Acid phosphatase in peripheral blood lymphocyte lysosomes in patients with diabetes mellitus.

In 55 patients with diabetes mellitus metabolically balanced the activity and intracellular localization of acid phosphatase has been estimated by means of semiquantitative method of Barka and Anderson [3] in peripheral blood lymphocytes. It has been shown that patients in question, both insulin-dependent and those with overweight, in comparison to control group, exhibit more than twofold lower numbers of lymphocytes having intact enzyme positive lysosomes and more than twofold higher numbers of that cells with damaged lysosomes and extralysosomal localisation of the enzyme. According to authors suggestion the above changes may be associated with disturbed lymphocyte functions in the patients with diabetes mellitus.

T and B cells and occupational exposure to benzene and its homologues (with regard to other blood cells).

In 106 workers (47 women and 59 men) having occupational contact with organic solvents containing benzene and its homologues during 1 to 122 months the quantitative evaluation of the white blood cell and the platelet system has been performed. In workers having longer time of service than 55 months the decreased T and the "non-T, non-B" lymphocyte count as well as increased counts of monocytes in the peripheral blood have been stated. Furthermore, the negative correlation between the T cell numbers and the exposure time has been stated what indicates the practical value of the E rosette test in evaluation of early biological effects of occupational exposure to solvents containing benzene, toluene and xylene. According to the authors' opinion a depressive effect of benzene and its homologues on the T cell-dependent immunity suggests an alternative mechanism of cancerogenic activity of that compounds via the immune system. The toxic effect of organic solvents containing benzene and its homologues on blood forming organs is among other reflected in quantitative alterations of the white blood cells and the platelet systems (6, 17). In workers occupationally exposed to benzene, toluene and xylene neutropenia (1, 2, 6, 17), eosinophilia (2), basophilia (2), monocytosis (14) and both lymphocytosis and lymphocytopenia (3, 14) has been observed. The occupational contact with above compounds may also results in thrombocytopenia (11, 12) or qualitative changes in platelets resembling that noted in patients with the Glanzman thrombasthenia (4).

Lithium carbonate effect on nitrotetrazolium blue (NBT) production in neutrophils of patients with granulocytopenia.

In 30 persons with essential granulocytopenia, lithium carbonate in doses of 12.5 mg/kg per body weight was applied during 10 days. This resulted in an increased absolute neutrophil count and total leukocyte count as well as increased percentage of cells reducing NBT in the spontaneous NBT test according to Park. Prior to the beginning of treatment, patients with granulocytopenia exhibited an elevated percentage of NBT-positive cells in comparison to the control group of 28 healthy persons. The significance of this observation for treating granulocytopenia by means of the agent mentioned above is discussed.

Enzymes of peripheral blood neutrophils in patients with cancer of the stomach.

In 25 patients (10 women and 15 men) aged 37 to 68 years activity of the following enzymes has been determined by the use of semiquantitative cytochemical methods: acid and alkaline phosphatase, beta-glucuronidase, N-acetyl-beta-glucosaminidase and myeloperoxidase. Additionally the content of glycogen and lipids has been determined in the cells studied. An increase of the myeloperoxidase and acid phosphatase activity and a decrease of the glycogen and the lipid content has been stated in neutrophils of the patients. Above alterations may reflect antitumor reactivity of neutrophils or nonspecific neutrophil response against inflammatory changes accompanying the tumor area.
