B- and T-cells and Ig-containing blast cells in healthy children.

Studies on rosette-forming cells and IgG, IgA, IgM and IgE-bearing lymphocytes were carried out in 70 healthy children aged 5 to 15 years. Furthermore, in 20 of these children, the percentages of IgG, IgA, IgM and IgE-containing blast cells after pokeweed mitogen stimulation of peripheral lymphocytes were evaluated. No differences in the percentages of rosette-forming cells, or in Ig-bearing and Ig-containing cells were found when three different age-groups were compared.

Transforamtion of twitching strains of Streptococcus sanguis.

Ninety-five strains of S. sanguis, 90 of which were twitching, were screened for competence in transformation with DNA from the "Challis" strain. Seventy-two strains, 68 of sero-group H and 4 of the provisional group 10043, were competent. Fourteen of the competent strains and all strains which appeared to be incompetent were tested with DNA from 3 other strains. The 14 competent strains were transformed by all the 3 DNAs. One of the apparently incompetent strains was transformed by autologous DNA only. Among 8 reference strains (including ATCC 10577, Type II of Washburn et al.) 5 were competent. Three of these did not show spreading or twitching. Among 16 non-spreading strains of alphahaemolytic streptococci which did not possess either the H or the 10043 group antigen, only one showed competence. The results indicate that twitching mobility is not a prerequisite for competence.

Cytogenetic studies in myelomatosis.

Cytogenetic studies have been carried out on bone marrow aspirates from 25 patients with myelomatosis. Abnormal stem lines were present in 7 of the patients; the remainder had a diploid chromosome complement. In most patients - also in those without an abnormal clone - some metaphases had a blurred appearance similar to that seen in bone marrow aspirates from patients with acute leukaemia. In many of the chromosome preparations obtained before cytostatic therapy some metaphases with structural aberrations on the chromosomes were seen. Evidence is presented that in the patients with abnormal stem lines in the bone marrow, the chromosome abnormalities are confined to the myeloma cells and are not found in the erythrocytic or granulocytic precursors, which thus do not seem to be involved by the neoplastic process. Based on the present resuts and on a review of the relevant literature some general cytogenetic features are emphasized which may contribute to a better understainding of the disorder. Especially, it is demonstrated that in myelomatosis the cytogenetic changes are much more uniform than in other malignant disorders with the exception of chronic myeloid leukaemia.
