Beta-glucuronidase-positive lymphocyte population with intact enzyme-positive lysosomes in patients with malignancies.

In 234 patients with various malignant diseases including cancer of the larynx, cancer of the lung, cancer of the stomach, cancer of large intestine as well as patients with Hodgkin's disease, plasma cell myeloma and precancerous states of the larynx, the count of peripheral blood lymphocytes showing intact beta-glucuronidase-positive lysosomes was determined using a semiquantitative histochemical method. All the patients studied, except patient with cancer of the lung, showed a tendency toward a low count of such cells significant in patients with precancerous states of the larynx, cancer of larynx, cancer of the larynx after radiotherapy and Hodgkin's disease and nonsignificant in cancer of the stomach, cancer of the large intestine and plasma cell myeloma. In patients with cancer of the lung the count of lymphocytes with intact beta-glucuronidase-positive lysosomes was significantly increased as compared with that in healthy subjects. The possible relationship between the low immune response in the patients and the functional state of the lymphocyte lysosomal system is discussed.

[Effect of tobacco smoking on various immunologic indicators].

The study conducted in 85 clinically healthy males--cigarette smokers--demonstrated a decrease of the total number of T lymphocytes related to smoking duration. There was no effect on the number of B cells while the number of non-B non-T lymphocytes increased. Smokers demonstrated decreased levels of serum IgA, IgG and IgM levels. The authors discuss these findings in relation to other controversial studies.

The enzymatic deficiencies in lymphocytes of smokers.

In the peripheral blood lymphocytes of 85 men aged 18 to 42 years having smoked cigarettes for 2 to 25 years, the activity of various intracellular enzymes was determined using the semiquantitative histochemical methods. The lymphocytes of people having smoked for less than 10 years were characterized by increased acid phosphatase (AP), leucine aminopeptidase (LAP) and lactic dehydrogenase (LDH) activities whereas that of N-acetyl-beta-D-glucosaminidase (NAG) were lowered. In subjects having smoked for more than 10 years lower acid phosphatase, beta-glucuronidase, N-acetyl-beta-D-glucosaminidase and lactic dehydrogenase activities have been observed. According to the authors' suggestions, the functional lymphocyte abnormalities noted in smokers by numerous investigators may be associated with the intracellular enzymatic deficiencies.

Histochemical evaluation of lysosomes in lymphocytes of workers having contact with organic solvents containing benzene and its homologues.

The count of lymphocytes having lysosomes containing acid phosphatase (AP), beta-glucuronidase (GR) and N-acetyl-beta-D-glucosaminidase (NAG) has been estimated in the peripheral blood of 108 workers occupationally exposed to organic solvents containing benzene and its homologues. It has been stated that the prolonged exposure is associated with a reduction in the number of cells having intact acid phosphatase-positive lysosomes. The negative correlation occurring in this connection indicates the practical value of histochemical evaluation of numbers of the above cells for screening the biological effect of occupational exposure to solvents studied.

The nonspecific esterase activity in lymphocytes of workers occupationally exposed to benzene and its homologues.

Activity of nonspecific esterase (NE) was determined in peripheral blood lymphocytes from workers occupationally exposed to organic solvents containing benzene, toluene and xylene, using a semiquantitative histochemical method. The benzene, toluene and xylene concentration at the workplace of the subjects studied, was equal to 0-370 0-580 and 0-560 mg/cu. m, respectively. In subjects having a working time longer than 54 months a decreased count of lymphocytes showing presence of NE in intact lysosomes and increased count of cells with presence of this enzyme both in the partially destroyed lysosomes and in the cell cytoplasm was observed. The above phenomenon demonstrates the lysosome damage and consecutive replacing of the enzyme activity from the lysosome into the cytoplasm. Since there was a positive correlation between the exposure time and numbers of cells with damaged NE-positive lysosomes it may be suggested that the semiquantitative histochemical estimation of NE activity within the lymphocytes can be used for monitoring the toxic effect of benzene and its homologues on lymphocytes.

The deficiency of B-glycosidases in leukocytes from patients with non-insulin dependent diabetes mellitus.

The activity and intracellular localization of two B-glycosidases i.e., N-acetyl-beta-D-glucosaminidase (NAG) and beta-glucuronidase (GR) was estimated by means of semi-quantitative histochemical methods in the peripheral blood neutrophils and lymphocytes from 52 patients with non-insulin dependent diabetes mellitus and compared with that in 67 healthy subjects of similar age and sex. The total neutrophil and lymphocyte count did not differ in the patients from that in the controls. The GR and the NAG activity indexes were significantly lower in the neutrophils from patients as compared with that in the control group. The number of lymphocytes with GR-positive and NAG-positive intact lysosomes were 200-fold and respectively 40-fold lower in the patients than in the healthy subjects. The authors discuss the significance of their observations with regard to the relationship between the enzymatic activity of the cells studied and their functional capabilities.

Long-term effect of radiotherapy on the lymphocyte lysosomal enzymes and serum immunoglobulins in patients with cancer of the larynx.

Thirty male patients ranging in age from 40 to 70 years, investigated 6--9 years after radiotherapy for cancer of the larynx, showed various alterations of the lysosomal apparatus of peripheral blood lymphocytes. These changes, consisting of diffusion of the lysosomal enzymes within the cytoplasm and disappearance of normal lysosomal granules, were related to a greater extent to the beta-glucuronidase and N-acetyl-beta-glucosaminidase positive-lysosomes than to the acid phosphatase-positive ones. Determination of the enzymatic activities was achieved by cytochemical techniques. The increased immunological reactivity of the patients was reflected in the frequently elevated immunoglobulin levels, particularly that of IgA, in the serum and an increase of the enzyme-positive lymphocyte count in the blood.
