N-acetyl-beta-glucosaminidase of peripheral blood lymphocytes in ontogenetic development of man.

The activity and intracellular localization of N-acetyl-beta-glucosaminidase in peripheral blood lymphocytes has been evaluated in 170 healthy subjects of varying age by means of semiquantitative cytochemical method. The groups of premature and term infants, children and adults have been compared with regard to cytochemical type of reaction and intracellular localization of that enzyme. It has been revealed that during last months of intrauterine life of man the granular type of reaction suggesting formation of enzyme-positive lysosomes becomes gradually more and more frequent. During further ontogenetic development starting from neonatal age up to fifth decade of life no significant alterations with regard to the numbers of enzyme-positive lymphocytes and the intracellular type of cytochemical reaction are noted. The last three decades of life are characterized by more frequent diffuse type of cytoenzymatic reaction within the lymphocytes suggesting a passage of the enzyme from lysosomes to the surrounding cytoplasm.

The lymphocyte lysosomes in ontogeny of man.

During intrauterine fetal development, the gradual formation of enzyme-positive lysosomes containing acid phosphatase, beta-glucuronidase, and N-acetyl-beta-glucosaminidase takes place within peripheral lymphocytes. During the first 5 decades of human life the lysosomal apparatus of lymphocytes containing the above enzymes exhibits relative stability whereas the final stages of man's ontogenetic development between 6th and 9th decade of life are characterized by the disintegration of enzyme-positive lysosomes.

The acid phosphatase activity in lymphocytes with regard to the ontogenetic development of man.

In 120 healthy subjects aged from 1 day of life to 90 years the activity and intracellular localization of acid phosphatase (AP) in the peripheral blood lymphocytes has been evaluated by using semiquantitative cytochemical method (Barka and Anderson 1962). The long lasting stability of the lysosomal apparatus of lymphocytes during the ontogenetic development between the first day of life and the fifth decade of life has been stated. The stability was expressed by relatively constant number of lymphocytes containing intact AP-positive lysosomal granules. During the last decades of life between 66 and 90 year lymphocyte lysosomes are destabilized and numerous cells with cytoplasmatic diffusion of the enzyme can be observed. According to the author's opinion the above phenomenon corresponds to the aging of the lymphocyte system.

Immunological and cytochemical indices of white blood cells in old age.

The group of aged subjects being 66 to 97 years old was compared with the middle-age group with regard to various immunological and cytochemical indices related to lymphocytes and neutrophils. The aged showed a lowered count and percentage of T cells, increased count and percentage of "non-B, non-T" lymphocytes, increased percentage of B cells. These alterations in the composition of lymphocyte subpopulation were associated with characteristic patterns of damage affecting the enzyme-positive lysosomal apparatus of lymphocytes with regard to acid phosphatase, beta-glucuronidase and N-acetyl-beta-D-glucosaminidase. There was a hundredfold smaller number of cells having intact enzyme-positive lysosomes in the aged than in the group of comparison. The changes mentioned above were also associated with the intracellular accumulation of glycogen in lymphocytes, decreased concentration of IgG and IgM in the serum and various changes in IgA concentration. Neutrophils of the aged were fewer in the blood of the aged than in younger subjects. However, an increased activity of myeloperoxidase, alkaline phosphatase, N-acetyl-beta-D-glucosaminidase, and an increased content of glycogen and lipids could be found in these cells. NBT-positive neutrophil numbers in the aged were lowered if the stimulated test was used and if there were no changes of the spontaneous test.

Effects of work environment contaminated with benzene, toluene and xylene on acid phosphatase activity in lymphocytes.

The activity of acid phosphatase (AP) in peripheral blood lymphocytes was determined by means of histochemical method in 108 workers having occupational contact with organic solvents containing benzene and its homologues. Benzene, toluene and xylene concentrations in the air at work stands were equal to 0-370, 0-580 and 0-560 mg/eu.m., respectively. A progressing reduction of numbers of lymphocytes having AP-positive intact lysosomes was stated in workers with a working time equal 31-122 months. This phenomenon was more expressed in workers exposed to greater concentrations of benzene and its homologues. The damage to the lysosomal apparatus of lymphocytes reflected by an increased permeability of lysosomal membranes can be regarded as one of the earliest symptoms of toxic effects of benzene, toluene and xylene on cells of the lymphocytic series.

Beta-glucuronidase of lymphocytes in ontogenetic development of man.

The activity and intracellular localization of lysosomal acid phosphatase in lymphocytes was studied in 170 healthy subjects of various age using semiquantitative cytochemical method. The groups studied included the premature and term infants, children and adults. During fetal development the gradual organization of enzyme-positive lysosomal granules within lymphocytes occurs and subsequent five decades of life are characterized by stabilization of lysosomal apparatus of these cells. In last decades of life, between 60 and 90, the destabilization and destruction of acid-phosphatase-positive lysosomal granules is observed.

Lysosomal enzymes of neutrophils in women with breast cancer.

Activity of acid phosphatase, beta-glucuronidase and N-acetyl-beta-D-glucosaminidase in peripheral blood neutrophils of 30 women with breast cancer has been studied by means of semiquantitative cytochemical methods. In comparison to 20 healthy women the patients showed an increased activity of acid phosphatase and beta-glucuronidase whereas that of N-acetyl-beta-D-glucosaminidase was significantly lowered. It has been suggested that observed intracellular enzymatic deficiency is of importance with regard to lowered antitumor activity of neutrophils. Analogous deficiency of N-acetyl-beta-D-glucosaminidase within the neutrophils has previously been observed by the authors in women with malignant tumors of reproductive organs.

Hematological indices of peripheral blood in workers occupationally exposed to benzene, toluene and xylene.

In 106 workers occupationally exposed to benzen, toluen and xylene through 1 to 122 months basic hematological indices of peripheral blood were evaluated. The benzene, toluene and xylene concentrations in the air at workplaces were equal to 0-370, 0-580 and 0-506 mg/m3, respectively. The workers were subdivided into three subgroups according to the service time corresponding to 1-29, 31-54 and 55-122 months. The first hematological changes noted in the workers studied consisted of diminishing the mean corpuscular hemoglobin and the mean corpuscular hemoglobin concentration in erythrocytes. Increased numbers of reticulocytes, lowered total count of leukocytes due to decreased numbers of T and "non-T, non-B" cells as well as increased numbers of monocytes were other signs of exposure investigated. Increased numbers of reticulocytes were noted in all workers independently of service time whereas other hematological alterations presented above were marked only in the subgroup of workers exposed to benzene, toluene and xylene through 55 to 122 months. It was stated that the T cell count decreased gradually in relationship with an extent of exposure time (negative correlation). Since laboratory examinations serving the evaluation of health state of workers exposed are only few it was postulated that the E rosette test may be of practical use for monitoring the toxic effect of benzene, toluene and xylene on the lymphocyte system.
