Further data on the development of red blood cell antigens Lua, Lub, and Cob.

One hundred and fifty-five cord cells were tested for the red blood cell antigens Lua, Lub and Cob in order to collect data on the early postnatal expression of these markers. Additionally, dosage studies were carried out in 8 - 10-month-old heterozygous children. Antigens Lua and Lub revealed to be significantly less expressed in children of both ages compared with those of their mothers, whereas no such differences could be demonstrated in the expression of the antigen Cob.

Polymorphism of the human phosphoglycolate phosphatase in Northrhine-Westphalia (F.R.G.) and its application to paternity testing.

Blood samples from 563 unrelated German and from 110 Turkish individuals living in the Düsseldorf area were studied for phosphoglycolate phosphatase polymorphism. The distribution of the observed phenotypes in the population genetic study did not diverge from the expected values according to Hardy-Weinberg law. In our series, the gene frequencies were calculated as follows: (a) Germans: PGP1 = 0.851, PGP2 = 0.118, PGP3 = 0.031, (b) Turks: PGP1 = 0.973, PGP2 = 0.018, PGP3 = 0.009. The assumed autosomal codominant mode of inheritance was confirmed by the examination of 109 mother-child pairs and by analysis of 70 cases of disputed paternity. The plausibility to exclude German non-fathers from paternity is 12.78%.

Size calculation of restriction enzyme HaeIII-generated fragments detected by probe YNH24 by comparison of data from two laboratories: the generation of fragment-size frequencies.

Restriction fragment-length polymorphism of locus D2S44 detected by the highly polymorphic probe YNH24 and restriction endonuclease HaeIII can be used to improve parentage testing when representative fragment-size frequencies can be obtained. By joining the results of different laboratories, it is possible to set up a meaningful databank. Therefore, the same randomly chosen samples were tested for the HaeIII RFLP detected by probe YNH24 in Düsseldorf (DUS) and Amsterdam (AMS). The results of the different fragment-size calculations obtained by using internal markers and a computerized system (DUS-cad and AMS-cad), and by using external markers and manual calculations (DUS-man), were analyzed. Comparing these results, no statistically significant differences were seen. The results obtained with probe YNH24 and enzyme HaeIII in Düsseldorf and Amsterdam can be used to attain a sufficient number of samples to generate relevant fragment-size frequencies.

Doppler energy: a new acquisition technique for the transthoracic detection of myocardial perfusion defects with the use of a venous contrast agent.

AIMS: This animal experiment was designed to study whether the new technique of Doppler energy imaging could display myocardial perfusion abnormalities with the use of a combination of transthoracic imaging and right atrial injection of a myocardial contrast agent. METHODS AND RESULTS: A series of 11 pigs were studied during (1) normal perfusion, (2) dipyridamole-induced coronary dilatation, and (3) during and after temporary occlusion of the left anterior descending or circumflex artery after a right atrial injection of 8 ml Levovist, 400 mg/ml. Short-axis views were obtained with the four following imaging modes: gray scale imaging (two-dimensional and M-mode), Doppler energy imaging (two-dimensional and M-mode). Visual inspection and off-line video densitometry (results expressed in arbitrary videointensity units 0 to 255) with digital background subtraction were performed. Doppler energy was significantly more sensitive in detecting the presence of contrast than gray scale imaging (background subtracted peak videointensity 32 +/- 17 versus 17 +/- 12, p < 0.001). Mean background-subtracted videointensity increased during dipyridamole-induced coronary hyperemia (40 +/- 14 versus 31 +/- 9, p < 0.003) using Doppler energy technique. Doppler energy imaging consistently detected absent perfusion (background subtracted videointensity -6 +/- 6) and immediate reperfusion (background subtracted peak videointensity 29 +/- 15, p < 0.001). CONCLUSIONS: With the use of a galactose-based contrast agent, Doppler energy data acquisition was superior to standard gray scale imaging in transthoracic evaluation of regional myocardial perfusion, absence of perfusion, and reperfusion.

Immunoglobulin allotypes in a Chinese population: comparison of haplotype frequencies with other Asian groups.

Two groups of Chinese individuals of the city of Wuhan in the People's Republic of China, consisting of 98 blood bank donors and 278 workers in a steel factory, respectively, have been typed for G1m, G2m, G3m, A2m and Km allotypes. The Gm-A2m haplotype frequencies as well as the Km gene frequencies appeared to be quite similar in these two groups of people, the main haplotypes being Gmaf;n;b A2m2 and Gmza;..;g A2m1. After joining the data of the two groups, the frequencies of the main Gm haplotypes and the A2m and Km gene frequencies have been compared with those of six other Asian populations. In Asia, going from the south to the north, an increasing frequency of Gmza;..;g and a decreasing frequency of Gmaf;n;b were observed. The frequencies found in the investigated Chinese population lie between the frequencies found in the Chinese from Taiwan and the Japanese.

Changes of intracellular calcium, fatty acids and phospholipids during miltefosine-induced apoptosis monitored by fluorescence- and 13C NMR-spectroscopy.

The alkylphosphocholine Miltefosine (hexadecylphosphocholine, HePC) induces apoptosis in human epithelial KB cells, whereas no such effect can be observed in a resistant clone (KBres). Its mode of action is mediated via the cell membrane, whereas the mechanism is still widely unknown. The use of various spectroscopic methods (fluorescence spectroscopy with Fura-2/AM on viable cells, 13C NMR spectroscopy on lipid extracts) reveals osmotic and metabolic changes in HePC treated sensitive cells. Intracellular free Ca(2+)-concentration increased over 300% of control in apoptotic cells, whereas KBres cells showed only a minor increase and no morphological response typical for apoptosis. The Ca(2+)-influx was mediated via calcium channels in the cell membrane. The HePC-induced influx is prevented by Gd3+, which blocks those calcium channels. Cells, grown in Ca(2+)-free medium, showed no apoptotic behaviour after treatment with HePC. If apoptosis was induced, an increased fatty acid and subsequent phospholipid biosynthesis was observed. This effect seems to be a specific marker of apoptosis in KB cells.

Peri-implant care of ailing implants with the carbon dioxide laser.

One of the many applications for which lasers have been proposed in implant dentistry is for the decontamination process. The purposes of this study were to assess possible alterations in titanium implants in vitro and in vivo by use of the carbon dioxide (CO2) laser and to determine whether new bone formation can occur on previously contaminated implants. In vitro, temperature changes at the bone-titanium implant interface were recorded during use of a CO2 laser-scanning system (Swiftlase). Additionally, the effects of laser irradiation on titanium implants at various power settings were examined. In 6 beagle dogs, a total of 60 implants and bony defects resulting from plaque accumulation were treated by air-powder abrasive (the conventional treatment), laser irradiation, or both. Depending on the parameters chosen, melting and other surface alterations were seen in vitro, especially in the superpulse mode. Otherwise, no alterations were found, even at high power settings in the continuous mode. In vivo, corresponding histologic examination of 4-month sections showed evidence of new direct bone-to-implant contact after laser-assisted therapy, especially when the implants had been treated concomitantly with submerged membranes. These results support the hypothesis that peri-implant defects can be treated successfully by laser decontamination without damaging the surrounding tissues in the dog model. Nevertheless, further investigations will be required to determine the clinical efficacy of the treatment.

[Significance and testimonial value of hemo-genetic legal identification with special reference to DNA analysis].

175 hemogenetic expertises on the identity of ostensibly jumbled blood samples were analysed. In 4.5% of the cases a genetically qualified non-identity was found, although an erronous mixing-up of samples could be excluded. A major problem may arise when the expert witness has to find out whether a non-identity is due to either genetic or to artificial reasons. A battery of conventional blood group systems as well as highly informative DNA polymorphisms is a powerful tool in discriminating between both reasons and enables the expert to reach a decision.

Combined extraction techniques of tumour cells and lipid/phospholipid assignment by two dimensional NMR spectroscopy.

As the cell membranes are one of the targets of drug treatment in cancer cells, their membrane composition and variations in this composition need to be analyzed. Gradient selected 2D-NMR inverse heteronuclear chemical shift correlations are have described, which offer optimum sensitivity combined with a high reliability for unequivocal signal assignment in proton, carbon and phosphorous spectra, concerning neutral lipids and in particular phospholipids. Additionally, an extraction procedure is presented to extract subsequently the water soluble (PCA extract) and lipophilic metabolites (chloroform/methanol extract) from the same cell batch.

Early stage monitoring of miltefosine induced apoptosis in KB cells by multinuclear NMR spectroscopy.

Synthetic ether lipids, like miltefosine (hexadecylphosphocholine), an alkylphosphocholine, are antineoplastic agents in vitro and in vivo. Their mode of action is mediated via the cell membrane, but the mechanism is still unclear. Miltefosine induces apoptosis in human epithelial KB cells, but slows down only proliferation in rat C6 glioma cells. NMR spectroscopy on lipid extracts reveals increased diacylglycerol and triacyglycerol biosynthesis in KB cells prior to DNA fragmentation indicating a CTP:phosphocholine-cytidylyl-transferase (CT) inhibition by the drug. Although C6 cells were morphologically affected by alterations in phospholipid composition and metabolism by a long term treatment (23 days) with the drug, no persistent diacylglycerol increase is observed.
