[Metabolic profile of newborn calves and levels of immunoglobulins in the first days of life].

After examination of the clinical state of 128 new-born calves, blood was collected from their vena jugularis for the determination of blood actual pH value, concentration of lactic acid, pCO2, base excess, buffer base and standard acid bicarbonate. The course and difficulty of parturition exerted a significant influence on the vitality of the calves and on the studied parameters of acid-base state. In the normally born calves, compared with those after dystocia, the following values were obtained: pH 7.20 +/- 0.03 : 7.11 +/- 0.07, pCO2 = 8.4 +/- 0.9 : 10.0 +/- 1.1 kPa, base excess -2.30 +/- 2.10 : 5.80 +/- 4.60 mmol/l, buffer base 43.0 +/- 2.4 : 39.5 +/- 6.5 mmol/l, standard acid bicarbonate 22.3 +/- 1.8 : 19.6 +/- 4.1 mmol/l and lactic acid concentration 5.6 +/- 2.0 : 10.7 +/- 5.1 mmol/l. The differences were statistically significant (P less than 0.05) and statistically highly significant (P less than 0.01). The continual study of the blood actual pH value and lactic acid concentration in the calves in the first 24 hours of life showed that with the same trend of changes in calves after dystocia the initial values were less favourable and that their normalization lasted longer. Attention is drawn to the importance of dystocia for the rise of respiratory metabolic acidosis and its effect on the vitality of newborn calves, and/or on their survival. The discussion deals with the importance of immunoglobulin levels in calves in the first days after birth for their further development. The determination of antibody content in colostral serum from the first milking in 33 and 29 cows on two farms showed great drawbacks in quality. A satisfactory level of IgG was found only in 36.36% and 58.62% of the cows, and a satisfactory level of IgM only in 12.12% and 24.13% of the studied cows. The determination of immunoglobulin content in their calves two to three days from birth (33 + 33 animals) showed normoglobulinemia only in 24.24% and 15.15% of cases. In 33 and 29 cows on two farms the colostrum serum from the first milking had an average content of immunoglobulins of class G amounting to 27.99 +/- 20.25 mg/ml and 36.95 +/- 21.62 mg/ml, and class M amounting to 3.64 +/- 1.25 and 2.04 +/- 1.42 mg/ml. Three days from birth, their calves had an IgG content of 4.25 +/- 2.57 mg/ml and 3.99 +/- 1.86 mg/ml and an IgM content of 0.30 +/- 0.20 and 6.38 +/- 0.25 mg/ml.

Cell lysis by induction of cloned lambda lysis genes.

The lysis gene region of bacteriophage lambda, including genes S, R, and Rz, was cloned into the plasmid pBH20. In the recombinant plasmid, the lysis genes are expressed under the control of the lacOP region. Induction of this "lysis operon" with the lac inducer, IPTG, under conditions where transcription from the lacOP region is not subject to catabolite repression, results in a sharply defined lysis after 35 min. Premature lysis can be accomplished by cyanide, chloramphenicol, or chloroform, exactly as in bacteriophage lambda infected cells. The lysis gene region of an S- mutant was also cloned into pBH20. Induction of the S- lysis operon has no apparent effect on culture growth; however, large quantities of bacteriolytic activity accumulate intracellularly. Neither cyanide nor chloramphenicol causes lysis in the induced S- clones. Thus premature lysis appears to be entirely an S-dependent phenomenon. A model for the control of lysis in bacteriophage lambda infections is presented in which it is the accumulation of the S gene product in competition with a host "anti-S" protein that determines lysis timing.

[Monitoring of udder health by quarterwise measurement of initial milking samples for electric conductivity--results of serial examinations of individual animals and herds].

More than 400 cows were involved in three experimental series in which quarterwise measurement was applied continuously, even daly in a number of instances, to initial milking samples, with the view to establishing the electric conductivity of the milk. Electric cell count determination, high-speed mastitis tests, clinical examination of udders and secretions, and bacteriological milk tests were complementarily conducted. The major purpose of the effort was to find out whether milk conductivity measurement would be helpful in detecting subclinical mastitis. Conductivity measurement, in addition to being in fair agreement with milk cell count data, proved to be suitable an approach to high-continuity monitoring of udder condition, with no disruption of the production process being required. Specification of limit values and diagnostic value of differences between quarter milking data from one and the same udder were additional subjects of this study.

[Observation on the course of arterial hypertension after kidney transplantation].

The behavior of blood pressure was investigated in 65 transplantation (Tx) patients in a nephrological outpatient service for an average of 18.6 months after Tx. Before Tx 2/3 of the patients exhibited hypertension, after Tx this was reduced to 1/2. No correlation was found between behaviour of blood pressure and such factors as renal function, incidence of rejection episodes, prednisolonee dose, diabetes mellitus and urinary function, incidence of rejection episodes, prednisolon dose, diabetes mellitus and urinary tract infection of the Tx. Only in patients with glomerulonephritis as original renal disease there was in increased incidence of hypertension before and after Tx. Causes of hypertension are discussed and demonstrated in the case of Tx arterial stenosis.

Identification of mutants of pyruvate kinase from red blood cells by means of trypsinization, electrophoresis, kinetic properties and immunological methods.

Enzymopathies of pyruvate kinase (PK) are characterized by polymorphism. 11 different mutants of PK were detected in 12 analysed cases. The frequency of double heterozygotes of the PK deficiency is very high. The mutant forms have been identified by electrophoretic, kinetic and immunologic methods in red blood cells of patients and members of their families. The effect of trypsin has greatly increased the sensitivity of the differentiation procedures. Differences in activity and quality of the proteolytic systems in liver and kidney in comparison to red blood cells are responsible for inactivation and degradation of PK mutants in both organs. Homozygotes and double heterozygotes show clinic manifestations. The rigidity of red blood cells increases with the severity of the nonspherocytic haemolytic anaemia.

In vivo immunomodulating properties of two synthetic agents: azimexon and tuftsin.

Mice were submitted to various immunologic tests at different times after a single intravenous (IV) injection of azimexon or tuftsin in order to determine the mode of action of these chemically defined immunomodulators. Azimexon, (BM 12,531) an aziridine derivative, potentiated antibody responses to both thymus-dependent (TNP-KLH) and thymus-independent (TNP-LPS) antigens and DTH reaction to oxazolone when injected at least 1 day before the antigen. It activated macrophages, rendering them cytostatic for tumor cells, but depressed ADCC activity of spleen cells directed against antibody-coated CRBC. Tuftsin, a basic tetrapeptide, potentiated antibody response to TNP-KLH when injected at least 3 days before the antigen. The response to TNP-LPS was stimulated on days 1 and 3, but was slightly depressed on day 7. It rendered macrophages highly cytostatic for tumor cells but, as observed with azimexon, the activation process required 7 days to develop. ADCC was enhanced throughout the period of observation.

[Aspects of myocardial infarction treatment in patients of higher age (author's transl)].

The influence of a 6 weekly physical training of the performance in patients of the age between 60 and 70 years who have had a definitive myocardial infarction has been tested in phase II or rehability. Dayly in patient and under physicinal control the conditioning were executed in the time of 30 minutes on a bicycle--hometrainer. The personal intensity of training was choosed so that anwunted to 60--70% of heart-frequency obtained by the maximal exercise. Additional a terrain-training was taken part. Under conditioning a different relation of performance was recognized by patients (12 patients--performance decreased, 15 patients--constant performance, 33 patients--performance increased). To prevent a negative development of physical performance the result of performance--pulse (N/Hf) should be observe especially.

[Measurement of electric conductivity in first quarter milking samples for comparative assessment of udder health in large dairy cow herds].

Described in this paper is an experimental milking kit by which to collect first quarter milking samples directly from the milk pipeline system during full operation of the milking parlor. Measurement of electric conductivity of milk samples thus collected during the milking process may provide information on the occurrence of subclinical mastitis in the herd concerned. Discussed are results obtained from comparative measurement of milk conductivity, indirect determination of milk cell counts, as well as from clinical udder checks and bacteriological milk tests in seven large herds.

[Identification of pyruvate kinase variants from red blood cells using trypsinization and electrophoresis].

Enzymopathies of pyruvate kinase (PK) are characterized by polymorphism. Nine distinct variants of the L-type from 10 patients suffering from nonspherocytic haemolytic anaemia have been identified by electrophoretic and kinetic methods. Typical changes of the electrophoretic mobility and kinetic properties of the L-type and K-type of PK can be produced by incubation of cytolysates and tissue homogenates in the presence of trypsin. After trypsinization the three distinct forms of the L-type from liver and erythrocytes show identical mobility. Trypsinization of haemolysates has proved to be a diagnostic tool for the differentiation of PK variants. The results also allow to distinguish whether the mutation is located in the region of the peptide chain which is split off by trypsin or not. In three cases the occurrence of two instead of one enzyme form after trypsinization indicates a double heterozygote heredity of PK-deficiency. The appearance of the K-type isoenzyme of PK in red blood cells may be considered as a physiological event during a limited period of erythropoiesis. The K-isoenzyme was found only in samples of red blood cells containing erythroblasts. One of the patients has shown a still unknown form of PK. The properties of this form resembled those of the major part of PK from human placenta.
