[A simple method of preparation of keratin filaments and production of a polyclonal broad-spectrum anti-cytokeratin antiserum for immunohistochemical application in fixed and unfixed epithelial tissues].

Rabbits were immunized with 10 nm filaments of a mixture of cytokeratins which has been isolated from human heel callus material and reconstituted to filaments in vitro. The antisera to keratins (ASK) have been tested histologically at fixed and unfixed tissue samples by means of the indirect immunofluorescence and PAP technique. The ASK recognized specifically only the epithelial cells of skin, of the mucous membranes of mouth and digestive tract, of salivary glands, sweat gland and mammary gland, but did not react with hepatocytes or kidney cells. The following tumors, tested till now, reacted with the antikeratin antisera: epithelial and lymphoepithelial carcinomas of skin, mouth and digestive tract, carcinomas of salivary glands, mammary gland and thyroid gland, adamantinoma, basalioma of skin, and metastases from carcinomas.

The properties of skeletal muscle.

The authors review the musculoskeletal system and the controversy that surrounds methods for improving and strengthening it. Disorders brought on by over utilization, deficient working habits, lack of appropriate maintenance care and intercurrent stress and fatigue from repetitious daily tasks and poor sleeping habits are recognized and discussed. Also discussed are muscle structure and its relation to the contractive state, muscle energy requirements, motor control, source of muscle strength and factors modulating it, training adaptations in skeletal muscle, methods of strength training, erogenic aids including anabolic steroids and electrical stimulation and the pathologic states in muscles.

Comparative study of the efficacy, tolerance and side-effects of dexchlorpheniramine maleate 6 mg b.i.d. with terfenadine 60 mg b.i.d.

Dexchlorpheniramine maleate 6 mg b.i.d. was compared with Terfenadine 60 mg b.i.d. for efficacy in controlling Ragweed Hay Fever symptoms, as well as tolerance and occurrence of adverse reactions. The study was a randomized, multicentric, parallel group, blind evaluator design which involved 174 patients equally divided, eighty-seven receiving dexchlorpheniramine and eighty-seven terfenadine, for a period of 2 weeks during the latter half of August and the first half of September 1983. The study indicated that dexchlorpheniramine at the doses tested, was significantly more effective in controlling hay fever symptoms than terfenadine.

[Computer tomography of tumors of the lateral and 3d ventricle].

In the diagnosis of tumours of the supratentorial ventricles, CT leaves some questions unanswered. This is true of the differential-diagnostic demarkation and the determination of the initial site of the tumour. Frequently, ventriculography cannot be renounced for an additional clearing-up of the last mentioned question.

Solid-phase methods for sequencing nucleic acids. III. Simultaneous sequencing of different long RNA fragments using DE 81 anion-exchange paper.

A solid-phase method for simultaneous sequencing of different long RNA fragments has been developed using Whatman DE 81 anion-exchange paper as the support. The approach involves 8 operations including: immobilization of heat-denatured 3'-end labeled RNA fragment on DE 81 paper; washing; modification reactions; washing; aniline reaction; washing; RNA desorption by salt and ethanol precipitation. For modifying the RNA, the following reactions were selected for the routine: G with dimethylsulfate at 90 degrees C/sodium borohydride at 0 degrees C, A + G with diethylpyrocarbonate at 90 degrees C, U + C with hydrazine at 0 degrees C and C with hydrazine/5M NaCl at 0 degrees C. The losses of RNA material during the reactions with large excess of reactants were 50% during the reduction with NaBH4 and 30% during C-reaction. Almost no losses were observed during aniline reaction. The RNA could be recovered by desorption with 2M NaClO4 in 50-70% yield. The whole solid-phase procedure up to the sequencing gel takes about 2 hours and is much faster and more convenient than chemical RNA sequencing in solution according to Peattie, especially if many fragments are to be processed.

Plasma levels of oxytocin and vasopressin before, during and after parturition in cows.

In cows, plasma oxytocin and vasopressin were determined by specific and sensitive radioimmunoassays before, during and after parturition. In no case, oxytocin is elevated until the forelimbs of the foetus were distending the vagina and presenting at the vulva, supporting the view that endogenous maternal oxytocin does not primarily induce parturition. This finding is confirmed by simultaneous recording of myometrial electrical activity in 3 animals. Maximal oxytocin levels of 60.4 to 116 pg/ml plasma were measured at delivery of the foetus. The elevation in plasma vasopressin (peak values at delivery: 4.74 to 41.5 pg/ml) might be due, at least partially, to the increase in plasma osmolality during parturition.
