[Hepatic arterial infusion of 32P-radionuclide microspheres for radiation therapy of hepatocellular carcinoma].

OBJECTIVE: To investigate the efficacy of internal radiation of (32)P-glass microspheres ((32)P-GMS) in unresected hepatocellular carcinoma (HCC) via subcutaneous arterial port. METHODS: Hepatic arterial (99)technetium-macroaggregate albumin ((99)Tc-MAA) scanning via subcutaneous arterial port was undertaken to measure lung/liver shunting ratio and tumor/liver ratio. Hepatic arterial infusion of (32)P-GMS was performed in 17 cases of HCC with a dose from 1.11 to 1.30 GBq. Twenty cases of HCC undergoing hepatic arterial chemoembolization (HACE) in the same period served as controls group. RESULTS: There was no treatment-related death in the 17 cases. In 7 of the 17 cases, AFP level and/or tumor size decreased by 50% after treatment, with a response rate of 64.7%. The median survival time was 5.5 months, and the 3-, 6-, 9-, 12-month survival rates were 94.1%, 44.1%, 31.0%, 24.4%, respectively. The therapeutic efficacy was better than that of HACE. The survival time was significantly longer in patients with T/N ratio >or= 2 than in those with T/N < 2 (P < 0.05). CONCLUSIONS: Hepatic arterial infusion of (32)P-GMS is an alternative treatment for unresected HCC.

[Protect intercellular communication of pigment epithelium after laser photocoagulation].

OBJECTIVE: To observe the protective effect on the intercellular communication of retinal pigment epithelial (RPE) cells of dexamethasone (Dex) and verapamil (Ver) at different concentrations after laser photocoagulation. METHOD: Ten pig eyes were used to culture pig pigment epithelium cells. Use different concentrations of Dex and Ver to incubate cultured RPE cells 2 hours before laser coagulation, and then measure the intercellular communication function of confluent cells surrounding the coagulation spots after photocoagulation. RESULT: 125 mg/L Dex and 20 mg/L Ver could protect the intercellular communication of RPE cells under laser photocoagulation. The high concentration of drugs had better result. CONCLUSION: Both Dex and Ver can preserve the intercellular communication in vitro. The protective capacity is related to the concentration of drug.

[Development of plasmid-based transgenic mice carrying with target gene xylE].

OBJECTIVE: To develop a transgenic mouse model carrying with plasmid pUC118NX integrated into its genomic DNA for detecting mutagenesis. METHODS: DNA of plasmid UC118NX in target gene xy1E was injected microscopically into male protonucleus of 376 mouse spermatova, and 225 survival spermatova were transferred into the oviducts in both sides of 11 pseudopregnant female mice to develop their offspring. The genomic DNA in survival young mice were analyzed with polymerase chain reaction (PCR)-Southern blot, plasmid transformation test and endonuclease-digestion. RESULTS: Seven pseudopregnant mice got pregnant, and 29 offspring were delivered and 25 survived of which 18 were identified with positive for PCR-Southern blot (72%). The two stout male mice with intact integration of plasmid pUC118NX in their genome were finally chosen as founders to detect gene mutation in vivo and establish transgenic mouse lineages. CONCLUSION: Transgenic C57BL/6J mice integrated with plasmid pUC118NX into their genomic DNA have been successfully developed.

[Affect of insulin-like growth factor I and estradiol on the growth of uterine leiomyoma].

To study roles of insulun-like growth factor 1(IGF-I) and estradiol(E2) in the growth of leiomyoma. Protein expressions of IGF-I receptor(IGF-I-R), estrogen receptor(ER), and cell proliferation associated nuclear antigen(ki-67) were analysed by streptavidin-peroxide method in 40 cases. Serum level and local concentration of IGF-I were determined by immunoradiometric assay(IRMA) in 20 cases. Serum level of estradiol was determined by radioimmunoassay in 20 cases. Twenty normal fertile women were studied as control. Protein expressions of IGF-I-R and ER in uterine leiomyoma were significantly higher than in myometrium(P < 0.05); and local concentration of IGF-I positively correlated with ki-67 (r = 0.6513, P < 0.05); and expression of estrogen receptor. The serum levels of IGF-I and E2 had no significant difference between leiomyoma patients and normal women. IGF-I and E2 promote the growth of leiomyoma and IGF-I may act as a mediator of estrogen.

[Comparative study of ventilatory functions of smokers with MTTp and F-V curve configuration].

To assess the clinical significance of partial mean transit time(MTTp) and F-V curve configuration for testing small airway functions in smokers, routine ventilatory functions, MTTp and F-V curve configuration were studied in 116 passive and active light, heavy smokers. Routine ventilatory functions and F-V curve configuration were determined with routine methods. MTTp were calculated with "convenient calculation". The results showed that the distribution of convex type of F-V curve rose with the degree of smoking. The decrease of V50, V25 and prologation of MTT45%-55%, MTT70%-80% in convex type were just correspondent with heavy smokers. The range of prolongation of MTT45%-55% and MTT70%-80% was 110%-120% of predicted value both in heavy smokers and convex type. These showed there were light disturbances in small airways. Therefore, the convex type, the prolongation of MTT45%-55% and MTT70%-80%, and the decrease of V50 and V25 should all be used as sensitive indices for testing small airway functions. The value of MTTp calculated with "convenient calculation" is accurate and easy-to-do so the method is appropriate for clinical application.

[Significance of specific time constant and F-V curve configuration in evaluating small airway function for smokers].

To evaluate the clinical significance of the specific time constant (S tau) and F-V curve configuration in testing small airway functions for smokers, routine ventilatory functions, S tau and F-V curve configuration were studied in 116 passive and light, heavy active smokers. Routine ventilatory functions and F-V curve configuration were determined with routine method. S tau were calculated from S tau = 1/V. The results showed that the distribution of convex type rose with the degree of smoking, and the decrease of V50, V25 and elongation of S tau 50, S tau 25 in convex type were just consistent with heavy smokers. The range of elongation of S tau 50 and S tau 25 was 120%-130% of predicted value in both heavy smokers and convex type. These showed there were slight disturbances in functions of small airways. Compared with COPD patients previously measured, their S tau 75, S tau 50, S tau 25 all prolongated to higher degree and meant both large and small airways were affected. Therefore, the convex type, elongation of S tau 50 and S tau 25, and decrease of V50 and V25 were sensitive indices for detecting small airway functions. They can be recommended for clinical application.

[Characteristics of chord time constant and F-V curve configuration in testing small airway functions for smokers].

UNLABELLED: To study the significance of chord time constant (tau ch) and F-V curve configuration in testing small airway function for smokers, the routine ventilatory functions, chord time constant (tau ch) and F-V curve configurations were studied in 116 passive and active (light, heavy) smokers. The tau ch was calculated with Knudson's method, tau ch = V/V. RESULTS: The distribution of convex type rose with the degree of smoking. The tau ch50 in heavy smokers and convex F-V curve prolongated to 130% of predicted value; tau ch50 in light smokers prolongated to 120%. These reflected that there were different degrees of disturbance in small airway functions of smokers and suggested that tau ch50 should be a sensitive and specific index for testing small airway functions. On the other hand, the tau ch75, tau ch50 and tau ch25 by the present authors previously measured in patients with chronic obstructive pulmonary diseases (COPD) all prolongated with the severity of the disease, which reflected that both large and small airways were affected. Therefore it is concluded that in terms of clinical implications the specificity of tau ch50 for small airway functions and the prolongation of tau ch with the severity of COPD are useful indexes.

[Vroman effect of plasma protein adsorption to biomaterials surfaces].

Fibrinogen adsorption from plasma to biomaterials surfaces passes through a maximum when studied as a function of adsorption time, plasma concentration, or column height in narrow spaces and these are called Vroman effect. Studies have demonstrated that Vroman effect is a general phenomenon of plasma proteins and reflects the competitive adsorption of plasma proteins for a limited number of surface sites. In this paper, the factors affecting Vroman effect, the relationship between contact activation and fibrinogen displacement, and the significance of the Vroman effect with respect to blood-material interactions are reviewed.

[Detection of hypodermal and hematopoietic necrosis baculovirus by PCR method].

For control of the explosive epidemic disease of Penaeid shrimp in China. We have developed a method for detection of Hypodermal and hematopoietic necrosis baculovirus (HHNBV) by polymerase chain reaction (PCR). Two pairs of PCR primers (DP1/DP2 and DP3/DP4) were prepared, based on the RFLP analysis between HHNBV and penaeid rod-shaped DNA virus. PCR using either DP1/DP2 or DP3/DP4 specified HHNBV genome. When PCR products amplified with DP1/DP2 were subjected to nested PCR with DP3/DP4, a high sensitivity about 0.04 pg was seem with the nested PCR.

[Expression of a HCV multi-epitopes antigen gene and study on its immunogenicity].

Due to the hypervariable character of hepatitis C virus (HCV), 5 conserved T and/or B cell epitopes from core, envelope, NS3 and NS5 protein of HCV were chosen to form a 270 bp multi-epitopes antigen gene. The gene was clone into a fusion vector pWR450-1 to express a beta-galactosidase-HCV hybrid protein GZ-PCX. The purified GZ-PCX protein was specifically recognized by human anti-HCV antibodies. These results show that the HCV hybrid multi-epitopes antigen has excellent immunogenicity, which might be able to be used as an effective diagnosis agent and to provide protectivity to any genotype of HCV which might partly solve the problems in the researches of HCV vaccines.

[Cloning of mitomycin C resistance gene from Streptoverticillium caespitosum ATCC27422 and studying its function].

Streptoverticillium caespitosum is the major producer of anti-cancer drug mitomycin. It demonstrated that there exists a strong restriction system for the transformation of foreign DNA in this strain. Using the blocked mutant S-6 which is sensitive to mitomycin C (MC) and does not produce mitomycin as a recepient and plasmid pIJ699 as vector, the gene library of the strain ATCC27422 was constructed. A clone of mitomycin C resistance gene(mcr) was obtained by screening from this library. The mitomycin C resistance gene cloned on the 6.2 kb fragment of plasmid pLX5 could be expressed in the Strptonmyces lividans TK54. It was the first time to introduce successfully the plasmid pLX5 into the wild type strain ATCC27422 by electroporation technique. And the transformant's resistance to mitomycin C has risen greatly: the MIC has gone up from 200 micrograms/ml to more than 1000 micrograms/ml. The result of fermentation showed that the yield of mitomycin of ATCC27422 harboring plasmid pLX5 was more than the wild strain ATCC27422's. It suggested that there is some relationship between the level of the resistance to mitomycin C and its biosynthesis.

[The visible spectra study of the conformation equilibrium of iron-metal-free porphyrin dimers].

A series of iron(III)-metal-porphyrin dimers linked with flexible alkoxyl chain at para position of two phenyl rings have been synthesized and characterized. An equilibrium of two conformations of porphyrin dimers (open and closed) has been studied by visible spectra. The intensity of absorption band 680 nm relates to equilibrium of two conformations. With the increase of the length of the alkoxyl chain, the closed conformation forms more easily. With the addition of axial ligand, the ferric ion of the dimers tends to bind with imidazole, and the intensity of the band 680 nm gradually decreases and disappears.

[Studies on tailed metalloporphyrin coordination compounds. IX. Synthesis of tailed porphyrin with covalently linked phenylalanine and its fluorescence property].

A new tailed porphyrin with covalently linked phenylalanine 2 and its zinc complex 3 were synthesized. Intramolecular energy transfer (from singlet excited phenylalanine to porphyrin) was observed in compound 2 and 3. The influence of coordination chemistry factor on the fluorescence of porphyrin was also discussed.

[Indirect determination of trace silicon in cobaltosic oxide by extraction-FAAS].

After the heteropolymobdate of silicon and phosphorus was formed, the heteropolymobdate of phosphorus was extracted into CH3COOH4H9 phase and P as interference element was separated. The heteropolymobdate of silicon was extracted by MIBK. Indirect determination of Si was achieved with Mo in MIBK determined by FAAS. Applying this method to the determination of silicon in cobaltosic oxide, the interferences of matrix cobalt and other coexistence elements were avoided. Compared to spectrophotometric method, the result is satisfactory.

[The hydrogen bond between 5-(2-carboxyphenyl)-10, 15, 20-triphenylporphyrin and amino acid ester--study with UV-Vis titration and 1H-NMR].

The hydrogen bonds between the hydrogen of pyrrols in 5-(2-carboxyphenyl)-10,15,20-triphenylporphyrin and the oxygen of the carboxyl group of amino acid ester were confirmed by UV-Vis spectroscopic titration method and 1H-NMR. When amino acid ester was added to o-TPPCO2H solution in CHCl3, the Soret and Q and red-shifted dramatically and several isobestic points could be observed in UV-Vis spectra. In 1H-NMR, the signal of the hydrogen atoms of pyrrole in o-TPPCO2H (-2.78 ppm) decreased and a new signal in 0.260 ppm emerged, when o-TPPCO2H mixed with L-PheOEt at 1 : 1 (mol/mol).
