[Pulmonary hydatidosis. Comparison of cytology and scanning electron microscopy].

The puncture of a hydatid cyst with a fine needle is not generally recommended as a procedure and may even be contra-indicated in the first instance. Sometimes, however, the cytologist will be surprised to discover some scolices in the aspirate when the radiology is misleading, or not suggestive, and the serology is negative. We report two cases where the diagnosis was made by the cytological examination of the aspirate. The cytological study of the liquids was compared with electron microscopy scanning, enabling the stages of development of the parasite in the tissue of the pulmonary parenchyma to be assessed.

Chromosomal changes in an ovarian granulosa cell tumor: similarity with carcinoma.

The chromosomal changes in an ovarian granulosa cell tumor were studied after short-term culture in defined medium. The abnormalities found included X monosomy, structural rearrangements of chromosome #1, and an interstitial deletion of the long arm of chromosome #6. This pattern is closely related to that found in ovarian carcinoma and argues for the malignant nature of such tumors at the cellular level.

Usefulness of basement membrane markers in tumoural pathology.

The distribution of basement membrane (BM) markers, type IV collagen, laminin (LM), heparan sulphate proteoglycan (HSP) and fibronectin (FN) has been studied by indirect immunofluorescence using specific antibodies, in tumoural pathology. The disrupted pattern of BM by these markers in severe dysplastic lesions of the breasts, the bronchi and uterine cervix provides evidence for malignancy. In invasive carcinomas, there is generally a loss of these BM components, with FN persisting in the stroma. The loss of these markers in BM is concomitant and superimposable in double staining studies. In embryonic tumours, the presence of BM markers is related to a mesenchymal differentiation of malignant cells with pericellular FN and/or maturation towards organoid structures with BM. In sarcomas, there is a loss of the pericellular BM staining around most transformed muscular and Schwann cells and adipocytes. The persistence of this labelling in some well-differentiated areas can help to diagnose the nature of the sarcoma. The persistence of intercellular filaments of FN corresponds to the mesenchymal and/or sarcomatous nature of undifferentiated anaplastic proliferations.

Simultaneous ultrastructural localization of Ag-NOR (nucleolar organizer region) proteins and ribonucleoproteins during mitosis, in human breast cancerous tissues.

The simultaneous ultrastructural localization of the Ag-NOR proteins and ribonucleoproteins (RNP) was carried out during mitosis in human breast cancerous tissues using a one-step silver staining method followed by acetylation. Owing to the good preservation of the cells and to the high precision of the Ag staining we were able to follow the behaviour of the argyrophilic nucleolar components from prophase to late telophase. During all the phases of mitosis the Ag-NOR proteins were associated with fibrillar RNP component (without visible fibrillar centre), which originated, during prophase, from the dense fibrillar component of the nucleolus and then integrated within the periphery of some metaphasic chromosomes as irregular structures. The Ag staining of these structures was homogeneous but lower compared to those of nucleoli during prophase. The RNP sheath that covered the chromosomes was never silver stained. From anaphase to early telophase the argyrophilic RNP structures disposed within the periphery of the chromatin mass formed roundish and then spherical structures in which the Ag-NOR proteins were clustered as centrally localized spheres. These findings showed the usefulness of our method for the precise study of the spatial relationship between the Ag-NOR proteins and the chromosomes. Thus we were able to show striking segregation of both the argyrophilic structures at the periphery of the chromatin mass and of the Ag-NOR proteins within these structures. This suggests that a high degree of the structural organization in the future nucleus is already present during anaphase. This could be related to the rapid resumption of ribosomal RNA synthesis.

[Experimental vaginitis in rats].

The authors present an experimental model of vaginal inflammation in female rats, through local instillation of different substances causing inflammation of varying degrees. The inflammatory reaction was studied macroscopically, histologically and biochemically. Using polysiloxane as a protective substance of vaginal mucosa was found to be of therapeutic interest.

Simultaneous high resolution localization of Ag-NOR proteins and nucleoproteins in interphasic and mitotic nuclei.

Silver stainable proteins of the Nucleolar Organizer Regions (Ag-NOR proteins) of human breast cancer tissues have been localized at the electron microscopical level with a new method which combines a simple and reproducible one step Ag-NOR staining method combined with an acetylation procedure. This new method allows the fine identification of nucleolar components, particularly those which are stained by silver. In order to determine the cytochemical nature of the components associated with Ag-NOR proteins, the EDTA regressive preferential staining procedure for ribonucleoproteins has been applied to sections. By this means the precise localization of the Ag-NOR proteins was studied simultaneously with that of ribonucleoprotein within interphasic nucleoli and mitotic chromosomes. In interphasic nucleoli, stainable Ag-NOR proteins were localized in fibrillar centres and part of the dense fibrillar component. No silver deposits were seen on perichromatin or interchromatin fibrils and granules. In metaphasic nuclei, Ag-NOR proteins were only found on roundish fibrillar ribonucleoprotein structures, which could correspond to secondary constrictions. No silver deposits were seen on the well defined ribonucleoprotein sheet surrounding the chromosomes. In telophasic nuclei, Ag-NOR proteins were seen on the central part of roundish ribonucleoprotein fibrillar structures integrated in decondensing chromosomes. These structures have been interpreted as the nucleolar organizer regions around which rRNA synthesis resumes. In interphasic and mitotic nuclei, Ag-NOR proteins were never found within condensed chromatin but always in association with ribonucleoprotein components. The new method proposed here appears to be a useful tool for the simultaneous study of the localization of ribonucleoprotein and Ag-NOR proteins during the cell cycle.

[Thymic carcinoid tumors. A clinico-anatomic study of 3 cases].

The carcinoid tumour of the thymus was first detailed by Rosai in 1972. The authors describe three cases of this tumour with particular reference to their histogenesis. It appears to be a tumour predominantly affecting males with an often unfavourable, though slow, progression. Some pathological associations are frequent and characteristic. In one third of cases there is an associated endocrine neoplasia. As a rule the tumour is sited in the anterior mediastinum. The precise diagnosis rests on histochemical criteria (specific granulations). These carcinoids are tumours of the APUD system of endodermal origin. The treatment is surgical.

[Basement membranes and tumor pathology].

The distribution of four basement membrane components, type IV collagen (C IV), laminin (LM), heparan sulfate proteoglycan (HSP) and fibronection (FN) has been studied by indirect immunofluorescence using specific antibodies, in benign and malignant proliferations of the mammary gland and in soft tissue tumors. In breast carcinomas, specially intraductal cancers, there is a progressive and concomitant loss of these macromolecules around tumoral cells, preceding an overt tumoral invasion. In sarcomas, FN is frequently seen between malignant cells but the regular pericellular labeling observed around normal muscular cells, Schwann cells and adipocytes is absent. Nevertheless, the persistance of some pericellular staining with anti-C IV, anti-LM, anti-HSP and anti-FN antisera, in most differentiated territories of liposarcomas, leiomyosarcomas and neurifibrosarcomas can help to the diagnosis of such lesions.

[Osteogenic metaplasia or residual embryonic endometrial ossification?].

There are only about 90 cases described in the literature of the presence of an intra-uterine osseous structure--so the condition is not a common one. Often the aetiology is unknown. The purpose of this work is to describe the clinical picture and the anatomo-pathological characteristics on one form which leaves practically no doubt as to how the presence of bony structures in the uterus arise. We call this residual endometrial-ossification (O.R.E.) and note osteogenic or osseous metaplasia. We define O.R.E. as the presence of an osseous structure inside the uterus which does not arise from metaplasia of the cells of the endometrium.

Fine structure of the frog palate mucociliary epithelium.

The mucociliary epithelium (MCE) of the frog palate is a pseudostratified columnar respiratory epithelium supported by connective tissue with no submucosal glands. Morphologically different types of cells have been identified using scanning and transmission electron microscopy: ciliated cells, mucous and serous cells, intermediate cells, basal cells, and patches of vomero-nasal epithelium with sensory receptors and supporting cells. The cilia of the MCE of the frog palate are 5 to 7 microns long and a cluster of 3 to 7 projections forms a ciliary crown at their tips. Using cationized ferritin, anionic charges are identified on their whole outer surface. The mucus discharged by the secretory cells forms a continuous blanket which lies at the tips of the cilia.

Ultrastructural localization of Ag-NOR proteins and nucleic acids in reticulated nucleoli.

Nucleoli with a thread-like fibrillar structure found in several human breast cancer cells were investigated for the localization of Ag-NOR proteins, RNA, DNA and associated nucleoproteins. Various cytochemical techniques were applied: the one-step silver method for Ag-NOR proteins, the Bernhard's staining preferential for RNP, the Feulgen-like osmium-ammine reaction for DNA and the RNase- and DNase-gold complexes for the demonstration of RNA and DNA respectively. Staining for Ag-NOR proteins was restricted to the edge of the threads or cords and to the nodes of the nucleolar network. The Bernhard's EDTA technique gave a positive staining of the entire nucleoli and on small peripheral granules which were linked to the nucleolar cords by fine RNP fibrils. Localization of RNA molecules by the RNase-gold complex showed a similar pattern, dense labeling by gold particles over the nucleoli and at the periphery of the cords. DNA molecules were revealed by the osmium-ammine technique in small nucleoplasmic clumps and at the edge of the nucleolar cords. Similar results were obtained by applying the DNase-gold complex which showed a labeling by gold particles over the dispersed chromatin and at the edge of the nucleolar cords. The consistency of the results obtained speaks in favor of their specificity and seems to demonstrate the simultaneous localization of certain Ag-NOR proteins, DNA and RNA molecules at the edge of the nucleolar cords. This edge of nucleolar cords becomes then the candidate for the site of nucleolar transcriptional activity.

[Pathogenesis of cysts of the external meniscus].

A classification of cysts of the menisci into three groups is proposed. The first is secondary to dystrophy of the meniscus. The second is secondary to traumatic lesions of the meniscus. Third is related to degenerative changes in the meniscus. The authors have studied 7 cases in the first group, 23 in the second group and 15 in the third group. They emphasize the role of a horizontal defect in the meniscus with subsequent formation of a cyst. They consider that, in monopodal weight-bearing, intra-articular pressure forces synovial fluid into the defect to initiate the formation of a cyst. The authors propose that the cyst and the damaged meniscus should be systematically removed to avoid recurrence.

[Colonic nerve network demonstrated by quinacrine].

Quinacrine can be used for fluorescence microscopy to visualize certain cell bodies and fibres in the myenteric plexus of the intestine, forming a network different from the cholinergic and adrenergic systems. In the Piebald mouse, animal model of congenital megacolon, the quinacrine positive plexus is thin in the distal narrowed segment and well developed in the large proximal zone. In Hirschsprung's disease, the quniacrine positive network has the same pathologic features but, contrary to the animal model, there is an increased number and size of extrinsic cholinergic and adrenergic fibres. So this mouse is an animal model of a pseudo-Hirschsprung's disease, which has not yet been reported in human pathology.
