Lens fodrin binds actin and calmodulin.

Fodrin was isolated from the chick lens and shown to bind to actin and calmodulin. Its localization to the cytoplasmic side of lens plasma membranes was demonstrated immunologically.

A clinical study of Laotian refugees infected with Clonorchis sinensis or Opisthorchis viverrini.

Twenty-six Laotian refugees infected with Clonorchis or Opisthorchis liver flukes, and 15 uninfected controls, were assessed clinically by history, physical examination, serum 5'nucleotidase level, and serum immunoglobulin E level. Ultrasound studies of the biliary tract were also done on a subset of six infected subjects. The results showed no differences between subjects and controls with respect to clinical symptoms, physical findings, 5'nucleotidase, or biliary ultrasound. Mean serum IgE levels were four times higher in fluke-infected individuals than uninfected controls (P less than 0.002). Even when those infected with other parasites in addition to Clonorchis were excluded from the analysis, serum IgE levels remained three times higher in fluke infected individuals (P = 0.06). This study provides some evidence that the fluke provokes a mild immune response in the asymptomatic human host but may not require eradication.

The distributions of N and F: measures of HLA haplotype concordance.

Nonrandom inheritance of the two HLA haplotypes of Chromosome 6, available from each parent among siblings affected by certain diseases, has afforded evidence of HLA-linked disease-susceptibility genes. Two algebraically equivalent measures of HLA haplotype concordance (that is, the excessive sharing of certain haplotypes among affected siblings) are used for family studies designed to test whether or not there is significant evidence of the existence of an HLA-linked disease-susceptibility gene or for inferring the mode of inheritance when this is already believed to apply. The distributions of these measures are derived under the null hypothesis of random inheritance of HLA haplotypes, and there is a short discussion of the case in which inheritance of a diseased gene causes a change, from the purely random case, in the distribution of haplotype concordance among affected siblings.

Renal tubular handling of potassium in patients with medullary sponge kidney. A model of renal papillectomy in humans.

Renal tubular function, with special emphasis on potassium excretion, was studied in three patients with medullary sponge kidney (MSK). Urinary acidification and concentration abilities were impaired, while glomerular filtration rates remained normal. After short-term intravenous (IV) potassium chloride loading, maximal excretion of potassium was lower in the patients with MSK than in the normal controls. The kaliuretic response to IV sodium sulfate and acetazolamide infusion was maintained. The capacity of the kidney to sustain potassium balance under conditions of either long-term potassium loading or depletion was preserved. We have concluded that the medullary and papillary segments of the human collecting duct, have an important role in the handling of short-term potassium loading. The integrity of these segments, however, is not essential for adaptation to prolonged potassium surfeit or dietary depletion.

Oxidation of dimethylamine and trimethylamine in methazotrophic yeasts by microsomal mono-oxygenases sensitive to carbon monoxide.

Whole cells of Candida boidinii grown on di- or tri-methylamine as sole nitrogen source readily oxidized both amines. The oxidation was potently inhibited by carbon monoxide. Cell-free extracts required the presence of 20 microM FAD before mono-oxygenase activity with both amines could be demonstrated. NADH was a better electron donor than NADPH. Activity was present in cells grown on secondary and tertiary amines but not on primary amines, and was detected in a number of different yeasts. Enzyme activity could be sedimented at 187 000 x g, and was associated with NADPH-cytochrome c reductase activity. It is thus probably microsomal. Activity was inhibited by cyanide, mercaptoethanol, carbon monoxide and proadifen hydrochloride (SKF 525-A).

Serological differences between the multiple amine oxidases of yeasts and comparison of the specificities of the purified enzymes from Candida utilis and Pichia pastoris.

1. Antiserum to purified methylamine oxidase of Candida boidinii formed precipitin lines (with spurs) in double-diffusion tests with crude extracts of methylamine-grown cells of the following yeast species: Candida nagoyaensis, Candida nemodendra, Hansenula minuta, Hansenula polymorpha and Pichia pinus. No cross-reaction was observed with extracts of Candida lipolytica, Candida steatolytica, Candida tropicalis, Candida utilis, Pichia pastoris, Sporobolomyces albo-rubescens, Sporopachydermia cereana or Trigonopsis variabilis. Quantitative enzyme assays enabled the relative titre of antiserum against the various methylamine oxidases to be determined. 2. The amine oxidases from two non-cross-reacting species, C. utilis and P. pastoris, were purified to near homogeneity. 3. The methylamine oxidases, despite their serological non-similarity, showed very similar catalytic properties to methylamine oxidase from C. boidinii. Their heat-stability, pH optima, molecular weights, substrate specificities and sensitivity to inhibitors are reported. 4. The benzylamine oxidases of C. utilis and P. pastoris both oxidized putrescine, and the latter enzyme failed to show any cross-reaction with antibody to C. boidinii methylamine oxidase. Benzylamine oxidase from C. boidinii itself also did not cross-react with antibody to methylamine oxidase. The heat-stability, molecular weights, substrate specificities and sensitivity to inhibitors of the benzylamine/putrescine oxidases are reported. 5. The benzylamine/putrescine oxidase of C. utilis differed only slightly from that of C. boidinii. 6. Benzylamine/putrescine oxidase from P. pastoris differed from the Candida enzymes in heat-stability, subunit molecular weight and substrate specificity. In particular it catalysed the oxidation of the primary amino groups of spermine, spermidine, lysine, ornithine and 1,2-diaminoethane, which are not substrates for either of the Candida benzylamine oxidases that have been purified. 7. Spermine and spermidine were oxidized at both primary amino groups; in the case of spermidine this is a different specificity from that of plasma amine oxidase. 8. Under appropriate conditions, P. pastoris benzylamine/putrescine oxidase (which is very easy to purify) can be a useful analytical tool in measuring polyamines.

Bacteria-specific IgE in patients with nasal polyposis. A preliminary report.

Sixty-one patients exhibiting chronic nasal polyposis had their conditions evaluated by radioallergosorbent tests to determine the presence of bacteria-specific serum IgE. Fifty-nine patients exhibited positive serum IgE to at least one of 11 bacteria tested. There are several implications of the collected data.

Human botulism (type F)--a rare type.

An unusual presentation of type F botulism (one of the world's three reported outbreaks) is described. The diagnosis was complicated by the presence of pre-existing Adie's myotonic pupil. There was also a history of self-inflicted small-caliber gunshot wound to the right brain 10 years before the onset of the botulism. Post-traumatic seizure disorder, also present in this patient, was under good control. No offending food was ever discerned, despite exhaustive study.

Detecting the hypersusceptible worker: genetics and politics in industrial medicine.

This paper examines the development of the concept of the hypersusceptible worker, as advanced by H. E. Stokinger and his associates. It is argued that this concept enabled industrial toxicologists to organize knowledge about workers' reactions to toxicity so as to sustain a professional and methodological ideology which reflects the relationship between industrial toxicology and capitalist industry. Moreover, it is argued that it is this aspect of hypersusceptibility, rather than its practical utility to specific corporations, that is most important. Constraints on implementation of screening programs based on the ideas of genetic hypersusceptibility are also discussed.
