Neutrophil alkaline phosphatase activity in patients with neoplasma during surgical treatment.

In 22 patients with alimentary tract neoplasma, granulocyte alkaline phosphatase activity was determined using the Kaplow's histochemical method. The enzyme activity was determined in peripheral blood before surgical intervention, in blood from vessel draining of tumor before its excision and in peripheral blood 2--3 weeks after excision of tumor. In paralleled tests the enzyme activity was estimated in peripheral blood of 22 healthy individuals. The study indicates that alkaline phosphatase activity of granulocytes is evidently lower in comparison to the controls. Decrease of this enzyme activity in granulocytes from vessels draining tumor and increase of this activity after removal of the tumor suggest that tumor may affect the activity of this enzyme. It seems possible that for decrease of bactericidal capacity of granulocytes in patients with neoplasma the reduction of alkaline phosphatase activity of granulocytes may partly be responsible.

Granulopoietic activity (GA) and colony stimulating factor (CSF) in myeloproliferative diseases.

Granulopoietic activity (GA) in vivo and colony stimulating factor (CSF) in vitro were measured in sera of patients with acute and chronic myeloid leukaemia, chronic lymphatic leukaemia and Hodgkin's disease. Before testing sera were dialysed for removing unspecific lipoproteid inhibitors of CSF. We have found a lower activity of CSF in these diseases than in the control group. GA was even below zero stimulation especially in chronic leukaemias. This confirms the presence of inhibitory effect of the tested sera on granulopoiesis in vivo and indicated that GA tested in vivo may be more favourable for the prognosis of leukaemias.

Bactericidal capacity of plasma and granulocytes against Escherichia coli in the small-for-dates newborn.

Infections due to Gram-negative bacteria in newborns and especially small-for-dates newborns sometimes run a dramatic course and have a high mortality rate. The bactericidal effect of plasma and granulocytes was determined by the method of Balch et al [1] in 27 small-for-dates term newborns and 19 normal term newborns. The number of killed bacteria was practically the same in th mixture of granulocytes and plasma in both groups. On the other hand, a threefold decrease of the bactericidal effect of plasma was observed when it was applied alone, without granulocytes. This may indicate that the defect is more in the plasma than in the granulocytes, and is probably due to the failure of class IgM antibodies to cross the placental barrier.

Nitroblue tetrazolium test in the term small-for-dates newborn.

The value of the nitroblue tetrazolium reduction test was studied using granulocytes obtained from 41 small-for-dates and 23 normal term newborns. The unstimulated test did not show any differences between the two groups, while after latex stimulation a statistically significant decrease was noted in the reduction by granulocytes obtained from small-for-dates newborns in comparison to those obtained from normal newborn infants. The results showed a wide scattering especially in the small-for-dates newborns. This may indicate that in some of these babies the granulocytic enzymes are defective to reduce the nitroblue tetrazolium to purple formazan.
