Novel glycosidase inhibitors, nojirimycin B and D-mannonic-delta-lactam. Isolation, structure determination and biological property.

A new aminosugar named nojirimycin B (1) has been isolated as its bisulfite adduct from the culture broth of Streptomyces lavendulae SF-425, together with nojirimycin. Microbiological oxidation of 1 with Gluconobacter suboxydans IAM 1829 gave a delta-lactam (2). The structures of 1 and 2 were determined to be 5-amino-5-deoxy-D-mannopyranose and D-mannonic-delta-lactam, respectively, on the basis of 1H NMR spectroscopy and X-ray structural analysis. Both 1 and 2 exhibited powerful inhibitory activity against rat epididymal alpha-mannosidase and apricot beta-glucosidase.

[An autopsy case of primary malignant melanoma of the esophagus--appraisal of radiation therapy].

A rare case of primary malignant melanoma of the esophagus is reported. Our case is of interest because an extraordinary effect was observed early during conventional radiation therapy. The primary lesion was irradiated with 60 Gy, it almost disappeared. Metastatic lesions were irradiated with 30 Gy, their volume decreased by more than 50%. At autopsy, these lesions were more or less unchanged. Histologically, cells from the main tumor were small and round. The growth of pleomorphic cells with large amounts of melanin granules was observed in the regions of irradiation. These findings suggest that the small round cells in malignant melanoma are radiosensitive and the pleomorphic cells with abundant melanin granules are radioresistant.

pMS76, a plasmid capable of amplification by treatment with chloramphenicol.

pMS76 is a nonconjugative, 5.54-megadalton plasmid. This plasmid is present in Escherichia coli K12 cells at about 20 copies per chromosome. In addition, the pMS76 plasmid can be mobilized by conjugative plasmids and it shares a number of other properties with the amplifiable ColE1 plasmid, including the ability to amplify copy number in the presence of chloramphenicol. However, pMS76 is compatible with ColE1-like replicons, pBR313, and with other multicopy plasmids, RSF1030 and pACYC184. Also the replication of pMS76 is rifampicin sensitive and requires DNA polymerase 1.

[Comparison between continuous arterial infusion chemotherapy and mitomycin C microcapsule administration in primary and metastatic liver cancer].

Continuous hepatic arterial infusion chemotherapy (HAI) and chemoembolization by microcapsulated Mitomycin C (MMC-m. c) were performed in patients with unresectable hepatoma or metastatic tumors of the liver. MMC-m. c showed significant antitumor effect and improvement of survival rate in unresectable hepatoma and liver metastasis of the breast cancer. Especially in liver metastasis of the breast cancer, MMC-m. c gained 80% of partial response rate and 11 months in 50% survival time. HAI was effective in multiple liver metastasis of colon and stomach cancer, showing 40% of one-year survival rate. A combination chemotherapy of HAI and MMC-m. c was performed in two cases of liver metastasis of the colon cancer. Tumor response was 100% in partial response rate. One of them died at 8 months and the other is alive at 8 months now.

[Detection of auditory impairment in the offsprings caused by drug treatment of the dams].

To study the auditory impairment induced by prenatal administration of aminoglycosides in the offspring, the shuttle box method to measure the auditory threshold of rats (Kameyama et al., Folia pharmacol. japon. 77, 15, 1981) was employed. Four groups of pregnant rats were administered 200 mg/kg kanamycin sulfate (KM), 200 mg/kg dihydrostreptomycin sulfate (DHSM), 100 mg/kg neomycin sulfate (NM), or 1 ml/kg saline intramuscularly from the 10th to the 19th day of pregnancy. The auditory threshold of the offspring could be measured by the shuttle box method in about 90% of the live born rats at the age of 100 days. The auditory thresholds of the groups were as follows (mean +/- S.E.): saline group, 53.8 +/- 0.6 dB (N = 36); KM group, 63.8 +/- 1.1 dB (N = 34); DHSM group, 60.0 +/- 1.2 dB (N = 29); NM group, 62.4 +/- 1.2 dB (N = 24). Auditory thresholds of drug-treated groups were significantly higher than that of the saline group. However, no increase in the auditory threshold of the mother rat was detected after treatment with aminoglycosides. In addition, the experimental procedure of the shuttle box method is very easy, and the auditory threshold of a large number of rats could be measured in a short period. These findings suggest that this method is a very useful one for screening for auditory impairment induced by prenatal drug treatment in rat offspring.

Effects of removal of the forestomach and caecum on the utilization of dietary urea in golden hamsters (Mesocricetus auratus) given two different diets.

1. Surgical removal of the forestomach, the caecum or both the forestomach and the caecum, was performed on growing male golden hamsters (Mesocricetus auratus) given a low-or high-fibre diet. 2. In Expt 1, 18 d after surgery, the hamsters which were given a concentrate before surgery were given an experimental low-fibre diet containing urea or [15N] urea (10 g/kg diet) for 9 d. In Expt 2, 5 d after surgery, the hamsters which were given a forage diet before surgery were adopted to an experimental high-fibre diet containing urea (10 g/kg) and dried Italian ryegrass (Lolium italicum) powder (400 g/kg diet) for 5 d, and 10 d after surgery, given a diet containing [15N]urea (10 g/kg diet) for 9 d. 3. In both experiments, removal of the caecum resulted in a significant lowering of both weight-gain and the digestibility of the dry matter, and removal of the forestomach resulted in a significant decrease of both accumulation of 15N in the body protein and proportion of 15N retained in the body protein as a proportion of the administered dosage. 4. The effect of removal of the caecum on the utilization of [15N] urea was not apparent in Expt 1, but the utilization of [15N] urea was significantly lowered by removal of the caecum in Expt 2. 5. These results suggest that the caecum may play a more important role in food utilization than the forestomach. However, the forestomach of the golden hamster plays a significant role in the utilization of dietary urea. It is presumed that the role of the caecum in the utilization of urea may vary under different dietary conditions.

Sarcoid myopathy with typical rash of dermatomyositis.

A 46-year-old woman had heliotrope erythema, poikiloderma, and proximal muscle weakness. Histologic examinations of the skin were consistent with dermatomyositis. Muscle biopsy revealed numerous noncaseating granulomatous lesions within the muscle fasciculi, confirming the diagnosis of sarcoid myopathy. No granulomatous lesion could be detected clinically in other organs. A combination of this skin and muscle disorder is rare and has not been reported previously.
