Urinary excretion of alpha-fetoprotein in rats on a cirrhogenic carbon tetrachloride regimen.

Alpha-fetoprotein was detected in the serum and urine of 2- to 9-mth-old rats subjected to protracted CCl4 poisoning. During the first 3 mth of the experiment, urinary excretion of AFP was found in 30-40 per cent. of the animals, increasing to 70 per cent. thereafter. The liver lesions progressed from acute parenchymal necrosis to cirrhosis, but hepatocellular carcinomas did not develop. Uptake of tritiated thymidine by the hepatocytes increased significantly but was constant throughout the experimental period. The findings are compared to the observations made during 3mDAB-induced hepatocarcinogenesis in the rat.

Excretion of alpha-foetoprotein in the urine of rats during exposure to 3'-methyl-4-dimethylaminoazobenzene.

Female rats were maintained on standard laboratory diet, Miller's diet or Miller's diet containing 3'MeDAB. Animals fed standard or Miller's diet did not excrete alpha-foetoprotein (AFP) in their urine. Early appearance of AFP was demonstrated by examining the urine of rats on the 3'MeDAB regimen. The incidence of positive urine samples was high between the 5th and 7th week of the experiment. It thereafter declined, but from the 3rd month it steadily rose and reached a maximum of 80% at about 10 months. Though urinary excretion of AFP was irregular in individual animals, several positive urine samples were obtained from all rats followed for more than a few months. The urine of 90% of hepatoma-bearing rats contained AFP at the time of killing. The incidence of elevated serum AFP levels as determined by immunodiffusion, increased with the duration of the experiment, but was still only 70 percent in rats fed 3'MeDAB for over 34 weeks. The severity of the hepatic alterations, as well as hepatocytic uptake of [3H]thymidine, increased with time. The serum of animals fed the standard diet was negative, whereas AFP was very infrequently detected in the serum of rats given Miller's hypoprotein diet. The results demonstrate that, in a population exposed to hepatocarcinogenic agent, the recurring detection of urinary excretion of AFP is a useful indicator of the high risk of developing hepatomas.

Experimental allergic sialoadenitis. X. Chronic destructive parotitis induced in immunized rats by daily intraductal challenges with antigen.

The right parotid duct of rats was instilled five to eight times with bovine serum albumin at daily intervals. Acute parotitis developed in non-sensitized animals, while chronic destructive sialoadenitis ensued in preimmunized rats. The morphological changes were reminiscent of those found in the salivary glands of patients with Sjögren's syndrome.

Acute Disease↗

Alveolar rhabdomyosarcoma presenting as subacute intravascular coagulation.

A patient with rhabdomyosarcoma is described in whom the presenting clinical and laboratory features were those of disseminated intravascular coagulation. The patient's rapid downhill course was primarily expressed by haemorrhagic tendency. An alveolar rhabdomyosarcoma, affecting many organs, including vascular and cardiac lumina, was found at necropsy and was considered to be the cause of the consumption coagulopathy.

alpha-fetoprotein in rats treated with anti-thymocytic serum.

Serum alpha-fetoprotein was detected by immunodiffusion in rats treated with anti-thymocytic serum. Augmented synthesis of AFP is related to compensatory hyperplasia of liver cells secondary to a state of hypoproteinemia in the proteinuric nephritic animals. The nephritis was produced by the anti-basement membrane antibodies present in the thymocytic antiserum.

Excretion of liver antigens in the urine of patients with hepatic diseases.

Liver antigens were detected in the urine of 4 of 42 patients with various liver diseases. The urine of 25 healthy subjects and patients with diseases not affecting the liver was devoid of antigens in detectable amounts. The presence of hepatic antigens in the urine did not correlate with severity of jaundice and SGOT levels but correlated with parenchymal necrosis and was associtated with a high mortaltiy.

Experimental allergic sialoadenitis. VIII. Acute parotitis following intraductal instillation of antiserum to salivary antigens in the rat.

Antiserum to ammonium sulphate-soluble proteins of rat saliva was raised in rabbits. The introduction of this antiserum, via the excretory duct, into the parotid gland of rats induced severe acute sialoadentis. It is suggested that the inflammatory response developed as a result of formation of immune complexes, composed of acinar epithelial antigens and the antibodies introduced from without.

Alpha-fetoprotein and liver cell proliferation in rats fed choline-deficient diet.

Liver 3H-thymidine labelling index and serum AFP were determined in rats fed a standard laboratory or choline deficient diet. A significant increase in the rate of liver cell proliferation was evidenced in animals on the deficient diet. Furthermore, elevated serum AFP levels were detected in a proportion of these rats. It is suggested that increased synthesis of AFP corresponds to liver cell regeneration secondary to hepatic damage.

Experimental allergic sialoadenitis. IX. The regional lymph nodes after antigenic challenges of the parotid glands in rats.

The regional lymph nodes of the parotid gland of rats were examined histologically. The nodes were removed from untreated animals or rats given instillations of BSA or saline into the parotid duct. Anti-BSA antibodies were assessed in the saliva and serum. Patterns of reactivity of the lymph nodes did not correlate with the presence or absence of salivary and serum antibodies. The occurrence of antibodies in the saliva appears to reflect the activity of immunocytes residing in the parotid gland. The morphological features of the lymph nodes correspond to the overall immunological experience.

Tritiated thymidine uptake by glomerular cells in proliferative glomerulonephritis of the rat.

A small but significant number of tritiated thymidine labelled cells were found, by autoradiography, in the glomeruli of rats with Masugi nephritis or chronic serum sickness nephritis. There were no labelled glomerular cells in sections of untreated animals. The findings favour the contention that in proliferative glomerulonephritis, glomerular hypercellularity is due to infiltration of monocytic cells into the tufts where they divide.

Tissue antigens in the saliva of rats. Demonstration of salivary gland specific and nonspecific antigens.

Antisera were raised in rabbits against the half-saturated ammonium sulfate soluble fraction of rat saliva. With these antisera, at least four antigens were demonstrated by immunodiffusion in the saliva and in a saline extract of the parotid gland; three of these antigens were also found in a saline extract of the submaxillary gland. Two of the four antigens were salivary gland specific, the other two being shared by the liver, pancreas and testis. The immunofluorescent pattern indicates that these antigens originate from the acinar epithelium of the salivary glands and probably constitute products of secretion and/or cellular turnover.

Antibodies in the saliva after antigenic challenge of the parotid gland of systemically sensitized rats.

Anti-BSA antibodies were detected in the parotid saliva of untreated rats. In non-sensitized animals, the parotid duct of which was repeatedly instilled with saline or BSA, the antibody titers rose significantly. Introduction of BSA into the parotid gland of sensitized animals was followed by a significant rise in salivary but not in circulating antibodies. The findings demonstrate the relative independence of the local from the systemic immune system.
