[Staging laparotomy for malignant lymphoma: interdisciplinary aspects and results (author's transl)].

Between 1963 and 1979 a total of 357 patients with malignant systemic lymphatic disease underwent splenectomy in the course of exploratory laparotomy. There were three groups: 1. Hodgkin's disease (215 patients), 2. non-Hodgkin's lymphoma (93), and 3. chronic lymphoid leukaemia (49). Comparing pre- and postoperative findings, primary exploratory laparotomy remained the most exact foundation for the differentiated treatment of Hodgkin's disease in stages I-III. Systematic staging laparotomy was of less importance for non-Hodgkin's lymphoma and chronic lymphoid leukaemia because of the much more frequent invasion of bone marrow, spleen and liver. However, it should be considered more often in the management of such cases.

Variations in normal electrocardiographic response to treadmill testing.

Forty healthy young men at low risk for coronary artery disease underwent progressive maximal treadmill testing. Four bipolar electrocardiographic leads including CM5, CC5, inferior-superior Y, anterior-posterior Z, and a standard V5 were recorded and later computer-processed. Measurements included amplitudes of the Q, R, S, J junction and T wave, R-T and Q-S intervals and S-T segment slope. These variables are presented as the 10th, 50th (median) and 90th percentiles throughout the testing procedure to define reference values for the electrocardiographic response to maximal treadmill testing. The medians are presented graphically so that the exercise-induced changes can be visualized. In addition, the percent change of R wave amplitude in V5 compared with the supine pretest value is displayed for each subject during and after testing.

Alteration of aminoglycoside antibiotic ototoxicity by hyper- and hypohydration.

The ototoxicities of tobramycin sulfate and gentamicin sulfate were investigated in guinea pigs under conditions of normal, increased, and decreased hydration. Increased hydration was associated with no decline in the amplitude of the cochlear microphonics, a lesser decline in the eighth nerve action potentials and lesser damage to the organ of Corti. Decreased hydration was associated with an increase in the threshold of the cochlear microphonics and the eighth nerve action potentials, a decline in the amplitude of the cochlear microphonics, a greater decline in the eighth nerve action potentials, and greater damage to the organ of Corti. Tobramycin sulfate was substantially less toxic than gentamicin sulfate with normal, increased and decreased hydration. These findings suggest the preferential use of tobramycin sulfate for patients with normal renal function, and especially patients with renal impairment.

[Cerebral vasospasm: clinical and biochemical aspects and treatment (author's transl)].

Cerebral vasospasm is caused by many factors. Primarily it is caused by Noradrenalin (NA) localised in the vessel walls and by platelet aggregation, with raising of the concentration of 5-HT in the intima. Its prolongation is based on blood breakdown products and or biochemical disturbances of brain metabolism. The most potent vasoconstrictors are acting through the vessel's own receptors (Serotonin, NE, Histamin), so the most effective therapy is based on the contractile system of the vessel. By blocking the alpha-receptors by Phenoxybenzamine, blockade of the Phosphodiesterase by Papaverin, Euphyllin to raise the concentration of cAMP and stimulation of Adenyl-Cyclase-System by beta-stimulation, it is possible to get good results in the treatment of cerebral arterial vasospasm.

[Accumulation of glycoproteins in activated neutrophilic granulocytes].

In contrast to the diffuse isotropic staining of control granulocytes activated ones by the 4th hour of experiment with phagocytosis show an intensive anisotropic staining with the topo-optical reaction of vicinal OH-groups i.e. ABT reaction. Appearance of the intensive birefringence is in a close connection with the significant increase of the glycoprotein content of granulocytes detectable also by other biochemical techniques. This phenomenon is not influenced neither by the nature of activating particules nor by the fact of their engulfing. Thus accumulation of glycoproteins may be considered as a phenomenon of general activation of granulocytes. Electronmicroscopically the main quantity of glycoproteins accumulated may be demonstrated in the primary granules. Intensive positive spheritic birefringence of these granules detected in semithin sections by ABT reaction seems to indicate that vicinal OH groups of the accumulated glycogen are aligned parallel to the surface of granules in highly linear molecular orientation which indicates the existence of a close connection between glycoproteins and lysosome membranes. Considering the increased glycogen content of Golgi membranes of activated cells it can be assumed that it is synthesized de novo in the first 4 hour of the activation process. Experimental model described makes possible a detailed morphological and biochemical study of accumulated glycogen and assessment of its functional importance.

Effect of pentobarbital on thalamo- and hippocampocortical processes in rats.

Experiments were carried out on 28 rats immobilized with d-tubocurarine. The effect of three successive doses of pentobarbital (PB--5, 15 and 40 mg.kg-1) on electrocorticographic manifestations (evoked responses, the rhythmic afterdischarge and barbiturate spindles, the self-sustained afterdischarge--SSAD) elicited by electrical stimulation of two thalamic nuclei with a different type of control projection and the hippocampus (CA3) was compared. We found no difference between the effect of PB on cortical electrographic manifestations elicited by stimulation of the dorsomedial part of the ventral nucleus (VDM) and the anterior lateral nucleus (LA) of the thalamus. Cortical responses elicited by stimulation of the hippocampus were heterogeneous and the effect of PB on them was smaller than on thalamocortical responses. Reduction of the hippocampocortical SSAD was likewise not significant until the deepest stage of barbiturate anaesthesia.

Structural analysis of hnRNP particles approached by in vitro phosphorylation using exogenous protein kinase and l gamma 32 P1 ATP.

Using an exogenous kinase, nuclear ribonucleoprotein complexes with sedimentation coefficients greater than 100S were phosphorylated in vitro before and after treatment with increasing concentrations of NaC1. The phosphorylation pattern of the proteins before raising the NaC1 concentration shows a major group of labelled proteins in the 30 000 to 40 000 MW range. Treatment of the complexes with 400 and 800 mM NaC1 produces a relative increase in the labelling of some polypeptides with the appearance of new labelled bands and the concomitant disappearance of several proteins. Even at the highest salt concentration used (1.2 M), it is still possible to identify a group of labelled polypeptides which are suggested to form the backbone structure of the nuclear RNP complexes.

A histochemical study with the aBt reaction of changes in glycoprotein content during bone formation and necrosis.

The glycoprotein content of the bone tissue was examined by means of the aldehyde-bisulphite-blue (ABT) reaction. It was found that the glycoprotein content showed variations during bone formation and necrosis. This was particularly characteristic in the osteocyte capsule where vicinal OH moieties changed in amount and orientation. The method allows to determine the relative age of the bone tissue.

Crystal structure of yeast tRNAAsp.

Two independent, three-dimensional structures of yeast tRNAAsp, mainly differing by the conformation of the D loop, have been obtained from a multiple isomorphous replacement (MIR) X-ray analysis at 3.5-A resolution. The folding of the ribose-phosphate backbone is similar to that found for tRNAPhe; major differences concern the relative positioning of the acceptor and anticodon stems, and the conformation of the loops in the two molecules. Crystal packing involves self-complementary GUC anticodon interactions.
