Factors acting upon differentiation of mouse teratoma in vivo: spontaneous differentiation and effects of interferon.

CE/44 mouse teratocarcinoma and its sublines isolated at the Institut Pasteur were studied comparatively in 129/Sv, hybrid F1 129 X NCS, and NCS strains of male and female mice with respect to the age of the animals and the type of the tumor graft (ascitic or subcutaneous solid tumor). The tumors grow more quickly and with more malignant characteristics in female mice. The tumor "takes" and growth are equally good in the hybrid F1 as in 129/Sv mice, but its maintenance in hybrid F1 is not so stable, for a tumor loss and a lessening of differentiation could be seen through the passages in this strain. In the NCS strain the tumor grows very rarely. Interferon slowed the subcutaneous tumor growth and favored neuroepithelial tissue differentiation, but allowed selective growth of one cellular type, the "trophoblastic" one, which is very malignant, and which, in the ascitic tumor, caused death in female animals by a hemorrhagic syndrome.

Immunological castration of male mice by a totally synthetic vaccine administered in saline.

It had been reported that immunological neutralization of the hypothalamic luteinizing hormone-releasing hormone (LH-RH) can be achieved by injection of the synthetic hormone, a decapeptide, linked to a carrier and administered with Freund's complete adjuvant. Data presented here demonstrate that the resulting immunological castration can be obtained by administering in an aqueous medium the synthetic decapeptide directly conjugated to the synthetic immunomodulator glycopeptide N-acetylmuramyl-L-alanyl-D-isoglutamine. In the group receiving the conjugate, a higher titer of anti-LH-RH antibodies was obtained than in the group receiving LH-RH in Freund's emulsion. Moreover, histological examination showed that seminiferous tubules were atrophied and that no spermatozoides could be observed. This procedure could lead to applications in the veterinary field and also serve as a useful model for other antiviral or antibacterial synthetic vaccines.

[Vitelline morphogenesis in a teratocarcinoma of mice].

Morphogenesis like cyto-differentiation is expressed in Mouse teratocarcinomas. The presence in certain tumours, of yolk-sac cavities combining trophoblast, Reichert's membranes and parietal distal endoderm in spite of the absence of included embryoïd structures, suggests that there is a precursor cell common to there extra-embryonic components.

Repair of large chest wall defects: experience with 23 patients.

The repair of large chest wall defects has been done on 23 patients who had 28 operations. Twenty-two patients had a neoplasm of the thoracic cage, while 1 had a large inflammatory mass. Nine patients had a partial lung and 3 a partial diaphragmatic resection done en bloc with the ribs. Seven resections were on the sternum. The repair was made either with a soft prosthesis (nylon mesh in 3 and Marlex mesh in 12 operations) or with a composite prosthesis of methyl methacrylate and metallic or Marlex mesh (13 operations). The association of methyl methacrylate and Marlex combines the solidity and the easy shaping of methyl methacrylate with the advantages of the mesh for an easy fixation and excellent incorporation. When the skin and the muscles are resected with the osteocartilaginous wall, an omentum flap is placed between the skin and the prosthesis to facilitate healing. There was one postoperative wound infection, which cleared with appropriate antibiotics. No prosthesis has extruded. The cosmetic and functional results are satisfactory. Repair of very large chest wall defects after resection can be done safely.

Differentiation of a teratocarcinoma line: preferential development of cholinergic neurons.

A line of embryonal carcinoma cells, PCC7-S, established in vitro from a spontaneous testicular teratocarcinoma, has been studied. Upon removing the cells from a low density monolayer culture system and permitting the cells to form aggregates in suspension, we observed a change of several physical and biochemical parameters: (a) reduction in average cell volume, (b) blockage and accumulation of cells in G1, (c) rise in secreted protease activity, (d) rise in acetylcholinesterase and choline acetyltransferase activities, and (e) disappearance of embryonic antigen F9. Although PCC7 aggregates did not undergo substantial morphological changes while suspended, when aggregates 4 or more days old were allowed to attach to plastic tissue culture dishes, substantial neurite outgrowth occurred over the next 1-3 d. This process was markedly enhanced by the addition to the growth medium of carboxymethylcellulose and inhibitors of DNA synthesis. Transmission electron microscopy disclosed a neurite ultrastructure consistent with that of neuronal processes. A veratridine-stimulated, tetrodotoxin-blocked sodium influx of 100 nmol/min per mg protein was also observed in these differentiated surface cultures. This cell line is discussed in terms of its utility for the study of early events leading to a commitment to cellular differentiation, as well as for the investigation of terminal differentiation to cholinergic neurons.

Cutaneous basophil hypersensitivity in atopic dermatitis.

The infiltration by basophils into delayed hypersensitivity skin test sites was examined in patients with atopic dermatitis, contact dermatitis and in normal healthy persons. Atopic dermatitis patients, with large amounts of IgE, injected intradermally with staphylococcal antigens showed reactions that were more transient and erythematous than those of normal persons. On histological examination there were numerous, degranulating basophils among the perivascular mononuclear cells. Normal persons, with small amounts of IgE showed typical mononuclear cell infiltration and few basophils. Patients with contact dermatitis, one with much IgE, responded to patch tests to potassium dichromate or to nickel sulphate by delayed type reactions, but on histology, two of the four patients showed a significant infiltration by basophils. It is considered that the erythematous response to the antigen in atopic dermatitis patients is related to the basophil infiltration.

Reactivity of monoclonal antibodies against intermediate filament proteins during embryonic development.

Monoclonal antibodies (mAbs) against a preparation of intermediate filaments from trophoblastoma cells were studied for their reactivity pattern during embryonic development and on adult tissue cells. Up to day 12 of embryonic development, epithelial cells of the three germ layers reacted with these mAbs. Later during development and in adult tissues, positive reactions could be observed only with epithelial cells derived from mesoderm and endoderm. Because of their tissue distribution, the proteins reacting with these mAbs might belong to the keratin family of intermediate filaments or they might represent a new group of intermediate filaments.
