Cells from adult mouse spleens were cultured in WEHI-3 cell-conditioned medium, which contains the lymphokine interleukin-3 (IL-3). Under these conditions, cells grow well for 4 to 8 weeks; the cultures contain a variety of cell types for the first 1 to 2 weeks but are subsequently composed largely of immune mast cells. We found that infection of these cultures with Harvey sarcoma virus (HaSV) profoundly enhanced the growth potential of the cells, resulting in the reproducible isolation of long-term cell lines. These HaSV-infected cells appeared to be phenotypically identical to the immune mast cells found in uninfected cultures as determined by biochemical, immunological, and cytological tests. Although the cells expressed protein p21Ha-ras at levels similar to those in HaSV-transformed fibroblasts, they continued to require IL-3 for growth in vitro. Similar IL-3-dependent, long-term mast cell lines were also cultured from the enlarged spleens present in HaSV-infected mice. These results suggest that high-level expression of an activated Ha-ras oncogene enhances growth in these cells, perhaps by stimulating the progression of the cells into S, without affecting differentiation or altering the requirements for normal growth factor.
A patient is presented with generalized blistering and scarring eruptions, showing immunopathological, histological and electron microscopical characteristics of disseminated cicatricial pemphigoid, successfully treated by combined therapy of systemic steroids and azathioprine.
The indirect pancreatic function test, the fluorescein dilaurate test, was performed twice on nine healthy volunteers. The test is based on the renal excretion of fluorescein liberated from an ingested fluorescein ester by pancreatic enzymes. As a control, fluorescein sodium was ingested under similar conditions. The results of the test are expressed as the ratio of the amount of fluorescein excreted in the test part and the control part, forming the test/control (T/C) ratio. The mean values of excreted fluorescein correlated well with those reported by other authors. The coefficient of variation was 35% in the repeated test and of the same order in the series of individuals. The test cannot be recommended as a quantitative test until further investigation has been performed. Its use as a screening test is found questionable.
Standardized osteochondral fractures in the left femoral condyle in 19 adult mongrel dogs were fixed with either fibrin sealant or Kirschner wire. Adaptation and mechanical strength after 4, 7 or 8, and 14 days were compared with an in vitro study of the initial strength of the fibrin sealant in osteochondral fractures. After 4 days, the maximum tensile strength was increased in the fibrin sealed group, whereas no difference in energy absorption at failure was found. Although the initial strength of the fibrin bond was low, the tendency to displacement was less in this group. Our results suggest that fibrin sealant can be used for fixation of small osteochondral fragments, provided that immobilisation is sufficient.

The comparative efficacy of molindone and haloperidol, given by injection for the first 2-3 days of hospitalization and then continued orally for up to 4 weeks, is reported from an ongoing double-blind study. Efficacy and side effects were assessed by the Brief Psychiatric Rating Scale, Clinical Global Impressions, Treatment Emergent Symptom Scale, and Target Symptom Ratings. Analyses based on the first 35 patients who entered the study indicate that both drugs were effective and well tolerated. There were slight advantages for molindone early during the injectable phase of treatment and for haloperidol late during the oral portion of the study, but these differences were not clinically significant. No significant differences in side effects were found between the two drugs.

A 49-year-old patient with concomitant solar urticaria (SU) and lichen planus (LP) is presented. The sunlight- and UV-A-provoked wheal and flare SU reactions were interpreted as an antigen-antibody reaction, probably involving a reaginic antibody, because SU was passively transferred by a serum factor utilizing the Prausnitz-Küstner reaction. The reduced percentage of suppressor cytotoxic T-cell subset in peripheral blood typical of active LP may facilitate the increased production of reaginic antibodies after presentation of antigen by sunlight. Not only was the usual immunological reaction of LP (similar to graft-versus-host reaction) suppressed by PUVA treatment, but increased tolerance to sunlight was also achieved.
In a single-blind trial 25 patients with progressive scleroderma and Raynaud's phenomenon intravenous infusions of iloprost, a prostacyclin derivative (carbaprostacyclin), were given daily for five hours during a six-day hospital stay, after a comparable initial single placebo infusion. Duration, frequency and intensity of Raynaud symptoms improved in more than 75% of the patients. This improvement was objectified by telethermometry which demonstrated acral hyperthermia and significantly briefer rewarming after standardized cooling of the hands. In addition, there was more rapid healing of ulcerations and necroses of the digital pulp. A significant inhibition of ADP- and collagen-dependent platelet aggregation was demonstrated during the iloprost infusion. Side effects, such as headache, nausea and tiredness occurred only transitorily during the infusion, were individually highly variable, and then only at higher concentrations. A dosage of 2 ng/kg X min was tolerated by all patients.

This report describes two cases of giant lymph node hyperplasia (GLNH) with unusual clinicopathologic features, both studied with immunohistochemical techniques. In the first case, mesenteric GLNH was associated with amyloidosis and the nephrotic syndrome. In the second, GLNH developed in a patient with previously treated Hodgkin's disease. In both cases, the GLNH was of the plasma cell variant. The plasma cells of the first case contained both kappa and lambda light chains, while those in the second case contained only the lambda light chain. The presence or absence of monoclonality was not predictable from the morphology alone. Cases of GLNH with unusual clinicopathologic features are reviewed and the significance of immunohistochemical studies discussed.

After a historical account and a brief description of the techniques of signal analysis used in their laboratory (visual evoked potentials (VEPs) with statistical calculations, Fast Fourier Transform, correlation function), the authors present the experiments they undertook concerning colored stimulations. These experiments were carried out in 3 steps: colored stimulations on a cathode-ray screen, with constant luminance, concerning 8 healthy volunteers (72 VEPs analyzed); colored stimulations on a 'structured' screen (pattern reversal), concerning 31 healthy volunteers (186 VEPs analyzed); colored and pattern stimulations on a new cathode-ray stimulator built in the 'Centre National des Arts et Métiers' enabling both preceding types of stimulations with increased reliability and offering new possibilities of stimulation (variation of contrast, luminance or chromaticity precisely controlled) with various patterns in the background. The results evidence two different groups of electrophysiological responses, distinctly related to the presence or absence of a morphological structure in the stimulation. As well as this, a significant increase in latencies is observed with blue stimulations.
Peripheral blood lymphocytes and different immunocompetent cells of the cutaneous inflammatory infiltrate were characterized by reactivity with monoclonal antibodies directed to surface antigens of T lymphocytes and Langerhans cells by means of the indirect immunofluorescence technique in 10 patients suffering from lichen ruber planus. In contrast to the controls (n = 10), lichen patients had a significantly reduced percentage of the suppressor/cytotoxic T cell subset in peripheral blood (p less than 0.002). At the dermoepidermal junction suppressor/cytotoxic T lymphocytes and Langerhans cells predominated. Some possible conclusions are discussed.
In order to minimize rerupture after closed treatment of Achilles tendon rupture, the period of immobilization was prolonged from 8 to 12 weeks and an above-knee plaster cast was used the first 3 weeks. In 37 consecutive patients, followed for an average of 3 years, rerupture had occurred in two patients. It is concluded that modified closed treatment is a good alternative to surgical treatment.
