[Quantitative determination of amino acids from blood-stained filter papers using high-performance thin layer chromatography and a TLC-scanner (author's transl)].

A method is described to determine amino acids by extraction from pieces of bloodstained and dried filter papers and following dansylation and separation by High-Performance Thin Layer Chromatography (HPTLC). Quantitative determination is possible by means of a TLC-Scanner measuring the fluorescent light of the dansylated amino acids. The possible use of this procedure as screening test is discussed.

[Nosocomial infection of hospitalised patients with chemotherapy-resistant strains of Klebsiella pneumoniae (author's transl)].

In a 500-bed general hospital the incidence of highly resistant strains of Klebsiella pneumonia was examined. About 20% of examined patients in the surgical and medical departments were carriers. Spread of the organism was via the hands of personnel and patients as well as various objects. The use of broad-spectrum antibiotics, especially ampicillin and cefalotin is thought to be the chief cause of the high incidence of Klebsiella emergence. About 90% of carriers had previously received antibiotics.

Effect of indomethacin on collagen metabolism of rat fracture callus in vitro.

The collagen metabolism of callus tissue from closed, non-immobilized rat femoral fractures was studied in vitro after in vivo treatment with indomethacin 2 mg/kg/day for 6, 9 and 12 days after fracture. Total hydroxyproline synthesis and incorporation of hydroxyproline into collagen were found to be significantly increased after indomethacin treatment, whereas no significant difference was found regarding collagen resorption. The results suggest that the recently demonstrated inhibition of fracture healing by indomethacin is not brought about by an inhibition of collagen synthesis.

Experiments on the detection of the carcinogenic N-nitroso-dimethylamine in the urine of rats after oral administration of the analgesic amidopyrine and nitrite.

N-Nitroso-dimethylamine (NDMA) was excreted unchanged in the urine of rats after oral administration of the compound in amounts ranging from 0.003% at a dose of 2 mg/kg to 0.08% at a dose of 15 mg/kg. By measurement of the NDMA excreted after oral administration of amidopyrine and nitrite, the extent of nitrosation in vivo was estimated to be about 30%. For clinical investigation of NDMA formation in patients treated with amidopyrine, however, we consider this method to be too insensitive.

Enhancement of tumor growth following immunization with Bacillus Calmette-Guérin cell walls.

The effect of preimmunization with Bacillus Calmette-Guerin cell walls (BCGcw) on tumor growth was studied with the use of transplantable Morris Hepatoma 3924a in inbred ACl rats. There was an increase in tumor incidence, an increase in the i.m. tumor growth rate, and an increase in the average i.m. tumor mass in animals that were preimmunized with BCGcw. The BCGcw-induced enhancement in tumor growth was dependent upon the amount of tumor present at the time of BCGcw immunization. No enhancement occurred in animals immunized with BCGcw 12 days after the inoculation of tumor cells.

Carcinogenic N-nitro-dimethylamine from the reaction of the analgesic amidopyrine and nitrite extracted from foodstuffs.

The reaction of the analgesic amidopyrine (100 mg) with nitrite extracted from cured meats and from spinach in varying degrees of spoilage was studied. Unde physiological conditions the carcinogenic dimethylnitrosamine was formed at milligram levels at nitrite concentrations as low as 4 mg (in 175 ml extracted from 100 g boiled ham). The rate of decrease in concentration in the human stomach after ingestion of amidopyrine and of nitrite contained in boiled ham or in a broth from boiled ham was also measured.

[Examinations of the possibilities to nitrosate nicotine and nornicotine and of the formation of N-nitrosonornicotine in the stomach of smokers (author's transl)].

It was examined how easily the tobacco-smoke constituents, nornicotine, and nicotine, are nitrosated in an acidic aqueous medium, and if a formation of nitrosamines in the stomach of smokers is possible after swallowing amine-containing saliva. More than 10% of the nornicotine present in tobacco-smoke is dissolved in saliva during smoking. While from the tertiary amine, nicotine, only a small yield of N-nitrosonornicotine was obtained, from nornicotine an about 50 times higher yield of nitrosocompound was formed than from N-methylbenzylamine, a secondary amine of moderate basicity and hence of moderate reactivity toward the nitrosating agents. The amounts of nitrite in the neutral stomach contents varied between 15 and 250 ppm; in the acidic stomach contents no nitrite was detected. No increase was observed during cigarette smoking. In 61 gastric juice of volunteers who smoked a total number of 280 cigarettes no nitrosonornicotine and no nitrosopyrrolidine (pyrrolidine is present in tobacco-smoke in similar amounts as nornicotine) was detected (detection limit: 5 ppb).

[Bacteriological study of redon drains in accident surgery (author's transl)].

After 'clean' operations, 121 Redon drains from 80 patients were cultured. 17.4% of Redon tips were contaminated, but microorganisms were found in only 7.4% of wound secretions aspirated from them before they were removed. Only Gram-positive cocci, most of them of questionable pathogenicity, were found. Although drains or wound secretions were found to be contaminated in 21 patients, clinical evidence of disturbed healing occurred in only two patients. The presence of microorganisms provided no pointer to later healing difficulties. Culturing Redon tips is, therefore not a reliable method for early recognition of wound infections. Antibiotics should not be used merely on evidence of bacterial contamination of Redon tips without any clinical signs of infection.

Transport and conjugation of 14C-bilirubin during acute and chronic cholestasis in the cholecystectomized dog.

The role of the lymphatics in the regurgitation of bilirubin during acute and chronic extrahepatic cholestasis in the dog was studied after intravenous injection of 14C-bilirubin. In acute cholestasis the injected labelled bilirubin was rapidly removed from plasma and reappeared in thoracic duct lymph as labelled bilirubin conjugates. No labelled conjugates were at any time demonstrated in plasma. This is most easily explained as a result of a bilio-lymphatic regurgitation. In chronic cholestasis the labelled bilirubin conjugates were delivered back into the blood, and no specific transport function of the lymph was found. Our results indicate a change in liver cell secretory direction in long-standing cholestasis.
