A polypeptide derived from mitochondrial dihydrolipoamide succinyltransferase is located on the plasma membrane in skeletal muscle.

Dihydrolipoamide succinyltransferase (DLST) is the core-enzyme of 2-oxoglutarate dehydrogenase complex which is located in mitochondria. In this study, several tissues from rat and human were immunostained with an affinity-purified anti-DLST antibody. Of the tissues examined, the plasma membrane of skeletal muscle was immunostained with the antibody besides mitochondria. Furthermore, subcellular fractionation analysis coupled with Western blotting demonstrated that the antigen of the anti-DLST antibody is distributed on the plasma membrane fraction in addition to the mitochondria fraction in skeletal muscle and that it is free from the complex. The molecular weight of the polypeptide bound to the plasma membrane was about 20 kilodaltons (kDa). The polypeptide was purified by immunoprecipitation and its N-terminal amino-acid sequence was determined. The amino-acid sequence exactly corresponded to a part of DLST. Northern blots revealed the presence of mRNA corresponding to the 20 kDa protein. We are the first to report that a mitochondrial protein is also present on the plasma membrane in skeletal muscle as well as in mitochondria.

Amplification of the mdm-2 gene and p53 abnormalities in uterine sarcomas.

The aim of this study is to address the role of mdm-2-gene amplification in the tumorigenesis of uterine sarcomas. Differential PCR with DNA from formalin-fixed paraffin-embedded specimens was employed in 12 patients with uterine sarcomas. We detected mdm-2-gene amplification in 4 out of 12 uterine sarcomas. The estimated copy number of the mdm-2 gene ranged from 4 to 13. Positive cases included 1 leiomyosarcoma and 3 carcinosarcomas, however, there was no correlation between mdm-2-gene amplification and clinicopathological characteristics. Over-expression of p53 protein was also immunohistochemically studied in the same series of patients: 4 out of 8 carcinosarcomas displayed p53 immunoreactivity. Taking these results together, only one carcinosarcoma was found to have both mdm-2-gene amplification and p53 over-expression. In contrast, half of the patients were found to have alterations either of mdm-2 or of p53. These findings support the notion that mdm-2-gene amplification might be an alternative mechanism for escaping from the regulatory pathway of p53 to suppress cell growth.

Advanced endometrial cancer detected at 7 months after childbirth.

It is very rare to find endometrial cancers arising within a short period following childbirth, presumably because pregnancy has a protective effect against endometrial cancer mediated by elevated secretion of progesterone. We present the case of a 30-year-old Japanese woman with stage IIIC endometrial cancer that was found 7 months after childbirth. The patient was treated with surgery followed by 6 cycles of intravenous combination chemotherapy and oral administration of medroxyprogesterone acetate. Histopathological examination of surgical specimen revealed poorly differentiated adenosquamous carcinoma of the endometrium with deep myometrial invasion, cervical stromal involvement, and pelvic and paraaortic lymph node metastases. The disease progressed rapidly and the patient died of the disease.

Immunohistochemical analysis of glutathione S-transferase mu expression in ovarian tumors.

OBJECTIVE: To explore the association between Glutathione S-transferase (GST) -mu expression and clinicopathologic features in ovarian tumors. STUDY DESIGN: Immunohistochemical study was made to investigate GST-mu expression in diverse ovarian tumors. RESULTS: All 75 ovarian tumors expressed GST-mu. There was no significant association between GST-mu immunopositivity and clinicopathological features. In serous cystadenocarcinoma, however, patients with weak GST-mu expression survived longer than those with moderate or strong GST-mu expression. Three of the 8 tumors that had expressed weak GST-mu initially increased GST-mu expression after chemotherapy. CONCLUSION: GST-mu expression is common in ovarian tumors and malignant tumors expressed more GST-mu than benign tumors. GST-mu might play a major role in the development of drug resistance in certain ovarian tumors and could be a useful marker of natural resistance and their future outcome.

The expression of vascular endothelial growth factor and transforming growth factor-beta associates with angiogenesis in epithelial ovarian cancer.

The object of this study was to clarify the association between microvessel density and clinicopathologic factors, as well as the association of angiogenic factors (vascular endothelial growth factor [VEGF] and transforming growth factor-beta [TGF-beta]) with tumor angiogenesis and patient survival in epithelial ovarian cancer. The expression of VEGF and TGF-beta was evaluated by immunohistochemical staining in 60 patients with epithelial ovarian cancer. The microvessel density, assessed by immunostaining for factor VIII-related antigen in the most neovascularized areas, varied depending on histological types but not on International Federation of Gynecology and Obstetrics stage. Patients with stage III carcinomas and positive TGF-beta had more extensive peritoneal dissemination and a worse outcome. The microvessel density of VEGF-rich and TGF-beta positive tumors was significantly higher than that of VEGF-poor and TGF-beta negative tumors. Angiogenesis appears to be an early event in epithelial ovarian cancer and may be induced differently in tumors of different histological types. The expression of VEGF and TGF-beta associates with the promotion of angiogenesis, and the expression of TGF-beta is a prognostic indicator in epithelial ovarian cancers.

Malignant melanoma of the uterine cervix: a case report.

We present case of a 57-year-old Japanese woman a 9 cm-size primary malignant melanoma of the uterine cervix. MRI demonstrated a low-signal T1-weighted spin echo image and hyperperfusion indicated by Gd-DTPA enhancement. The sequence analysis of the MTS1/CDK4I gene revealed no deletion or mutation. The results of cytological, pathological, and electron microscopy tests are also presented.

Placenta growth factor is abundantly expressed in human cervical squamous cell carcinoma.

Tumor angiogenesis is essential for solid tumor growth. The object of this study was to investigate the presence of newly identified angiogenic factor, placenta growth factor (P1GF) in human cervical cancers. The expression of P1GF mRNA was assessed by RT-PCR in 29 patients with cervical cancer. Fifteen out of 29 cervical cancers expressed a certain level of P1GF mRNA. In addition, the expression levels of P1GF mRNA in squamous cell carcinomas were significantly higher than those in adenocarcinomas. There was no correlation between the expression of P1GF mRNA and FIGO stage. These results indicate that the expression of P1GF may be implicated in the promotion of angiogenesis in human cervical squamous cell carcinomas, but not in human cervical adenocarcinomas.

Reverse correlation between P-glycoprotein expression and proliferative activity in endometrial adenocarcinoma.

Immunohistochemical study was employed using a monoclonal antibody (C219) to investigate P-glycoprotein expression in 23 normal endometria and 40 endometrial adenocarcinomas. P-glycoprotein immunopositivity was observed in the mid- to late-proliferative phase and the whole secretory phase of normal endometrium. In contrast, no P-glycoprotein was detected in endometrium of early proliferative phase nor post-menopausal endometrium. We also investigated the expression of P-glycoprotein in endometrial cancers to find 16 out of the 40 endometrial adenocarcinomas (40%) expressed P-glycoprotein. P-glycoprotein immunopositivity was often observed in gland growing parts but not in solid growing parts in a given tumor specimen. Proliferative activity of the tumor, as measured by PCNA labeling, was significantly higher in the P-glycoprotein-negative group than that in P-glycoprotein-positive group (P < 0.005). Thus, there was a strong reverse relation between P-glycoprotein expression and proliferative activity in endometrial adenocarcinomas.

Effect of denervation on morphogenesis of the rat fungiform papilla.

In an attempt to elucidate the effects of denervation on development and maintenance of the structure of the fungiform papilla, unilateral neurectomy of the chorda tympani-lingual nerve of rats was performed at day 1 and at weeks 1, 2, 3, 4, 7, and 10 after birth. Specimens were obtained at days 3, 7 and 10, weeks 2, 3, 4, 6, and 8, and months 3 and 4 after neurectomy for examination by light and scanning electron microscopy. At first, the fungiform papillae were atrophic, then progressed to forms resembling filiform papillae. When an immature fungiform papilla was denervated, it eventually changed to papilla identical to normal filiform papillae. The elicited changes differed according to the time of neurectomy; it was found that early neurectomy resulted in a more rapid and marked morphological change of the fungiform papillae. The filiform-like papillae derived from the fungiform ones showed various shapes, sizes, and orientations and were rarely present on the unoperated control side of the lingual dorsum. Sections of the filiform-like papillae revealed that they had no taste buds. These findings suggest: (1) Morphogenesis and structural maintenance of the fungiform papillae require the presence of the chorda tympani and/or lingual nerve. (2) Completion of differentiation and maturation differ in time among fungiform papillae. (3) Fungiform papillae may be transformed filiform papillae induced and maintained by a neurotrophic factor of factors coming from the chorda tympani and/or lingual nerve. (4) Fungiform papillae are rarely innervated contralaterally.

Immunohistochemical analysis of P-glycoprotein expression in diverse histological types of epithelial ovarian tumors.

P-glycoprotein is a transmembrane protein which acts as an energy-dependent drug efflux pump for a variety of anti-cancer drugs. The mdr-1 gene which encodes P-glycoprotein was successfully cloned in 1986. To investigate P-glycoprotein expression in diverse ovarian tumors, including benign, low malignant potential and malignant, immunohistochemical study was done using a monoclonal antibody (C 219). Overall, 8 out of the 59 epithelial ovarian tumors (13.6%) expressed P-glycoprotein. It was noted that 5 of the 12 mucinous tumors were found to express P-glycoprotein, while none of the 31 serous tumors were immunohistochemically positive. In 10 malignant ovarian tumors, P-glycoprotein immunostaining was examined both prior to and after chemotherapy. Nine of them did not express any P-glycoprotein before or after chemotherapy. However, one tumor expressed P-glycoprotein after six courses of multidrug resistance-related drug administration. These findings indicate that P-glycoprotein expression is not so common in ovarian tumors, regardless of their malignant potential. Nevertheless, the results suggest a strong association between P-glycoprotein expression and certain histological cell types in epithelial ovarian tumors. It is also possible that P-glycoprotein appears as a result of chemotherapy, but such a phenomenon can not occur unless chemotherapy is administered at high doses for a long period of time.

Oxidative stress during recovery from muscle atrophy.

Single ankle joints of male Wistar rats (15-week-old) were immobilized in the extended position for 7 days and remobilized for 5 days after the immobilization period. Atrophic and contralateral soleus, typical slow red muscles, were collected and their levels of thiobarbituric acid-reactive substance (TBARS) and glutathione were measured. Five-day remobilization did not increase muscle weight significantly. However, there were significant increases in TBARS and oxidized glutathione in the recovering muscle, which strongly suggested that enhanced oxidative stress occurred during the recovery from disuse muscle atrophy. Vitamin E injection accelerated the recovery from atrophy, thus showing that oxidative stress slowed it down.

Incidence and histology of human accessory parotid glands.

The parotid glands of 228 Japanese human cadavers were examined to determine the incidence and histological features of accessory parotid glands. The incidence was found to be 56% with no differences between right and left sides or between sexes. Thirty parotid glands and their associated accessory glands were examined histologically: eight of these accessory glands were found to be mixed secretory glands (i.e., containing both serous and mucous acini). Thus, the pattern of differentiation of a significant fraction of accessory glands differs from that of the main parotid gland: it appears that mixed acini, present in the early stages of development, persist into later life, and their presence may be related to tumors developing at these sites.

Role of iron in oxidative stress in skeletal muscle atrophied by immobilization.

To clarify the role of iron in oxidative stress in skeletal muscle atrophied by immobilization, we investigated the effect of deferoxamine--an iron-chelating agent. Deferoxamine, iron-saturated deferoxamine and double-distilled water (control) were administered subcutaneously from the 4th day after immobilization via osmotic pumps to male Wistar rats (14 weeks old), one ankle joint of which was immobilized in the extended position. After 12 days' immobilization, soleus--typical slow red muscles were collected from both hind limbs and their levels of thiobarbituric acid-reactive substance (TBARS) and glutathione were measured. Deferoxamine suppressed the increase of TBARS and glutathione disulfide in atrophied muscle while iron-saturated deferoxamine did not, which strongly suggests that the iron-chelating action of deferoxamine suppressed the increased oxidative stress. This means that iron plays a very important role in increasing oxidative stress in atrophied muscle. In addition, deferoxamine decreased the degree of atrophy, an effect thought to be mediated by the suppression of oxidative stress.

Anatomical study of the accessory mental foramen and the distribution of its nerve.

We examined the range of the accessory mental foramen [AMF] and its accessory mental nerve in three Japanese cadavers. The diameters of the AMF were relatively small: 0.74 mm, 0.80 mm and 0.89 mm. The distances between the mental foramen and AMF were: 0.67 mm, 2.1 mm and 5.74 mm. The distribution of the accessory mental nerve was different in the three cases. These nerves communicated with the branches of the facial and buccal nerves.

[A study on the proliferation of the uterine endometrium].

UNLABELLED: A proliferative activity study of the endometrium was evaluated in three different parts by means of an immunohistochemical approach and flow cytometry (FCM). The materials were 29 cases of hysterectomies due to uterine myoma. Twenty-one cases were in the proliferative phase, and 8 cases were in the secretory phase. The three parts of the endometrium obtained from the upper, middle and lower part of the uterine body were investigated by an immunohistochemical technique with Ki-67 and anti-DNA polymerase alpha antibody. Single staining with Propidium Iodide (PI) was used for cell cycle analysis in FCM. RESULTS: (1) The rate of positive Ki-67 cells was 25.9 +/- 6.3% in the upper part, 26.0 +/- 3.3% in the middle part, and 25.5 +/- 5.8% in the lower part. And the rate of positive DNA polymerase alpha cell was 27.2 +/- 5.8% in the upper part, 24.0 +/- 2.9% in the middle part, and 24.8 +/- 4.0% in the lower part. The growth potential of each part was not significant. (2) FCM did not detect a significant difference among the three parts of the endometrium in cell cycle analysis. (3) Hematoxylin-eosin staining showed that the upper part was thicker than the lower part and the number of glands in the upper part was more abundant than in the lower part. This difference was significant (p less than 0.01). (4) Ki-67 was recognized even in the early secretory phase. [A case report of emergency surgical repair of traumatic transection of thoracic descending aorta].

The injury to the thoracic aorta caused by blunt chest trauma is often fatal. This case is 22-year-old male suffering from transection of the thoracic descending aorta caused by traffic accident. He was transported to our emergency room by an ambulance 15 minutes after the accident. Hundred fifty minutes after arrival to the hospital, we were rush to bring him to the operation theater suspecting serious injury of the thoracic organs in association with left hemothorax. The left standard thoracotomy disclosed the injury of the thoracic descending aorta. Simple cross clamp was applied to the thoracic descending aorta distal to the left subclavian artery for 20 minutes. Completely transected aorta was reapproximated using monofilament 3-0 polypropylene sutures with running manner. He tolerated the procedure well without any complication. His postoperative course was uneventful. He was followed up at the orthopedic department for associated hip fracture thereafter.
