Granulocyte-mediated airway edema in guinea pigs.

To determine the role of polymorphonuclear leukocytes (PMNs) in the airway edema that accompanies airway inflammation, we studied the effects of a 1-h exposure to 2 ppm toluene diisocyanate (TDI) on tracheal extravasation of Evans blue dye and on the concentration of PMNs in the tracheal wall. Tracheal Evans blue content was significantly increased by TDI exposure (53.6 +/- 8.0 micrograms/g tracheal tissue (mean +/- SE) for animals exposed to TDI and 16.3 +/- 2.0 for animals exposed to air, P less than 0.0025) as were both the intravascular and extravascular concentration of PMNs in tracheal sections (intravascular PMNs were 28.0 +/- 8.4 X 10(3) cells/mm3 for TDI and 1.5 +/- 1.5 X 10(3) for air, P less than 0.025, extravascular PMNs were 10.9 +/- 4.5 X 10(3) for TDI and 0 for air, P less than 0.05). PMN depletion with vinblastine or with hydroxyurea abolished both the increase in tracheal Evans blue extravasation and the increase in the concentration of intravascular and extravascular PMNs in animals exposed to TDI. PMN depletion with hydroxyurea did not significantly inhibit the increase in tracheal Evans blue extravasation caused by intravenous histamine. Administration of donor PMNs to animals depleted of PMNs with hydroxyurea reconstituted the TDI-induced increase in tracheal Evans blue extravasation (80.4 +/- 17.3 micrograms/g tissue (mean +/- SE) in animals exposed to TDI vs. 21.3 +/- 2.9 in animals exposed to air, P less than 0.025) and in the intravascular concentration of PMNs in tracheal sections [18.5 +/- 3.4 X 10(3) cells/mm3 (mean +/- SE) in animals exposed to TDI vs. 1.3 +/- 1.3 X 10(3) in animals exposed to air, P less than 0.0025].(ABSTRACT TRUNCATED AT 250 WORDS)

[Correlation of clinical and computed tomographic findings for therapy and prognosis of acute pancreatitis].

A prospective study was performed in 156 patients with acute pancreatitis to find out the value of computed tomography for the diagnosis and early prognosis. The CT finding leads to a diagnosis and differentiation in 3 risk groups similar to the clinical staging. The prognostic value of CT depends on the reproducible grading of pancreatic swelling and the grade of extrapancreatic fluid collections. CT finding correlates with early prognostic factors and the clinical staging, leading to different risk groups which allows an adequate stage-depending therapy of patients with acute pancreatitis.

Pathological changes in the heterologous phase of antibasement membrane antibody mediated disease in the rat.

The immunological and structural changes during the heterologous phase of experimental antibasement membrane antibody mediated disease was sequentially studied in the rat following single i.v. injections of rabbit antibodies to basement membrane antigens prepared from kidney, lung and salivary gland tissues. Although each of the anti-bodies bound strongly to GBM, structural changes were initially subtle accompanied by proteinuria and hematuria. More severe structural changes related to dose and duration of the disease did not appear for several weeks.

Multiple-dose nonlinear regression analysis program for the microcomputer.

An adaptation of a previously published program for nonlinear regression analysis of serum drug concentrations which can utilize data obtained during multiple-dose administration is described. Specific programs have been developed for aminoglycosides, digoxin, and theophylline. The programs provide initial parameter estimates (for a one-compartment linear model) for each drug based on mean population parameters and refined estimates based on observed concentration data. Clinical examples which demonstrate the flexibility of these programs are provided. The programs may also be employed for fitting data for any drug that can be adequately described by a one-compartment linear model.

Transluminal angioplasty of the abdominal aorta.

Transluminal angioplasty of the abdominal aorta was performed in 6 women between 49 and 69 years of age with histories of cigarette smoking. Five had a relatively small aorta. This procedure is effective in treating focal aortic stenoses.

Influence of insulin and substrate concentration on protein synthetic rate in fetal tissues.

The influence of fetal plasma substrate and insulin concentrations on the fractional synthesis rate or turnover of the mixed proteins in the organs of fetal lambs was calculated from measurements of the rate of uptake of L-14C lysine by the protein in the steady state in utero. The control values for protein synthesis rate in the fetal organs of Welsh mountain ewes, 130 to 140 days' pregnant, were similar to those previously observed in the fetuses of Border Leicester ewes: the mean turnover rates were 38, 72, 40 and 63 per cent per day for the brain, liver, cardiac muscle and the placenta respectively; that for skeletal muscles was 6.8 per cent per day; the corresponding half lives were 2.15, 1.27, 1.8 and 1.42 days respectively and 11.3 days for skeletal muscle. A fivefold increase in the plasma glucose levels had no influence on protein turnover rate over a six hour period. A fourfold increase in the concentration of some amino acids, following a mixed infusion, decreased turnover rate in brain, heart and liver. The half life was increased by 30 per cent, but a small reduction in half life was observed in skeletal muscle. Insulin caused a fall in intracellular lysine concentration in skeletal muscle and in plasma, which suggests that the hormone can reduce muscle protein catabolism in the fetal lamb in the last trimester of gestation. Enhancement of synthesis rate occurred in the presence of the amino acid infusions only; the half life of the proteins was reduced by a factor of three. Insulin also increased placental protein synthesis rate by 50 per cent but had no influence on the other tissues studied.

[Ileostoma as part of a surgical therapy concept in acute pancreatitis].

14 patients with severe acute hemorrhagic pancreatitis underwent surgical procedure. In all patients besides intensive care and operative necrosectomy, lavage and drainage a loop ileostoma was installed. Pathophysiological considerations concerning the value of the loop ileostoma in the treatment of acute pancreatitis are discussed.

Biochemical markers in a species endangered by introgression: the red wolf.

The red wolf (Canis rufus), native to much of the southeastern United States, is endangered by man's activities and by hybridization with other species of the genus Canis. The absence of diagnostic morphological markers to distinguish the red wolf from its hybrids has led to the application of the methods of biochemical genetics to this problem. The finding of a unique electrophoretically determined allele with a distribution congruent with the geographical distribution of the remaining red wolf population is reported.
