[Hemorheological study on the process of tumor invasion and lymphatic metastasis].

Mouse transplantable hepatoma (H22) was used as an experimental model for lymphatic metastasis. The tumor cells were transplanted subcutaneously into right footpads of inbred 615-strain mice. The hemorheological changes during invasion and lymphatic metastasis in mice were observed. The results of histological examination showed that tumor invasion was divided into early, middle and late invasive stages, and the metastasis was divided into early, middle, late and final metastasis periods. Tumor invasion began 3 days after tumor transplantation, and the late stage of invasion was found at 9 days. Metastasis began 6 days after transplantation; at 19 days metastasis to lymph nodes in the right side reached grade IV. Metastasis to lymph nodes in the left side appeared 14 days after tumor transplantation. In the early and middle invasive stages only plasma viscosity increased significantly, while other hemorheological values were unchanged. The blood viscosity and aggregation of red blood cells decreased significantly after the metastatic late stage. The hematocrit of red blood cells began to decrease in the final stage of metastasis, and in the meantime, the rigidity of red blood cells began to increase, while plasma viscosity showed no changes after the late metastatic stage. The results and mechanism of hemorheological changes are discussed.

TCM treatment of adolescent functional uterine hemorrhage--a clinical report of 105 cases.

105 cases of adolescent functional uterine hemorrhage were refractory to western medicine, but successfully cured with Chinese herbal drugs during the period from April 1982 to July 1986. At the bleeding stage of the disease, the Triple Action Hemostatic Decoction [( symbol: see text]) was used, and at the re-adjusting stage when bleeding stopped, the Decoction for Strengthening Kidney and Reinforcing the Constitution [( symbol: see text]) was given. The rate of complete cure was 13.3%, of marked improvement 76.2%, of some improvement 8.6%, with a total effective rate of 98.1%. The author also discussed the etiological mechanism of the disease and the theory that guided him in designing the prescription in the light of TCM.

Hidden thermodynamics of mutant proteins: a molecular dynamics analysis.

A molecular dynamics simulation method is used to determine the contributions of individual amino acid residues and solvent molecules to free energy changes in proteins. Its application to the hemoglobin interface mutant Asp G1(99) beta----Ala shows that some of the contributions to the difference in the free energy of cooperativity are as large as 60 kilocalories (kcal) per mole. Since the overall free energy change is only -5.5 kcal/mole (versus the experimental value of -3.4 kcal/mole), essential elements of the thermodynamics are hidden in the measured results. By exposing the individual contributions, the free energy simulation provides new insights into the origin of thermodynamic changes in mutant proteins and demonstrates the role of effects beyond those usually considered in structural analyses.

A comparative study of Tam3 and Ac transposition in transgenic tobacco and petunia plants.

Transposition of the Anthirrinum majus Tam3 element and the Zea mays Ac element has been monitored in petunia and tobacco plants. Plant vectors were constructed with the transposable elements cloned into the leader sequence of a marker gene. Agrobacterium tumefaciens-mediated leaf disc transformation was used to introduce the transposable element constructs into plant cells. In transgenic plants, excision of the transposable element restores gene expression and results in a clearly distinguishable phenotype. Based on restored expression of the hygromycin phosphotransferase II (HPTII) gene, we established that Tam3 excises in 30% of the transformed petunia plants and in 60% of the transformed tobacco plants. Ac excises from the HPTII gene with comparable frequencies (30%) in both plant species. When the beta-glucuronidase (GUS) gene was used to detect transposition of Tam3, a significantly lower excision frequency (13%) was found in both plant species. It could be shown that deletion of parts of the transposable elements Tam3 and Ac, removing either one of the terminal inverted repeats (TIR) or part of the presumptive transposase coding region, abolished the excision from the marker genes. This demonstrates that excision of the transposable element Tam3 in heterologous plant species, as documented for the autonomous element Ac, also depends on both properties. Southern blot hybridization shows the expected excision pattern and the reintegration of Tam3 and Ac elements into the genome of tobacco plants.

[Electron microscopy of the angioarchitecture of the human iris and ciliary body].

With the corrosion cast technique and electron microscopy, the major arterial circle of the iris, formed by the terminal branches of the medial and lateral long posterior ciliary arteries and of the anterior ciliary arteries, is shown not an anastomosed vascular ring but divided small branches of the arteries to supply the iris and ciliary body. The iris vessels demonstrate a radiate distribution of about 180-200 branches. The calibres of the iris arteries are homogeneous and measure about 50-120 microns. The branching portion of a precapillary arteriole begins with a constriction. The vessels of the ciliary processes account for more than half the volume of the ciliary body and have a rich capillary network of at least two layers. The draining veins are located on the surface and at the base of the ciliary body. There are 7-10 draining veins between two ciliary processes.

Observation of growth and invasiveness of human uterine cervix carcinoma cell line (CC801) in the testes of nude mice.

A human uterine cervix carcinoma cell line (CC801) was implanted subcapsularly in the testes of nude mice and its growth and invasiveness was observed. The results showed that the taking rate of CC801 tumor cells was 70%. Two weeks after implantation, the tumor cells were surrounded by pseudocapsule and showed no invasiveness, but 4 weeks after transplantation, they began to proliferate and to penetrate the pseudocapsule. Thereafter, the tumor cells continued to invade the neighboring tissues, causing degeneration and atrophy of seminiferous tubules at later stages. After 12 weeks, the tumor tissues occupied the whole testis. The process of invasion could be divided into 5 stages: latency, proliferation, early invasion, intermediate invasion and late invasion. The rate of lymphatic metastasis was 6.6%, but no lung metastasis was found.

[Prostaglandin E2, F2 alpha,6-keto-prostaglandin F1 alpha and thromboxane B2 in the placenta of normotensive women and women with pregnancy induced hypertension].

Prostaglandin E2 (PGE2), Prostaglandin F2 alpha (PGF2 alpha),6-keto-prostaglandin F1 alpha (6-keto-PGF1 alpha) and thromboxane B2 (TXB2) were measured with radioimmunoassay in placentae of 18 normotensive and 22 moderate or severe PIH gravidae. The content of PGE2 and PGF2 alpha did not change significantly in placentae of PIH patients suggesting that PGE2 and PGF2 alpha in placenta were not important in causing PIH. The content of placental 6-keto-PGF1 alpha also showed no significant difference, while TXB2 levels were significantly higher in PIH patients. The 6-keto-PGF1 alpha/TXB2 in PIH patients was remarkably lower than in normotensive women. These results indicate that an increase in TXB2 level and thus a decrease in the ratio of 6-keto-PGF1 alpha to TXB2 of placenta may be a pathogenetic factor in PIH.

[Haemorheologic changes during tumor development after subcutaneous transplantation of mouse forestomach carcinoma (FC)].

After transplantation of FC, there was a latent period of about 3 days until local invasion was first observed. Active local tumor invasion occurred on day 6 (invasion stage). From day 9 when the invasion became extensive lung metastasis developed (early metastatic stage). The lung metastasis was more severe from day 12 (mid metastatic stage) and became widespread from day 15 (late metastatic stage). Lung metastasis was observed in 42.8%, 50% and 73.3% of the mice during early, mid and late metastatic stages, respectively. There was no haemorheologic change observed in the latent period. In the invasion and early metastatic stages, an increase in plasma and whole blood viscosity was evident but the hematocrit and red cell aggregation were normal. As lung metastasis became more and more extensive, all the haemorheologic parameters were markedly decreased.

[Study on the invasiveness of mouse uterine cervix carcinoma cell (U14) implanted in mouse testis].

A good invasion and metastasis pattern was observed after implanting of U14 tumor cells subcapsular in testis, which provided a new model in vivo for studying tumor invasion and metastasis. The transplantability of U14 tumor-inoculation was 90%, retaining high metastasis in lymph-node (88.8%) and lung (77.7%). On the first day after implantation, U14 tumor cells were in a state of relative rest. On the 3rd day, tumor cells began to proliferate and invade the neighboring tissue along the natural space between seminiferous tubules. Degenerative and atrophic changes of seminiferous tubules appeared in the invasion areas. The process of invasion can be divided into 4 stages, namely latency, proliferation, early invasion and late invasion.

Haemorheological study during the process of tumor development after subcutaneous transplantation of mouse forestomach carcinoma (FC) cells.

A metastatic tumor model of mouse forestomach carcinoma (FC) was used in this study. Tumor cells were subcutaneously transplanted into right costal region of inbred 615-strain mice. Animals were killed on the 3rd, 6th, 9th, 12th and 15th day. Histological and haemorheological examinations were performed. Based on the histological findings the development of the tumor was divided into 5 stages; i.e. latent stage, invasive stage, early stage of metastasis, middle stage of metastasis and late stage of metastasis, corresponding with 3rd, 6th, 9th, 12th and 15th day after transplantation of mouse forestomach carcinoma. Results of haemorheological studies indicated that plasma and blood viscosity elevated in the invasion and early metastatic stages and the increased viscosity of blood was able to promote metastasis during the early stage of tumor development, while the tumor developed in the middle and late metastatic stages when the values of plasma viscosity, blood viscosity, aggregation of red blood cells and haematocrit decreased but the rigidity of red blood cells increased in these periods.

Pregnancy interruption with RU 486 in combination with dl-15-methyl-prostaglandin-F2 alpha-methyl ester: the Chinese experience.

In a multicenter study taking place in four centers in Beijing (People's Republic of China), pregnancies up to 49 days of amenorrhea (DA) were interrupted with RU 486 (RU 38486, mifepristone, 600 mg orally once), followed 36-60 hours later by administration of dl-15-methyl-PGF 2 alpha-methyl ester (PG05, 1 mg vaginal suppository). One-hundred-and-sixty women were included in the study, three of whom being excluded from efficacy assessment because of non-compliance to the protocol. Complete pregnancy interruption without additional surgical procedure (success) was obtained in 136 women (86.6%, 95% confidence interval: 81.3-91.9%). The success rate was significantly (P = 0.013) higher for pregnancies below (91.3%), than for pregnancies above 42 days of amenorrhea (DA) (76.6%). The time elapsed between RU 486 intake and complete expulsion was 2.8 +/- 1.5 (sd) days (range: 1-12 days). Expulsion took place at the latest 4 days after RU 486 in 125 women (94.7%), and in 107 of these women, it occurred 3.1 +/- 1.7 (sd) hours after PG05 administration. Uterine bleeding occurred in all women after RU 486 intake whatever the outcome of treatment and lasted 11.5 +/- 4.8 (sd) days (range: 3-36 days). It was judged more or much more abundant than usual periods in 6.15% of the women. It led to a slight but significant decrease in hemoglobin as measured eight and 14 days after RU 486 intake. In five women, hemoglobin decreased by 4 g/dl or more, but no patient required a blood transfusion.(ABSTRACT TRUNCATED AT 250 WORDS)

Experimental study of invasion and metastasis of cancer cells and establishment of the morphological pattern in the process of invasion and metastatic models of cancer cells.

For the study of invasion of tumor cells in vitro and in vivo methods were used to observe the invasion process and morphological characteristics of tumor cells. The different animal tumor cells and cell lines from human tumors were used. The process of cancer cells in vitro invasion can be divided into 5 stages. We suggest that filopodia protruding from the tumor cell margin seem to play an important role in the initiation of cell invasion. For experimental study of cancer metastasis we established three spontaneous metastatic models, including spontaneous lymphatic and hematogenous metastasis of mouse uterine cervix carcinoma No 14(U14) and No 27(U27), and third spontaneous lung metastasis in mouse forestomach carcinoma (FC). Two tumor cell lines are established: One is MFC cell line which is derived from FC tumor; second cell line is MUCC which is derived from the U27 tumor. These metastasis models and their cell lines are very useful for studying the mechanism of metastasis, tumor heterogenicity and antimetastatic agents of tumor. The results are discussed.

Establishment and characterization of a continuous murine uterine cervix cancer cell line metastatic to lymph nodes and lungs.

The murine uterine cervix cancer (MUCC) cell line was derived from a chemically induced Kunming mouse uterine cervix cancer (U27) and maintained in culture on solid substrates for over 100 passages. Cultures were morphotypically heterogeneous and heteroploid, with a modal number of chromosomes = 80. Each cell showed at least two abnormal chromosomes. Immunogold-silver staining was positive for keratin, vimentin, and laminin but not for desmin. The population doubling time was 27.8 h with a saturation density of 3.2 X 10(5) cells/cm2 and a peak mitotic index of about 6%. MUCC cells produced colonies on tissue culture plastic (68%) and in soft agar (8%). MUCC cells were fully malignant inasmuch as they produced in syngeneic mice invasive tumors that reproducibly were metastatic to lymph nodes and lungs. The MUCC cell line is the first mouse cervix cancer cell line useful for the study of invasion and metastasis.

Pathology of tumours produced in syngeneic Fischer rats by fibroblast-like cells before and after transfection with oncogenes.

Fischer rat cells before and after transfection with immortalizing and transforming genes produced tumours after s.c., i.p., or i.v. injection of cell suspensions and after s.c. implantation of cellular aggregates in the tail of syngeneic rats. Tumours were described histologically as fibrosarcoma-like. Virtually all tumours were considered macroscopically to be invasive because they adhered to the neighbouring tissues; in many tumours invasion was confirmed microscopically. All types of cells produced lung colonies (artificial metastases) after i.v. injection. Spontaneous metastases (from a primary tumour) were found with some tumours produced by cells before as well as after transfection. Differences in metastasis between various cell types could not be ascribed to variations in the periods of observation, in the minimum tumour-bearing periods, in the latency periods, or in the volume of primary tumours. We concluded that local invasion and spontaneous metastasis are usefull for the characterization of malignancy in experimental fibrosarcoma-like tumours. Since Fischer rat cells produced invasive and sometimes metastatic tumours before transfection, the present data do not show a rôle of immortalizing and transforming genes in the acquisition of invasiveness and metastatic capability.
