocyte precursor cells (OPCs) persist in substantial numbers in the adult brain in a quiescent state suggesting that they may provide a source of new oligodendrocytes after injury. To determine whether adult OPCs have the capacity to divide rapidly, we have developed a method to highly purify OPCs from adult optic nerve and have directly compared their properties with their perinatal counterparts. When cultured in platelet-derived growth factor (PDGF), an astrocyte-derived mitogen, perinatal OPCs divided approximately once per day, whereas adult OPCs divided only once every 3 or 4 d. The proliferation rate of adult OPCs was not increased by addition of fibroblast growth factor (FGF) or of the neuregulin glial growth factor 2 (GGF2), two mitogens that are normally produced by retinal ganglion cells. cAMP elevation has been shown previously to be essential for Schwann cells to survive and divide in response to GGF2 and other mitogens. Similarly we found that when cAMP levels were elevated, GGF2 alone was sufficient to induce perinatal OPCs to divide slowly, approximately once every 4 d, but adult OPCs still did not divide. When PDGF was combined with GGF2 and cAMP elevation, however, the adult OPCs began to divide rapidly. These findings indicate that adult OPCs are intrinsically different than perinatal OPCs. They are not senescent cells, however, because they retain the capacity to divide rapidly. Thus, after demyelinating injuries, enhanced axonal release of GGF2 or a related neuregulin might collaborate with astrocyte-derived PDGF to induce rapid division of adult OPCs.

Molecular cloning and tissue expression of porcine beta-defensin-1.

Beta-defensins constitute an emerging family of cysteine-rich antimicrobial peptides, which are particularly prominent at mucosal epithelial sites in mammals. Here we report the identification of a novel beta-defensin from porcine tissues, porcine beta-defensin-1 (pBD-1). The cDNA sequence of pBD-1 encoded a 64 amino acid prepro-peptide, which contained the beta-defensin consensus sequence of six invariantly spaced cysteine residues. Northern blot analysis showed that pBD-1 was expressed abundantly in tongue epithelia and that the expression was regulated developmentally. Using RT-PCR, pBD-1 mRNA was detected throughout the respiratory and digestive tracts and also in thymus, spleen, lymph node, brain, liver, kidney, urinary bladder, testis, skin, heart, muscle, bone marrow, peripheral blood neutrophils, alveolar macrophages, and umbilical cord. The wide expression of pBD-1 suggests that this endogenous peptide antibiotic may contribute to both mucosal and systemic host defenses in pigs, which may have implications for the use of porcine tissues and organs in xenotransplantation.

Effects of gender and estradiol treatment on focal brain ischemia.

The present studies were undertaken to investigate the effects of gender and estrogen treatment on focal cerebral ischemia in male and female rats. Focal ischemia was created by inserting a 3-0 surgical suture through the left cervical internal carotid artery to obstruct the blood flow into the middle cerebral artery (MCA). The MCA was reperfused by removing the suture in 40 min. All rats were sacrificed for measurement of infarct area after 24 h. In the first study, mortalities from MAC occlusion were 12.5% (2/16) each for intact male rats and intact female rats, and 23.5% (4/17) for ovariectomized (OVX) female rats. The coronal infarct area (mean+/-S. E.M.) was 9.5+/-1.0% for intact female rats, 16.6+/-1.6% for intact male rats (p=0.0001 vs. intact female rats), and 16.0+/-1.4% for OVX female rats (p=0.0002 vs. intact female rats). In a second experiment, OVX-female rats were administrated either 17beta-estradiol (E2) or its vehicle, hydroxypropyl-beta-cyclodextrin (HPCD), at 40 min after the onset of MCA occlusion. Mortalities were 40% (4/10) for vehicle treated OVX rats and 0% for E2 treated OVX rats. The coronal infarct area (mean+/-S.E.M.) was 19.3+/-1.8% for vehicle treated rats vs. 8.0+/-1. 2% for E2 treated rats (p<0.01). Serum estrogen levels for vehicle treated OVX rats were 14.5+/-1.2% pg/ml vs. 142.7+/-23.6 pg/ml for E2 treated OVX rats (p<0.01). These results strongly suggest that the level of circulating estrogens play an important role in protecting brain tissues against ischemia induced by MCA occlusion.

Evidence of hepatocyte apoptosis in rat liver after the administration of carbon tetrachloride.

In acute liver injury induced by the injection of CCl4, cell death has been attributed to the necrosis of hepatocytes in the centrilobular area. In the present study, we re-examined the hepatic injury evoked by CCl4 in rats and explored the possibility that apoptosis may also contribute to its pathogenesis. Apoptotic hepatocytes were identified and quantified by light and electron microscopy, the in situ immunohistochemical labeling of nuclear DNA fragmentation, flow cytometry, and DNA gel electrophoresis. We found that a substantial number of hepatocytes underwent apoptosis. Apoptotic changes were also observed in ballooned hepatocytes. Apoptotic hepatocytes increased in number at 3 hours and peaked at 6 hours after the CCl4 injection. Apoptotic bodies were sequestrated in the adjacent hepatocytes and sinusoidal cells. Double staining of the cells with immunostaining for phagocytes and terminal deoxynucleotidyl transferase-mediated dUTP nick end-labeling staining for labeling of DNA fragmentation showed that the majority of apoptotic hepatocytes were phagocytosed by Kupffer cells and macrophages. The results indicated that apoptosis occurs in the ballooned and injured hepatocytes of the centrilobular area. What occurs after CCl4 administration may be important in reducing inflammation, shortening the course of acute hepatic injury, and preventing the development of fibrosis.

Energy dependence of a new solid state diode for low energy photon beam dosimetry.

Any newly introduced radiation dosimeters must be evaluated for clinical use. This paper reports an evaluation of the energy response of a newly fabricated solid state diode for low-energy photon beam dosimetry. The diode, which has minimal buildup, is one of five models designed for in vivo patient dosimetry. Measurements were made using this diode for x-ray beams with kilovoltage potential from 20 to 100 kVp from a superficial x-ray treatment unit. In addition, measurements were also made using calibrated ion chamber designed for low-energy x-ray beams. A cone size of 20 x 20 cm was used and measurements were taken with diode and ion chamber placed at 35 cm from the source. The energy response was determined by taking the ratio of the diode measurement to the ion chamber measurement. The results show that the response of this new diode increases as the quality of the x-ray beam increases at low energy range; however, the percent difference of the detector response between the 70 kVp and 100 kVp x-ray beam is less than 1%, suggesting that this detector can be used as a dosimetry tool for the typical superficial x-ray treatments.

Neurovascular flow simulation review.

Computer simulation of the cerebral circulation has been carried out for three decades. We review the developments of cerebral circulation computer models of the last 30 years. Existing models are discussed in terms of fluid dynamics and possible clinical usage. Also the status and new achievements in some related research areas are summarized. Any new achievements in such areas would contribute to the progress of building a better model. A patient specific, highly predictive and reflective model could be a powerful clinical tool which would greatly benefit neurosurgeons and patients.

Synthesis and biological properties of the four optical isomers of 5-o-carboranyl-2',3'-didehydro-2',3'-dideoxyuridine.

The four isomers of the 5-o-carboranyl-2',3'-didehydro-2',3'-dideoxyuridine (d4CU) were synthesized and their antiviral activity and cytotoxicity in normal and cancer human cells determined. Coupling of silylated 5-o-carboranyluracil with the protected D/L 2,3-dideoxy-2-phenylselenenylribosylacetates provided after oxidative elimination and deprotection, the desired compounds. The presence of the electron deficient 5-o-carboranyl moiety on uracil influenced the yield of the various isomers. In general, the compounds demonstrated weak anti-human immunodeficiency virus activity in primary human lymphocytes. No marked difference in the biological profile was noted for the various optical isomers, suggesting that the high lipophilicity of these nucleosides imparted by the carboranyl moiety overrides stereochemical considerations in the 2',3'-didehydro-2',3'-dideoxyaglycon moiety.

Electrophysiological and biochemical responses of mouse vomeronasal receptor cells to urine-derived compounds: possible mechanism of action.

Receptor cells of the vomeronasal organ (VNO) are thought to detect pheromone-like molecules important for reproductive physiology. Several compounds derived from male mouse urine have been demonstrated to affect endocrine events in female mice. In the present study, the ability of these compounds to affect VNO activity was tested. In dissociated VNO cells held under voltage clamp conditions, application of dehydro-exo-brevicomin (DHB) evoked an outward current at negative holding potentials and an inward current at positive holding potentials. Under current clamp, DHB reduced action potential firing. Since DHB application caused a decrease in membrane conductance, this compound appeared to act by reducing inward current through closing an ion channel. Biochemical experiments tested the effects of DHB and 2-(sec-butyl)-4,5-dihydrothiazole (SBT) on cAMP levels in the VNO. A mixture of DHB and SBT decreased cAMP levels in VNO sensory tissue and had no effect on VNO non-sensory tissue. The results suggest that pheromones have an inhibitory influence on action potential generation and on cAMP levels in receptor cells of the VNO.

The role of protegrins and other elastase-activated polypeptides in the bactericidal properties of porcine inflammatory fluids.

The mammalian host response to infection includes the production and secretion of antimicrobial peptides from phagocytes and epithelial cells. Protegrins, a group of broadly microbicidal peptides isolated originally from porcine neutrophil lysates, were found to be stored as inactive proforms in porcine neutrophil granules but could be activated extracellularly by neutrophil elastase. We assessed the biological role of protegrins and other elastase-activated polypeptides in the microbicidal activity of neutrophil secretions and inflammatory fluids. When stimulated with phorbol myristate acetate (PMA), neutrophils generated stable microbicidal activity against both Escherichia coli and Listeria monocytogenes under normal-salt conditions and in the presence of 0 to 10% serum. The generation of these antimicrobial substances was dependent on neutrophil elastase, since it was inhibited by 1 mM N-methoxysuccinyl-Ala-Ala-Pro-Val chloromethyl ketone when it was present during activation, but not when this inhibitor was added afterwards. However, elastase-dependent activation of proprotegrins to protegrins in PMA-stimulated neutrophils was not inhibited by the presence of 1 to 2% serum. Porcine neutrophils also released antibacterial activity during phagocytosis of latex beads, and this too was dependent in large part on elastase-activated polypeptides, including protegrins. Moreover, protegrins were found at bactericidal concentrations in cell-free abscess fluid from naturally infected pigs. Taken together, these studies show that protegrins and other elastase-activated polypeptides are important stable antibacterial factors in porcine neutrophil secretions. The potential host defense role of elastase as an activating enzyme for the precursors of microbicidal peptides must be taken into account when therapeutic inhibitors of neutrophil elastase are evaluated for clinical use as anti-inflammatory agents.

[Ultrastructural study on neuromuscular junction of patients with lambert-Eaton myasthenia syndrome or mitochondrial encephalomyopathy].

OBJECTIVE: To study the ultrastructural changes of neuromuscular junction (NMJ) in patients with lambert-Eaton myasthenia syndrome (LEMS) and mitochondrial encephalomyopathy (MEMS). METHODS: The MMJ of internal intercostal muscles from patients with LEMS, MEMS or cardiac disease (control group) was labelled by HRP-BuTx and was quantitatively studied using image analysis system on electron microscopic photo. RESULTS: The ratio of nerve terminal area with postsynaptic membrane of NMJ was less in the MEMS group than controls. Both the length of presynaptic membrane and the ratio of the length of postsynaptic memberane to presynaptic membrane was larger in the LEMS group than in the MEMS group and controls, but the ratio of nerve terminal area with postsynaptic memberane area was less in the LEMS patients than in the MEMS group and controls. CONCLUSION: The areas of nerve terminal in the MEMS patients shrink because of mitochondrial damage. In LEMS patients however, the major change is the elongation of postsynaptic memberane and increasing of Ach receptors.

[Preliminary observation of immunotherapy on asthmatic guinea pig].

OBJECTIVE: To observe the effect of the anti-IL-5 monoclonal antibody(McAb) on eosinophils of the guinea pigs with asthma. METHOD: Sensitized guinea pigs by OVA were divided into two groups randomly, which were treated with the McAb and normal saline (NS), respectively. Difference of peripheral blood eosinophil count (PBEC) among basal pre-treatment and post-treatment, and the difference of bronchoalveolar lavage fluid eosinophil count (BALFEC) between two groups above were compared. RESULT: The PBEC after treatment with the McAb [(1.63 +/- 0.26) x 10(9)/L] was lower than that before treatment [(1.76 +/- 0.27) x 10(9)/L] (P < 0.05) and the BALFEC of treatment group with the McAb was lower than that with NS [(0.41 +/- 0.06) x 10(9)/L vs. (0.46 +/- 0.07) x 10(9)/L] (P < 0.05). CONCLUSION: The anti-IL-5 McAb has the ability to reduce the PBEC and the BALFEC.

[Analysis and toxicological evaluation of hazardous gases in sealed cabin].

82 volatile organic compounds (VOCs) of eight organic sorts and 3 target inorganic compounds in a sealed cabin that simulating the flying spaceship were identified and quantified for 5 d, the law of hazardous gas concentration variation was discussed, and the atmosphere toxicology was evaluated preliminarily. It provides a basis for detecting gas compounds and evaluating the atmosphere toxicology in the spaceship.

[Determination of ferulic acid and peoniflorin in siwu decoction prepared by different methods of yellow rice wine].

OBJECTIVE: To compare the influence of different methods with YRW (yellow rice wine) on the composition of Siwu decoction. METHOD: TCL-SM was used to determine the ferulic acid, paeoniflorin and matters dissolved in ether in Siwu decoction. RESULT: The Siwu decoction with YRW preparation has the highest contents, while the Siwu decoction containing Chinese Angelica root and Rehmammia root prepared with YRW has the lowest contents. CONCLUSION: The Siwu decoction prepared with YRW or added with YRW is more helpful for ferulic acid and paeoniflorin to be dissolved out than the Siwu decoction containing Chinese Angelica root and Rehmannia root prepared with YRW.

[Research of optic canal decompression by transnasal endoscopic approach].

OBJECTIVE: To find a better method to treat traumatic optic neuropathy. METHODS: Fourteen cases were treated by transnasal endoscopic sphenoethmoidal optic canal decompression in clinic. RESULTS: Nine of them had their visual acuity improved from NLP, HM, FC/50 cm, HM, NLP, LP, NLP, HM, NLP to 0.2, 0.3, 0.3(+2), 0.4, 0.06, 0.3, 0.1, 0.7, 0.12 after the operation. The clinical anatomy, attentions during operation, merits of operative approaches were discussed. CONCLUSION: Transnasal endoscopic sphenoethmoidal optic canal decompression is a good method for treatment of post-traumatic optic neuropathy.

[Effect of bovine plasma fibronectin on rat papilla cells in vitro].

OBJECTIVE: To observe the effect of exogenous fibronectin (FN) on the rat dental papilla tissues and cells. METHODS: Tooth germs of first mandibular molars obtained from 17-day-old rat (SD) embryos were cultured for 3, 6, 9 days on millipore filters in DMEM supplemented with 10% FCS, 50 mg/L Vitamin c. The dental papilla were isolated by trypsin treatment of homologous tooth germs and cultured on uncoated (control) and FN coated millipore filters for 3, 6 days, bovine plasma FN deposited on millpore filters by treated for 2 hours with 80 mg/L FN. RESULTS: As shown by histological and cytological data, some mesenchymal cells differentiated into odontoblasts and deposited predentine in cultured tooth germs. In cultured dental papillas, polarized functional cells were observed in FN coated group, odontoblast-like cells never differentiated in control cultures. CONCLUSION: FN may be involved in odontoblast polarization.

[Periodontal regeneration enhanced by BMP and GTR technique].

OBJECTIVE: To evaluate the effects of BMP on the repair of animal periodontal defects by using GTR technique. METHODS: 30 experimental mandibular buccal alveolar periodontal defects were produced surgically around premolar and molar teeth in 8 young mongrel dogs. The exposed root surfaces were curetted to remove the periodontal ligament and the cementum. The defects were devided into 3 equal groups (collagen, collagen + BMP, control). The results were examined histologically 1 month and 3 months respectively. RESULTS: In experimental groups the JE downgrowth was inhibital in the early healing, and the new attachment formation was promoted while comparing with the controls. Compound membrane (with BMP) showed the capacity to support more new bone and cementum formation than the other two groups. CONCLUSION: It is suggested that exogenous BMP can be used in GTR technique to promote periodontal regeneration.

[Endovascular treatment of carotid-cavernous sinus fistulae].

OBJECTIVE: To treat carotid-cavernous sinus fistulae mmunications between 43 patients with endovascular procedure. METHOD: Of the 43 patients 37 were male and 6 female. Traumatic and spontaneous CCF were noted in 39 and 4 patients respectively. 36 patients with CCF were given ICA alone, both internal and external carotid arteries in 7. Fistulae occlusion in 31 patients and ICA obliteration in 12 patients were achieved with transarterial detachable balloon technique. External carotid feeding arteries were embolized with silk suture in 7 patients. RESULT: Angiographic and clinical cure wer obtained in 39 patients and improved in 4. One patient was complicated by hemiplegia postoperatively. CONCLUSION: Endovascular balloon embolization is the first method of choice in dealing with carotid-cavernous fistulae.

[RP-HPLC method for determination of isorhynchophylline in plasma and its pharmacokinetics in rabbits].

A reversed phase HPLC method for determination of isorhnchophylline (IRHY) in rabbit plasma was developed. A Shimadzu LC-10A liquid chromatograph equipped with a Shimadzu SPD-10A multiwavelength detector and a Chromatopac LC-10A data processor were used. The analytical column was a 4.6 mm x 150 mm CLC-ODS. The mobile phase was a mixture of methanol-water (95:5), the flow rate was 1 ml.min-1. Detection was performed at UV 254 nm. The calibration curve was linear in the range from 0.016 to 16 micrograms.ml-1 with gamma = 0.9990. The lowest detection limit was 0.016 microgram.ml-1. The precision and accuracy of within-day and day-to-day ranged from 3.8% to 6.9%. The recoveries were 80.5% to 85.1%. The drug concentrtion-time curves of IRHY in rabbits after i.v. of 2 mg.kg-1 and 5 mg.kg-1 were shown to fit a two-compartment open model with half-times of 1.32 +/- 0.6 h and 1.25 +/- 0.09 h respectively. The absorption was fast after intraduodenal administration of IRHY (2 mg.kg-1 and 5 mg.kg-1) to rabbits with half-times of 1.75 +/- 0.18 h and 1.26 +/- 0.20 h, respectively. The bioavailability of IRHY was about 42.4% to 69.4%. The results showed that the method was simple and fast. The absorption and elimination were fast atfer intraduodenal administration or i.v. of IRHY to rabbits.
