Tissue inhibitors of granulocyte-macrophage (GM) colony formation.

The effect of mice tissue homogenates on the inhibition of granulocyte-macrophage colony formation in vitro was studied. The potent inhibitory activity was displayed by homogenate from liver. The lowest values were found in the kidney and heart. An interrelationship between the rate of inhibition and protein concentration in the homogenates was noted. Biological role and the type of inhibitory activity are briefly discussed.

Serum lysozyme activity in patients with breast cancer treated by mastectomy and radiation.

In 37 patients with breast cancer, lysozyme activity in undiluted and diluted sera was determined. Blood was collected from peripheral vein before surgical intervention, during surgery from vein draining when radiotherapy was finished. In parallel experiments the lysozyme activity was tested in peripheral blood of 40 healthy persons. Increased lysozyme activity both in undiluted and diluted sera was observed and radiotherapy did not change the activity of the tested enzyme.

Inhibition of granulocyte-macrophage (GM) colony formation by sera of patients with neoplasma of digestive tract in the course of surgical treatment.

The effect of sera from patients with neoplasm of the digestive tract obtained during intervention has been tested on granulocyte-macrophage (GM) colony formation. Independently of the source of blood (peripheral or tumour draining vein) and the time of sampling (during or 2-3 weeks after operation) the tested sera suppressed GM colony formation. The highest decrease was caused by blood sampled from the vein draining the tumour. It is concluded that tumour cells may be a source of the GM colony formation inhibitory activity. The possible character of the activity is discussed.

Neutrophil acid phosphatase activity in patients with gastric or rectum carcinoma during surgical treatment.

In 22 patients with alimentary tract carcinoma granulocyte acid phosphatase activity was determined using the histochemical method of Suzuki. Enzyme activity was determined in peripheral blood 3 times, viz. before surgical intervention, in blood from a vessel draining the tumour before its excision, and in peripheral blood 2-3 weeks after excision of the tumour. In parallel tests, enzyme activity was established in peripheral blood of 22 healthy individuals. The results showed that acid phosphatase activity of granulocytes is lower than in the controls, especially in granulocytes collected from the vessel draining the tumour. The lower acid phosphatase activity in granulocytes from patients with carcinoma was mainly due to the lower percentage of positive cells. After removal of the tumour acid phosphatase activity of the granulocytes increased in the majority of cases. It is assumed that an enhanced exocytosis of lysosomal enzymes in the course of tumour disease was responsible for the decrease of activity in the granulocytes.

The activity of leukocytic alpha-amylase in leukemias.

The alpha-amylase activity has been determined in leukocytes and sera of patients with acute myeloblastic leukemia, chronic myeloid leukemia, chronic lymphoid leukemia and in a control group. The leukocyte alpha-amylase activity was calculated in units per mg of protein. In all groups of patients an increase of the tested enzyme could be observed. The enhancement of alpha-amylase activity was correlated with the rate of leukocytic maturation. The normal activity of this enzyme in the sera of tested patients should be stressed. This statement that seems to be contradictory is briefly discussed.

Acid phosphatase and non-specific alpha-naphthol acetate esterase activities of monocytes in patients with cancer of the digestive tract during surgical treatment.

In 22 patients with cancer of the alimentary tract the activities of acid phosphatase and non-specific alpha-naphthol acetate esterase in monocytes were tested. The enzyme activity was tested in the peripheral blood before surgical intervention, in blood from vessels draining the tumour before its excision and in the peripheral blood before surgical intervention, in blood from vessels draining the tumour before its excision and in the peripheral blood 2--3 weeks after tumour excision. In parallel tests the enzyme activity was estimated in the peripheral blood of 22 healthy individuals. The study indicates that the non-specific alpha-naphthol acetate esterase activity of monocytes derived from patients with cancer and control group did not show a marked difference. The acid phosphatase activity in monocytes derived from a tumour efferent vessel was found to be higher in majority of the cases than the activity of this enzyme in monocytes derived from the peripheral blood. After removing the tumour the acid phosphatase activity of monocytes was elevated in half of the cases. It seems possible that the increase of acid phosphatase activity in monocytes derived from cancer patients may be due to the activation of monocytes in contact with cancer antigens or antigen-antibody complexes.

Nitroblue tetrazolium test in patients with neoplasma during surgical treatment.

In 20 patients with alimentary tract neoplasma, nitroblue tetrazolium test (NBT) with resting and stimulated granulocytes was done using Park's et al. [14] method. NBT test was determined in peripheral blood before surgical intervention, in blood from vessel draining of tumor before its excision and in peripheral blood 2-3 weeks after excision of tumor. In parallel experiments the NBT test was estimated in peripheral blood of 21 healthy individuals. We have shown that NBT dye reduction was evidently diminished in patients with cancer disease especially in blood samples from vein draining of tumor both in resting and stimulated granulocytes. The excision of tumor caused improvement of NBT dye reduction. The importance of determination of NBT test in patients with cancer disease is briefly discussed.

Peroxydase activity of the granulocytes in the course of surgical treatment of neoplasm of the alimentary tract.

Granulocyte peroxidase activity of patients with alimentary tract neoplasms has been investigated by using the histochemical method. Blood samples were collected before surgical treatment and anaesthesia, during surgical intervention from vessels draining the neoplasms, and 2-3 weeks after surgery from peripheral vessels. The activity of myeloperoxidase in the polymorphonuclear granulocytes of patients was evidently lower in comparison to the activity of this enzyme in the granulocytes of normal individuals. The excision of the tumour did not evidently increase the tested activity in blood from peripheral vein in comparison to the same sample and to the blood from vein draining of tumour. We can suggest that granulocytes may release myeloperoxidase to the surrounding media in the course of neoplasmatic diseases and in this case the cells are exhausted in this enzyme.

The effect of somatostatin on haemostasis in dogs.

The effect of somatostatin on haemostasis in the dogs was tested. The following data could be found: marked shortening of clotting time, decrease of platelet count, decrease of fibrinogen, level, an increase of plasma euglobulin fibrinolytic activity, prolongation of thrombin time and consumption of plasminogen. The obtained results indicate a biphasic haemostatic reaction, first hypercoagulability and then hypocoagulability. The presented results are briefly discussed with some remarks to clinical practice.
