Menstrual blood loss and hematologic indices in healthy Chinese women.

Menstrual blood loss, cyanmethemoglobin and serum ferritin were determined in 421 healthy, noncontracepting Chinese women. The range of menstrual blood loss (MBL) was 4.1-273.6 mL, the mean value was 54.2 mL, and the median was 42.9 mL. The range of hemoglobin was 8.3-16.7 g/dL, and the mean value was 13.2 g/dL. The range of ferritin was 1.2-180.0 ng/mL, the mean value was 22.8 ng/mL, and the geometric mean was 17.1 ng/mL. The upper normal limit of MBL in Chinese women was set at 80 mL.

[Establishment of a mouse forestomach carcinoma cell line (MFC) with spontaneous hematogenous metastasis and preliminary study of its biological characteristics].

A mouse forestomach carcinoma cell line (MFC) prone to blood-born metastasis to lung was established in vitro. This cell line has been subcultured for more than 132 passages. MFC cells lacked in contact inhibition and, morphologically, appeared as round, polygonal and spindle cells. Ultrastructurally, there were abundant microvilli on the cellular surface and extensive filopodia in the cytoplasm; nuclei showed irregular shape with an increased nucleus-cytoplasm ratio; desmosomes, hemidesmosomes as well as a small number of tonofibrils were observed. The doubling time of MFC cell population was 24.7 hours. The average mitotic index was 32.9% and the maximum was 62% with 70-76 mode. The homotransplant efficiency was 100% (11/11). In morphology, the solid tumors induced were similar to FC tumor, from which the MFC cell line was derived and 81.8% (9/11) developed spontaneous lung metastasis. It is indicated that this cell line still retains the primary tumor's feature of spontaneous hematogenous metastasis. MFC cell line might be very useful in experimental treatment, study of tumor metastatic mechanism and isolating subpopulation.

A randomized comparative clinical evaluation of the Steel Ring, the VCu200 and the TCu220c IUDs.

A randomized comparative clinical study of the Steel Ring, VCu200 and TCu220c IUDs was carried out in Beijing. Six-hundred subjects were recruited and each group of 200 was inserted with one of the three devices. The TCu220c and the VCu200 had statistically significantly lower pregnancy and expulsion rates at one and two years of use than the Steel Ring. However, the Steel Ring consistently had the lowest rate of removal for pain and/or bleeding. At two years, the early differences of use-related discontinuation and continuation between the higher rates of the Steel Ring and the lower rates of the VCu200 and TCu220c were significantly reduced. There were no statistically significant differences of net event rates between VCu200 and TCu220c.

Menstrual blood loss, haemoglobin and ferritin concentration of Beijing women wearing steel ring, VCu 200, and TCu 220c IUDs.

Menstrual blood loss (MBL), serum ferritin and whole blood cyanmethemoglobin were measured at pre- and 1, 3, 6, 12, 18 and 24th postinsertion cycles in 60 women wearing the Steel Ring, the Copper V (VCu 200) or the Copper T (TCu 220c). The MBL, incidence of menorrhagia and iron deficiency anemia were lower among users of the Steel Ring than among users of the Copper V and T. Anemia occurred later and the duration of menstruation was shorter with the Steel Ring than with the Copper T. There were no statistically significant differences between the Copper V and T.

Invasiveness and metastatic capability of rat fibroblast-like cells before and after transfection with immortalizing and transforming genes.

Invasion in vitro and in vivo and spontaneous metastasis was investigated in cell lines before and after introduction of immortalizing (polyoma large-T and activated myc) genes and of transforming (polyoma middle-T and activated ras) genes in Fischer rat cells. Invasion in vitro was tested by confrontation of rat cells with embryonic chick heart fragments in organ culture. Invasion in vivo and metastasis was evaluated in nude mice and in syngeneic rats after injection of cells i.p. or s.c. in the flank and after implantation of cell aggregates s.c. in the tail. Rat cells were also analyzed for the presence of myc oncogenes, and for the expression of ras oncogenes. Cells from primary or low passage rat embryo (REF) cells were not invasive in vitro and did not produce tumors in vivo. Cell lines (LTRAT1, LTaRAT1) derived from REF cultures after transfection with plasmids encoding polyoma large-T antigens, behaved like REF cells. Cell lines (REFpEJgpt4, REFpEJmycN7) established from REF cultures after transfection with either a plasmid encoding an activated human ras protein or with the latter plasmid plus one containing an activated myc gene, were invasive in vitro and in vivo and produced invasive and metastatic tumors in syngeneic rats. Cell lines (FR3T3) established in an apparently spontaneous way were invasive in vitro and produced invasive tumors in vivo without metastasis. Derivatives of FR3T3 (FRLT1, MTT4, MMC1, and PyT21) transfected with plasmids encoding one or more of the polyoma antigens, differed from FR3T3 cells by a shorter latency period of tumor formation (less than 1 versus 1 to 3 weeks). Like FR3T3 tumors, FRLT1, MTT4, MMC1, and PyT21 tumors were invasive but not metastatic. Other spontaneously established lines (Rat1) were invasive and metastatic. Cells (Rat1pEJ6.6) derived from Rat1 cultures after transfection with a plasmid encoding an activated ras protein, showed shorter tumor latency periods (less than 1 versus 7 weeks). A thymidine kinase deficient Rat1 derivative (Rat2) was not invasive in vitro but produced invasive and metastatic tumors in vivo with long (9 to 21 weeks) latency periods. Rat2pT24B4 cells derived by us from Rat2 cells after transfection with a plasmid containing a mutated human ras gene (pT24), were invasive in vitro and in vivo as were cells derived from Rat2 tumors. We conclude from our experiments that invasiveness and metastatic capability are often acquired by established REF-derived cell lines in an apparently spontaneous way.(ABSTRACT TRUNCATED AT 400 WORDS)

Clinical investigation of a low-dose levonorgestrel-releasing vaginal ring.

Contraceptive effectiveness and adverse effects were observed in 108 women who used the levonorgestrel-releasing vaginal ring. One-year cumulative net rates per 100 women were pregnancy, 3.7; expulsion, 4.6; use-related discontinuation, 26.9; and continuation, 71.2. The main adverse effects were menstrual disturbances, which improved after longer usage. The menstrual blood loss (MBL), hemoglobin (Hb), and ferritin concentrations were determined in 20 women. The MBL gradually declined after insertion of the vaginal ring and the Hb concentration was significantly increased in the 6th and 12th periods after insertion (P less than 0.05).

[Establishment and biologic properties of a mouse uterine cervix cancer cell line (MUCC) with lymphatic and hematogenous metastatic capacities].

For study of tumor metastatic mechanism, a mouse uterine cervix cancer cell line (MUCC), as derived from a transplantable mouse uterine cervix cancer No. 27 (U 27) with metastatic capacities through lymphatics and blood, was established in vitro. It has been successfully maintained for more than 24 months through more than 100 passages. MUCC cells are epithelioid with definite malignant morphological features. The doubling time of the cell population is 27.8 hours. The average mitotic index is 30.6%, with a maximum of 58.9%. Chromosomes show aberration in number and structure. The homotransplant rate is 75% (9/12). The solid tumor, produced by homotransplant, is similar to the tumor U 27. Metastases are present in the lymph nodes and lungs in these 9 animal models. It is evident that this cell line retains the capacity of inducing daughter tumor lines with spontaneous lymphatic and blood metastases. MUCC cell line might be very useful in isolating subpopulations which possess the different metastatic abilities and the study of metastatic mechanism.

[Experimental study on tumor metastasis after treatment by hematoporphyrin derivative and laser].

The purpose of the present experiment is to observe whether metastasis will develop after the tumor is treated by hematoporphyrin derivative (HPD) and pulse laser. Mouse transplantable forestomach carcinoma (FC) was used of which the spontaneous metastasis rate was 33% to 83%. 179 inbred "615" mice were divided into six groups for repeated experiments. The first repeated experiment included three groups: group 1, pulse laser with HPD; group 2, continuous laser with HPD; group 3, control. The second repeated experiment also included three groups: group 4, pulse laser without HPD; group 5, pulse laser with HPD and group 6, control. On day 5 after FC transplantation, the animals of group 1, 2 and 5 were injected intraperitoneally with HPD (50mg/kg) and then group 1, 2, 4 and 5 were treated with laser on day 7. From group 1, 2 and 3, 10 animals were sacrificed on day 12 and the rest animals on day 16. The lymph nodes and lungs of all animals were taken out for gross and histological examination. It was found that the tumor metastasis rates of the six groups were: group 1, 32.3% (10/31); group 2, 21.8% (7/32); group 3, 38.1% (8/22); group 4, 64.0% (16/25); group 5, 74.1% (20/27) and group 6, 83.3% (35/42). It shows that laser does not increase the incidence of tumor metastasis.
