The possibility that Ca(2+)-ATPase from the plasma membrane-rich fraction of bovine parotid gland is ecto-Ca(2+)-dependent nucleoside triphosphatase.

1. Parotid plasma membrane nonpump low-affinity Ca(2+)-ATPase, which possesses high-affinity (Ca2+ + Mg2+)-ATPase activity, was characterized. 2. Purified Ca(2+)-ATPase hydrolyzed the nucleoside triphosphates, GTP, ITP, CTP, UTP, TTP (67-93% of ATP) and nucleoside diphosphates, ADP, GDP, IDP, CDP, TDP (12-40% of ATP) but not AMP and p-NPP. 3. The maximum activities of Ca(2+)- and (Ca2+ + Mg2+)-ATPases were obtained in the presence of 1 mM and 0.13 microM Ca2+, respectively. 4. The Km values for Ca2+ in Ca(2+)- and (Ca2+ + Mg2+)-ATPases were 0.2 mM and 22 nM, respectively. 5. The activities of both Ca(2+)- and (Ca2+ + Mg2+)-ATPases were found in the right-side-out-vesicles obtained from the plasma membrane-rich fraction. 6. These features suggest that Ca(2+)-ATPase is an ecto-Ca(2+)-dependent nucleoside triphosphatase.

Flow cytometric analysis of nuclear DNA content in tongue squamous cell carcinoma: relation to cervical lymph node metastasis.

The relationship between DNA ploidy and the incidence of cervical lymph node metastasis in 36 patients with tongue squamous cell carcinoma (SCC) was investigated. The aneuploidy rate of tongue carcinomas was 15/36 (42%), and the mean DNA index (DI) was 1.23, with a range from 0.87 to 3.54. Histologically identified cervical lymph node metastasis was observed in 11 cases, and the incidence of the cervical lymph node metastasis was significantly (P < 0.02) higher in the aneuploid cases (8/15) than in the diploid cases (3/21). Recurrence of the primary lesions was seen in nine cases 0.3-2.5 years after the initial treatment. No obvious difference in the incidence of the recurrence was noted, however, between the diploid (5/21) and the aneuploid (4/15) cases. These results indicate a significant relationship between aneuploidy and incidence of the regional lymph node metastasis, in contrast to the absence of a positive relationship between aneuploidy and recurrence of tongue SCC.

Pharmacokinetics of mefenamic acid in preterm infants with patent ductus arteriosus.

The pharmacokinetics of mefenamic acid (MA), 2 mg/kg, were studied in 17 preterm infants with symptomatic patent ductus arteriosus. They were given this dosage orally at 24 h intervals. There were marked inter-individual differences in some of the pharmacokinetic parameters after the first dose; peak plasma concentration (Cmax) varied from 1.2 to 6.1 micrograms/mL with a mean of 3.8 micrograms/mL, time to reach Cmax (tmax) varied from 2 to 18 h with a mean of 7.7 h and plasma half-life (t1/2) varied from 3.8 to 43.6 h with a mean of 18.7 h. The group of infants (10/17) who had ductus closure after the first dose had significantly lower clearance (P < 0.01), longer t1/2 (P < 0.01) and higher 24 h plasma concentration (P < 0.001) compared to the group of infants (7/17) who had no ductus closure after the first dose. It appeared that the plasma concentration of MA had to be above 2.0 micrograms/mL and maintained at this concentration for at least 12 h for MA associated with ductus closure in preterm infants to take effect. In view of the inter-individual variation of plasma MA concentration and the effective plasma concentration, we suggest that measurement of the plasma concentration should be done 24 h after the first dose. This might be useful for safe and effective therapy for infants with ductus closure failure after the first dose.

Structural modification of cell wall mannans of Candida albicans serotype A strains grown in yeast extract-Sabouraud liquid medium under acidic conditions.

The cell wall mannans of two Candida albicans serotype A strains, NIH A-207 and J-1012 (abbreviated as A and J strains, respectively), cultured in yeast extract-Sabouraud liquid medium at pH 2.0, contained neither a phosphate group nor a beta-1,2-linked mannopyranose unit (H. Kobayashi, P. Giummelly, S. Takahashi, M. Ishida, J. Sato, M. Takaku, Y. Nishidate, N. Shibata, Y. Okawa, and S. Suzuki, Biochem. Biophys. Res. Commun. 175:1003-1009, 1991). In this study, the mannans obtained from A and J strains grown in pH 2.0 medium (abbreviated as mannans A2 and J2, respectively) exhibited quite different reactivities against rabbit anti-C. albicans and anti-Saccharomyces cerevisiae sera compared with those of mannans from the corresponding strains cultured in conventional medium at pH 5.9 (abbreviated as mannans A and J, respectively). Namely, mannans A2 and J2 lost reactivity against the former serum but reacted with the latter serum to a higher extent than mannans A and J. In order to account for these difference in more detail, mannans A2 and J2 were subjected to acetolysis. Elution profiles of the acetolysates were completely different from those of acetolysates obtained from mannans A and J reported in our previous papers. The 1H nuclear magnetic resonance spectra of the oligosaccharides from mannans A2 and J2 obtained by this procedure indicate that the side chains are composed of alpha-linked mannopyranose units densely linked to the alpha-1,6-linked backbone. The long side chains containing one alpha-1,3-linked mannopyranose unit are markedly increased.

Oscillatory pressure-flow relationships of canine airways: a steady-state model for different gases.

We measured the airway pressure drop (delta Paw) between the trachea and the alveolus in three normal anesthetized paralyzed dogs, together with flow (V) at the trachea. Alveolar pressure was measured using a closed-chest modification of the standard alveolar capsule technique. Measurements were made during a period of sinusoidal ventilation at 0.83 Hz with a tidal volume of 35 ml/kg, after a 30-s period of apnea. We found that the amplitude of the swings in delta Paw decreased transiently after onset of sinusoidal ventilation. We established that this decrease was due to a reflex bronchodilatation by making additional measurements on vagotomized dogs. We developed a mathematical model to account for the steady-state data that included both laminar and turbulent flow, airway wall elastance, airway gas inertance, and the Bernoulli effect at the site of tracheal pressure measurement. The model accurately described the data obtained from each dog with four different gas mixtures containing air, helium, neon, and sulfur hexafluoride. We also constructed Moody plots from the measured delta Paw-V relationships, after removing the estimated contributions of gas inertance and the Bernoulli effect from delta Paw, and found evidence of the effects of changing airway dimensions throughout a breath, V unsteadiness, and differences between inspiratory and expiratory resistances.

Frequency dependence of pulmonary and chest wall mechanics in young and adult cats.

The frequency (f) dependence of pulmonary and chest wall mechanics was assessed in nine kittens and four cats. Kittens and cats were anesthetized, paralyzed, and mechanically ventilated at various f between 0.13 and 1.6 Hz and 0.09 and 0.79 Hz, respectively. Resistance and dynamic compliance pertaining to the respiratory system (Rrs and Cdyn,rs), lungs (RL and Cdyn,L), and chest wall (RW and Cdyn,W) were estimated by fitting a single-compartment model to data obtained from regular ventilation. Static lung and chest wall compliances (Cst,L and Cst,W) were computed from quasi-static pressure-volume data. Lung tissue resistance (Rti) was estimated with alveolar capsules in open-chest animals. The f dependence of the two-compartment viscoelastic model of the respiratory system was assessed by computing the effective resistance [Rmod,rs(omega)] and compliance [Cmod,rs(omega)] from data obtained at the lowest experimental f. Both Cdyn,L and Cdyn,W decreased with increasing f in all animals. Cdyn,L/Cst,L and Cdyn,W/Cst,W were lower in kittens than in cats. RL and RW decreased markedly with f in all animals. Rti/RL showed a marked f dependence, its values being similar in both young and adult cats at their respective resting f. CstW/Cst,L ratio was higher in kittens than in cats. A better agreement was found between Cmod,rs(omega) and Cdyn,rs than between Rmod,rs(omega) and Rrs.

Incomplete suppressive effects of acetylsalicylic acid on the heat sensitization of canine testicular polymodal receptor activities.

1. Using canine testis-spermatic nerve preparations in vitro, we studied the effects of acetylsalicylic acid (ASA), an inhibitor of cyclooxygenase, on the polymodal receptor response to heat stimulation itself and on the sensitizing effects on the heat and bradykinin responses induced by strong heat stimulation (55 degrees C for 30 s). 2. Greater heat responses tended to be strongly suppressed by ASA (550 microM), with a significant correlation observed between the response magnitude and the magnitude of the suppression (r = 0.88 for the response at 45 degrees C; r = 0.83 for the response at 48 degrees C). The change in the heat response at 48 degrees C from 1.18 +/- 0.19 to 0.62 +/- 0.11 imp/s was statistically significant (n = 21, P < 0.01). 3. After stimulation at 55 degrees C in the presence of ASA, resting discharges at 34 degrees C appeared in only 2 of the 21 units tested. The responses at 45 and 48 degrees C, however, were significantly potentiated after this stimulation (those at 45 degrees C from 0.12 +/- 0.05 to 2.19 +/- 0.51 imp/s, mean +/- SE; those at 48 degrees C from 0.62 +/- 0.11 to 2.42 +/- 0.49 imp/s; P < 0.01, n = 21 for both). Sensitization was observed up to 50 min after stimulation at 55 degrees C.(ABSTRACT TRUNCATED AT 250 WORDS)

Germline mutations of the p53 tumor suppressor gene in children with osteosarcoma.

PURPOSE: We investigated the possibility that a significant proportion of children with osteosarcoma harbor germline mutations of the p53 tumor suppressor gene and, therefore, this subgroup of pediatric cancer patients should be considered for large-scale predictive testing. PATIENTS AND METHODS: Genomic DNA extracted from peripheral-blood leukocytes from 235 unselected children with osteosarcoma from 33 institutions were screened for the presence of germline p53 mutations using constant denaturant gel electrophoresis (CDGE). Exons 5 through 8 were evaluated in all patients and exon 2 and exon 9 were analyzed in 59 and 95 patients, respectively. Those samples that showed aberrant migration on CDGE were sequenced or analyzed by restriction enzyme digestion of polymerase chain reaction (PCR) products to confirm the nature of the gene alteration. RESULTS: In 18 samples, CDGE showed fragments of the p53 gene with altered electrophoretic mobilities compared with wild-type p53. DNA sequencing showed that 11 samples had an identical, previously described polymorphism. The other seven contained heterozygous p53 mutations located in exon 5 (n = 3), exon 6 (n = 1), exon 7 (n = 1), and exon 8 (n = 2). Six alterations were missense mutations and one was a nonsense mutation. Three of these patients had first-degree relatives with cancer. One of these three kindreds had a family history consistent with Li-Fraumeni syndrome (LFS). CONCLUSION: We identified germline p53 mutations in seven of 235 (3.0%) children with osteosarcoma. Four of these mutations were found in patients who did not have first-degree relatives with cancer. Although genetic transmission of the altered p53 gene could not be tested in this survey because of how it was designed, it is possible that predictive testing for p53 mutations could identify unaffected relatives of gene carriers who also have a high risk for the development of cancer. This study provides evidence for the importance of considering children with osteosarcoma for predictive testing for germline p53 mutations.

Enhancement of the anti-tumor activity of recombinant interleukin-2 (rIL-2) by immunocomplexing with an F(ab')2 fragment of murine monoclonal antibody against rIL-2.

We have investigated the biological properties of an immune complex composed of recombinant interleukin-2 (rIL-2) and an F(ab')2 fragment of a monoclonal antibody against rIL-2 in mice for the induction of killer cells and anti-tumor activity, as well as the pharmacokinetic properties of the immune complex injected subcutaneously into mice. The immune complex demonstrated sustained serum rIL-2 levels, with a 2.4-times longer "mean-residence-time" than free rIL-2. A more significant portion of rIL-2 was detected in lymph nodes after the subcutaneous injection of the immune complex than with rIL-2 alone. Splenic lymphocytes from mice given the immune complex showed higher killer cell activity against YAC-1 and P815 cells than those from mice given rIL-2 alone. The immune complex also exerted a more significant anti-tumor effect in a dose-dependent manner in Meth-A fibrosarcoma-bearing mice than dit rIL-2 alone.

An ex vivo assay for estimating the antiviral state of hepatocytes.

An ex vivo antiviral assay was established which uses hepatocytes from mice given recombinant mouse interferon-beta (rmIFN-beta). Assay results were compared with results obtained with a 2',5'-oligoadenylate synthetase (2-5AS) assay. rmIFN-beta was intraperitoneally administered to C3H mice and the antiviral state of their liver parenchymal cells was evaluated in an in vitro cytopathic effect assay. In this assay, cells are infected with vesicular stomatitis virus (VSV) and surviving cells are determined colorimetrically. The antiviral state was measured as the resistance of hepatocytes to VSV infection with increasing doses of rmIFN-beta. The antiviral state correlated well with the dose-dependent increase in hepatic 2-5AS activity. This good correlation suggests that induction of 2-5AS mediates the antiviral action of interferon in liver tissue. This ex vivo assay could be a useful tool for estimating the ability of hepatocytes to resist hepatitis virus infection.

Pharmacokinetic alteration in rats of recombinant interleukin-2 (rIL-2) by immunocomplexing with a monoclonal antibody against rIL-2.

We have investigated the pharmacokinetic alteration in rats of recombinant interleukin-2 (rIL-2) by immunocomplexing with a monoclonal antibody against rIL-2. Serum rIL-2 levels after the intravenous administration of the immune complex at a dose of 100 micrograms/rat as rIL-2 were significantly higher than those after intravenous administration of rIL-2 alone at the same dose. Pharmacokinetic analysis indicated that the distribution volume of rIL-2 decreased from 74.0 to 10.3 ml/rat, while the elimination rate of rIL-2 was little changed by immunocomplexing with the antibody. On the other hand, serum rIL-2 levels after the subcutaneous administration of the immune complex at a dose of 100 micrograms/rat as rIL-2 were sustained longer than those after the subcutaneous administration of rIL-2 alone at the same dose, and Tmax shifted from 0.83 to 3.0 h by immunocomplexing with the antibody. Pharmacokinetic analysis also revealed that the mean-residence-time of rIL-2 increased from 1.98 to 6.52 h, and the area-under-the curve of rIL-2 decreased slightly, from 834 to 548 ng.h/ml, by immunocomplexing with the antibody.

[An epidemiological study of urogenital cancer in Gunma Prefecture].

The incidence and mortality rate of urogenital cancers in Japan are both low compared to those in western countries. However, the incidence and mortality patterns of cancer in Japan are currently becoming closer to those of western countries, and the importance of urogenital cancers is increasing. We conducted an analysis of urogenital cancers in Gunma Prefecture. The subjects were newly detected urogenital cancer patients living in Gunma Prefecture diagnosed between 1985 and 1992. Details were as follows: prostate cancer 1411, bladder cancer 1253 (male 937, female 316), renal cell carcinoma 411 (male 287, female 124), renal pelvic and ureter cancer 187 (male 127, female 60) and testicular cancer 162. Incidence rate was calculated by year, district and age, and was expressed per 100,000/year and was adjusted to world population. Regarding the incidence rate per year for males, that of prostate cancer and renal cell carcinoma increased dramatically from 8.3 to 13.6 and from 1.1 to 3.2, respectively. Incidence rate of other cancers in males showed a slight increase or remained almost stable. Incidence rate by year for females showed a slight increase or remained almost stable as a whole. Gunma Prefecture was divided into 10 districts by the range of daily life of people and the incidence rates of prostate cancer, bladder cancer and renal cell carcinoma for each district were calculated. Incidence rate of prostate cancer tended to be higher in the northern parts of the prefecture, while that of bladder cancer showed no detectable trend.(ABSTRACT TRUNCATED AT 250 WORDS)

Clinical significance of prostate specific antigen for early stage prostate cancer detection.

The characteristics of serum prostate specific antigen (PSA) in normal Japanese men were studied in 1480 subjects examined by mass screening (MS) for prostate cancer (Pca) in Gunma Prefecture in 1992. The serum PSA concentration was correlated with patient age. The average serum PSA level increased by 0.04 ng/ml/year. The upper normal limits (95 percentiles) of age specific PSA for normal men are 1.33 ng/ml for those aged 39-49 years, 3.65 ng/ml for those aged 50-59 years, 4.06 ng/ml for those aged 60-69 years, 5.09 ng/ml for those aged 70-79 years and 5.66 ng/ml for those aged 80-89 years. Among 227 normal men examined by our MS in 1991 and 1992, the PSA velocity (PSAV) was calculated to be 0.05 ng/ml/year. Among 10 Pca patients with normal PSA levels (< 6 ng/ml) detected previously by our MS, three had an abnormal PSAV. We demonstrated the possibility that PSA density could distinguish between Pca and benign prostate hypertrophy. The significance of PSA as a Pca screening modality should be evaluated across multiple age ranges and in combination with previous PSA data and/or prostate volume estimated by sonography.

[Concentrations of new quinolone agents in serum and prostate tissue].

Twenty five patients with benign prostate hypertrophy were administered ofloxacin (OFLX) simultaneously with norfloxacin (NFLX) in 6 patients, ciprofloxacin (CPFX) in 10, tosfloxacin (TFLX) in 5 and enoxacin (ENX) in 4 patients. The dose of these new quinolones was 100 mg (except 150 mg of TFLX). Their concentrations in the serum and the prostate tissue were measured by high performance liquid chromatography (HPLC). The serum concentration of OFLX was significantly higher than that of NFLX, CPFX, TFLX or ENX. The prostate tissue concentration of OFLX was significantly higher than that of CPFX or TFLX. The ratio of tissue versus serum concentration of each new quinolone agent was not significantly different. The high OFLX tissue concentration appeared to be caused by the high serum concentration.

[Usefulness of newly developed 99mTc-labeled STA-R12 for tumor imaging].

We synthesized four new tumor imaging agents, 99mTc-labeled metalloporphyrins (99mTc-STA-R12, -STA-R21, -STA-RN101 and -ATN-12) for the tumor imaging. We compared the differences of tumor imaging potency among these agents in CDF1 mice implanted with colon 26 tumor. Tumor images with these agents were obtained by using digital gamma-camera (RC135-E, Hitachi, Tokyo) and the biodistributions were analyzed by computerized medical radionuclide imageanalyser (RP-200, Hitachi, Tokyo). The highest tumor organ ratio and the excellent tumor image were obtained by 99mTc-STA-R12 (13,17-Bispropanyl aspartic acid-3-ethenyl-8-[N,N',N",N"-tetrakis (carboxylmethyl-2,7,12,18-tetramethyl-porphyrinato]-manga nes e(III). The maximum concentration of 99mTc-STA-R12 in tumor tissue was observed at around 120 minutes after i.v. injection. On the contrary, the uptake rates of other organs and tissues such as liver, brain, muscle, lung, bone and blood continuously decreased. The rapid accumulation of STA-R12 in cancer tissue and the clearance from other tissue suggested a potential usefulness of this compound for tumor imaging agent.

Comparative analysis of engraftment after cryopreservation of peripheral blood stem cell autografts by controlled- versus uncontrolled-rate methods.

Peripheral blood stem/progenitor cells (PBSC) were cryopreserved by different methods and their clonogenic viabilities were compared. The traditional cryopreservation method involves controlled-rate freezing with 10% dimethyl sulfoxide (DMSO) and a programmed freezer (PF). The alternative method incorporates 6% hydroxyethyl starch and 5% DMSO without PF. In this non-randomized study, we analyzed the data of 24 patients (aged 1-17 years) who were in their first complete remission and who had undergone PBSC autograft (PBSCT) for high-risk acute lymphoblastic leukemia, to determine the effects of these two different methods of cryopreservation on time to engraftment and transfusion requirements after PBSCT. In all patients, PBSC were collected within 5 months of diagnosis and all had been conditioned with the high-dose MCVAC regimen (MCNU, cytosine arabinoside, etoposide and cyclophosphamide) without total body irradiation. Twelve patients received PBSC that had been cryopreserved by the uncontrolled method without PF and the data were compared by actuarial analysis to those of 12 historical controls whose cells had been frozen by PF. No difference was observed in the time to engraftment of granulocytes and platelets or the transfusion requirements between the two groups. Our results indicate that, in terms of preserving engraftment potential, the simplified uncontrolled-rate cryopreservation method is at least as effective as the traditional controlled-rate freezing procedure with PF.
