The role of E receptors in the attachment of thymocytes and T lymphocytes to human target cells.

Human thymocytes, activated T lymphocytes, and neuraminidase-treated T cells possess the distinct capacity of forming conjugates with various human cell lines. The present study investigated whether E receptors, which endow human T cells with their capacity to bind sheep red blood cells (SRBC), are involved in this phenomenon. Monoclonal antibodies to human T cells and various simple sugars were studied for their effect on the attachment of human T cells to target cells. A-22, a monoclonal antibody to the E receptor, inhibited the formation of E rosettes by T cells and SRBC, and reacted in immunofluorescent-staining assays with the majority of human thymocytes and peripheral T cells, and with T-cell lines capable of forming E rosettes. When human thymus cells were treated with A-22 antibody they showed a reduction of up to 70% in their capacity to attach to the GM-4762 lymphoblast cell line and the K-562 myeloid line. Antibody treatment of the target cells, rather than of the thymus cells, had no effect on the formation of conjugates between thymus cells and target cells. Treatment of thymus cells with various monoclonal antibodies to T cells which do not react with the E receptor had no inhibitory effect. The exposure of human thymus cells to various simple sugars (D-mannose, D-fucose, galactose, and lactose) markedly reduced their capacity of forming conjugates with target cells. Exposure of neuraminidase-treated peripheral blood lymphocytes and of activated T cells to A-22 antibody inhibited their attachment to human target cells. The present study suggests that E receptors play a role in the attachment of human thymus cells and activated T cells to other human cells, and raises the possibility that these T-cell receptors may be involved in the process of recognition of "self" structures by human T lymphocytes.

Asymmetry of perception in free viewing of chimeric faces.

We have devised a new free-vision task to index functional cerebral asymmetry for processing facial characteristics. Confirming its sensitivity to properties of lateralized hemispheric functions, left- and right-handers were clearly differentiated on this task with respect to several aspects of performance that conform with known differences between handedness groups in hemispheric asymmetry. Additionally, there were highly reliable and stable individual differences in perceptual asymmetries within handedness. Analyses of items in the task revealed that most of the differences between items in the asymmetries they elicited were random.

Are variations among right-handed individuals in perceptual asymmetries caused by characteristic arousal differences between hemispheres?

We propose that much of the variance among right-handed subjects in perceptual asymmetries on standard behavioral measures of laterality arises from individual differences in characteristic patterns of asymmetric hemispheric arousal. Dextrals with large right-visual-field (RVF) advantages on a tachistoscopic syllable-identification task (assumed to reflect characteristically higher left-hemisphere than right-hemisphere arousal) outperformed those having weak or no visual-field asymmetries (assumed to reflect characteristically higher right-hemisphere than left-hemisphere arousal). The two groups were equal, however, in asymmetries of error patterns that are thought to indicate linguistic or nonlinguistic encoding strategies. For both groups, relations between visual fields in the ability to discriminate the accuracy of performance followed the pattern of syllable identification itself, suggesting that linguistic and metalinguistic processes are based on the same laterally specialized functions. Subjects with strong RVF advantages had a pessimistic bias for rating performance, and those with weak or no asymmetries had an optimistic bias, particularly for the left visual field (LVF). This is concordant with evidence that the arousal level of the right hemisphere is closely related to affective mood. Finally, consistent with the arousal model, leftward asymmetries on a free-vision face-processing task became larger as RVF advantages on the syllable task diminished and as optimistic biases for the LVF, relative to the RVF, increased.

Bioactivity of gentamicin in purulent sputum from patients with cystic fibrosis or bronchiectasis: comparison with activity in serum.

Two mechanisms of potential biologic antagonism of gentamicin in purulent sputum from patients with cystic fibrosis or bronchiectasis were studied: reduction of activity by ions and antibiotic binding. Antagonism by ions was assessed by examination of the activity of gentamicin against Pseudomonas aeruginosa in dialysates of serum or sputum in ion-depleted broth. The ionic content of the dialysates increased and reflected differences in the ion content of serum and sputum. Gentamicin had significantly less activity against P aeruginosa in sputum or serum dialysates than in ion-depleted broth alone. When gentamicin was mixed with serum or sputum before dialysis, the level of antipseudomonas activity of the sputum dialysates was significantly lower than that of the serum dialysate; this finding was correlated with greater binding by sputum. Thus, both binding and antagonism by ions evidently reduce the level of bioactivity of gentamicin in serum and in sputum. Purulent sputum, whether from children with cystic fibrosis or adults which bronchiectasis, is more inhibitory than serum; the greater degree of binding, rather than differences in the composition or quantity of cations, explains this difference.

Correction by bromocriptine of hypothalamic dysfunction and post-prandial hypoglycaemic symptoms in a 31-year-old woman.

A 31-year old female presented with recurrent episodes of post-prandial hypoglycaemic symptoms. Basal serum levels of ACTH, cortisol, GH, insulin and glucagon were normal. An adrenaline test demonstrated a normal peripheral response. An exercise test failed to produce ACTH, cortisol or FFA responses. Insulin (0.1 u/kg)-induced-hypoglycaemia failed to elevate serum ACTH, cortisol or GH. Metyrapone and ACTH tests were normal, demonstrating adequate hypophyseal and adrenal function. These findings suggested that the patient suffered from hypothalamic dysfunction. Bromocriptine (Parlodel, 7.5 mg/d for 5 weeks) resulted in an improved general condition, accompanied by a decrease in sugar consumption. Following treatment, FFA, ACTH and cortisol responses to exercise test were normal, as were ACTH, cortisol and GH responses to insulin-induced hypoglycaemia. It is concluded that bromocriptine may be useful in the treatment of post-prandial hypoglycaemic symptoms associated with hypothalamic dysfunction.

Isolation and characterization of casein-producing and -nonproducing cell populations from 7,12-dimethylbenz(a)anthracene-induced rat mammary carcinomas.

Previous studies from our laboratory have demonstrated that hormone-dependent 7,12-dimethylbenz(a)anthracene-induced rat mammary carcinomas possess only a limited ability to synthesize the mammary gland-specific milk proteins, and this expression of normal differentiated function is limited to a small subpopulation of tumor cells (S. C. Supowit and J. M. Rosen, Cancer Res., 42: 1355-1360, 1982). In an effort to define the molecular defect in hormone-regulated casein gene expression in these tumors, a method has been developed which separates the 7,12-dimethylbenz(a)anthracene-induced rat mammary carcinoma into two subpopulations of cells. Collagenase-dissociated tumor cell suspensions were fractionated by both continuous and discontinuous Percoll gradient centrifugation, and the separated cell populations were analyzed following plating on collagen-coated dishes. Two fractions of differing densities were isolated. The top fraction (rho = 1.05), containing approximately 75% of the dissociated tumor epithelial cells, is depleted in casein-producing cells, while the bottom fraction (rho = 1.07) is enriched 5- to 10-fold in casein-producing cells as assayed by both indirect immunofluorescence, using a specific anticasein antibody, and direct measurement of casein messenger RNA levels. These two subpopulations were further characterized by scanning and transmission electron microscopy, with respect to their cell morphology and ultrastructure, for the presence of hormone receptors; the arrangement of the epithelial cell marker, keratin; and their cell cycle distributions and ploidy. Prolactin, insulin, and estradiol binding were found to be uniform throughout each cell subpopulation and qualitatively similar between fractions. Both fractions appeared to be epithelial-like in morphology, containing numerous surface microvilli as assessed by scanning electron microscopy and containing numerous surface microvilli as assessed by scanning electron microscopy and contained keratin filaments as determined by indirect immunofluorescence. Furthermore, they had similar cell cycle distributions and ploidies of 2n. Differences in cell ultrastructure were observed between these two tumor epithelial cell fractions, reflecting an increased development of rough endoplasmic reticulum in the bottom fraction and a higher concentration of lipid-containing vesicles and free polysomes in the top fraction. The failure of the majority of the cells within the 7,12-dimethylbenz(a)anthracene-induced mammary tumors to produce casein in response to prolactin is not due to a receptor-negative phenotype or general cell type difference.

Epstein-Barr virus and infectious mononucleosis in children.

The presence of IgG antibodies to Epstein-Barr virus (EBV) has been investigated in two groups of children aged 3 to 35 months. The test group consisted of 31 children from an institution in which a high incidence of suspected infectious mononucleosis (IMN) was found. The control group comprised 54 children, aged 1 to 26 months, with noninfectious diseases. In the test group, 29 of the 31 children were positive for IgG anti-EBV antibodies in the serum, whereas only 5 children were positive in the control group. None of the children in the test group had detectable heterophile antibodies, and most of them did not show the typical symptoms of IMN. The reliability of diagnosing IMN in early childhood, by means of detection of anti-EBV serum antibody levels, is discussed.

Cell surface receptors involved in the attachment of murine cytotoxic T lymphocytes to target cells - a study with xenoantisera to T-cell antigens.

In previous studies it was demonstrated that rabbit anti-mouse thymus sera (RAT) but not rabbit anti-brain serum (RABR), block the activity of cytotoxic T lymphocytes (CTL). The aim of the present study was to determine to what extent inhibition of the lytic activity of CTL reflects inhibition of the attachment of CTL to target cells. Following heat inactivation RAT and RABR were absorbed with mouse liver and kidney and tested for their capacity to inhibit the attachment of sensitized peritoneal exudate T lymphocytes (PEL) to various target cells. The exposure of sensitized PEL to RABR markedly reduced their capacity to form conjugates with either allogeneic or syngeneic target cells. In contrast, RAT inhibited only the formation of conjugates of sensitized PEL with the respective target cells against which they were immunized. Treatment with RAT had no effect on the formation of conjugates with irrelevant target cells. Additive experiments in which PEL were exposed to a mixture of both RAT and RABR indicated that the two antisera blocked different types of attachment. The inhibitory activity of RAT on conjugate formation could be removed by absorption with B-lymphoma cells but not with myeloma cells. Analysis of the correlation between the inhibitory effect of RAT on cell-mediated lysis (CML) and on conjugate formation revealed that the serum was about twice as effective in its capacity to inhibit CML as to inhibit the attachment of PEL to target cells. The results indicate that while RABR inhibits non-specific attachment of CTL which does not lead to cell lysis. RAT exerts its effect by interfering with immunologically specific functional receptors on CTL.

Cervical pregnancy analysis: a review and report of five cases.

Five cases of cervical pregnancy are presented, with an incidence of 1:2400. Four of the cases, proved after hysterectomy and histopathologic confirmation, occurred in 1981. One was clinically most probable and was treated conservatively by curettage and packing. The distinction between proved and clinically probable cases of cervical pregnancy is defined. A review and analysis of the literature reveals an increasing incidence of cervical pregnancy. This may be due largely to increased numbers of induced abortions and sharp curetting. The maternal mortality rate has decreased from approximately 40 to 45% to zero in the past 30 years. Approximately 70% are treated necessarily by total hysterectomy. The remaining 30% are treated conservatively; however, ensuing pregnancies are rare in such women. Only three cases of pregnancy following probable cervical pregnancy have been reported. The last reported case was in 1953.

Interaction of estrogen receptors with drugs that affect prolactin secretion.

The competition of bromocriptine and lisuride hydrogen maleate (LIM) with estradiol binding to various tissues was evaluated by the dextran coated charcoal method. Bromocriptine and LIM competitively inhibited the binding of [3H] estradiol to its cytosolic receptors in rat uterine, pituitary and hypothalamic tissue and in DMBA induced mammary tumors. Ki was 2 X 10(-5) M for bromocriptine and 2 X 10(-4) M for LIM. Metoclopramide, dopamine and L-dopa had no significant effect on [3H] estradiol binding. The interaction of bromocriptine and LIM was specific for estrogen receptors. There was no interaction with progesterone receptors from rat uterus and pituitary and with testosterone receptors from rat epididymis and testis. When tested for estrogenity in the immature rat uterus, bromocriptine and LIM induced specific estrogen inducible proteins such as cytosolic estrogen and progesterone receptors. However, they do not affect the uterine/body weight ratio and peroxidase activity. A clear interaction of inhibitors (bromocriptine and LIM) of prolactine secretion, with cytosolic estrogen receptors from various tissues was shown. Some in vivo estrogenic effect was also demonstrated in the immature rat uterine system.

Regulation of protein kinases activity by quercetin in Ehrlich ascites tumor cells.

Cyclic AMP-independent protein kinase activities from Ehrlich ascites tumor cells, partially purified by DEAE-cellulose and phosphocellulose chromatography were inhibited by quercetin. The cyclic AMP in the tumor ascites cells and the cyclic AMP-dependent protein activity from this tumor and from bovine and mouse tissues were unaffected by this drug. Since we reported that quercetin elevates cyclic AMP level in Ehrlich ascites tumor cells, this bioflavonoid may have a dual effect on the protein kinase activities in these cells, thus, increasing the cyclic AMP-dependent and decreasing the cyclic AMP-independent protein kinase activities.
