Characterization of a region of the lutropin receptor extracellular domain near transmembrane helix 1 that is important in ligand-mediated signaling.

The lutropin receptor (LHR), a member of the G protein-coupled receptor family, contains a relatively large N-terminal extracellular domain, accounting for about half of the receptor and responsible for high affinity ligand binding, and a standard heptahelical portion with connecting loops and a C-terminal tail. LHR and the other two glycoprotein hormone receptors, i.e. the follitropin and TSH receptors, contain an invariant 10-amino acid residue sequence, FNPCEDIMGY (residues 328-337 in rat LHR), in the extracellular domain separated by only a few amino acid residues from the beginning of transmembrane helix 1. In view of the invariant nature of this region in the three glycoprotein hormone receptors and preliminary data in the literature on the importance of Glu332 and Asp333 in signal transduction, we undertook a systematic investigation of all 10 amino acid residues because this region may function as a switch or trigger for communicating ligand binding to the extracellular domain with a conformational change of the membrane-embedded C-terminal half of the receptor to activate G proteins, particularly Gs. A total of 36 single, double, and multiple replacements, as well as two deletions, of LHR were prepared and characterized in transiently transfected COS-7 cells. Of these mutants LHRs, 26 expressed on the cell surface in sufficient numbers that quantitative assessments could be made of human choriogonadotropin binding and ligand-mediated cAMP production. Replacements of Cys331 abolished ligand binding to intact cells, although binding could be detected after solubilization of the cells. Replacements of the other nine amino acid residues that did not interfere with receptor folding or trafficking had no significant effect on ligand binding affinity; however, replacements of Pro330, Glu332, and Asp333 resulted in diminished signaling, especially for the two acidic residues. An interesting observation was made in which replacement of Tyr337 with Ala or Asp, while having no profound change on receptor function, could overcome to some extent limited expression of replacements at positions 332 and/or 333, thus permitting a more definitive analysis of signaling. Replacement of the decapeptide sequence with Gly10 prevents expression, whereas deletion of all 10 residues and deletion of Glu332-Asp333 prevents functional expression at the cell surface. Thus, this invariant sequence in the glycoprotein hormones is required for proper folding, trafficking, and ligand-mediated signaling, but not ligand binding, in LHR. Amino acid residues, Glu332, Asp333, and to a limited extent, Pro330, are important in ligand-mediated signaling but not ligand binding.

Essential role of biliary glycoprotein (CD66a) in morphogenesis of the human mammary epithelial cell line MCF10F.

Normal mammary epithelial cells express the cell surface protein biliary glycoprotein (BGP or CD66a) in a polarized manner, suggesting that this protein may play a role in the formation of mammary acini. In order to test this hypothesis, we interrupted the expression of BGP in the mammary epithelial line MCF10F when cultured in or on Matrigel, a source of extracellular matrix (ECM). When analyzed by immunofluorescence confocal microscopy, the BGP staining is confined to the lumenal surface and colocalizes with actin. Sequential scanning electron microscopy demonstrates that the MCF10F cells migrate to form clusters, followed by apoptotic cell death within the center, resulting in lumen formation. Transmission electron micrographs reveal the presence of tight junctions and desmosomes between the cells, microvilli along the lumenal surface, and typical apoptotic bodies within the lumen. When the MCF10F cells are transfected with the BGP antisense gene and grown in Matrigel, they exhibit reduced acini formation (12% and 20%) compared to untransfected cells (52%) or to cells transfected with vector only (62%). Acini formation is also significantly reduced when MCF10F cells grown in Matrigel are treated with anti-BGP antibody (18% at 100 microgram/ml), or recombinant soluble BGP (18% at 0.4 microM). In contrast, the BGP-negative MCF7 breast tumor cell line, which does not form acini when grown in matrigel, exhibits >60% cell death with the occasional formation of acini, when transfected with the BGP sense gene and grown in Matrigel. These results support the hypothesis that BGP plays a role in the normal differentiation program of mammary epithelial cells, indicating that its expression is essential to the formation of the lumen. Furthermore, and as shown by others, the differentiation program depends on the presence of ECM. The lack of expression of BGP in the MCF7 breast cancer cell line suggests that the downregulation of BGP expression confers a growth advantage to these cells in ECM. In addition, we found that the MCF10F cells could be separated into a BGP-positive epithelial fraction (MCF10F-e), and a BGP-negative myoepithelial fraction (MCF10F-m). When the myoepithelial cell-enriched fraction is grown on Matrigel, web-like structures are formed. These cells have a typical spindle shape cell morphology and express keratin, alpha-smooth muscle actin and vimentin, markers of the myoepithelial cell phenotype. When MCF10F-m cells are treated with IFNgamma, they express CEA (carcinoembryonic antigen) but not BGP. Since breast carcinomas, especially in situ carcinomas, express CEA, this finding may suggest a heretofore unappreciated relationship between myoepithelial cells and breast cancer.

The cellular basis of corneal transparency: evidence for 'corneal crystallins'.

In vivo corneal light scattering measurements using a novel confocal microscope demonstrated greatly increased backscatter from corneal stromal fibrocytes (keratocytes) in opaque compared to transparent corneal tissue in both humans and rabbits. Additionally, two water-soluble proteins, transketolase (TKT) and aldehyde dehydrogenase class 1 (ALDH1), isolated from rabbit keratocytes showed unexpectedly abundant expression ( approximately 30% of the soluble protein) in transparent corneas and markedly reduced levels in opaque scleral fibroblasts or keratocytes from hazy, freeze injured regions of the cornea. Together these data suggest that the relatively high expressions of TKT and ALDH1 contribute to corneal transparency in the rabbit at the cellular level, reminiscent of enzyme-crystallins in the lens. We also note that ALDH1 accumulates in the rabbit corneal epithelial cells, rather than ALDH3 as seen in other mammals, consistent with the taxon-specificity observed among lens enzyme-crystallins. Our results suggest that corneal cells, like lens cells, may preferentially express water-soluble proteins, often enzymes, for controlling their optical properties.

Specific intervention for multiplication skill with learning-disabled students.

A multiple baseline design was used to assess the intervention of individual tutoring to help two learning-disabled students (15 and 16 years old) improve their multiplication of 1-digit numbers over nine days of 10-min. instruction. Observation or visual analysis indicated 40 to 50% improvement after the intervention, and a nonparametric test of randomization showed that the students' improvement was retained during the 2-wk. follow-up.

Transforming growth factor(beta)-mediated corneal myofibroblast differentiation requires actin and fibronectin assembly.

PURPOSE: Recent studies indicate that transforming growth factor (TGF)beta is a potent inducer of corneal myofibroblast differentiation and expression of smooth muscle-specific, alpha-actin (alpha-SMA). Although TGFbeta is known to enhance synthesis of extracellular matrix proteins and receptors, little is known about how it modulates the expression of smooth muscle proteins in nonmuscle cells. The purpose of this study was to identify the role of Arg-Gly-Asp (RGD)-dependent tyrosine phosphorylation in regulating alpha-SMA gene expression and ultimately myofibroblast development. METHODS: Because cell culture in serum-containing media mimics myofibroblast transformation, all experiments were performed on freshly isolated rabbit keratocytes plated in defined, serum-free media. Cells were exposed to TGFbeta (1 ng/ml), Gly-Arg-Gly-Asp-D-Ser-Pro (GRGDdSP, 50 microM), Gly-Arg-AL-Asp-Ser-Pro (GRADSP; 100 microM), or herbimycin A (0.1-10 nM) at 24 hours (sparse) or 7 days (confluent). Cells were evaluated by immunocytochemistry and proteins and RNA collected for western and northern blot analyses using antibodies specific for alpha-SMA, fibronectin, focal adhesion proteins, and phosphotyrosine (clones 4G10 and PY20); and probes directed against rabbit alpha-SMA. All experiments were repeated at least three times. RESULTS: Keratocytes exposed to TGFbeta showed expression of alpha-SMA that coincided with the intracellular reorganization of the actin cytoskeleton and the extracellular assembly of fibronectin fibrils. Addition of RGD containing but not control peptides blocked the organization of intracellular actin, extracellular fibronectin, and alpha-SMA protein and mRNA. Immunoprecipitation of cell proteins with 4G10 or PY20 identified the TGFbeta-associated tyrosine phosphorylation of paxillin, pp125fak, p130, PLCgamma, and tensin, which was blocked by addition of GRGDdSP. Addition of herbimycin A to keratocytes exposed to TGFbeta showed a dose-dependent loss of alpha-SMA protein and mRNA which correlated with loss of tyrosine phosphorylation, absence of actin reorganization, and fibronectin assembly. CONCLUSIONS: The data suggest that TGFbeta-mediated alpha-SMA gene expression leading to myofibroblast transformation may involve an RGD-dependent phosphotyrosine signal transduction pathway.

Tubby-like protein 1 homozygous splice-site mutation causes early-onset severe retinal degeneration.

PURPOSE: To characterize the disease expression of an autosomal recessive human retinal degeneration associated with a mutation in TULP1 (tubby-like protein 1), a gene with currently unknown function. METHODS: Homozygotes and heterozygotes from an extended Dominican kindred with a TULP1 splice-site gene mutation (IVS14+1,G-->A) were studied clinically and with visual function tests. Sequence analysis of TULP1 was also performed in unrelated patients with severe retinal degeneration from a North American clinic population. RESULTS: Homozygotes had nystagmus, visual acuity of 20/200 or worse, color vision disturbances, bull's eye maculopathy, and peripheral pigmentary retinopathy. Younger patients had a relatively wide extent of kinetic visual fields; older patients had only peripheral islands. No rod function was measurable by psychophysics in any of the patients; markedly reduced cone function was detectable across the visual field of younger patients and in the remaining peripheral islands of older patients. Rod and cone electroretinograms (ERGs) were not detectable using standard methods; microvolt-level cone ERGs were present in some patients. Heterozygotes had normal visual function. No putative pathogenic sequence changes in TULP1 were observed in North American patients with comparably severe retinal phenotypes, mainly in the diagnostic category of Leber congenital amaurosis. CONCLUSIONS: This TULP1 splice-site mutation in homozygotes causes early-onset, severe retinal degeneration involving macular and peripheral cones and rods. The constellation of phenotypic findings suggests that the TULP1 gene product is critically important for normal photoreceptor function and may play a role in retinal development.

[Significance of maximal midexpiratory time and F-V curve configuration in testing small airway function for smokers].

To evaluate the clinical significance of maximal midexpiratory time (MET) in testing small airway functions for smokers, the routine ventilatory functions, MET and F-V curve configurations were studied in 116 passive and light, heavy active smokers. Routine ventilatory functions and F-V curve configuration were determined with LR80 flow volume recorder. The MET was measured with FJD80 spirometer. The results showed that the distribution of convex type rose with the degree of smoking. In heavy smokers, the MET elongated to 127% of the predicted value and in the convex type F-V curve, it elongated to 120% of the predicted value; both were consistent with the decrease of corresponding V50, V25. This implied when MET elongated to 120%-130% of the predicted value in smokers, there had been slight disturbances in small airway functions. It is therefore suggested that MET and convex type F-V curve both are sensitive indices for testing small airway functions, thus it should be of clinical significance and be appropriate for general application.

Portal hemodynamics in cirrhotics with portal hypertension using color Doppler velocity profile.

OBJECTIVE: To investigate portal hemodynamics and its correlation with esophageal variceal bleeding (EVB) in cirrhotics with portal hypertension by using a newly-developed technique, color Doppler velocity profile (CDVP). METHODS: Hemodynamics of portal trunk (PT), right anterior branch (RAB) and splenic vein (SV) were evaluated in 48 cirrhotics with portal hypertension and 35 normal volunteers by CDVP. The parameters included maximum cross-sectional velocity (CS-Vmax), flow volume, congestion index (CI), profile parameter N and pattern of flow curve. Stepwise logistic regression model was employed to determine EVB-relating factors in cirrhotics. RESULTS: CS-Vmax in PT and RAB was significantly lower in cirrhotic group than that of normal group, being 14.91 +/- 3.08 cm/s, 9.44 +/- 2.70 cm/s vs 30.52 +/- 6.75 cm/s, 12.82 +/- 2.69 cm/s, respectively (P < 0.01 for both). Flow volume of PT and SV was significantly higher in cirrhotic group compared with that of normal group, being 25.16 +/- 10.48 ml.min-1.kg-1, 15.83 +/- 9.18 ml.min-1.kg-1 vs 20.43 +/- 5.57 ml.min-1.kg-1, 5.81 +/- 2.04 ml.min-1.kg-1, respectively (P < 0.01 for both). CI of PT, RAB and SV was significantly higher in cirrhotic group than in normal group, being 0.142 +/- 0.0654, 0.105 +/- 0.0496, 0.0884 +/- 0.0431 vs 0.0326 +/- 0.0142, 0.0757 +/- 0.0342, 0.0483 +/- 0.0230, respectively (P < 0.01 for all). In dynamic variation of flow volume over time, RAB and SV in cirrhotic group increasingly presented flat pattern and periodically changed pattern, respectively (P < 0.01 for both). Between cirrhotic subgroups without and with EVB history, there were significant differences in flow volume, CI and N value of SV, splenic size, degree of esophageal varices (EV) and portal hypertensive gastropathy (PHG), and stepwise logistic regression revealed that N value of SV, splenic size, degree of EV and PHG were four independent factors in relation to EVB. EVB scores calculated from the regression equation had a close correlation with EVB. In patients with EVB score > 0, 88.9% of them had EVB, and in those with EVB < 0, 76.9% of them had no history of EVB. CONCLUSION: In cirrhotics with portal hypertension, portal venous system has the features of elevated vascular resistance and hyperdynamics, and the latter mainly results from increased blood flow in SV. EVB score may become a valuable parameter in predicting occurrence of EVB.

[Mutation of human mismatch repair genes in hereditary nonpolyposis colorectal cancer (HNPCC) families].

OBJECTIVE: To investigate germline mutations of mismatch repair genes hMLH1 and hMSH2 in 29 HNPCC families. METHODS: Mutations were detected by PCR-SSCP analysis, and confirmed by DNA sequencing. RESULTS: (1) In 29 families, the overall mutation rate of these two genes was 31.0% (9/29). Mutation detected in all of the 9 families was located in the hMLH1 gene, indicating that the hMLH1 gene was the main responsible gene in these families. An obvious difference was observed when compared to the mutation rate in the control group (P < 0.01); (2) All of the 10 family members who developed colorectal cancer (CRC) harbored the same mutation as detected in their proband, indicating a strong association between gene mutation and development of CRC. In 29 healthy family members, 5 asymptomatic carriers of mutation were found who were considered as the high-risk populations of CRC. CONCLUSION: A certain number of HNPCC families can be benefited from the genetic screening for mutation of the mismatch repair genes.

[Dynamic enhanced spiral CT characteristics of recurrent nasopharyngeal carcinoma and radiofibrosis].

OBJECTIVE: To study the characteristics of dynamic enhanced density-time curves of recurrent nasopharyngeal carcinoma(RNPC) and radiofibrosis(RF) in spiral CT. METHODS: Fifty cases of RF and 46 cases of RNPC were examined both before and after contrasting in spiral CT. The CT values of the regions of interest (ROI) at various acquisition time (0, 20, 30, 40, 50, 60, 90, 120, 150, 180, 210, 240, 270, and 300 sec after the beginning of the contrast injection) were collected. The difference in CT value of various acquisition time was analysed statisticaly and the dynamic enhanced density-time curves were established. RESULTS: The typical curve in RF group was a slowly ascending-slowly descending one. Its peak CT value as seen at 210 sec postcontrast and measured 60.1 +/- 2.57 Hu. That of the RNPC was of rapidly ascending-slowly ascending-slowly descending one. Its peak CT valne was seen at 150 sec postcontrst and measured 75.4 +/- 7.2 Hu. The differences of CT values were significant between RNPC and RF from 40 to 300 sec postcontrast (P < 0.001). CONCLUSION: The different characteristics of the dynamic enhanced desity-time curves and CT values of the ROI may provide important informations for differential diagnosis of RNPC and RF.

[The effect of memory T lymphocytes on IgE production by B lymphocytes in patients with bronchial asthma].

OBJECTIVE: To study the role of CD(4)(+) CD(45)RO(+) T lymphocytes in the pathogenesis of bronchial asthma. METHODS: CD(4)(+) CD(45)RO(+) T lymphocyte subsets were isolated from peripheral blood of patients with bronchial asthma and healthy donors. Then they were cultured with self-B lymphocytes and designated as pokeweed mitogen (PWM)-stimulated and non-stimulated group. Secreted IgE was measured in the supernatant. RESULTS: CD(4)(+) CD(45)RO(+) T lymphocytes had a positive role on IgE production by B lymphocytes from patients of bronchial asthma in non-stimulated group, but the positive role did not exit in the presence of PWM. The effect of activated and non-activated CD(4)(+) CD(45)RO(+) T lymphocytes from patients with bronchial asthma on the secretion of IgE by B lymphocytes were further investigated. CONCLUSION: The function of activated CD(4)(+) CD(45)RO(+) T lymphocytes returned to normal level. It is suggested that CD(4)(+) CD(45)RO(+) T lymphocytes have adverse effects on IgE production in patients with bronchial asthma; which may be related to the pathogenesis of asthma.

[Evaluation of therapeutic effects of portoenterostomy: reported of 426 cases].

OBJECTIVES: To investigate the selection of the types of anastomosis and to evaluate the therapeutic effects of portoenterostomy. METHODS: From 1993 to 1998, 426 patients with obstructive jaundice underwent portoenterostomy. The patients included 236 cases (56%) of hilar strictures in hepatolithiasis, 98 cases (23%) of end-stage periampullary tumors, 64 cases (15%) of proximal cholangiocarcinoma, and 28 cases (6%) of congenital choledochus cyst. Reconstruction of biliary tract was performed after resection of affected duct or palliative bypass for end-stage carcinoma. The types of anastomosis included extra or hilar hepatic bilioenterostomy (160 cases, 37%), and intra-hepatic duct anastomosis with different type of hepatectomy (266 cases, 63%), in which anastomosis was performed by round ligament approach (34 cases, 8%), and anastomosis through gallbladder fossa (15 cases, 4%). RESULTS: Short- and 1-6 year follow-up showed that jaundice of patients was completely relieved by suitable type of bilioenterostomy. CONCLUSIONS: The good therapeutic effects of bilioenterostomy are based on the correct selection of anastomosis methods.

[Effect of wear particles on cell-to-cell communication of synoviocyte system in vitro and its significance in aseptic loosening of prosthesis].

OBJECTIVE: To study the biological reason of abnormal fiber proliferation at bone-implant interface of aseptically loosened prosthesis by observing cell-to-cell communication of synoviocyte system in vitro under the stimulation of wear particles. METHODS: The synoviocyte system of normal human hip joint was established in vitro and the change of cell-to-cell communication was monitored by fluorescence redistribution after photobleaching technique (FRAP) and confocal laser scanning microscope (CLSM) after Ti alloy, CoCr alloy or UHMWPE particles suspension (1.5 mg/ml, W/V) was added into the system. RESULTS: Cell-to-cell communication of synoviocyte system significantly decreased under the stimulation of three kinds of particles (P < 0.01). he decreased degree was maximal in UHMWPE group and minimal in Ti alloy group (P < 0.01). The cell-to-cell communication level of fibroblast-like-cells (FCs) was significantly lower than that of macrophage-like-cells (MCs) (P < 0.05). CONCLUSIONS: Besides the effect of cytokines promoting fiber proliferation, the great amount of fibrous granuloma at bone-implant interface which has relations with aseptic loosening may be caused by the decrease of cell-to-cell communication of fibroblast under direct stimulation of wear particles.
