[Studies on tannin and hydrolysate in three species of Chinese Caesalpinia plants].

OBJECTIVE: To get Chinese Caesalpinia plants that can replace the imported Tara to produce tannin and gallic acid. METHOD: Extract tannin with water, condense and hydrolyze the extracts with alkali, the hydrolysates were identified. Furthermore, an enlarged scale experiment was conducted. RESULT: Tannin contents of the plants were 32.2%, 46.6% and 58.5% respectively, the converted ratios of raw material to hydrolysate were 154:1, 11.8:1, 3.09-2.79:1 respectively. CONCLUSION: We got the Tara-replacing plants in Chinese Caesalpinia plants for the first time.

[Identification of medicinal and edible tender shoots of Aralia in China].

OBJECTIVE: To Study the identification of medicinal and edible tender shoots of Aralia in China. METHODS: Morphological identification and differential thermal analysis(DTA). RESULTS: Important morphological and DTA features for identification were found. CONCLUSION: It is difficult to identify these tender shoots merely on morphological characteristics, but the combination of morphological features with distinctive aspects of DTA curve helps distinguish these species and give an identification index.

[Species and distribution of Chinese medicinal plant Caesalpinia].

OBJECTIVE: To gain a clear idea on the resources of medicinal plant Caesalpinia in China. METHOD: Conducting field investigation and consulting related specimens and data. RESULT: The distribution, growing environment and medicinal parts of 14 species of Caesalpinia have been clarified, and a key for their identification is given. CONCLUSION: A scientific basis for further study of the medicinal plant Caesalpinia in China has been provided.

[Investigation on the fruit output and ecological environment of Caesalpinia in China].

OBJECTIVE: To estimate the fruit output of Caesalpinia in China and understand its ecological environment. METHOD: On-the-spot investigation in the producing area. RESULT: The output of 8 species of Caesalpinia was estimated and their ecological environment was described. CONCLUSION: The natural resources of Chinese Caesalpinia, especially C. decapetala and C. minax, are rich, and thus worth exploring and developing.

[Phenotypic changes of mandibular condylar cartilage cells during subculture].

OBJECTIVE: To demonstrate the phenotypic changes of mandibular condylar cartilage(MCC) cells during subculture in vitro. METHODS: MCC cells were harvested from human fetus by digestion of collagenase and cultured in DMEM supplemented with 10% newborn calf serum, then they were passaged when reached confluence. Morphological changes were observed under phase-contrast microscope. Type II collagen, proteoglycan synthesis and type I, II and III procollagen mRNA levels were also studied in serial monolayer cultures. RESULTS: During subculture, type II collagen and proteoglycan synthesis were decreased, being accompanied by a lower type II collagen mRNA level. In contrast, type I and III procollagen mRNA levels were elevated progressively. The morphology of polygonal-shaped chondrocytes was lost to the bipolar fibroblastic cells. CONCLUSION: Passage and culture can affect the phenotypic property of MCC cells.

[Determination of dyes in cosmetic by high performance liquid chromatography].

A reversed-phase high performance liquid chromatographic method was used to detect 5 dyes including p-phenylenediamine in cosmetrics. A Zorbax C8 column was used and mobile phase was V (triethanolamine):V(water):V(acetonitrile) = 0.95:94.05:5 adjusted to pH 7.7 by phosphoric acid. The wavelength 280 nm was selected. Samples were extracted with 95% ethanol by ultrasonic method. The recoveries were 87%-107% and the relative standard deviations were 2.3%-6.4%.

[Determination of eleven preservatives including bronopol in cosmetic by reversed-phase ion-pair chromatography].

Reversed-phase ion-pair chromatography was used to detect 11 preservatives including bronopol, esters of p-hydroxybenzoic acid (PHBA) in cosmetics. Zorbax C8 column was used and mobile phase was V (0.05 mol/L NaH2PO4):V(methyl alcohol):V(acetonitrile) = 50:35:15 with addition of hexadecyltrimethylammonium chloride (2 mmol/L) and the pH adjusted to 3.5 by using phosphoric acid. The wavelength 254 nm was selected for detection. Samples were extracted with methanol by ultrasonic method. The recoveries were 85%-107% and the relative standard deviations were 2.2%-7.5%.

[Activated T lymphocytes in epiretinal membranes from eyes of patients with proliferative diabetic retinopathy].

PURPOSE: To investigate the potential contribution of immune-mediated processes to the development of proliferative diabetic retinopathy(PDR), an immunohistochemical study was undertaken to characterize the infiltrating immune cells in epiretinal membrances from the eyes of patients with PDR. METHODS: A total of 15 PDR epiretinal membrane specimens obtained surgically from pars plans vitrectomy were studied by using a panel of monoclonal antibodies against T lymphocytes (CD4), interleukin-2(IL-2) and interleukin-2 recpetors(IL-2R). RESULTS: Twelve of 15 specimens (80%) contained CD4-positive cells. IL-2 was found in 12 of 15 samples (80%), of which 11 also contained CD4-positive cells, and IL-2R was detected in 10 of 15 membranes(67%), of which 9 contained CD4-positive cells and released IL-2. Most of the IL-2R-positive membranes were from type I diabetic patients, 40% of them are younger than 40 years. CONCLUSION: Our study demonstrated the involvement of activated immune cells and release of lymphokine(s) in more than half of the diabetic epiretinal membranes tested and revealed that the processes of immune responses and the biological effects of lymphokines(s) may play an important part in the development of epiretinal membranes of PDR, especially in young-onset and type I diabetes.

[Expression of CD44v and nm23-H1 proteins in laryngeal carcinoma and its correlation with clinical significance].

OBJECTIVE: To investigate the correlation between the expression of two gene proteins and their clinical significance in laryngeal carcinoma. METHODS: Sixty fresh laryngeal carcinoma samples were analyzed and 20 para-carcinoma tissue and 12 normal laryngeal mucosa samples were studied as controls. All specimens were collected from patients operated on from 1996 to 1998 in the same department (including 52 males and 8 females, age ranged from 41 to 77). The quantitative expression of CD44v and nm23-H1 proteins was analyzed by flow cytometry (FCM) and immunofluorescence technique. RESULTS: 1. Higher level expression of CD44v and nm23-H1 proteins was detected in the laryngeal carcinoma specimen than in normal laryngeal mucosa. 2. The expression of CD44v protein was positively correlated with the metastasis, clinical staging and pathological classification; but not correlated with sex, age, tumor size, course of the disease, T classification and category of laryngeal cancer. 3. The expression of nm23-H1 protein was inversely correlated with metastasis and clinical staging while positively correlated with age and tumor size; no significant relations were found between the expression and the sex and course of the disease, T classification and category of laryngeal cancer. 4. There was an inverse correlation between the protein expression of CD44v and nm23-H1. CONCLUSION: CD44v and nm23-H1 probably play an important regulating role in the metastasis of laryngeal carcinoma.

[Study of a new kind of spectrometer coupled with flow injection and CCD diode array detection for multi-components simultaneous determination].

A new kind of spectrophotometric instrument combined with a flow injection device, a prism spectrography, a CCD diode array detection system and a popular microcomputer was established for multi-components simultaneous determination. The measurements of lead and cadmuium with meso-tetra (4-trimethylammoniumphenyl) porphine were completed with the instrument. The detection limit was 0.014 microg/mL for Pb(II) and 0.015 microg/mL for Cd(II). The sampling rate was 60 samples/h.

[Study on the interaction of Cu2Zn2SOD with CoCl2, Co(His)n by spectral and magnetic resonance analysis].

Direct interaction of Cu2Zn2SOD with inorganic metal compound (CoCl2) and organic metal compound [Co(II)(His)n] was studied by ICP, VIS, NMR and measurement of enzyme activity. It has been found that in aqueous solution, there exists a direct interaction of the metal ions of the active center in the metalloenzyme (Cu2Zn2SOD) with external added CoCl2, Co(His)n. As a result, part of the metal ions in metalloenzyme were replaced and the corresponding metalloenzyme derivatives were produced and the catalytic activity of enzyme were affected. Moreover, the interaction of Co(II)(His)n with Cu2Zn2SOD is stronger and quicker, than that of CoCl2 with this enzyme, as well as the Co(II) from Co(II)(His)n is easier to enter the active center of enzyme.

[Surface-enhanced Raman spectra studies on a novel dipyridophenazine complexes of ruthenium(II) and it's effect on DNA].

SERS is better method at analysing the samples which are prone to emit fluorescence or photodecomposition than other methods. Raman scattering of a novel complexes of Ruthenium (II): Ru(bpy)2dppz and it's effects on DNA were studied with surface-enhanced Raman spectra(SERS). We find that when this complex binds to DNA, only the ligand dppz intercalate into the double helix of DNA, and form a larger conjugate system, affect the metal-to-ligand charge transfer(MLCT), produce strong photoluminescence. All these cause shifting of some Raman peaks of the complex and changex of the relative intensity of some other peaks. And on the charge absorbed spectra, after the addition of DNA, the compex' absorbed peaks of Ru(bpy)2dppz solvent shift to long wavelength, this is also one of the character of the intercalation.

[Study on the interaction of copper-zinc superoxide dismutase with cobalt (II)-histidine by spectral analysis--II. Effect of amount of external added Co(His)n, phosphate].

The effect of changing the amount of external added Co(His)n and phosphate on the interaction of Cu2Zn2SOD with organic metal compound (Cobalt (II)-Histidine) have been studied by means of ICP, VIS and measurement of enzyme activity. It has found that in aqueous solution, there exists a direct interaction of the metal ions of the active center in the metalloenzyme (Cu2Zn2SOD) with external added Co(His)n. As a result, part of the metal ions in metalloenzyme were replaced and the corresponding metalloenzyme derivatives (Co (II)-substituted derivatives of SOD) were produced and the catalytic activity of enzyme were affected. It has also studied the intensity of interactions in different molar ratios (Cobalt (II)-Histidine) and of the presence of phosphate and got some results under the effect of corresponding factors.

[Study on the interaction of copper-zinc superoxide dismutase with cobalt (II)-histidine by spectral analysis. II. Effects of pH, interaction time].

The direct interaction of Cu2Zn2SOD with organic metal compound (Cobalt (II)-Histidine) was studied by ICP, VIS and the measurement of enzyme activity, and also investigated the effect of pH values and different interaction times for this interaction. The results showed that with the increased in pH values and the more longer interaction times, the intensity of interaction were increased and the corresponding catalytic activity of enzyme were affected.

[Determination of methanol in hair spray by GC method].

This report improved the chromatographic conditions of present standard method for testing methanol in hair spray GB7917.4-87. To avoid error detection on peak overlapping between methanol and dimethyl ether of original standard method, this method used column GDX-102 packed with 25% polyglycol 1540. The recoveries were in the range of 90.4%-107.0%, and the relative standard deviation was between 1.8% and 5.2%.

Inhibition of beta-amyloid cytotoxicity by midkine.

Midkine (MK) is a neurotrophic and angiogenic growth factor whose expression occurs mainly in fetus. It was reported that MK was present in senile plaques of Alzheimer's disease (AD). To investigate the role of MK during amyloid plaques formation in AD, we examined the in vitro effect of MK on Abeta aggregation and Abeta-induced cytotoxicity. We found that incubation of MK with Abeta resulted in the formation of MK/Abeta complexes. The C-terminus of MK (60-121) played a similar role as the full length MK in complex formation. This interaction of MK and Abeta demonstrated significant inhibition on Abeta self-aggregation. MK also inhibited the cytotoxicity of Abeta on PC12h cells. These findings suggest that MK protects the cells from Abeta-induced cytotoxicity through its complex formation with Abeta. MK is probably expressed to prevent cell death in AD.
